Coordinated regulation of trimethylamine catabolism in abundant marine bacteria.

Methylated amines (MAs) are ubiquitous in the marine environment, constituting an important part of the marine dissolved organic nitrogen pool and contributing to the formation of cloud condensation nuclei in the marine atmosphere. The aerobic trimethylamine (TMA) oxidation pathway represents the primary route for TMA utilization in marine environments and has been characterized in the model marine Roseobacter group bacterium Ruegeria pomeroyi DSS-3. However, the regulatory mechanism underlying this pathway has remained unclear. Transcriptome analysis revealed coordinated induction of the enti

Phage-bacteria network analysis and its implication for the understanding of coral disease.

Multiple studies have explored microbial shifts in diseased or stressed corals; however, little is known about bacteriophage interactions with microbes in this context. This study characterized microbial 16S rRNA amplicons and phage metagenomes associated with Montastraea annularis corals during a concurrent white plague disease outbreak and bleaching event. Phage consortia differed between bleached and diseased tissues. Phages in the family Inoviridae were elevated in diseased or healthy tissues compared with bleached portions of diseased tissues. Microbial communities also differed between d

Oxidation state of bioavailable dissolved organic matter influences bacterioplankton respiration and growth efficiency.

Oxygen consumption by oceanic microbes can predict respiration (CO2 production) but requires an assumed respiratory quotient (RQ; ΔO2/ΔCO2). Measured apparent RQs (ARQs) can be impacted by various processes, including nitrification and changes in dissolved organic matter (DOM) composition, leading to discrepancies between ARQ and actual RQ. In DOM remineralization experiments conducted in the eastern North Atlantic Ocean, ARQs averaged 1.39 ± 0.14, similar to predictions for complete consumption of plankton biomass. DOM removed with an elevated nominal oxidation state (i.e., more oxidized DOM)

Simultaneous removal of chlorobenzene and Cr(VI) from groundwater using microbial fuel cell with low-cost Si modified ferrihydrite electrodes.

Aromatic chlorinated compounds and Cr(VI) in groundwater pose significant challenges due to their resistance. This study explores microbial fuel cells using low-cost Si-modified ferrihydrite (SiFh) electrodes for simultaneous chlorobenzene and Cr(VI) removal. The voltage output of MFC assembled with SiFh modified electrode was approximately 1.63 times higher than the bare electrode, achieving 1.23 times higher in chlorobenzene degradation efficiency. CF-SiFh loaded MFC had the highest power generation and best EET efficiency, which was positive to greatest and fastest chlorobenzene removal. Mi

Methionine synthesis and glycine betaine demethylation are intricately intertwined in cosmopolitan marine bacteria.

Across all domains of life, cobalamin-dependent methyltransferases have diversified to perform a range of crucial functions, such as methionine synthesis and the demethylation of various reduced nitrogen and sulfur compounds. These large modular enzymes typically possess three substrate-binding domains, two binding either the methyl donor or methyl acceptor, as well as a cobalamin-binding domain. Here, by challenging the current paradigm of glycine betaine (GBT) catabolism, we have identified a unique methyltransferase in aerobic environmental bacteria that has a dual function both as a methio

Syngas biological transformation into hydroxyectoine.

Syngas from the gasification of organic wastes represents a promising feedstock for fostering a sustainable bioeconomy. However, its potential is currently constrained by the low-value products generated. Osmolytes, such as hydroxyectoine, are high-value compounds, however, their biological production as isolated osmolytes is not yet cost-effective. This study utilized shotgun genomics and laboratory validation to find a carboxydotrophic, halotolerant bacterium, Hydrogenibacillus schlegelii, that could produce hydroxyectoine using H2, CO and CO2 as the sole source of energy and carbon. Algal-Bacterial Interaction-Driven Secondary Metabolites From Phaeobacter inhibens and Their Anti-Allergic Effects on Th2 Cell Immune Response.

The Roseobacter clade, a versatile Rhodobacteraceae lineage, comprises up to 20% of marine bacteria and drives key biogeochemical cycles. Phaeobacter inhibens, a representative model species, is associated with the alga Emiliania huxleyi, exhibiting a dual lifestyle that alternates between promoting symbiotic growth and displaying pathogenicity during algal senescence. In this study, we investigated the metabolic responses of P. inhibens cultured with sinapic acid, an algal-derived lignin catabolite known to modulate algal-bacterial interactions. Detailed LC-MS/UV-guided analysis of the sinapi

Re-examining the iron paradox: the influence of growth medium on regulation and functionality of the tropodithietic acid (TDA) biosynthetic pathway in Phaeobacter piscinae.

Tropodithietic acid (TDA) is a secondary metabolite with antimicrobial and iron-binding properties, produced by several marine Roseobacter group bacteria. Although classified as a siderophore, TDA is also produced under iron-replete conditions. To address this paradox, we fused the tdaCDE promoter region of Phaeobacter piscinae S26 with a promoter-less gfp reporter gene to monitor the transcription of TDA biosynthetic genes under different conditions and validated its accuracy using RT-qPCR. In both complex ($\frac{1}{2}$YTIO) and low-complexity (IOCGH) marine media, iron supplementation repre

Bisphenol F induces liver-gut alteration in zebrafish.

The unease of consumers with bisphenol A has led to the increased industrial usage of bisphenol F (BPF), which is a new hazard to environmental health. Here, zebrafish were exposed to three BPF concentrations (0.5, 5, and 50 μg/L) from the embryonic stage for 180 days. Results showed that zebrafish body length and weight decreased and hepatosomatic index values increased, even at environmentally relevant concentration. Histological analysis identified the occurrence of hepatic fibrosis and steatosis in 5 and 50 μg/L groups, which indicated the liver injury caused by BPF. Based on the untargete

Enhancement of Cr(VI) reduction by indigenous bacterial consortia using natural pyrite: A detailed study to elucidate the mechanisms involved in the highly efficient and possible sustainable system.

Pyrite was applied to Cr(VI) bioremediation as an inorganic electron donor due to the ability to provide electrons, while the role of pyrite in Cr(VI) bioremediation where organics as electron donors remains unknown. Herein a pyrite-based Cr(VI) bioreduction process in the sediment system containing lactate was demonstrated to be effective to detoxify Cr(VI): over 2200 mg L-1 Cr(VI) was continuously removed within 210 h with high reactivity (10.5 mg/(L·h)) all along. High-throughput 16S rDNA gene sequencing indicated that the pyrite could shape a functioning community that electrochemically ac

Intratumoral microbial networks as biomarkers for second primary oral cancer risk in esophageal squamous cell carcinoma.

Esophageal squamous cell carcinoma (ESCC) survivors remain at elevated risk of developing second primary oral cancer (SPOC), yet the role of intratumoral microbiomes in SPOC emergence is not fully understood. We performed 16 S rRNA V3-V4 sequencing on tumor brushings from 28 ESCC patients (20 SPOC-negative, 8 SPOC-positive) to profile microbial diversity, taxonomic composition, functional potential, and interaction networks. Alpha diversity metrics (Chao1, Shannon) did not differ significantly between groups (p > 0.05), whereas sparse partial least squares-discriminant analysis of beta diversi

Bacterial periphytic communities related to mercury methylation within aquatic plant roots from a temperate freshwater lake (South-Western France).

Macrophyte floating roots are considered as hotspots for methylmercury (MeHg) production in aquatic ecosystems through microbial activity. Nevertheless, very little is known about periphyton bacterial communities and mercury (Hg) methylators in such ecological niches. The ability to methylate inorganic Hg is broadly distributed among prokaryotes; however, sulfate-reducers have been reported to be the most important MeHg producers in macrophyte floating roots. In the present work, the periphyton bacterial communities colonizing Ludwigia sp. floating roots were investigated through molecular met

A case of Stappia indica-induced relapsing peritonitis confirmed by 16S ribosomal RNA gene sequencing analysis in a patient undergoing continuous ambulatory peritoneal dialysis.

A 69-year-old woman with 26-year history of systemic lupus erythematosus and 4-year history of peritoneal dialysis was hospitalized for treatment of bacterial peritonitis. On admission, peritoneal dialysate was collected and subjected to bacterial culture. Cell count in the cloudy peritoneal dialysate was 4194/μL, and Gram-negative bacilli were detected. Vancomycin (1 g/day) and ceftazidime (1 g/day) were administered intraperitoneally, which resulted in rapid decrease in cell count in the peritoneal dialysate. However, on the 7th hospital day, peritonitis relapsed with abdominal pain and clou

Description of Sulfitobacter gelatinilyticus sp. nov. and Sulfitobacter weihaiensis sp. nov., isolated from marine sediment.

Two Gram-stain-negative, yellowish-white, facultative anaerobic and rod-shaped bacterial strains, designated F26169LT and F26204T, were isolated from coastal sediment of Jingzi Port, Weihai. Based on phenotypic, physiological, biochemical, chemotaxonomic and phylogenomic analyses, the two strains were affiliated with the genus Sulfitobacter. They showed obvious differences in phenotypic, chemotaxonomic and genomic characteristics compared with closely related taxa in this genus, and the ANI, AAI and dDDH values between them and related species were all lower than the standard thresholds for ba

A denitrifying community associated with a major, marine nitrogen fixer.

The diazotrophic cyanobacterium, Trichodesmium, is an integral component of the marine nitrogen cycle and contributes significant amounts of new nitrogen to oligotrophic, tropical/subtropical ocean surface waters. Trichodesmium forms macroscopic, fusiform (tufts), spherical (puffs) and raft-like colonies that provide a pseudobenthic habitat for a host of other organisms including marine invertebrates, microeukaryotes and numerous other microbes. The diversity and activity of denitrifying bacteria found in association with the colonies was interrogated using a series of molecular-based methodol

Bacterial communities associated with Porites white patch syndrome (PWPS) on three western Indian Ocean (WIO) coral reefs.

The scleractinian coral Porites lutea, an important reef-building coral on western Indian Ocean reefs (WIO), is affected by a newly-reported white syndrome (WS) the Porites white patch syndrome (PWPS). Histopathology and culture-independent molecular techniques were used to characterise the microbial communities associated with this emerging disease. Microscopy showed extensive tissue fragmentation generally associated with ovoid basophilic bodies resembling bacterial aggregates. Results of 16S rRNA sequence analysis revealed a high variability between bacterial communities associated with PWP

Prokaryotic Novelty and Metabolic Diversity in the Tagoro Shallow Hydrothermal System (Canary Islands, Spain).

The shallow hydrothermal system of the Tagoro submarine volcano (Canary Islands, Spain) has been discharging low-temperature, nutrient-rich, acidic fluids high in CO2 and H2S since March 2012. Despite influencing local microbial communities, the effects of these hydrothermal fluids on community structure and diversity across spatial gradients remain poorly understood. Using 16S rRNA gene sequencing, we observed significant differences in prokaryotic community structure from epipelagic waters to benthic habitats affected by hydrothermal activity. Prokaryotic richness and diversity decreased clo

Microbial metacommunity of salt marshes rebuilds along an elevational gradient after initial disturbance.

Salt marshes are ecologically important ecosystems with dynamic nutrient exchange between land and sea. Their zonation along an elevation gradient supports specific communities exhibiting successional patterns. Previous studies have mainly focused on individual domains, with limited attempts to explore interdependencies of community assemblies across domains. Here, we investigated the co-occurrence of prokaryotes and microeukaryotes in natural salt marsh sediments and experimental islands placed in the adjacent tidal flat. The islands contained originally bare and transplanted plots at three d

A New Microbial Pathway for Organophosphonate Degradation Catalyzed by Two Previously Misannotated Non-Heme-Iron Oxygenases.

The assignment of biochemical functions to hypothetical proteins is challenged by functional diversification within many protein structural superfamilies. This diversification, which is particularly common for metalloenzymes, renders functional annotations that are founded solely on sequence and domain similarities unreliable and often erroneous. Definitive biochemical characterization to delineate functional subgroups within these superfamilies will aid in improving bioinformatic approaches for functional annotation. We describe here the structural and functional characterization of two non-h
