Dopamine transporters, D2 receptors, and dopamine release in generalized social anxiety disorder.

BACKGROUND: Dopamine D2 receptor and dopamine transporter (DAT) availability in the striatum (STR) have each been reported abnormal in generalized social anxiety disorder (GSAD) in studies using single photon emission computerized tomography (SPECT). D2 receptors and DAT have not previously been studied within the same GSAD subjects, however, and prior GSAD studies have not assessed dopamine release or subdivided the STR into functional subregions. METHODS: Unmedicated adults with GSAD (N=17) and matched healthy comparison (HC) subjects (N=13) participated in this study. Of these, 15 GSAD and

Bidirectional regulation of dopamine D2 and neurotensin NTS1 receptors in dopamine neurons.

Several lines of evidence suggest a close association between dopamine (DA) and neurotensin (NT) systems in the CNS. Indeed, in the rodent brain, abundant NT-containing fibres are found in DA-rich areas such as the ventral tegmental area and substantia nigra. Moreover, it has been shown in vivo that NT, acting through its high-affinity receptor (NTS1), reduces the physiological and behavioural effects of DA D2 receptor (D2R) activation, a critical autoreceptor feedback system regulating DA neurotransmission. However, the mechanism of this interaction is still elusive. The aim of our study was

Psychosis pathways converge via D2high dopamine receptors.

The objective of this review is to identify a target or biomarker of altered neurochemical sensitivity that is common to the many animal models of human psychoses associated with street drugs, brain injury, steroid use, birth injury, and gene alterations. Psychosis in humans can be caused by amphetamine, phencyclidine, steroids, ethanol, and brain lesions such as hippocampal, cortical, and entorhinal lesions. Strikingly, all of these drugs and lesions in rats lead to dopamine supersensitivity and increase the high-affinity states of dopamine D2 receptors, or D2High, by 200-400% in striata. Sim

Leukocytic Dopamine D2 Receptors as Biomarkers for Brain Dopamine Levels in Parkinson Disease.

Having the ability to objectively index dopamine (DA) levels in the brain with a peripheral biomarker of brain DA would enable the objective monitoring of the progression of Parkinson disease (PD) and other DA-dependent disorders. This study investigates this potential biomarker using a DA-depletion approach, the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) neurotoxin model and subjects with PD, which are well-known models of DA depletion in the midbrain of rodents and humans, respectively. MPTP-induced DA depletion in the substantia nigra compacta resulted in a significant decrease in

Partial agonistic action of clozapine at dopamine D2 receptors in dopamine depleted animals.

In the present experiments, the locomotor and stereotypic stimulant effects of the atypical antipsychotic agent, clozapine were studied in mice depleted of their brain dopamine by reserpine and alpha-methyl-p-tyrosine pretreatment. Clozapine at a dose of 1 mg/kg produced slight stimulation of locomotor activity, but failed to produce any stereotypy. However, clozapine at 0.5 or 2 mg/kg failed to show increase in locomotor activity or stereotypy. Clozapine potentiated the locomotor activity and stereotypy induced by selective dopamine D1 receptor agonist, SKF 38393 but blocked locomotor activit

Studies on vascular dopamine receptors with the dopamine receptor agonist: SK&F 82526.

The vascular effects of a new dopamine receptor agonist, SK&F 82526, were evaluated in isolated rabbit splenic arterial ring segments. In this preparation, previously shown to possess dopamine receptors, SK&F 82526 produced a stereoselective relaxation with the R-enantiomer more active (ED50 1 X 10(-6) M) than the S-enantiomer (ED50 7 X 10(-6) M). Because SK&F 82526 lacks alpha agonist adrenoreceptor activity, a unique feature of these studies was the ability to examine the relaxant action of a dopamine receptor agonist using norepinephrine to contract the tissue in the absence of phenoxybenza

The physiology, signaling, and pharmacology of dopamine receptors.

G protein-coupled dopamine receptors (D1, D2, D3, D4, and D5) mediate all of the physiological functions of the catecholaminergic neurotransmitter dopamine, ranging from voluntary movement and reward to hormonal regulation and hypertension. Pharmacological agents targeting dopaminergic neurotransmission have been clinically used in the management of several neurological and psychiatric disorders, including Parkinson's disease, schizophrenia, bipolar disorder, Huntington's disease, attention deficit hyperactivity disorder (ADHD(1)), and Tourette's syndrome. Numerous advances have occurred in un

[Effect of (+/-)12-chloroscoulerine on brain dopamine receptors].

AIM: To assess potencies of tetrahydroprotoberberines (THPB) and hydrobenzyltetrahydroisoquinolines (HBTI) on DA receptors. METHODS: The receptor binding assay with calf striatum to D1 and D2 receptors, and the animal behavior tests were used. RESULTS: (+/-) 12-Chloroscoulerine (CSL) was the most potent one among the THPB and HBTI. The affinities of CSL to D1 and D2 receptors were 13 and 51 nmol.L-1, respectively. In rats, CSL showed an antagonistic effect on the stereotypy and induced catalepsy. In the 6-OHDA lesioned rats, however, CSL exerted the agonistic effect to DA receptors. CONCLUS

Peripheral and central dopamine receptors in respiratory control.

The role of peripheral and central dopaminergic mechanisms in respiratory control was studied in anesthetized cats. In one series, we simultaneously measured carotid chemoreceptor and ventilatory responses to hypoxia and hypercapnia before and after a saturation dose of intravenous domperidone, a peripheral dopamine (D2) receptor antagonist. Both carotid chemoreceptor and ventilatory responses were augmented by domperidone essentially in proportion, suggesting that they reflected the increase of peripheral chemoreceptor activity. Haloperidol which crosses into the brain from blood, given subse

The pathophysiological functions mediated by D1 dopamine receptors.

The administration of the D1 DA receptor antagonist SCH 23390 produces catalepsy in rat, and this behavior can be abolished by pretreatment with selective D2 DA receptor agonists. The administration of mixed D1/D2 DA agonists, but not of selective D2 DA agonists, produces SMB behavior in monkeys with surgical unilateral VMT lesions of the brain stem. The DA agonist-induced SMB behavior is abolished by pretreatment of the monkeys with either the D1 DA antagonist SCH 23390 or with the mixed D1/D2 antagonist fluphenazine, but not with the selective D2 antagonist (+/-) sulpiride. Dopamine receptors in pancreatic acinar cells from dog.

Dispersed acini from dog pancreas were used to characterize dopamine receptors in a study on the stimulation of cellular cyclic AMP formation and the binding of [3H]dopamine. Dopamine elicited concentration-dependent stimulation of cellular cyclic AMP with a maximal increase occurring at a concentration of 0.1 mM (EC50 = 1 microM). Epinine produced a potent stimulation similar to that by dopamine; the alpha-adrenoceptor agonists (amidephrine and noradrenaline) were less potent. Isoproterenol and apomorphine were ineffective. The effect of dopamine was potently blocked by dopamine receptor anta

Cardiovascular dopamine receptors: physiological, pharmacological and therapeutic implications.

Dopamine receptor activation can lead to pronounced changes in cardiovascular function. The myriad of effects produced by dopamine receptor agonists results from the activation of dopamine receptors located at different anatomical sites in the cardiovascular system. Further basic research is required to better characterize these dopamine receptors so as to allow the development of more specific dopamine receptor agonists. Endogenous dopamine may be involved in the physiological control of fluid and electrolyte balance and continuing research efforts in this area should provide for a better und

Detection of dopamine receptors in the area postrema.

[3H]Spiroperidol labeled dog area postrema membranes with high affinity (KD, 0.1 nM) and stereospecificity. The displacement of the radioligand by different dopamine agonists and antagonists indicated that the labeled sites were dopaminergic in nature. Guanine nucleotides decreased agonist affinity for the labeled sites without affecting the affinity for antagonists. Adenylate cyclase in the area postrema was stimulated by guanine nucleotides and sodium fluoride but not by dopamine. The results suggest that the dopamine receptor in the area postrema can be classified as D2-type receptors.

Elevation of brain neuroleptic/dopamine receptors in schizophrenia.

To test the dopamine hypothesis of schizophrenia the authors measured specific 3H-neuroleptic/dopamine binding sites in three dopamin-rich regions of 59 postmortem normal human brains and 50 postmortem brains from schizophrenic patients using 3H-haloperidol and 3H-spiperone. The binding of 2 nM 3H-haloperidol and of 1 nM 3H-spiperone was significantly elevated in the brains from schizophrenic patients. The brain regions from patients who had no history of being treated with neuroleptic drugs also exhibited significantly higher binding of the 3H-neuroleptics. These results are compatible with t

Nucleus Accumbens Acetylcholine Receptors Modulate Dopamine and Motivation.

Environmental reward-predictive cues can motivate reward-seeking behaviors. Although this influence is normally adaptive, it can become maladaptive in disordered states, such as addiction. Dopamine release in the nucleus accumbens core (NAc) is known to mediate the motivational impact of reward-predictive cues, but little is known about how other neuromodulatory systems contribute to cue-motivated behavior. Here, we examined the role of the NAc cholinergic receptor system in cue-motivated behavior using a Pavlovian-to-instrumental transfer task designed to assess the motivating influence of a

Dopamine receptors--physiological understanding to therapeutic intervention potential.

There are two families of dopamine (DA) receptors, called D1 and D2, respectively. The D1 family consists of D1- and D5-receptor subtypes and the D2 family consists of D2-, D3-, and D4-receptor subtypes. The amino acid sequences of these receptors show that they all belong to a large superfamily of receptors with seven transmembrane domains, which are coupled to their intracellular signal transduction systems by G-proteins. The implications of DA receptors in neuropsychiatry and cardiovascular and renal diseases are discussed. Neuropsychiatry indications include Parkinson's disease, schizophre

Clozapine occupies high levels of dopamine D2 receptors.

An important discrepancy has been noted concerning the number of dopamine D2 receptors which must be occupied in patients by clozapine, in contrast to other antipsychotic drugs, in order to achieve an antipsychotic effect. For example, when D2 receptors are labelled by radioactive raclopride or spiperone congeners in patients, psychosis-controlling doses of all antipsychotic drugs occupy about 70% or more of the D2 receptors in patients. However, equi-effective psychosis-controlling doses of clozapine (approximately 400 mg/day; approximately 70-130 nM in spinal fluid) only occupy between 20% a

Chronic pharmacological manipulation of dopamine receptors in brain.

Dopamine receptors in the brain play an important role in the treatment of schizophrenia and in the development of tardive dyskinesia. In Parkinson's disease the loss of dopamine innervation and the use of chronic administration of L-DOPA or therapy with dopamine agonists also affects the function of dopamine receptors in brain. Subacute administration of neuroleptic drugs to rodents for a few weeks followed by the withdrawal of the drug induces supersensitivity of dopamine receptors in the striatum. However, the long-term administration of neuroleptic drugs to rodents shows that typical neuro
