The cannabinoid CB1 receptor and the endocannabinoid anandamide: possible antidepressant targets.

BACKGROUND: Major depression has the highest rate of prevalence and incidence of morbidity among all mental disoders. The limited efficacies of current antidepressant treatments necessitate the development of alternative pharmacotherapies. Recent preclinical findings suggesting that cannabinoid CB(1) receptor agonists and endocannabinoid enhancers possess antidepressant-like properties, and clinical evidence that the CB(1) antagonist rimonabant increases the risk of depression and suicidality, support the notion that the endocannabinoid system represents a novel target in the treatment of mood

Alterations in behavioral flexibility by cannabinoid CB1 receptor agonists and antagonists.

RATIONALE: Cannabinoid CB1 receptors are expressed in the prefrontal cortex, but their role in mediating executive functions such as behavioral flexibility is unclear. OBJECTIVE: The present study examined the effect of pharmacological activation or blockade of the cannabinoid CB1 receptors on behavioral flexibility using a strategy set-shifting task conducted on a cross maze. MATERIALS AND METHODS: In experiment 1, rats initially were trained to turn left or right while ignoring the visual cue to obtain a food; on the second test day, rats had to inhibit the previously learned rule and appr

Cannabinoid receptors CB1 and CB2 form functional heteromers in brain.

Exploring the role of cannabinoid CB(2) receptors in the brain, we present evidence of CB(2) receptor molecular and functional interaction with cannabinoid CB(1) receptors. Using biophysical and biochemical approaches, we discovered that CB(2) receptors can form heteromers with CB(1) receptors in transfected neuronal cells and in rat brain pineal gland, nucleus accumbens, and globus pallidus. Within CB(1)-CB(2) receptor heteromers expressed in a neuronal cell model, agonist co-activation of CB(1) and CB(2) receptors resulted in a negative cross-talk in Akt phosphorylation and neurite outgrowth

Binding of the hemopressin peptide to the cannabinoid CB1 receptor: structural insights.

Hemopressin, a bioactive nonapeptide derived from the α1 chain of hemoglobin, was recently shown to possess selective antagonist activity at the cannabinoid CB(1) receptor [Heimann, A. S., et al. (2007) Proc. Natl. Acad. Sci. U.S.A. 104, 20588-20593]. CB(1) receptor antagonists have been extensively studied for their possible therapeutic use in the treatment of obesity, drug abuse, and heroin addiction. In particular, many compounds acting as CB(1) receptor antagonists have been synthesized and subjected to experiments as possible anti-obesity drugs, but their therapeutic application is still

Roles of cannabinoid CB1 and CB2 receptors in the modulation of psychostimulant responses.

Addiction to psychostimulant drugs, such as cocaine, D-amphetamine, and methamphetamine, is a public health issue that substantially contributes to the global burden of disease. Psychostimulant drugs promote an increase in dopamine levels within the mesocorticolimbic system, which is central to the rewarding properties of such drugs. Cannabinoid receptors (CB1R and CB2R) are expressed in the main areas of this system and implicated in the neuronal mechanisms underlying the rewarding effect of psychostimulant drugs. Here, we reviewed studies focusing on pharmacological intervention targeting ca

Cannabinoids inhibit emesis through CB1 receptors in the brainstem of the ferret.

BACKGROUND & AIMS: Marijuana and other cannabinoids are effective anti-emetics. Despite ongoing controversy over their usage, the receptor distribution and the site of the anti-emetic action of these compounds are not known. Our aim was to investigate whether the cannabinoid 1 receptor (CB1r) and endocannabinoids play a role in the anti-emetic action of cannabinoids. METHODS: Ferrets were given an emetic stimulus and the number of episodes of retching and vomiting were observed after administration of CB1r agonists and a CB1r antagonist. Localization of cannabinoid CB1 receptors in the human anterior eye and retina.

PURPOSE: To determine the presence and distribution of CB1 cannabinoid receptors within the human eye. METHODS: A subtype-specific affinity-purified polyclonal antibody to the cannabinoid CB1 receptor was used to determine CB1 localization. Postmortem human eyes were fixed in methacarn and embedded in paraffin. Sagittal sections were mounted on slides and immunostained using antibodies to the CB1 receptor. Antibody binding was detected either by using peroxidase conjugated secondary antibodies and developing with diaminobenzidine or by using fluorescent secondary antibodies. RESULTS: Strong

Cannabinoid CB1 receptor and mu-opioid receptor interaction: new insights from conditional knockout mice.

Co-administration of cannabinoids and opioids has been shown to enhance analgesic effects. However, the underlying mechanisms remain unclear. One hypothesis suggests that cannabinoid CB1 receptors (CB1R) and mu-opioid receptors (MOR) interact at the cell membrane or intracellular signaling level. This study aimed to test this hypothesis by examining CB1R-MOR colocalization and assessing whether the deletion of one receptor affects the other's response to its ligand. Results from RNAscope in situ hybridization revealed that CB1R and MOR mRNAs exhibit distinct regional distributions in the mouse

Envisioning Cannabinoid CB1 Receptor Biased Signaling as a Therapeutic Target for Schizophrenia.

The relationship between the endocannabinoid system and the emergence and treatment of schizophrenia-related symptoms continues to be a topic of significant interest within psychiatry. Cannabis is the most widely used recreational drug worldwide, and individuals with schizophrenia use it at a much higher rate than the general population. The shared genetic risk for schizophrenia and cannabis use may partially account for this phenomenon. However, the exposure to cannabis in individuals at risk of schizophrenia appears to not only represent the manifestation of overlapping risks, but also pharm

Negative allosteric modulation of CB1 cannabinoid receptor signaling suppresses opioid-mediated reward.

Blockade of cannabinoid type 1 (CB1)-receptor signaling decreases the rewarding properties of many drugs of abuse and has been proposed as an anti-addiction strategy. However, psychiatric side-effects limit the clinical potential of orthosteric CB1 antagonists. Negative allosteric modulators (NAMs) represent a novel and indirect approach to attenuate CB1 signaling by decreasing affinity and/or efficacy of CB1 ligands. We hypothesized that a CB1-NAM would block opioid reward while avoiding the unwanted effects of orthosteric CB1 antagonists. GAT358, a CB1-NAM, failed to elicit cardinal signs of

Propofol Attenuates Brain Ischemic Injury Through the Cannabinoid Type 1 (CB1) Receptor.

Background and objective Although the anesthetic propofol has been shown to reduce ischemic brain injury, its precise neuroprotective mechanisms remain unclear. Many studies have reported that propofol injection can increase endocannabinoid levels in vivo and in vitro. This study investigates the cerebroprotective pathways mediated by propofol during ischemic insults. Methods In vivo studies used a rat middle cerebral artery occlusion (MCAO) model to simulate stroke-related cerebral ischemia/reperfusion injury. In vitro, oxygen-glucose deprivation/reoxygenation (OGD/R) models were used to mimi

Cannabinoid CB1 receptor overactivity contributes to the pathogenesis of idiopathic pulmonary fibrosis.

BACKGROUND: Breast cancer is the most prevalent malignancy among women worldwide and in Iraq. The rising incidence and mortality are mostly attributed by late diagnosis and inadequate screening. Although mammography, ultrasound, and magnetic resonance imaging are essential for early detection, their diagnostic performance varies by age, breast density, and population characteristics. Most available guidelines are based on Western populations and may not be fully applicable to younger Iraqi women with dense breasts, particularly in resource-limited settings. METHODS: This prospective comparati

The CB1 cannabinoid receptor mediates excitotoxicity-induced neural progenitor proliferation and neurogenesis.

Endocannabinoids are lipid signaling mediators that exert an important neuromodulatory role and confer neuroprotection in several types of brain injury. Excitotoxicity and stroke can induce neural progenitor (NP) proliferation and differentiation as an attempt of neuroregeneration after damage. Here we investigated the mechanism of hippocampal progenitor cell engagement upon excitotoxicity induced by kainic acid administration and the putative involvement of the CB1 cannabinoid receptor in this process. Adult NPs express kainate receptors that mediate proliferation and neurosphere generation i

Analysis of the endocannabinoid system by using CB1 cannabinoid receptor knockout mice.

The endocannabinoid system has been involved in the control of several neurophysiological and behavioural responses. To date, three lines of CB1 knockout mice have been established independently in different laboratories. This chapter reviews the main results obtained with these lines of CB1 knockout mice in several physiological responses that have been previously related to the activity of the endocannabinoid system. Studies using CB1 knockout mice have demonstrated that this receptor participates in the control of several behavioural responses including locomotion, anxiety- and depressive-l

A photoswitchable ligand for opposite control over cannabinoid receptors CB1 and CB2.

Photopharmacology offers a powerful strategy for the spatiotemporal modulation of protein function with molecular precision. While all the current photoswitchable ligands developed to date act on single targets, we report Azo23, the first bifunctional photoswitchable ligand that exerts opposing efficacy modulation toward two closely related G protein-coupled receptors, cannabinoid receptors CB1 and CB2. Upon ultraviolet irradiation, Azo23 isomerizes from its trans to cis form, resulting in a potent CB1 agonist and CB2 antagonist, with a remarkable activity shift exceeding 50-fold. Molecular do

Synergistic and additive terpene-THC interactions in cannabinoid CB1 and CB2 receptors.

Cannabis terpenes are widely recognized for shaping the plant's aromatic and sensory profiles. Yet, their pharmacological role in cannabis is less established. Recent work demonstrated that several cannabis terpenes can activate cannabinoid receptors type 1 and 2 (CB1R and CB2R) as low-efficacy agonists, raising the possibility that they may also modulate the activation at these receptors by Δ9-tetrahydrocannabinol (THC), through orthosteric or allosteric mechanisms. Here, we systematically investigated the effects of individual terpenes and of terpene mixtures on THC-evoked activation at CB1R

A light and electron microscopic study of the CB1 cannabinoid receptor in primate brain.

The immunohistochemical distribution and subcellular localization of the cannabinoid CB1 receptor was determined in the adult monkey using a polyclonal antiserum raised against the amino terminus of the rat CB1 receptor. At the level of light microscopy, our results generally parallel earlier studies investigating CB1 distribution in rodent brain with a few differences. In particular, high levels of receptor were found in the cortex, hippocampus, amygdala, cerebellum. However significant differences were also noted. The most striking differences were high levels of CB1 receptor in the monkey s

Don't Sweat It: Cannabinoid CB1 Receptors Reduce Sweating in a Mouse Model.

Numerous exocrine glands play key physiological roles in the body that include tearing, salivation, and lactation, as well as the control of body temperature via sweating. Malfunction of sweat glands can be deeply problematic or-in the case of anhidrosis-life-threatening. The prevalence of sweating disorders is high, affecting millions. The few available therapies are generally of limited effectiveness. Several lines of evidence point to regulation of sweating by the cannabinoid signaling system, an arrangement that would mirror cannabinoid regulation of tearing and salivation. Mice sweat in t
