Central cholinergic receptors and antidiuresis due to hypertonicity in sheep.

Injection of 10 microgram of carbachol into a lateral cerebral ventricle caused a large reduction in urine flow and free water clearance and an increase in urine osmolality in water-loaded sheep. This antidiuretic effect persisted for at least 1 h, and was prevented by a prior intracerebroventricular (ICV) injection of atropine (100 microgram). ICV injections of atropine (100 microgram) given 10 min before and again midway throgh an intracarotid infusion of 2 M NaCl at 0.8 ml/min for 10 min did not alter the antidiuretic response which normally follows this hypertonic stimuli. It was concluded

Expression of muscarinic cholinergic receptors on lymphocytes during rheumatoid arthritis.

The level of muscarinic receptors on lymphocytes from rheumatoid arthritis (RA) patients in comparison with healthy individuals was determined by the binding studies of a specific muscarinic ligand [3H]-QNB in the presence of the competitive antagonist atropine. The response to phytohaemagglutinin and the influence of acetylcholine on this process in patients' lymphocytes was also studied. We have found direct correlation among PHA stimulation indices, the level of muscarinic receptors, and the influence of acetylcholine on PHA stimulation indices. Differences in these parameters occurring in

Dispersed rat parotid acinar cells. III. Characterization of cholinergic receptors.

The in vitro characterization of cholinergic receptors in dispersed rat parotid acinar cells was accomplished through investigations of the net transmembrane fluxes of K in response to exposure of the cells to selected cholinergic agonists and antagonists. Interaction of acetylcholine bromide (ACh) and carbamylcholine (carbachol) with the cholinergic receptors resulted in rapid net efflux of K from the cells. This cellular response was demonstrable in concentrations of carbachol as low as 10(-8) M. With gradual increase in the concentrations of the agonist an increase in the K efflux was obser

In vitro and in vivo modulation of cholinergic muscarinic receptors in rat lymphocytes and brain by cholinergic agents.

A binding site for 3H-quinuclidinyl benzylate (QNB) has been identified in rat lymphocytes which has the characteristics of a cholinergic muscarinic receptor (Costa, L. G., Kaylor, G. & Murphy, S. D. (1988). Muscarinic cholinergic binding sites on rat lymphocytes. Immunopharmacology, 16, 139-149.) Here we show that prolonged exposures to cholinergic compounds in vitro and in vivo modulate muscarinic receptor binding in lymphocytes as well as in brain tissue. Exposure of rat splenic lymphocytes in vitro to oxotremorine caused a time- and concentration-dependent decrease in the density of 3H-QNB

BACKGROUND: In the gastrointestinal tract of several species, facilitating 5-HT4 receptors were proposed on myenteric cholinergic neurons innervating smooth muscle by in vitro study of the effect of the selective 5-HT4 receptor agonist prucalopride on submaximal cholinergic contractions. This was not yet established in the murine gastrointestinal tract. METHODS: In circular smooth muscle strips from murine fundus, jejunum and colon, contractions were induced by electrical field stimulation in the presence of guanethidine, L-NAME and for colon also MRS 2500. Submaximal contractions were induce

Alterations of striatal cholinergic receptors after lesioning of the substantia nigra.

Dopamine deficiency syndrome is known to cause cholinergic hyperactivity. Therefore, it was hypothesized that the said phenomenon may be due to enhanced cholinergic receptor functions. In the present study we examined the changes in striatal dopaminergic and cholinergic receptors in unilateral substantia nigra lesioned rats that showed vigorous ipsilateral rotation (total turns > 300) in response to apomorphine (1 mg kg(-1) ip). [3H] Spiperone ([3H]-SP) and [3H]-quinuclidinyl benzilate ([3H]-QNB) bindings were performed in the striata of the lesioned animals. There was no significant differenc

Trk neurotrophin receptors in cholinergic neurons of patients with Alzheimer's disease.

Besides cortical pathology, Alzheimer's disease (AD) is characterized by a loss of cholinergic neurons in the basal forebrain but not in the caudate nucleus, putamen or mesencephalon. Since cholinergic neurons which degenerate in AD are sensitive to nerve growth factor (NGF), a link between NGF sensitivity and the vulnerability of cholinergic neurons has been suspected. Levels of NGF are not altered in patients with AD, however. Thus, cholinergic nerve cell death in AD could not result from a deficiency in NGF receptors. Using sequential immunohistochemistry with antibodies that recognize pref

Adrenergic and cholinergic receptors in isolated non-pigmented ciliary epithelial cells.

Methods were developed to isolate non-pigmented ciliary epithelium from bovine eyes (BCBE) free from other cell types in the ciliary process. Once background binding due to pigment was minimized, it was possible to assay specific beta adrenergic receptors in these preparations. The BCBE beta adrenergic receptors showed appropriate stereospecificity and an order of binding affinities compatible with a beta 2 adrenergic selectivity. Interestingly, specific high affinity muscarinic cholinergic receptors were detected in both BCBE and primate non-pigmented ciliary epithelium. These muscarinic rece

Muscarinic cholinergic receptors in the piebald mouse model for Hirschsprung's disease.

The contracted segment of terminal large intestine in both Hirschsprung's disease in humans and in the piebald-lethal strain of mice with congenital megacolon may result from muscarinic cholinergic receptor denervation supersensitivity. This hypothesis was tested by direct binding studies using the muscarinic receptor ligand, tritiated quinuclidinyl benzilate. There were no differences in either the receptor density, dissociation constant, or agonist/antagonist Hill coefficients in the proximal, middle, or distal colon of mice with congenital megacolon as compared with normal littermates. Striatal cholinergic transmission. Focus on nicotinic receptors' influence in striatal circuits.

The critical role of acetylcholine (ACh) in the basal ganglia is evident from the effect of cholinergic agents in patients suffering from several related neurological disorders, such as Parkinson's disease, Tourette syndrome, or dystonia. The striatum possesses the highest density of ACh markers in the basal ganglia underlying the importance of ACh in this structure. Striatal cholinergic interneurons (CINs) are responsible for the bulk of striatal ACh, although extrinsic cholinergic afferents from brainstem structures may also play a role. CINs are tonically active, and synchronized pause in t

Distribution of cholinergic and dopaminergic receptors in rainbow trout pineal gland.

The involvement of multiple receptors in modulating the function of the pineal gland was investigated by searching for dopaminergic and cholinergic receptors in trout pineal gland. Dopamine D1 and D2 receptors were measured using [3H]SCH23390 and [3H]spiperone, respectively. Muscarinic and nicotinic cholinergic receptors were measured using quinuclidinyl benzilate ([3H]QNB) and [3H]methylcarbamyl choline, respectively. High-affinity choline uptake sites were measured using [3H]hemicholinium-3. The distribution of dopaminergic receptors varied throughout the pineal gland in that the density of

Agonist-regulated alteration of the affinity of pancreatic muscarinic cholinergic receptors.

Heterologous desensitization is a term that indicates that exposure of a cell to an agonist attenuates the response of that cell to other agonists. We examined heterologous desensitization of muscarinic cholinergic receptors of pancreatic acini and characterized mechanisms that might be responsible for desensitization. Muscarinic cholinergic receptor binding was measured by using N-[3H]methscopolamine bromide ([3H]NMS). N-Methscopolamine bromide (NMS), a receptor antagonist, bound to a single class of receptors with an affinity of 0.22 +/- 0.04 nM and a capacity of 61.5 +/- 5.1 fmol/mg of prot

Mediation of contraction by cholinergic muscarinic receptors in the ureterovesical junction.

1. The muscarinic receptor profile of the sheep ureterovesical junction has been studied by means of in vitro techniques. The relative potency (pD2 = -log EC50) and maximum effect (Emax) observed with carbachol were 51-fold and 25% greater than with acetylcholine respectively. This could be due to the presence of active acetylcholinesterase in this tissue. 2. The pA2 values obtained with the muscarinic antagonists were pirenzepine (8.52), AF-DX 116 (8.05), 4-DAMP (9.41) and hexahydroxiladifenidol (8.66). The slope values of Schild plots were not significantly different from unity, indicating c

Nicotinic and muscarinic cholinergic receptors in Alzheimer's disease and related disorders.

Alzheimer's disease (AD) and Parkinson's disease (PD) and to a lesser extent progressive supranuclear palsy are characterized by dysfunction of the cholinergic basal forebrain neurons which send projections to cortex and other areas of the telencephalon. In this paper we demonstrate that this cholinergic dysfunction is associated with consistent reductions in density of cortical nicotinic but not muscarinic cholinergic receptors in these disorders. These cholinergic abnormalities are not found in Huntington's disease.

Muscarinic cholinergic receptors in area postrema and brainstem areas regulating emesis.

Central cholinergic pathways modulate both the perception of excessive motion stimuli and the expression of motion sickness symptoms, such as nausea and vomiting. Specific brainstem areas which mediate motion-induced emesis include the area postrema (AP), vagal nuclear complex (VNC), reticular formation (RF) at the site of the vomiting center, and the vestibular complex (VC). In this report, histological studies indicated the cellular organization of brainstem structures mediating emesis was similar in bovine and squirrel monkey brain. The objective of this study was to characterize biochemica

Cholinergic augmentation of lymphocyte-mediated cytotoxicity. A study of the cholinergic receptor of cytotoxic T lymphocytes.

Cholinergic agonists have previously been shown to augment the ability of sensitized lymphocytes to injure cells bearing the sensitizing alleantigens. The cholinergic receptor of the attacking lymphocyte population has been studied with pharmacological manipulation of an in vitro system that quantitates the injury mediated by sensitized attacking cells upon target cells. The data reveal that muscarinic ligands are several orders of magnitude more potent than nicotinic agents in altering cytotoxicity.

Biochemical indentification of the mammalian muscarinic cholinergic receptor.

The muscarinic cholinergic antagonist 3-quinuclidinyl benzilate (QNB) binds avidly but reversibly to the muscarinic cholinergic recptor of mammalian brain and peripheral tissues. [3H]QNB binding provides a simple, sensitive, specific assay for the muscarinic cholinergic receptor binding. Inhibition of [3H]QNB binding to homogenates of brain and guinea pig ileum by muscarinic drugs correlates with their pharmacologic potencies, while nicotinic agents and noncholinergic drugs have neglibible affinity. The regional distribution of [3H]QNB binding throughout rat and monkey brain parallels to a maj

Narcolepsy: cholinergic receptor changes in an animal model.

An inbred colony of narcoleptic doberman pinschers has been analyzed for muscarinic receptor levels in 19 discrete brain regions. In comparison to age-matched controls, receptors were generally elevated in the brainstem and reduced in forebrain areas. No changes in receptor binding affinity were detected. The increased receptor levels found in the brainstem suggest that cholinoceptive neurons in this region are hypersensitive and may be involved in the initiation of cataplexy and other aspects of the narcolepsy syndrome.
