Involvement of NO-guanylyl cyclase pathway for the depression of spinal monosynaptic reflex by Mesobuthus tamulus venom in neonatal rat in vitro.

AIMS: The present study was undertaken to evaluate the role of nitric oxide (NO) in Mesobuthus tamulus (MBT) venom-induced depression of spinal reflexes. MAIN METHODS: Experiments were performed on isolated hemisected spinal cords from 4 to 6day old rats. Stimulation of a dorsal root with supramaximal strength evoked monosynaptic (MSR) and polysynaptic reflex (PSR) potentials in the corresponding segmental ventral root. KEY FINDINGS: Superfusion of MBT venom (0.3μg/ml) depressed the spinal reflexes in a time-dependent manner and the maximum depression was seen at 10min (MSR by 63%; PSR by 79

Noise-Limited Failure of OGY Chaos Control in Regulating Monosynaptic Reflex Variability in the In Vivo Cat Spinal Cord.

Monosynaptic reflexes (MSRs) elicited by constant-intensity group I afferent stimulation exhibit marked amplitude variability, commonly attributed to stochastic presynaptic modulation and dynamic postsynaptic excitability. Here, we tested whether this variability could be attenuated using the Ott-Grebogi-Yorke (OGY) chaos-control algorithm, which stabilizes unstable periodic orbits in low-dimensional nonlinear systems. In spinalized, anesthetized cats, real-time implementation of the OGY method failed to reduce MSR amplitude variability, as quantified by the coefficient of variation, and the r

[Long term depression of the recurrent inhibition of monosynaptic spinal reflexes after sciatic nerve crush in adult rats].

Sciatic nerve injury is a common disease of peripheral nerve in clinic. After nerve injury, there are many dysfunctions in motoneurons and muscles following regeneration. Previous studies mostly investigated the aspects related to the injured nerve, and the effect on the recurrent inhibition (RI) pathway of spine following regeneration was not fully understood. Following reinnervation after temporary sciatic nerve crush, the functional alteration of RI was studied. In adult rats, RI between lateral gastrocnemius-soleus (LG-S) and medial gastrocnemius (MG) motor pools was assessed by conditioni

Cholinergic mechanisms related to REM sleep. III. Tonic and phasic inhibition of monosynaptic reflexes induced by an anticholinesterase in the decerebrate cat.

The depression of the postural activity induced by intravenous injection of eserine sulphate (0.1 mg/kg), an anticholinesterase, has been studied in precollicular decerebrate cats. The extensor and flexor monosynaptic reflexes elicited by single shock stimulation of the GS, P1-FDHL and DP nerves are tonically depressed during the episodes of postural atonia induced by the anticholinesterase. A further phasic depression of the monosynaptic reflexes occurs during the bursts of rapid eye movements (REM) typical of these episodes. These changes in spinal reflex activity closely resemble the tonic

Daily passive cycling attenuates the hyperexcitability and restores the responsiveness of the extensor monosynaptic reflex to quipazine in the chronic spinally transected rat.

Activity-based interventions such as locomotor training or passive cycling have a positive influence on the spinal circuitry and recovery following a spinal cord injury (SCI). The use of quipazine in combination with exercise training has demonstrated a greater functional recovery than has exercise training alone. However, the influence of exercise or training on the responsiveness of the spinal cord to quipazine has not been examined following a chronic spinal transection. The purpose of this study was to characterize the flexor and extensor monosynaptic reflex (MSR) response pre- and post-qu

Involvement of AMPA receptors for Mesobuthus tamulus Pocock venom-induced depression of monosynaptic reflex in neonatal rat spinal cord in vitro.

Glutamate is a putative neurotransmitter at Ia-alpha motoneuron synapse in the spinal cord and mediate the action via N-methyl-D-aspartate (NMDA) and a-amino-3-hydroxy-5-methyl-4-isoxazole-propionate (AMPA) receptors. Since NMDA receptors are not involved in M. tamulus Pocock (MBT) venom-induced depression of spinal monosynaptic reflex (MSR), the present study was undertaken to evaluate the role of AMPA receptors in mediating the depression of MSR by MBT venom. The experiments were performed on isolated hemisected spinal cord from 4-6 day old rats. Stimulation of a dorsal root with supramaxima

Changes in segmental reflexes following chronic spinal cord hemisection in the cat. I. Increased monosynaptic and polysynaptic ventral root discharges.

The effect of a chronic spinal cord hemisection on segmental reflex transmission was studied in cats. Recordings of ventral root responses were made after a terminal transection below the initial lesion to eliminate descending influence through the intact spinal half. Procedures to ensure comparability between sides, that are lacking in earlier work on this experimental model, were introduced in the present work. It was demonstrated that there was an increase in reflex size on the lesioned (left) side, relative to the other side, of both mono- and polysynaptic reflexes. The reflex changes were

Early phase of vincristine neuropathy in man. Electrophysiological evidence for a dying-back phenomenon, with transitory enhancement of spinal transmission of the monosynaptic reflex.

Ten patients with Hodgkin's disease were examined before and after each administration of vincristine sulfate (2 intravenous injections of 1.4 mg/m2 of body surface during the first week of each month for 3 months). The motor conduction velocity of the peroneal nerve, the conduction velocity in palmar sensory fibres of the median nerve, and the conduction velocity in the H reflex pathway remained unchanged. The amplitude of distal muscle (extensor digitorum brevis) and sensory nerve (median) potentials decreased, while the maximal response of more proximal muscles (soleus) was not significantl

Strychnine-like action of the convulsant barbiturate, CHEB.

The effect of 5-(2-cyclohexylideneethyl)-5-ethyl barbituric acid (CHEB) on the isolated spinal cord of the immature rat was examined using extracellular recording. At concentrations less than 20 microM CHEB increased the monosynaptic reflex (MSR) but depressed the reflex at greater concentrations (30-100 microM). At concentrations which enhanced the monosynaptic reflex, CHEB reduced the responses of motoneurones to glycine and to a lesser extent to those of L-glutamate. In the presence of strychnine (5 microM), which enhanced both mono- and polysynaptic reflexes, CHEB produced only slight enha

Contribution of polysynaptic pathways in the mediation and plasticity of Aplysia gill and siphon withdrawal reflex: evidence for differential modulation.

The gill and siphon withdrawal (GSW) reflex of Aplysia is centrally mediated by a monosynaptic and a polysynaptic pathway between sensory and motor neurons. The first objective of this article was to evaluate quantitatively the relative importance of these two components in the mediation of the GSW reflex. We have used an artificial sea water (ASW) solution containing a high concentration of divalent cations to raise the action potential threshold of the interneurons without affecting the monosynaptic component of the reflex (2:1 ASW). Compound EPSPs induced in gill or siphon motor neurons by

Neurophysiological mechanisms in abnormal reflex activities in cerebral palsy and spinal spasticity.

Dorsal root stimulation, carried out during operation in 80 spastic cerebral palsy patients and in one spastic quadriplegic patient, allowed a study of abnormal monosynaptic and polysynaptic reflexes. Frequency-related depression of the monosynaptic reflex was not present; increased activity through non-suppressed polysynaptic pathways was shown.

Spinal reflexes and the concentrations of 5-HIAA, MHPG, and HVA in lumbar cereborspinal fluid after spinal lesions in man.

Descending bulbospinal pathways that employ specific neurotransmitter substances are known to be capable of modulating segmental reflex activity in the experimental animal. To determine whether this might also occur in man correlations have been sought between the activity in spinal reflex pathways and the lumbar cerebrospinal fluid (CSF) concentrations of 5-hydroxyindolacetic acid (5-HIAA), 3 methoxy-4-hydroxyphenylglycol (MHPG), and homovanillic acid (HVA) in 12 patients with complete or virtually complete spinal lesions. The concentrations of 5-HIAA and MHPG in lumbar CSF ARE REDUCED AFTER

Differing actions of convulsant and nonconvulsant barbiturates: an electrophysiological study in the isolated spinal cord of the rat.

The effects of various pairs of convulsant and nonconvulsant barbiturates on mono- and polysynaptic activity were studied in the isolated spinal cord of the immature rat, using extracellular recording. The convulsant barbiturates, 5-ethyl-5-(3-methylbut-2'-enyl) barbituric acid (3M2B), 5-ethyl-5-(1,3-dimethylbut-1'-enyl) barbituric acid (1,3M1B) and (+)-5-(1,3-dimethylbutyl)-5-ethyl barbituric acid [(+) DMBB] all increased the monosynaptic reflex at concentrations between 5 and 50 microM with no change in polysynaptic activity. When the concentration was raised to between 100 and 300 microM, h

The action of local anaesthetics on the spinal cord of the cat.

The effects of intravenously injected lignocaine, procaine and amethocaine on the monosynaptic and polysynaptic reflexes and the cord dorsum potentials elicited by selective electrical stimulation of appropriate afferent fibres have been investigated in decerebrate cats before and after strychnine. The three drugs depress the amplitude of the reflex responses in the basal preparation for 40 to 50 min. Smaller doses eliminate the reflex augmentation produced by strychnine for comparable periods. Very small doses selectively abolish the augmentation of the P component of the cord dorsum response

Locomotor training modifies soleus monosynaptic motoneuron responses in human spinal cord injury.

The objective of this study was to assess changes in monosynaptic motoneuron responses to stimulation of Ia afferents after locomotor training in individuals with chronic spinal cord injury (SCI). We hypothesized that locomotor training modifies the amplitude of the soleus monosynaptic motoneuron responses in a body position-dependent manner. Fifteen individuals with chronic clinical motor complete or incomplete SCI received an average of 45 locomotor training sessions. The soleus H-reflex and M-wave recruitment curves were assembled using data collected in both the right and left legs, with s

Diazepam effect on reflex activity in patients with complete spinal lesions and in those with other causes of spasticity.

The effects of diazepam on reflex pathways in patients having complete spinal lesions and in patients having incomplete lesions of the spinal cord or multiple sclerosis are compared to determine whether diazepam has an action at spinal level. The drug produced no significant alteration in the excitability of the monosynaptic arc in the patients with complete spinal lesions. In contrast, in the group with incomplete spinal lesions or multiple sclerosis, diazepam reduced the excitability of the monosynaptic arc. This action did not appear to result from a reduction in fusimotor drive. Diazepam a

Botulinus toxin: effect on the central nervous system of man.

Electrical stimulation of multiple peripheral nerves, elicited "H" reflexes in a patient, 61 years old, with botulism. These reflexes are extremely suggestive of some central release or failure of inhibitory control of a monosynaptic or polysynaptic spinal reflex arc. This "central" action of botulinus toxin is similar to that suggested for tetanus toxin.
