Regulated cell death (RCD) is a basic biological phenomenon associated with cell and tissue homeostasis. Recent studies have enriched our understanding of RCD, and many novel cell death types, such as ferroptosis and pyroptosis, have been discovered and defined. Aortic aneurysm and dissection (AAD) is a life-threatening condition, but the pathogenesis remains largely unclear. A series of studies have indicated that the death of smooth muscle cells, endothelial cells and inflammatory cells participates in the development of AAD and that corresponding interventions could alleviate disease progre

BACKGROUND: Altered metabolism contributes to disease progression and variable treatment response, but metabolite abundance alone rarely determines whether stressed cells recover or undergo regulated cell death (RCD).
SCOPE OF REVIEW: This review proposes mitochondrial quality control (MQC) as an intermediate gating layer that links metabolic signals to cell-fate execution through coordinated redox control, mitochondrial integrity, organelle turnover, and renewal. Four representative metabolites-β-hydroxybutyrate (β-BHB), lactate, succinate, and acetate-span complementary substrate-, receptor

Regulated cell death (RCD) encompasses a variety of genetically controlled cell death programs that are essential for maintaining tissue homeostasis and have emerged as promising therapeutic targets in oncology. When used to prepare whole-tumor-cell (WTC) vaccines, dying cancer cells provide broad tumor-antigen repertoires together with damage-associated molecular patterns (DAMPs) that can function as endogenous adjuvants. This review examines how distinct RCD pathways shape antitumor immunity and how they can be exploited to engineer next-generation WTC vaccines. We summarize the molecular an

Renal cell carcinoma (RCC) is the third most prevalent urological malignancy, characterized by histological heterogeneity and complex molecular landscapes. Although established regimens, including surgical intervention, targeted therapies and immunotherapy, effectively manage early-stage disease, patients with advanced RCC still have a poor prognosis. The expanding spectrum of regulated cell death (RCD), ranging from established apoptosis and ferroptosis to emerging modalities like cuproptosis, disulfidptosis and ammonia-induced cell death, offers novel mechanistic insights into tumor vulnerab

Endocrine tumors arise from highly differentiated hormone-secreting cells and are characterized by strong hormone dependence, high metabolic activity, and marked phenotypic plasticity. During tumor progression and therapeutic intervention, endocrine tumor cells can adapt to adverse microenvironmental conditions and treatment pressure through remodeling of differentiation states, reprogramming of signaling pathways, and metabolic adaptation. Regulated cell death (RCD) is a key determinant of cell fate and an important mediator of anticancer treatment responses, while also being closely linked t

Chronic obstructive pulmonary disease (COPD) is a progressive inflammatory airway disorder, with emerging evidence highlighting the central role of regulated cell death (RCD) in its pathogenesis. However, the regulatory mechanisms, crosstalk between different RCD pathways, and their role in intercellular communication remain poorly understood. This review examines major forms of RCD (apoptosis, necroptosis, ferroptosis, pyroptosis, NETosis, and PANoptosis) in COPD, exploring their regulation, crosstalk, role in intercellular signaling, and potential as therapeutic targets. Mechanistically, RCD

Reactive oxygen and nitrogen species (RONS) constitute a unifying molecular axis across various cancer therapy modalities and are primary regulators of regulated cell death (RCD). Generally, cancer cells function under high oxidative stress to maintain proliferation, making them vulnerable to therapeutic approaches that push RONS levels above their survival threshold. The purpose of this review is to consolidate mechanistic evidence linking redox modulation to therapeutic efficacy. We analyzed current literature regarding standard and emerging anticancer modalities, including radiotherapy, pro

Ischemic stroke (IS) globally threatens human health due to its limited treatment options and the severity of its complications, which often result in long-term disability. SIRT1, a key member of the sirtuin family, plays a pivotal role in regulating various cellular processes by facilitating the deacetylation of both histone and non-histone substrates. As a critical regulator, SIRT1 is involved in multiple pathophysiological processes, including neuroinflammation, regulated cell death (RCD), oxidative stress, and more, which are central to the progression of IS. In IS, SIRT1 exerts its effect

Therapy resistance and disease recurrence remain major challenges across cancer types despite substantial advances in targeted therapy, chemotherapy, and immunotherapy. In many clinical settings, treatments successfully engage stress and death signaling pathways and produce rapid tumor regression, yet complete and durable eradication is uncommon. A consistent pattern emerges in which most tumor cells are eliminated while a small fraction survives, persists as residual disease, and later drives relapse. These observations suggest that failure is not always due to the absence of death pathways,

Myopathies are a heterogeneous group of skeletal muscle disorders caused by genetic mutations or acquired insults, including inflammation, infection, endocrine imbalance, and toxic exposure. Myopathies affect a substantial number of individuals worldwide and are a significant cause of chronic muscle weakness and disability. Despite diverse etiologies, progressive myofiber injury and degeneration underlie the functional decline across disease subtypes. Accumulating evidence indicates that innate immune activation and regulated myofiber death pathways, including apoptosis, necroptosis, and pyrop

Regulated cell death (RCD) is well-known as a controlled form of cell death regulated by one or more cascading signaling pathways. Over the past few decades, increasing evidence has implicated various non-apoptotic forms of RCD in neurons-including ferroptosis, parthanatos, necroptosis, pyroptosis, autophagic cell death, paraptosis, and cuproptosis-in the pathogenesis of neurodegenerative diseases (NDs) and their associated clinical manifestations. We provide an in-depth analysis of the associations between these RCDs and NDs, including Alzheimer's disease (AD), Parkinson's disease (PD), Hunti

Autoimmune diseases develop from a breakdown in immune tolerance and arise through complex interactions among genetic susceptibility, environmental factors, immune dysregulation, and tissue damage. Although apoptosis has traditionally been recognized as a central mechanism for maintaining immune homeostasis, increasing evidence has identified several non-classical forms of regulated cell death (RCD) that also contribute to autoimmune disease pathogenesis. These pathways include ferroptosis, pyroptosis, necroptosis, PANoptosis, NETosis, parthanatos, and cuproptosis. Unlike classical apoptosis,

Regulated cell death (RCD) is a fundamental biological process essential for tissue homeostasis, and the elimination of damaged or malignant cells. In cancer, dysregulation of RCD is closely linked to tumor initiation, progression, therapeutic resistance, and remodeling of the tumor microenvironment (TME). In this review, we classify RCD in cancer into three broad groups. Classical cell death types include apoptosis, autophagy, necroptosis, and pyroptosis, which have well-established roles in controlling cell fate. Metal-dependent pathways, represented by ferroptosis, and cuproptosis, highligh

Nigratine (also known as 6E11), a flavanone derivative of a plant natural product, was characterized as highly specific non-ATP competitive inhibitor of RIPK1 kinase, one of the key components of necroptotic cell death signaling. We show here that nigratine inhibited both necroptosis (induced by Tumor Necrosis Factor-α) and ferroptosis (induced by the small molecules glutamate, erastin, RSL3 or cumene hydroperoxide) with EC50 in the µM range. Taken together, our data showed that nigratine is a dual inhibitor of necroptosis and ferroptosis cell death pathways. These findings open potential new

Twelve regulated cell death programs have been described. We review in detail the basic biology of nine including death receptor-mediated apoptosis, death receptor-mediated necrosis (necroptosis), mitochondrial-mediated apoptosis, mitochondrial-mediated necrosis, autophagy-dependent cell death, ferroptosis, pyroptosis, parthanatos, and immunogenic cell death. This is followed by a dissection of the roles of these cell death programs in the major cardiac syndromes: myocardial infarction and heart failure. The most important conclusion relevant to heart disease is that regulated forms of cardiom

BACKGROUND: Sepsis is characterized by dysregulated inflammation leading to organ dysfunction. While immune activation and metabolic stress are central features, accumulating evidence suggests that regulated cell death programs actively influence inflammatory trajectories rather than serving as passive end-stage events. Apoptosis, pyroptosis, necroptosis, ferroptosis, and neutrophil extracellular trap (NET) formation have each been implicated in sepsis; however, their relative hierarchy, temporal dynamics, and compartment-specific relevance remain incompletely defined.
OBJECTIVE: To synthesiz

Regulated cell death (RCD) plays an important role in the pathophysiology of sepsis. Traditionally dominated by the concept of apoptosis, the understanding of cell death has significantly expanded to include a range of highly regulated, non-apoptotic mechanisms such as ferroptosis, necroptosis, pyroptosis, parthanatos, and NETosis. This review describes recent advances in the molecular pathways underpinning these RCDs and explores their contributions to inflammatory tissue damage during sepsis and ischemia/reperfusion injury. We highlight the key molecular drivers and signalling cascades for e
