Melatonin protects inner retinal neurons of newborn mice after hypoxia-ischemia.

Retinopathy of prematurity is a vision-threatening disease associated with retinal hypoxia-ischemia, leading to the death of retinal neurons and chronic neuronal degeneration. During this study, we used the oxygen-induced retinopathy mice model to mimic retinal hypoxia-ischemia phenotypes to investigate further the neuroprotective effect of melatonin on neonatal retinal neurons. Melatonin helped maintain relatively normal inner retinal architecture and thickness and preserve inner retinal neuron populations in avascular areas by rescuing retinal ganglion and bipolar cells, and horizontal and a

mTORC1 and mTORC2 expression in inner retinal neurons and glial cells.

The retina is one of the most metabolically active tissues, yet the processes that control retinal metabolism remains poorly understood. The mTOR complex (mTORC) that drives protein and lipid biogenesis and autophagy has been studied extensively in regards to retinal development and responses to optic nerve injury but the processes that regulate homeostasis in the adult retina have not been determined. We previously demonstrated that normal adult retina has high rates of protein synthesis compared to skeletal muscle, associated with high levels of mechanistic target of rapamycin (mTOR), a kina

Brimonidine prevents axonal and somatic degeneration of retinal ganglion cell neurons.

BACKGROUND: Brimonidine is a common drug for lowering ocular pressure and may directly protect retinal ganglion cells in glaucoma. The disease involves early loss of retinal ganglion cell transport to brain targets followed by axonal and somatic degeneration. We examined whether brimonidine preserves ganglion cell axonal transport and abates degeneration in rats with elevated ocular pressure induced by laser cauterization of the episcleral veins. RESULTS: Ocular pressure was elevated unilaterally by 90% for a period of 8 weeks post- cauterization. During this time, brimonidine (1mg/kg/day) or

Foxn3 is required to suppress aberrant ciliogenesis in nonphotoreceptor retinal neurons.

The retinal photoreceptors possess specialized sensory cilia critical for phototransduction while the nonphotoreceptor cells typically exhibit simpler primary cilia or lack them altogether. This dichotomy in ciliary architecture underpins the functional specialization of retinal cell types, but how this dichotomy arises and is maintained remains elusive. This study explores the role of the transcription factor Foxn3 in establishing and maintaining this divergence. We generated retina-specific Foxn3 conditional knockout (Foxn3CKO) mice, which show that Foxn3 is essential for repressing ciliary

Early Diabetes Induces Changes in Mitochondrial Physiology of Inner Retinal Neurons.

Diabetic retinopathy, a leading cause of vision loss, was considered as a solely vascular disorder but some recent studies suggest that retinal neurons may be affected much before the appearance of vascular lesions. However, the cellular processes involved in diabetes-induced degeneration of retinal neurons are poorly understood. Calcium (Ca2+) signaling plays a key role in normal functioning of neurons, and its dysregulation may lead to degeneration of neurons. Mitochondria are crucial components involved in the regulation of intracellular Ca2+ signaling. In this study, we have investigated t

Repetitive light stimulation inducing glycine receptor plasticity in the retinal neurons.

Neurotransmitter receptor plasticity is a mechanism that can regulate the temporal and intensity encoding of a synapse. While this has been extensively studied as a mechanism of learning, less is known about such processes in sensory systems. This study examines modulation of glycine receptor function at the first synapse in the retina. It was found that horizontal cells, which are postsynaptic to photoreceptors, have glycine receptor currents that are enhanced when internal calcium is elevated. This can be achieved by glutamatergic synaptic input or by activation of voltage-gated calcium chan

Retinal Neuronal Changes and Kidney Dysfunction in Diabetes Mellitus.

BACKGROUND: Diabetic kidney disease (DKD) and diabetic retinopathy (DR) share common microvascular pathophysiology, yet the relationship between retinal neuronal alterations and kidney dysfunction remains unclear. This cross-sectional study examines the association between individual retinal layer thickness and kidney indicators to identify novel optical coherence tomography (OCT) biomarkers for DKD. METHODS: A total of 410 type 2 diabetes (T2D) patients (815 eyes) were recruited from the outpatient clinics of a tertiary eye centre in Singapore. Retinal layer thickness was measured using OCT

Retinal clues for selective neuronal loss in multiple sclerosis.

OBJECTIVE: The relationship between the cell body layer and the dendritic network layer of the retina and cognitive performance (CP) in MS patients has not been examined separately. The objective of this study is to predict cognitive impairment (CI) in RRMS patients and to examine the relationship between CP and ganglion cell layer (GCL), inner plexiform layer (IPL), and GCL divided by IPL (GCL/IPL). METHODS: Ophthalmological evaluation, retinal segmentation, and Symbol Digit Modalities Test (SDMT) were performed on 102 RRMS patients and 54 healthy subjects. The relationships of GCL, IPL, and

Neuronal Bmal1 regulates retinal angiogenesis and neovascularization in mice.

Circadian clocks in the mammalian retina regulate a diverse range of retinal functions that allow the retina to adapt to the light-dark cycle. Emerging evidence suggests a link between the circadian clock and retinopathies though the causality has not been established. Here we report that clock genes are expressed in the mouse embryonic retina, and the embryonic retina requires light cues to maintain robust circadian expression of the core clock gene, Bmal1. Deletion of Bmal1 and Per2 from the retinal neurons results in retinal angiogenic defects similar to when animals are maintained under co

Ontogenesis of the pinealo-retinal neuronal connection in albino rats.

It was accepted for a long time that in mammals there is only retinofugal neuronal connection between the eye and the pineal body (PB). In our previous paper we described that nerve cells were present in hamster PB and these neurons could establish a reverse connection with the retina through a transsynaptic pathway. In adult albino rats neuronal perikarya were not found. In this present experiment it was examined whether the lack of these nerve cells in the PB of adult rats is the result of an apoptotic phenomenon or the lack of migration during the fetal period. Green fluorescence protein ex

Stem cells: a source for neuron repair in retinal disease.

Several features of the eye make it an excellent candidate for cell-based therapy to treat corneal and retinal degenerative disease. Corneal and retinal stem cells have been identified in the adult human eye. Advances in the stem cell field, including methods for cell purification, culture, and molecular characterization, have made it easier to manipulate eye stem cells. Moreover, delivery, sequestration, tracking, and measurement of outcomes of transplanted cells are both feasible and easier for the eye than for other tissues. These factors, together with our understanding of the molecular pa

Retinal ultrastructure of neuronal ceroid-lipofuscinosis in the dalmatian dog.

Ultrastructural studies of the retinae in two NCL-affected Dalmatian dogs revealed ubiquitous accumulation of lipopigments in numerous cell types of the retina, the fine structure of which closely resembled that seen in NCL-affected English setters. Photoreceptors and other retinal cell types were largely intact. These findings show that the retinal involvement in NCL of our Dalmatian dogs is identical to that of NCL-affected English setters. It also shows that in canine NCL a severe retinopathy, regularly encountered in human childhood NCL, does not develop. Thus, the NCL of Dalmatian dogs--a

Neuronal ceroid lipofuscinosis. A retinal trypsin digest study.

Neuronal ceroid lipofuscinosis, also known as the Batten-Vogt syndrome, is clinically manifested by seizures, loss of intellect, and deterioration of vision. The retinal histopathologic and ultrastructural changes have been well-described. This report adds the findings of trypsin digest preparation of the retinae in three such patients with varying duration of this disease. Acellular retinal capillaries, or ghost vessels, were observed, and the degree of this change correlated with the duration of clinical symptoms. These changes, as well as the histopathologic abnormalities, are discussed.

Microtubule dynamics in adult retinal ganglion cells and dorsal root ganglion neurons.

INTRODUCTION: While axon regeneration is very limited in the adult central nervous system (CNS) in vivo, this is not the case in the peripheral nervous system (PNS). Indeed, both CNS and PNS neurons can regenerate in vitro although to varying degrees. Given the role of microtubule stabilization in promoting regeneration, we have examined microtubule polymerization during the regeneration of two types of adult neurons in vitro, retinal ganglion cells (RGCs) from the CNS and dorsal root ganglion (DRG) neurons from the PNS. METHODS: In order to compare microtubule dynamics between these cell typ

Distribution of GABA(C)-like responses among acutely dissociated rat retinal neurons.

GABAergic responses of acutely dissociated rat retinal neurons, including both bipolar cells (BCs) and other, morphologically round cells (RCs), were assayed with the fluorescent (FL), voltage-sensitive probe oxonol DiBaC4(5). Using intensified video microscopy and simultaneous recording, GABA responses were identified in one-third of cells in a typical microscope field; of these 85% hyperpolarized (0.05-0.3 log unit FL decreases) while the remainder depolarized (0.05-0.2 log unit FL increases). GABA-sensitive cells were also TACA-sensitive (trans-4-Aminocrotonic acid), and these ligands appea

Postsynaptic neuronal activity promotes regeneration of retinal axons.

The wiring of visual circuits requires that retinal neurons functionally connect to specific brain targets, a process that involves activity-dependent signaling between retinal axons and their postsynaptic targets. Vision loss in various ophthalmological and neurological diseases is caused by damage to the connections from the eye to the brain. How postsynaptic brain targets influence retinal ganglion cell (RGC) axon regeneration and functional reconnection with the brain targets remains poorly understood. Here, we established a paradigm in which the enhancement of neural activity in the dista

Adult type of neuronal ceroid-lipofuscinosis with retinal involvement.

A rare case of Kufs' disease with retinal lesions is reported in which the first symptom of visual disturbances later resulted in total blindness. Various neurologic symptoms and mental deterioration also developed. The total duration of the illness was 32 years. Neuropathologic examination showed extensive ballooned nerve cells filled with lipopigments in the CNS. In the retina there was a thinning with severe loss of rods, cones, and outer nuclear and outer plexiform layers. The remaining ganglion cells of the retina were also ballooned and accumulated with lipopigments. Histochemical and el

Interpreting the retinal neural code for natural scenes: From computations to neurons.

Understanding the circuit mechanisms of the visual code for natural scenes is a central goal of sensory neuroscience. We show that a three-layer network model predicts retinal natural scene responses with an accuracy nearing experimental limits. The model's internal structure is interpretable, as interneurons recorded separately and not modeled directly are highly correlated with model interneurons. Models fitted only to natural scenes reproduce a diverse set of phenomena related to motion encoding, adaptation, and predictive coding, establishing their ethological relevance to natural visual c
