Lack of influence of histamine H1-, and H2-receptor antagonists on histamine level in rat brain.

The intraperitoneal administration of three antihistaminic drugs of H1 type: mepyramine, diphenyhydramine (each 5 and 30 mg/kg) and danitracen (2 and 10 mg/kg) did not modify histamine level in the rat brain after 2 h. Histamine H2-receptor antagonists: cimetidine (25-250 microgram) and metiamide (100 microgram) given directly into the brain had no significant effect on histamine content after 1.5 h. L-histidine (0.5 g/kg i.p.) increased after 1.5 h histamine level in various experiments by 50-75% above control. All antihistaminic drugs did not change significantly histamine level in the brain

A novel membrane sensor for histamine H1-receptor antagonist "fexofenadine".

The construction and general performance of thirteen new polymeric membrane sensors for the determination of fexofenadine hydrochloride based on its ion exchange with reineckate, tetraphenylborate and tetraiodomercurate have been studied. The effects of membrane composition, type of plasticizer, pH value of sample solution and concentration of the analyte in the sensor internal solution have been thoroughly investigated. The novel sensor based on reineckate exchanger shows a stable, potentiometric response for fexofenadine in the concentration range of 1 x 10(-2) - 2.5 x 10(-6) M at 25 degrees

Increased expression of histamine H1 receptor mRNA in allergic rhinitis.

BACKGROUND: Histamine plays an important role in producing nasal symptoms via histamine H1 receptor (H1R) in allergic rhinitis. It is reported that the minimum histamine concentration that induces sneezing is lower in allergic patients than in normal control subjects. Previous studies by binding assay on H1R gave divided results on whether the number of H1Rs is increased in allergic rhinitis or not. OBJECTIVE: The objective of this study was to examine if H1R mRNA expression is increased in patients with allergic rhinitis compared with normal healthy volunteers. METHODS: We extracted RNA fro

Systemic mastocytosis treated with histamine H1 and H2 receptor antagonists.

A case of systemic mastocytosis with skin characteristics of telangiectasia macularis eruptiva perstans is reported. Systemic involvement was confirmed as a combination of highly increased urinary excretion of methyl imidazole acetic acid (Melm AA) and increased amounts of mast cells in skin, liver and colon transversum. Treatment with cimetidine (H2 receptor antagonist) resulted in diarrhea, however, the addition of cyproheptadine (H1 receptor antagonist) was beneficial in amelioration of the cutaneous symptoms of mastocytosis. The excretion of Melm AA was unchanged during treatment. After 7

Trifluoperazine inhibits phosphoinositide hydrolysis as a histamine H1-receptor antagonist.

In human astrocytoma cells (1321N1), trifluoperazine potently inhibited histamine-induced phosphoinositide hydrolysis, while it slightly inhibited carbachol-induced phosphoinositide hydrolysis. Trifluoperazine as well as diphenhydramine inhibited [3H]mepyramine binding in astrocytoma cell membranes with Ki values of 0.052 microM and 0.005 microM, respectively. However, trifluoperazine only slightly inhibited [3H]quinuclidinyl benzilate binding with a Ki value of 3 microM. These results indicate that trifluoperazine specifically inhibits histamine-induced phosphoinositide hydrolysis as a histam

Is protein kinase C involved in histamine H1-receptor desensitization?

Histamine H1-receptor mediated effects in guinea-pig lung and intestine appear to desensitize homologously rather rapidly. Within a few minutes of exposure to a high concentration of histamine (30-100 microM) the c-GMP production in guinea-pig lung and the contraction of guinea-pig jejunum are markedly attenuated. In both tissues the responses to other stimulating agents (e.g. muscarinic agent, calcium ionophore) are not affected. The protein kinase C (PKC) activating phorbolester phorbol-12,13- dibutyrate (PDB) concentration-dependently depresses H1-receptor responses in both tissues. Yet, PD

Cerebrospinal inflammatory response following scorpion envenomation: role of histamine H1 and H3 receptors.

BACKGROUND: The mechanism of the inflammatory process induced by scorpion venom in the cerebrospinal tissues has not yet been completely elucidated. Therefore, we aimed to investigate the role of histamine through its H1 and H3 receptors in this process. METHODS: Histamine H1 and H3 receptor antagonists, Hydroxyzine (10 mg/kg) and Betaserc (20 mg/kg), respectively, were administered by intraperitoneal route to mice 1 h before subcutaneous envenomation with a subletal dose (0.5 mg/kg) of Androctonus australis hector venom. Cerebrospinal inflammation response was assessed 24 h after envenomatio

Imaging histamine H1 receptors in the living human brain with carbon-11-pyrilamine.

The brain distribution and kinetics of the H1 receptor antagonist, carbon-11-pyrilamine (11C-pyrilamine) were examined in vivo in two baboons and one human by positron emission tomography. After i.v. administration of the tracer, brain activity peaked within 20 min after injection and subsequently decreased, reflecting reversible binding to the receptor. Pretreatment with 1 mg/kg diphenhydramine reduced the brain activity at 70 min by 33%, 29%, 26%, and 23% of the control values in frontal cortex, temporal cortex, hippocampus, and cerebellum, respectively. Coinjection of 1 and 5 mg/kg cold pyr

Histamine stimulates ion transport by dog pancreatic duct epithelial cells through H1 receptors.

Histamine affects pancreatic secretion, but its direct action on ion transport by pancreatic duct epithelial cells (PDEC) has not been defined. We now characterize the secretory effects of histamine on cultured, well-differentiated, and nontransformed dog PDEC. Histamine stimulated, in a concentration-dependent manner (1-100 microM), a cellular 125I- efflux that was inhibited by 500 microM 5-nitro-2-(3-phenylpropylamino)benzoic acid, 2.5 mM diphenylamine-2-carboxylate, and 500 microM DIDS and thus mediated through Ca2+-activated Cl- channels. Histamine-stimulated 125I- efflux was 1) inhibited

Antidiabetic Properties of Histamine H1 Receptor Antagonists: A Systematic Review.

Allergies and diabetes mellitus are highly prevalent, frequently co-occurring diseases. Antihistamines, the first-line treatment for allergies, block H1 receptors but also exhibit anti-inflammatory and antioxidant properties that may improve glucose metabolism. This systematic review examined the evidence for the potential antidiabetic effects of commonly used first- and second-generation antihistamines. A comprehensive search of the PubMed database (1978-May 2026) yielded 17 eligible in vivo, in vitro, clinical, and in silico studies. Most reports indicated that antihistamines improve glucose

Short-term desensitization of phosphatidylinositol turnover via muscarinic acetylcholine receptors and histamine H1-receptors in smooth muscle.

Smooth muscle of guinea-pig taenia caecum was desensitized by treatment with 10(-4)M carbachol or 10(-4)M histamine for 30 min in Ca-free solution containing 2mM EGTA. Phosphatidylinositol turnover stimulated by carbachol was not reduced by desensitization with either carbachol or histamine, while the turnover stimulated by histamine was reduced by desensitization with histamine, but not with carbachol. These results are consistent with our previous report (1) that heterologous desensitization induced by carbachol occurs at intracellular Ca stores and homologous desensitization by histamine oc

Ameliorative Effect of Hesperidin Against Motion Sickness by Modulating Histamine and Histamine H1 Receptor Expression.

Motion sickness (MS) is the visceral discomfort caused due to contradicting visual and vestibular inputs to the brain leading to nausea and vomiting. Sensory conflict theory which proves histamine elevations as the primary reason for MS provides a path for an effective pharmaco-therapy. We aimed to evaluate the anti-MS effect of hesperidin (HSP) by modulating histamine and histamine receptor H1 (HRH1) expression. The inhibitory effect of HSP on histamine release was studied in KU812 cells treated with 10 µM calcium ionophore. The in vivo anti-MS effect of HSP was evaluated in Balb/c mice. Thir

[The effect of histamine and histamine H1 and H2 receptor blockers on the tone of the longitudinal muscles in the human appendix].

The aims of this study were to determine whether the effect of histamine on a smooth musculature of the human appendix is mediated by histamine H1-receptors only or whether histamine H2-receptors are involved, as well. The obtained results indicate histamine increases tonus in musculature; however, histamine H1-receptor antagonist (promethazine) decreases tonus. Histamine H2-receptor antagonist (cimetidine) has caused a mild increase in tonus of musculature what indicates that it has a certain intrinsic activity. By blocking histamine receptors with H1 and H2 antagonists which were used either

Histamine induces peripheral and central hypersensitivity to bladder distension via the histamine H1 receptor and TRPV1.

Interstitial cystitis/bladder pain syndrome (IC/BPS) is a common chronic pelvic disorder with sensory symptoms of urinary urgency, frequency, and pain, indicating a key role for hypersensitivity of bladder-innervating sensory neurons. The inflammatory mast cell mediator histamine has long been implicated in IC/BPS, yet the direct interactions between histamine and bladder afferents remain unclear. In the present study, we show, using a mouse ex vivo bladder afferent preparation, that intravesical histamine enhanced the mechanosensitivity of subpopulations of afferents to bladder distension. Role of brain histamine H1- and H2-receptors in neostigmine-induced hyperglycemia in rats.

We previously reported that when neostigmine, an inhibitor of acetylcholine esterase, was injected into the third cerebral ventricle, the concentration of hepatic venous plasma glucose was increased via central muscarinic receptors in anesthetized rats. To determine whether brain histamine receptors are involved in cholinergic system transmission with regard to central nervous system (CNS)-mediated glucoregulation, we examined the effects of the H1 receptor antagonist pyrilamine and the H2 receptor antagonist ranitidine on neostigmine-induced hyperglycemia in anesthetized rats. The injection o

Effects of histamine on coronary hemodynamics in humans: role of H1 and H2 receptors.

To evaluate whether histamine exerts a direct effect on coronary hemodynamics in humans, and to investigate the role played by H1 and H2 receptors in this response, intracoronary saline solution or histamine (4 micrograms) was administered in 10 patients with normal coronary arteries during diagnostic cardiac catheterization. Histamine injection was repeated after intravenous cimetidine (400 mg) and diphenhydramine (10 mg). The electrocardiogram, arterial pressure and thermodilution coronary blood flow were continuously monitored during and for 40 seconds after each injection. Immediately afte

Histamine H1- and H2-receptors in canine renal artery in vivo and in vitro.

The present work investigates (a) the modification by pretreatment with selective H1- and H2-receptor antagonists on the dose-response curves (DRC) to histamine for heart rate, blood pressure, renal arterial blood flow and renal vascular resistance in anesthetized dogs, and (b) the characteristics of the DRC to histamine in canine isolated renal artery. In vivo, pretreatment with metiamide (10 mg/kg i.v.) did not modify the DRC to histamine. In contrast, significant rightward shift of the DRC to histamine for all the hemodynamic parameters was observed after diphenhydramine (5 mg/kg i.v.). Com

[125I]Iodobolpyramine, a highly sensitive probe for histamine H1-receptors in guinea-pig brain.

[125I]Iodobolpyramine is a novel 125I-ligand for histamine H1-receptors, synthesised using the 125I-Bolton Hunter reagent (2000 Ci/mmol) for acylation of an aminopentyl analogue of mepyramine. Its specific binding varied linearly with the concentration of guinea-pig cerebellar membranes and represented about 80% of the total. Selective interaction with H1-receptors was demonstrated by estimation of Ki values of known agonists and antagonists and confirmed by the low affinity of histamine H2- and H3-receptor antagonists and of non-histaminergic agents. At 25 degrees C, [125I]iodobolpyramine exh

Histamine H1 receptor: A target to treat pancreatic ductal adenocarcinoma (PDAC) by repurposing approved H1-antihistamines?

New efficacious and safe therapies are needed for patients with pancreatic ductal adenocarcinoma (PDAC), the most common type of pancreatic cancer. A bioinformatic approach comparing human PDAC tumors to normal pancreas identified differentially expressed G protein-coupled receptors (GPCRs). The GPCRs with higher differential expression in PDAC included the histamine H1 receptor (HRH1), which, on average, is ~20-fold higher expressed in PDAC tumors. HRH1 is readily targetable by approved H1-antihistamines. Studies with human and murine PDAC tumor cells revealed that HRH1 agonists increase intr
