Horizontal cell density and mosaic regularity in pigmented and albino mouse retina.

The present study has examined the density and mosaic regularity of the population of horizontal cells in the pigmented and albino mouse retina. Retinal wholemounts were immunostained for calbindin, and labeled cells within sampled fields were analyzed to determine horizontal cell soma size and density. The X-Y positional coordinates of each cell were determined, from which the geometrical properties of the mosaic were examined using nearest neighbor and Voronoi domain analyses, and regularity indices were derived from those measures. Autocorrelation and density recovery profile analyses were

Decoding the roles of bipolar, amacrine, and horizontal cells.

The retina consists of several specialized cell types, crucial for visual processing. Bipolar cells, amacrine cells, and horizontal cells are retinal interneurons, playing pivotal roles in transmitting and modulating visual information within the retina. In this chapter, we first highlight the significance of bipolar cells in relaying signals from photoreceptors to ganglion cells, initiating visual signaling. Recent consensus indicates the existence of different types of bipolar cells in various species, leading us to focus on their functional diversity and morphologic characteristics. Second,

Role of melatonin and its receptors in the vertebrate retina.

Melatonin is a chemical signal of darkness that is produced by retinal photoreceptors and pinealocytes. In the retina, melatonin diffuses from the photoreceptors to bind to specific receptors on a variety of inner retinal neurons to modify their activity. Potential target cells for melatonin in the inner retina are amacrine cells, bipolar cells, horizontal cells, and ganglion cells. Melatonin inhibits the release of dopamine from amacrine cells and increases the light sensitivity of horizontal cells. Melatonin receptor subtypes show differential, cell-specific patterns of expression that are l

Necessity and Sufficiency of Ldb1 in the Generation, Differentiation and Maintenance of Non-photoreceptor Cell Types During Retinal Development.

During mammalian retinal development, the multipotent progenitors differentiate into all classes of retinal cells under the delicate control of transcriptional factors. The deficiency of a transcription cofactor, the LIM-domain binding protein Ldb1, has been shown to cause proliferation and developmental defects in multiple tissues including cardiovascular, hematopoietic, and nervous systems; however, it remains unclear whether and how it regulates retinal development. By expression profiling, RNA in situ hybridization and immunostaining, here we show that Ldb1 is expressed in the progenitors

Spatiotemporal coordination of Slit-Robo repulsion and neurturin-Gfrα attraction guides multipolar migration during retinal lamination.

Multipolar migration is a conserved neuronal migration mode in the developing brain, enabling emerging neurons to navigate in crowded environments and reach precise laminar positions. Yet, how these cells interpret external cues to guide their migration is not fully understood. We investigate this question using retinal horizontal cells as a model. Combining transcriptomics, targeted CRISPR screening, and live imaging, we reveal the spatiotemporal guidance system underlying horizontal cell lamination: repulsive Slit1b/2-Robo2 signaling in the amacrine cell layer initiates apical horizontal cel

Oral proton pump inhibitors disrupt horizontal cell-cone feedback and enhance visual hallucinations in macular degeneration patients.

PURPOSE: Visual hallucinations (VHs) occur in macular degeneration patients with poor vision but normal cognitive function. The underlying mechanisms are poorly understood. We report the identification of pharmaceutical agents that enhance VH and use these agents to examine the contribution of retinal neurons to this syndrome. METHODS: We detail clinical observations on VH in five macular degeneration patients treated with proton pump inhibitors having the core structure, 2-pyridyl-methylsulfinyl-benzimidazole. We tested possible retinal mechanisms using paired whole cell recordings to examin

Machine Learning Identifies Key Gene Markers Related to Fetal Retina Development at Single-Cell Transcription Level.

PURPOSE: The retina is part of the central nervous system, and its function is vision. During the embryonic development of the fetus, it diversifies the seven major types of cells from a retinal progenitor cell. The purpose of this study is to investigate essential features that are necessary for the development of the fetal retina. METHODS: We generated a comprehensive single-cell transcriptional atlas of the human retina by leveraging datasets from the Chan Zuckerberg Initiative Single-Cell Biology collection of human fetal retinas. The eight critical types of retinal cells were investigate

The absence of BBSome function decreases synaptogenesis and causes ectopic synapse formation in the retina.

Photoreceptors possess ribbon synapses distinct from the conventional synapses in the brain. Little is known about the function of the BBSome, a complex integral in ciliary and intracellular trafficking, in ribbon synaptic formation. We performed immunohistochemistry using retinas from Bardet-Biedl Syndrome (BBS) mouse models and found that BBS mutant animals have significantly fewer ribbon synapses in the outer plexiform layer and increased ectopic synapses in the outer nuclear layer compared to controls. Many ectopic synapses in BBS mutant retinas are associated with horizontal cell axonal p

Utilizing a culture system for horizontal cells to study neural circuit assembly in the developing mouse retina.

The precise wiring of the nervous system relies on neurons extending their processes at the right time and place to find their appropriate synaptic partner. The mechanisms that determine when and where neurons extend their neurites during synaptogenesis remains a central question in the field. In the present study, we used a cell culture system coupled with live imaging to investigate the wiring mechanisms in the developing mouse retina. We focused on horizontal cells which are a class of interneurons in the outer mouse retina known to synapse selectively to the distinct types of photoreceptor

Horizontal cells of the rat retina are resistant to degenerative processes induced by ischemia-reperfusion.

The fate of calbindin D28k (calbindin)-labeled horizontal cells in the ischemic rat retina induced by increasing intraocular pressure was investigated by immunocytochemistry using an antiserum against calbindin. In the normal retina, strong calbindin-like immunoreactivity was seen in the horizontal cells, and the density of the labelled horizontal cells was 815.3+/-15.2 per mm2. The thickness of the treated retina became progressively thinner with increasing reperfusion time after the ischemic insult due to a loss of retinal cells in the inner and outer nuclear layers. However, the densities o

Joro spider venom: glutamate agonist and antagonist on the rod retina of the dogfish.

The venom of the Joro spider, reported to block glutamate action selectively at some glutaminergic synapses, was applied focally to rod horizontal and on-bipolar cells in dark-adapted retinal slices. The venom hyperpolarized horizontal cells in the dark and blocked their responses to light. Depolarization of horizontal cells by ionophoretic pulses of L-glutamate, L-aspartate or kainate was antagonized by the toxin in a slowly reversible manner. These results are consistent with other evidence for glutamate as the rod neurotransmitter acting on a single postsynaptic receptor type on horizontal

Isolated horizontal cells from carp retina demonstrate dopamine-dependent accumulation of cyclic AMP.

Horizontal cells of the carp retina were separated from other retinal cell types by using enzymatic dissociation and velocity sedimentation at unit gravity. Fractions containing horizontal cells were tested for their ability to accumulate cyclic AMP in the presence of various putative neurotransmitters. Micromolar concentrations of dopamine, when added in the presence of 3-isobutyl-1-methylxanthine, stimulated cyclic AMP accumulation in these isolated cells. The dopamine-dependent accumulation of cyclic AMP in intact isolated horizontal cells was blocked by nanomolar concentrations of dopamine

Investigation of tissue cysts in the retina in a mouse model of ocular toxoplasmosis: distribution and interaction with glial cells.

The conversion of tachyzoites into bradyzoites is a way for Toxoplasma gondii to establish a chronic and asymptomatic infection and achieve lifelong persistence in the host. The bradyzoites form tissue cysts in the retina, but not much is known about the horizontal distribution of the cysts or their interactions with glial cells in the retina. A chronic ocular toxoplasmosis model was induced by per oral administration of T. gondii Me49 strain cysts to BALB/c mice. Two months after the infection, retinas were flat-mounted and immunostained to detect cysts, ganglion cells, Müller cells, astrocyt

Selective retinal neuron loss and impaired neurovascular support underlie myopic retinopathy in RPE-specific Lrp2-deficient mice.

Pathologic myopia is a major cause of irreversible visual impairment worldwide and is characterized by excessive axial elongation accompanied by progressive retinal degeneration. Whether vision loss results primarily from passive retinal stretching or selective neurodegeneration remains unclear, hindering the development of effective neuroprotective and regenerative therapies. Here, we investigated retinal neuronal, vascular, and glial alterations in retinal pigment epithelium (RPE)-specific Lrp2 knockout (Best1-Cre/Lrp2fl/fl conditional knockout, CKO) model of pathologic myopia. The CKO mice

Horizontal cell connectivity in the anchovy retina-a 3D electron microscopic study.

BACKGROUND: Block-face scanning electron microscopy has opened a new era of connectomics research, in which it is possible to make dense reconstructions of all cells in a clipping of a neuronal network, such as the retina, resolving synaptic contacts. Anchovies, exceptionally abundant marine teleosts, have retinae with regions for triple cone-based color vision and a region with specialized cone photoreceptors, so-called polycones, made of long and short cones with axially oriented outer segment lamellae for polarization contrast vision. This modality, discovered in the 1970s, is unique in ver

Early B-cell factors are required for specifying multiple retinal cell types and subtypes from postmitotic precursors.

The establishment of functional retinal circuits in the mammalian retina depends critically on the proper generation and assembly of six classes of neurons, five of which consist of two or more subtypes that differ in morphologies, physiological properties, and/or sublaminar positions. How these diverse neuronal types and subtypes arise during retinogenesis still remains largely to be defined at the molecular level. Here we show that all four family members of the early B-cell factor (Ebf) helix-loop-helix transcription factors are similarly expressed during mouse retinogenesis in several neur

Analysis of vertebrate eye movements following intravitreal drug injections. III. Spontaneous nystagmus is modulated by the GABAa receptor.

1. Turtle eye movements were recorded in response to horizontal motion of patterned stimuli and intravitreal injections of selective GABAergic drugs by using a contact lens search-coil technique. Similar to results from rabbit and cat, injection of picrotoxin into the turtle's eye results in a spontaneous horizontal nystagmus, with its slow-phase movement in a temporal-to-nasal direction with respect to the injected eye. In contrast, there were no prominent vertical eye movements in response to either horizontal optokinetic stimuli or drug injections. 2. Injections of bicuculline or bicucullin

Immuno-histological detection of resistant columnar units and vulnerable networks in the rat retina after asphyxia-induced transient cardiac arrest.

BACKGROUND: Stroke-related loss of vision is one of the residual impairments, restricting the quality of life. However, studies of the ocular manifestations of asphyxia cardiac arrest/resuscitation (ACA/R) have reported very heterogeneous results. OBJECTIVE: We aimed to evaluate the ACA/R-induced degeneration pattern of the different retinal cell populations in rats using different immuno-histological stainings. METHODS: The staining pattern of toluidine blue and the ganglion cell markers β-III-tubulin and NeuN; the calcium-binding protein parvalbumin, indicating ganglion, amacrine, and hori

Rod photoreceptors control the ON vs OFF polarity of cone-signaling neurons.

A fundamental feature of the visual system is its ability to detect image contrast. The contrast processing starts in the first synapse of the retina where parallel pathways are established to compute contrast to bright (ON pathway) and dark (OFF pathway) objects, separately transferred to morphologically identified ON and OFF cells throughout the visual system. Here, we found that response polarity in ON and OFF neurons is not fixed but rather switches dynamically to the opposite polarity. The switch was not observed in rod-knockout mice, indicating that rods generate the polarity switch. We

The neuronal chaperone proSAAS is highly expressed in the retina.

The many layers of the neuroretina contain a complex, interconnected network of specialized neurons that both process visual stimuli and conduct processed information to higher brain areas. Neural networks rely on proteostatic control mechanisms to maintain proper protein homeostasis both in cell bodies as well as within synapses; protein chaperones play an important role in regulating and supporting this process. ProSAAS is a small neuronal chaperone that functions as an anti-aggregant in in vitro assays and is released upon depolarization in neuronal primary cultures. We here report a potent
