Peritoneal carcinomatosis (PC) is considered incurable, and more effective therapies are needed. Herein we test the hypothesis that GPA33-directed intracompartmental pretargeted radioimmunotherapy (PRIT) can cure colorectal peritoneal carcinomatosis. Nude mice were implanted intraperitoneally with luciferase-transduced GPA33-expressing SW1222 cells for aggressive peritoneal carcinomatosis (e.g., resected tumor mass 0.369 ± 0.246 g; n = 17 on day 29). For GPA33-PRIT, we administered intraperitoneally a high-affinity anti-GPA33/anti-DOTA bispecific antibody (BsAb), followed by clearing agent (in

This manuscript reviews current advances in the use of radioimmunotherapy (RIT) for the treatment of B-cell non-Hodgkin's lymphoma (NHL). RIT has been in use for more than 20 years and has progressed significantly with the discovery of new molecular targets, the development of new stable chelates, the humanization of monoclonal antibodies (MAbs), and the use of pretargeting techniques. Today, two products targeting the CD20 antigen are approved: (131)I-tositumomab (Bexxar(®)), and (90)Y-ibritumomab tiuxetan (Zevalin(®)). (131)I-tositumomab is available in the United States, and (90)Y-ibritumum

BACKGROUND: Radioimmunotherapy (RIT) is a safe and effective therapeutic option for patients with indolent B-cell non-Hodgkin lymphomas (NHL), in both up-front and relapsed/refractory settings. Two approved agents (90Y-ibritumomab tiuxetan and 131I-tositumomab) are available in the United States. Both target CD20 with similar clinical outcomes but with unique clinical considerations and radiation precautions due to the use of varying radioisotopes.
METHODS: This paper reviews the available evidence for these approved RIT agents and examines the recently published and ongoing clinical trials o

Radioimmunotherapy (RIT) combines the targeting advantage of a monoclonal antibody with the radiosensitivity of non-Hodgkin lymphoma (NHL) cells. There are now two radioimmunoconjugates (RICs) - ibritumomab tiuxetan (Zevalin) and tositumomab (Bexxar) - that are approved by the FDA in the US for relapsed low-grade or follicular B-cell NHL. Both agents target the CD20 antigen on B-cell lymphoma cells. In relapsed disease, single doses of RIT produce an 80% overall response rate, with approximately 20% of patients achieving durable responses. RIT is very well tolerated and is delivered on an outp

Colorectal cancer (CRC) is a deadly form of cancer worldwide. Patients with locally advanced rectal cancer and metastatic CRC have a poor long-term prognosis, and rational and effective treatment remains a major challenge. Common treatments include multi-modal combinations of surgery, radiotherapy, and chemotherapy; however, recurrence and metastasis rates remain high. The combination of radiotherapy and immunotherapy (radioimmunotherapy [RIT]) may offer new solutions to this problem, but its prospects remain uncertain. This review aimed to summarize the current applications of radiotherapy an

INTRODUCTION: Corona Virus Disease of 2019 (COVID-19) pandemic has renewed interest in monoclonal antibodies for treating infectious diseases. During last two decades experimental data has been accumulated showing the potential of radioimmunotherapy (RIT) of infectious diseases. In addition, COVID-19 pandemic has created a novel landscape for opportunistic fungal infections in post-COVID-19 patients resulting from severe immune suppression.
AREAS COVERED: We analyze recent results on targeting "pan-antigens" shared by fungal pathogens in mouse models and in healthy dogs; on developing RIT of

Approximately 66,000 Americans develop non-Hodgkin lymphoma (NHL) each year. Although the use of unlabeled antibodies such as rituximab has significantly improved survival when combined with standard chemotherapy regimens, approximately two-thirds of lymphoma patients eventually develop disease recurrence and die of their disease. Novel treatments are urgently needed to cure these patients. One strategy involves the use of radiolabeled immunoconjugates that specifically localize radiation delivery to sites of lymphoma while minimizing toxicity to normal tissues. A growing number of studies sup

Elevated intracellular glutathione, intratumoral hypoxia, and excessive polyamines severely compromise the efficacy of radiotherapy (RT) by attenuating radiation damage and maintaining an immunosuppressive tumor microenvironment (ITME). Strategies that concurrently address these obstacles are urgently needed to enhance RT outcomes. Herein, we fabricated a biomimetic nanoplatform (PCuP) via the self-assembly of copper ions and piceatannol into CuP nanometal-polyphenol cores, followed by surface camouflage with platelet membranes (PM). Owing to the platelet membrane coating, PCuP preferentially

Background/Objectives: Malignant mesothelioma has a poor prognosis and limited therapeutic options. C-ERC/mesothelin is highly expressed in mesotheliomas and is a potential target for radioimmunotherapy (RIT). This study evaluated the radiolabeled anti-C-ERC/mesothelin antibody mAb 22A31 as a therapeutic agent. Methods: C-ERC/mesothelin expression in mesothelioma cell lines was assessed by Western blotting, and the specific binding of 125I-labeled mAb 22A31 was examined. Biodistribution of 111In-labeled mAb 22A31 was evaluated in a mesothelioma cell line, MSTO-211H tumor-bearing mice. The ther

Bacterial outer membrane vesicles (OMVs) with catalase deposition have significant potential for cancer immunotherapy, yet their therapeutic efficiency is limited by insufficient immune activation. Metal ions with a high atomic number could act as radiosensitizers to increase radiation-dose deposition at tumor sites and boost the immunogenic effects of radiotherapy (RT). Herein, we report metal-phenolic OMVs by cloaking OMVs with hafnium (Hf)-phenolic networks (Hf-OMVs) for enhanced cancer radioimmunotherapy. In tumor microenvironment, catalase deposited in the envelope of OMVs initiated the d

In cancer treatment, traditional radiotherapy (RT) is routinely conducted in local tumors and limited by radioresistance, while recently emerged immunotherapy is confronted with low response rate, high cost, and cytokine release syndrome. Logically, the combination of the two therapeutic modalities into radioimmunotherapy is promising to complement with each other for systemic elimination of cancer cells with high specificity, efficiency, and safety. Especially, RT-induced immunogenic cell death (ICD) plays an imperative role in radioimmunotherapy to evoke systemic immune response against canc

Radioimmunotherapy (RIT) represents a paradigm shift in precision cancer therapy by integrating the molecular specificity of monoclonal antibodies with the cytotoxic potential of therapeutic radionuclides. However, the therapeutic success of RIT is not merely associated with targeted radiation delivery but also depends on complex interplay of radiobiology, tumor immunology, antibody pharmacology, and patient-specific dosimetry. This review explores the molecular and cellular mechanisms, including DNA damage response, immunogenic cell death, tumor microenvironment remodeling, cytokine signaling

PURPOSE: Increased angiogenesis is a marker of aggressiveness in many cancers. Targeted radionuclide therapy of these cancers with angiogenesis-targeting agents may curtail this increased blood vessel formation and slow the growth of tumors, both primary and metastatic. CD105, or endoglin, has a primary role in angiogenesis in a number of cancers, making this a widely applicable target for targeted radioimmunotherapy.
METHODS: The anti-CD105 antibody, TRC105 (TRACON Pharmaceuticals), was conjugated with DTPA for radiolabeling with 177Lu (t 1/2 6.65 days). Balb/c mice were implanted with 4T1 m

HIV/AIDS remains an enormous public health burden. Advances in anti-retroviral therapy (ART) have greatly reduced mortality and morbidity but HIV remains incurable, with patients suffering numerous disease- and treatment-related side effects. Any curative strategy for HIV must selectively eliminate existing infected cells. Radioimmunotherapy (RIT) is an established clinical modality in cancer treatment and has been shown to be effective in multiple infectious diseases models. We have recently demonstrated that RIT using a gp41-targeting antibody was effective and safe in eliminating HIV-infect

Therapeutic monoclonal antibodies (MoAbs) have been the major breakthrough in the treatment of both indolent and aggressive B-cell non-Hodgkin's lymphoma (NHL), and have become standard of care for these diseases. However, patients may not respond to antibody therapy, or resistance can develop. Radioimmunotherapy (RIT) makes use of continuous low dose radiation emitted by radioisotopes targeted directly to the lymphoma cells. Both the (131)I-labeled murine CD20 MoAb tositumomab (Bexxar®) and (90)Y-labeled ibritumomab-tiuxetan (Zevalin®) have been shown to be more effective than unlabeled MoAbs

Prostate cancer is a common cancer in men and continues to be a major health problem. Imaging plays an important role in the clinical management of patients with prostate cancer. An important goal for prostate cancer imaging is more accurate disease characterization through the synthesis of anatomic, functional, and molecular imaging information. Positron emission tomography (PET)/computed tomography (CT) in oncology is emerging as an important imaging tool. The most common radiotracer for PET/CT in oncology, (18)F-fluorodeoxyglucose (FDG), is not very useful in the imaging of prostate cancer.
