Triatomines are vectors of American Trypanosomiasis also known as Chagas´ disease where several reservoirs including dogs are involved in the transmission cycle of the causal agent (Trypanosoma cruzi). Considering that the prevalence of American trypanosomiasis in dogs is higher than in humans and that dogs in addition are susceptible of this disease, and are involved in peridomestic transmission to humans, the search for new alternatives for vector control of the triatomines responsible for transmission in dogs is required. Over the 20 weeks the study lasted, 600 individual female, adult of R

Incorporation of 3H-thymidine by splenic lymphocytes was studied in CBA mice that were bitten by the triatomine bug Rhodnius prolixus (Stål) 14, 7, 4, and 2 d before lymphocyte isolation, respectively. In bitten mice, both spontaneous and mitogen-induced proliferative responses were reduced. The most pronounced effect was observed 4 d after exposure when lymphocytes were significantly suppressed in proliferative response to the mitogens concanavalin A, phytohemagglutinin, and bacterial lipopolysaccharide. The maximum inhibition caused an 85% reduction of proliferation and was observed in phyto

Bloodsucking bugs use infrared radiation (IR) for locating warm-blooded hosts and are able to differentiate between infrared and temperature (T) stimuli. This paper is concerned with the neuronal coding of IR in the bug Rhodnius prolixus. Data obtained are from the warm cells in the peg-in-pit sensilla (PSw cells) and in the tapered hairs (THw cells). Both warm cells responded to oscillating changes in air T and IR with oscillations in their discharge rates. The PSw cells produced stronger responses to T oscillations than the THw cells. Oscillations in IR did the reverse: they stimulated the l

Chagas disease remains an important public health concern across the Americas, affecting both human and animal populations. The triatomine bug Rhodnius prolixus, an obligate hematophagous vector of Trypanosoma cruzi, is commonly associated with environments where domestic and working animals are housed. Military working dogs (MWDs) deployed or stationed in endemic or expansion zones are particularly vulnerable to infection, which can lead to severe clinical disease and reduced operational readiness. This study evaluated the residual knockdown efficacy of esfenvalerate- and dinotefuran-treated

Triatomines (Hemiptera: Reduviidae) are hematophagous insects that transmit Trypanosoma cruzi, the etiological agent for Chagas disease, to humans and other mammals. As medically important vectors, species such as Rhodnius prolixus (Hemiptera: Reduviidae) have long been used as a model organism for physiological studies. Laboratory rearing of triatomines is needed to support vector and parasite research. Many environmental conditions, such as suitable housing containers, light source and duration, temperature, humidity, and density, must be addressed when adapting triatomines from a natural ha

BACKGROUND: Rhodnius prolixus is a vector of Chagas disease and has become a model organism to study physiology, behavior, and pathogen interaction. The publication of its genome allowed initiating a process of comparative characterization of the gene expression profiles of diverse organs exposed to varying conditions. Brain processes control the expression of behavior and, as such, mediate immediate adjustment to a changing environment, allowing organisms to maximize their chances to survive and reproduce. The expression of fundamental behavioral processes like feeding requires fine control i

Rhodnius prolixus is an insect vector of two flagellate parasites, Trypanosoma rangeli and Trypanosoma cruzi, the latter being the causative agent of Chagas disease in Latin America. The R. prolixus neuroendocrine system regulates the synthesis of the steroid hormone ecdysone, which is essential for not only development and molting but also insect immunity. Knowledge for how this modulates R. prolixus midgut immune responses is essential for understanding interactions between the vector, its parasites and symbiotic microbes. In the present work, we evaluated the effects of ecdysone inhibition

BACKGROUND: Triatomine bugs are vectors of the protozoan parasite Trypanosoma cruzi, which causes Chagas disease. Rhodnius pallescens is a major vector of Chagas disease in Panama. Understanding the microbial ecology of disease vectors is important in the development of vector management strategies that target vector survival and fitness. In this study we examined the whole-body microbial composition of R. pallescens from three locations in Panama.
METHODS: We collected 89 R. pallescens specimens using Noireau traps in Attalea butyracea palms. We then extracted total DNA from whole-bodies of

Within invertebrates, the kinin family of neuropeptides is responsible for the modulation of a host of physiological and behavioural processes. In Rhodnius prolixus, kinins are primarily responsible for eliciting myotropic effects on various feeding and diuresis-related tissues. Here, the R. prolixus kinin receptor (RhoprKR) has been identified, cloned and sequenced from the central nervous system (CNS) and hindgut of R. prolixus. Sequence analyses show high similarity and identity between RhoprKR and other cloned invertebrate kinin receptors. The expression profile of RhoprKR shows the RhoprK

Jack Bean Urease (JBU) is a urease isoform from the plant Canavalia ensiformis with recognized insecticidal properties, although its full entomotoxic potential remains poorly understood. In this study, we used Rhodnius prolixus, a vector of Chagas disease, as a model to investigate the effects of JBU on reproduction. Injection of a sublethal dose in vitellogenic females significantly reduced oviposition and delayed ovarian development, with ovarioles displaying degenerating follicles. Microscopy analyses revealed signs of apoptosis, autophagy, and necrosis in the ovarian tissue of JBU-treated

Olfactory proteins mediate a wide range of essential behaviors for insect survival. Odorant binding proteins (OBPs) are small soluble olfactory proteins involved in the transport of odor molecules (=odorants) through the sensillum lymph to odorant receptors, which are housed on the dendritic membrane of olfactory sensory neurons also known as olfactory receptor neurons. Thus, a better understanding of the role(s) of OBPs from Rhodnius prolixus, one of the main vectors of Chagas disease, may ultimately lead to new strategies for vector management. Here we aimed at functionally characterize OBPs

BACKGROUND: Rhodnius prolixus is a blood-sucking bug vector of Trypanosoma cruzi and T. rangeli. T. cruzi is transmitted by vector feces deposited close to the wound produced by insect mouthparts, whereas T. rangeli invades salivary glands and is inoculated into the host skin. Bug saliva contains a set of nitric oxide-binding proteins, called nitrophorins, which deliver NO to host vessels and ensure vasodilation and blood feeding. NO is generated by nitric oxide synthases (NOS) present in the epithelium of bug salivary glands. Thus, T. rangeli is in close contact with NO while in the salivary

Trypanosoma cruzi is a single-celled eukaryotic parasite responsible for Chagas disease, a major cause of morbidity and mortality in Central and South America. While the host-pathogen interactions of T. cruzi have been extensively studied in vertebrate models, investigations into its interactions within its insect host remain limited. To address this gap and establish a genetically tractable system for studying parasite-vector dynamics, we conducted quantitative kinetic infection studies using the Y strain of T. cruzi and the model vector Rhodnius prolixus. We began by comparing parasite infec

The phenylalanine/tyrosine degradation pathway is frequently described as a catabolic pathway that funnels aromatic amino acids into citric acid cycle intermediates. Previously, we demonstrated that the accumulation of tyrosine generated during the hydrolysis of blood meal proteins in Rhodnius prolixus is potentially toxic, a harmful outcome that is prevented by the action of the first two enzymes in the tyrosine degradation pathway. In this work, we further evaluated the relevance of all other enzymes involved in phenylalanine/tyrosine metabolism in the physiology of this insect. The knockdow

Genetic relationships among 10 species of bugs belonging to the tribe Rhodniini (Hemiptera: Reduviidae), including some important vectors of Chagas disease, were inferred from allozyme analysis of 12 enzyme loci (out of 21 enzyme systems examined), using agarose gel electrophoresis. These species formed two clusters: one comprising Rhodnius brethesi, R. ecuadoriensis, R. pallescens and R. pictipes; the other with Psammolestes tertius, Rhodnius domesticus and the Rhodnius prolixus group comprising R. nasutus, R. neglectus, R. prolixus and R. robustus. The resulting tree was [((R. ecuadoriensis,

Rhodnius prolixus is a hematophagous insect and the primary vector of Trypanosoma cruzi, the causative agent of Chagas disease. Blood gorging represents an epidemiologically critical event because it is a requirement for development throughout the life cycle and for successful vector reproduction, contributing to population spread. Blood gorging also triggers diuresis, a physiological process that can expel parasites from the hindgut onto the host's skin, thereby facilitating transmission. Serotonin, a biogenic amine, acts through five G-protein-coupled receptors (5-HT1aR, 5-HT1bR, 5-HT2aR, 5-

Chagas disease control relies on chemical interventions, currently compromised by insecticide resistance. Triatoma virus (TrV) emerges as a biological candidate, requiring standardized methodologies to purify, quantify, and relate viral doses to biological effects in triatomines. We established a comprehensive protocol for the quantitative study of TrV using Rhodnius prolixus as a model, combining physical purification via sucrose gradients and absolute quantification by qPCR targeting the TrVgp1 gene to link viral load with host survival. Primers were validated, and a plasmid standard curve w

Oocyte polarity establishment is a conserved and crucial phenomenon for embryonic development. It relies on the precise spatial localization of maternal factors deposited during oocyte development, which is essential for establishing and maintaining cell polarity and subsequently specifying embryonic axes. The heterogeneous nuclear ribonucleoprotein (hnRNP) encoded by the squid (sqd) gene has been implicated in mRNA localization and embryonic axis establishment in Drosophila melanogaster. Comparative genomics allowed for the identification of a homologue in Rhodnius prolixus. In this study, we

In insects, lipids are stored in the fat body mainly as triacylglycerol. Lipids can be directly provided by digestion and incorporated from the hemolymph, or synthesized de novo from other substrates such as carbohydrates and amino acids. The first step in de novo lipid synthesis is catalyzed by acetyl-CoA carboxylase (ACC), which carboxylates acetyl-CoA to form malonyl-CoA. Rhodnius prolixus is a hematophagous insect vector of Chagas disease and feeds exclusively on large and infrequent blood meals. Adult females slowly digest the blood and concomitantly accumulate lipids in the fat body. In

Host-seeking behaviour displayed by the haematophagous triatomine Rhodnius prolixus Stål, 1859 is influenced by different cues involving heat, moisture and odours. Concentrations of volatile odours present in human sweat and breath that activate, attract or non-attract triatomines were tested behaviourally with a double choice olfactometer in adults of R. prolixus. Attraction of a mixture of substances carried by a controlled air-stream was established and compared with volatiles from human extracts obtained by rubbing face, hands or feet. Volatiles from face and feet extracts showed a higher
