ICRF-187 was given to 62 evaluable patients with advanced solid tumors in a Phase I clinical trial. Weekly infusions were given in dosages ranging from 0.85 g/m2 to 7.42 g/m2 for a total of four weeks with a two week rest period between courses. Dose-limiting hematological toxicity was seen in heavily pretreated patients at a dose of 3.8 g/m2/week. All patients also developed reversible SGOT elevations. In a double-blind trial the effect of ICRF 159 or placebo associated with radiotherapy was compared in 70 patients with cervical carcinoma. No significant differences were found on histopathological examination of surgical specimens obtained 6 weeks after the end of the treatment. Clinical evaluation after a follow-up of 46 +/- 5 months, also showed no significant effect of ICRF 159 with radiotherapy.

The tolerance of cardioxan (JCRF-187, dexrazoxan), an ethylenediamine tetra-acetic acid analog and a Chiron Company product, administered to pediatric oncohematological patients, is discussed. The drug was used in 37 cases of acute lymphoblastic and non-lymphoblastic leukemia, non-Hodgkin's lymphoma and Hodgkin's disease treated by polychemotherapy including anthracycline antibiotics administration. No untoward side-effects or inhibition of the therapeutic effect were observed.

The cooperative group included 18 provincial and district hospitals. A retrospective study was carried out on 140 cases of therapy related leukemia (TRL) caused by bimolane (BML) for psoriasis from 1984 to 1992. This series of BML-TRL consists of 90 male and 50 female patients. 87.1% of them were from 20 to 50 years old. Annual incidence varied from 4 to 24 cases, and was maintained at this level through the period from 1986 to 1991 without any declining tendency. ICRF-187 is the D-enantiomer of the racemic antitumor agent ICRF-159 and was selected for clinical trials on the basis of aqueous solubility suitable for iv administration. Eighteen patients with refractory advanced solid tumors received ICRF-187 by a 48-hour continuous iv infusion in 5% Dextrose in Water, USP. The total dose ranged from 200 to 1000 mg/m2/48 hours. PURPOSE: Anthracyclines, such as doxorubicin and daunorubicin, continue to be widely used in the treatment of cancer, although they share the adverse effect of chronic, cumulative dose-related cardiotoxicity. Dexrazoxane is a derivative of the powerful metal-chelating agent ethyl enediamine tetra acetic acid. Its cardioprotective effect is thought to be due to its ability to chelate iron and reduce the number of metal ions complexed with anthracycline and, consequently, decrease the formation of superoxide radicals.
