Inducible galanin and GalR2 receptor system in motor neuron injury and regeneration.

Galanin has been ascribed several physiological roles that are thought to be mediated via multiple galanin receptors. Recently, two galanin receptors--galanin receptor-1 (GalR1) and galanin receptor-2 (GalR2)--have been cloned and characterized and shown to have differences in amino acid sequence, pharmacology, and second messenger signaling systems. Previous studies have demonstrated an up-regulation of galanin expression in damaged neurons of several different types. Using in situ hybridization histochemistry this study investigated whether adult cranial motor neurons express mRNAs encoding

Galanin receptor-mediated inhibition of glutamate release in the arcuate nucleus of the hypothalamus.

It is thought that galanin, a 29 amino acid neuropeptide, is involved in various neuronal functions, including the regulation of food intake and hormone release. Consistent with this idea, galanin receptors have been demonstrated throughout the brain, with high levels being observed in the hypothalamus. However, little is known about the mechanisms by which galanin elicits its actions in the brain. Therefore, we studied the effects of galanin and its analogs on synaptic transmission using an in vitro slice preparation of rat hypothalamus. In arcuate nucleus neurons, application of galanin resu

Preferential Gs protein coupling of the galanin Gal1 receptor in the µ-opioid-Gal1 receptor heterotetramer.

Recent studies have proposed that heteromers of µ-opioid receptors (MORs) and galanin Gal1 receptors (Gal1Rs) localized in the mesencephalon mediate the dopaminergic effects of opioids. The present study reports converging evidence, using a peptide-interfering approach combined with biophysical and biochemical techniques, including total internal reflection fluorescence microscopy, for a predominant homodimeric structure of MOR and Gal1R when expressed individually, and for their preference to form functional heterotetramers when co-expressed. Results show that a heteromerization-dependent cha

Increased galanin-galanin receptor 1 signaling, inflammation, and insulin resistance are associated with affective symptoms and chronic fatigue syndrome due to long COVID.

BACKGROUND: Patients with Long COVID (LC) often experience neuropsychiatric symptoms such as depression, anxiety, and chronic fatigue syndrome (CFS), collectively referred to as the physio-affective phenome of LC. Activated immune-inflammatory pathways and insulin resistance significantly contribute to the physio-affective phenome associated with LC. METHODS: In a cohort of 90 individuals, categorized into those with and without LC, we evaluated, 3-6 months following acute SARS-CoV-2 infection, the correlations between the Hamilton Depression (HAMD), Hamilton Anxiety (HAMA), and Fibro-Fatigue

Molecular identification and coexpression of galanin and GalR-1 galanin receptor in the human ocular ciliary epithelium: differential modulation of their expression by the activation of alpha2- and beta2-adrenergic receptors in cultured ciliary epithelial cells.

Here we report the coexpression of the neuropeptide galanin and GalR-1 galanin receptors in the human ciliary epithelium, a bilayer of neuroepithelial cells [nonpigmented (NPE) and pigmented (PE)] with neuroendocrine functions, and in a cell line (ODM-2) derived from the NPE cells. Stimulation of ODM-2 cells with phorbol ester [phorbol 12-myristate 13-acetate (PMA)] or forskolin resulted in an up-regulation (two- to threefold) of galanin mRNA expression. Procaterol, a selective beta2-adrenergic agonist, and the catecholamine isoproterenol exerted a long-term down-regulation on galanin mRNA exp

Solution structure by 2D 1H-NMR of a chimeric peptide recognized by galanin and neuropeptide Y receptors.

The 25 amino acid residue chimeric peptide M32, galanin(1-13)-neuropeptide Y(25-36)-amide, was synthesized. The peptide was found to be recognized by both galanin and NPY receptors. The solution structure in 30% (v/v) 1,1,1,3,3,3-hexafluoro-2-propanol was examined by 2-D 1H-NMR and by CD. Proton resonance assignments were made, and structures were calculated using DIANA and refined by restrained energy minimization and molecular dynamics. The obtained structures contain an alpha-helical part in the NPY portion of the peptide including residues 13-20, and in some structures it continues to the

Evidence for endogenous inhibition of autotomy by galanin in the rat after sciatic nerve section: demonstrated by chronic intrathecal infusion of a high affinity galanin receptor antagonist.

We have studied the effect of M-35 [Galanin(1-12)-Pro-bradykinin(2-9)-amide], a newly developed high affinity antagonist for galanin receptors, on self-mutilation (autotomy) behavior of the deafferented limb in rats after unilateral section of sciatic nerves. M-35 (1.3 micrograms/microliters) or saline was applied to the lumbar spinal cord through a chronically implanted intrathecal catheter at a rate of 0.5 microliter/h for 10 days post axotomy via an osmotic minipump. Axotomized rats infused with M-35 autotomized significantly more than those perfused intrathecally with saline or those axoto

Spexin disrupts migrating myoelectric complex in the rat small intestine: The role of galanin-2 and muscarinic receptors.

Spexin (SPX) is an endogenous peptide expressed throughout the gastrointestinal tract. Although its impact on postprandial intestinal motility has been examined, its effect during fasting remains unclear. This study aimed to investigate the direct effects of SPX on jejunal and ileal segments in vitro, its effects on fasting small intestinal motility in vivo, and to determine the roles of galanin-2, muscarinic, and 5-hydroxytryptamine-3 (5-HT₃) receptors in these actions. The contractile responses of rat jejunal and ileal segments to SPX (10⁻⁹-10⁻⁶ M), with or without pretreatment with galanin-

Impact of galanin receptors 2 and 3 double-knockout on neuroinflammation and functional recovery following traumatic brain injury.

Traumatic brain injury (TBI) is one of the world's leading causes of death and disability in young individuals and the mechanism underlying TBI-associated neuroinflammation is poorly understood. The regulatory neuropeptide galanin (GAL) and its three receptors (GAL1-3R) are assumed to modulate the neuroinflammatory response following TBI, especially by signalling via GAL2R and GAL3R. Therefore, the role of GALRs in acute neuroinflammation and functional recovery following moderate Controlled Cortical Impact TBI was studied using GAL2/3R-double-KO (GAL2/3R-KO) mice. Effects of trigeminal nerve injury on the expression of galanin and its receptors in the rat trigeminal ganglion.

In the spinal nervous system, the expression of galanin (GAL) and galanin receptors (GALRs) that play important roles in the transmission and modulation of nociceptive information can be affected by nerve injury. However, in the trigeminal nervous system, the effects of trigeminal nerve injury on the expression of GAL are controversy in the previous studies. Besides, little is known about the effects of trigeminal nerve injury on the expression of GALRs. In the present study, the effects of trigeminal nerve injury on the expression of GAL and GALRs in the rat trigeminal ganglion (TG) were inve

Neuroanatomical characterization of the G protein-coupled receptor activity evoked by galanin-related ligands.

Galanin neuropeptide is distributed throughout the mammalian nervous system modulating a plethora of diverse physiological functions, including nociception, cognition and neuroendocrine regulation. The regulation of the galaninergic system is an interesting approach for the treatment of different diseases associated to those systems. Nevertheless, the pharmacological selectivity and activities of some galanin receptor (GalR) ligands are still in discussion and seem to depend on the dose, the receptor subtype and the second messengers to which they are coupled at different brain areas. The acti

Galanin receptor 2: A multifunctional modulator in neurological diseases - Structure, mechanisms, and therapeutic prospects.

Galanin receptor 2 (GALR2), as a subtype of neuropeptide galanin receptor, is predominantly expressed in the central and peripheral nervous systems. Accumulating evidence suggests that GALR2 is involved in the pathogenesis of depression, Alzheimer's disease, epilepsy, neuropathic pain, multiple sclerosis, as well as in neurological tumors. Here, we summarize the current knowledge on the molecular architecture, signal transduction mechanisms, and diverse biological functions of GALR2. In addition, we also discuss the involvement of GALR2 in neurological diseases, with a particular emphasis on e

The Effect of Stress on the Galaninergic System in the Rat Adenohypophysis: mRNA Expression and Immunohistochemistry of Galanin Receptors.

The neuropeptide galanin is a widely distributed neurotransmitter/neuromodulator that regulates a variety of physiological processes and also participates in the regulation of stress responses. The effect of stress is dependent on the activity of the hypothalamic-adenohypophyseal-adrenal axis. Although the adenohypophysis is a crucial part of this axis, galanin peptides and their receptors have not yet been identified in this part of the pituitary after activation of the stress response. Since there are many controversies about the occurrence of individual galanin receptor subtypes in the aden

Antinociceptive roles of galanin receptor 1 in nucleus accumbens of rats in a model of neuropathic pain.

It has been reported that galanin and its receptors might be involved in the modulation and transmission of nociception in the central nervous system. Our previous research has also demonstrated that galanin induces antinociception in the nucleus accumbens (NAc) of intact rats. However, the interaction between galanin and its receptors in the NAc and the underlying mechanism of suppressing pain transmission remain unclear. The present study seeks to determine the antinociception induced by galanin receptor (GalR)-1 stimulation in the NAc of rats with neuropathic pain. The left sciatic nerve of

Galanin receptor binding sites in the temporal and occipital cortex are minimally affected in Alzheimer's disease.

Galanin receptor binding sites were examined in the inferior temporal and medial occipital gyri of patients with Alzheimer's disease (AD) and matched control subjects using quantitative autoradiography. In the inferior temporal gyrus, galanin binding was reduced selectively in layers V-VI of the AD cases compared to controls, the magnitude of the reduction (45%) being similar to that of choline acetyltransferase activity (40%) in this region. In the medial occipital gyrus, galanin binding in the AD cases was not different from controls in any cortical layer despite a reduction in choline acety

Receptor subtype-specific pronociceptive and analgesic actions of galanin in the spinal cord: selective actions via GalR1 and GalR2 receptors.

Galanin is a 29-aa neuropeptide with a complex role in pain processing. Several galanin receptor subtypes are present in dorsal root ganglia and spinal cord with a differential distribution. Here, we describe a generation of a specific galanin R2 (GalR2) agonist, AR-M1896, and its application in studies of a rat neuropathic pain model (Bennett). The results show that in normal rats mechanical and cold allodynia of the hindpaw are induced after intrathecal infusion of low-dose galanin (25 ng per 0.5 microl/h). The same effect is seen with equimolar doses of AR-M1896 or AR-M961, an agonist both

Epigenetic inactivation of galanin receptors in salivary duct carcinoma of the parotid gland: Potential utility as biomarkers for prognosis.

Salivary duct carcinoma (SDC) constitutes one of the most aggressive cancers in the salivary gland and is associated with a poor prognosis; however, no established systemic therapy options are available. SDC exhibits biological similarity to prostate and breast cancers, therefore anti-hormone therapy and molecular target therapies are available, however with limited beneficial effects. Galanin and galanin receptors (GALRs) are well established as molecular biomarkers to predict the survival rate and risk of recurrence of head and neck squamous cell carcinoma. The present study investigated the

Association of Galanin and Its Receptor Gene Polymorphism with Opioid Dependence: Preliminary Findings from India.

BACKGROUND: Genetic factors contribute to the development of opioid dependence syndrome (ODS), with evidence suggesting that the neuropeptide galanin, which plays a role in the stress response, may influence addiction risk through its receptor galanin receptor 1 (GALR1). However, this area is largely unexplored in the Indian context. METHODS: This case-control study included 85 opioid-dependent patients and 85 healthy controls, all males, recruited from a tertiary care hospital in North India. All participants were assessed using a socio-demographic proforma, a case record form for substance

The activation of galanin receptor 2 plays an antinociceptive effect in nucleus accumbens of rats with neuropathic pain.

Our previous research has shown that galanin plays an antinociceptive effect via binding to galanin receptors (GalRs) in nucleus accumbens (NAc). This study focused on the involvement of GalR2 in galanin-induced antinociceptive effect in NAc of neuropathic pain rats. The chronic constriction injury of sciatic nerve (CCI) was used to mimic neuropathic pain model. The hind paw withdrawal latency (HWL) to thermal stimulation and hind paw withdrawal threshold (HWT) to mechanical stimulation were measured as the indicators of pain threshold. The results showed that 14 and 28 days after CCI, the exp
