Red, Gold and Green: Microbial Contribution of Rhodophyta and Other Algae to Green Turtle (Chelonia mydas) Gut Microbiome.

The fitness of the endangered green sea turtle (Chelonia mydas) may be strongly affected by its gut microbiome, as microbes play important roles in host nutrition and health. This study aimed at establishing environmental microbial baselines that can be used to assess turtle health under altered future conditions. We characterized the microbiome associated with the gastrointestinal tract of green turtles from Guinea Bissau in different life stages and associated with their food items, using 16S rRNA metabarcoding. We found that the most abundant (% relative abundance) bacterial phyla across th

Effects of marine heatwaves on haploid and diploid isomorphic phases from two distant populations of Gracilaria caudata (Gracilariales, Rhodophyta).

Marine heatwaves (MHWs) are becoming more common and intense, forcing the physiological limits of marine organisms and causing major ecological impacts throughout the world. This work used a well-studied species of Rhodophyta as a model to understand how different thermal ecotypes and isomorphic life history phases respond to MHWs. The thermal ecotypic populations of Gracilaria caudata in Brazil constitute an adaptive strategy for the species to remain in a wide temperature range. In this study, tetrasporophytes and unfertilized female gametophytes of G. caudata from the Tropical Southwestern

Population genetics of the freshwater red alga Batrachospermum gelatinosum (Rhodophyta) I: Frequent intragametophytic selfing in a monoicous, haploid-diploid species.

Life cycles with a prolonged haploid phase are thought to be correlated with greater rates of selfing and asexual reproduction. In red algae, recent population genetic studies have aimed to test this prediction but have mostly focused on marine species with separate sexes. We characterized the reproductive system of the obligately monoicous (i.e., hermaphroditic) freshwater red alga Batrachospermum gelatinosum and predicted that we would find genetic signatures of uniparental reproduction because of its haploid-diploid life cycle. We sampled 18 sites and genotyped 311 gametophytes with 10 poly

Antihypertensive Effects of Two Novel Angiotensin I-Converting Enzyme (ACE) Inhibitory Peptides from Gracilariopsis lemaneiformis (Rhodophyta) in Spontaneously Hypertensive Rats (SHRs).

A variety of biologically active products have been isolated from Gracilariopsis lemaneiformis. In the present study, two novel angiotensin-converting enzyme (ACE) inhibitory peptides, FQIN [M(O)] CILR, and TGAPCR, were screened and identified from G. lemaneiformis protein hydrolysates by LC-MS/MS. The IC50 values of FQIN [M(O)] CILR and TGAPCR were 9.64 ± 0.36 μM and 23.94 ± 0.82 μM, respectively. In the stability study, both peptides showed stabilities of pH, temperature, simulated gastrointestinal digestion, and ACE hydrolysis. The Lineweaver⁻Burk plot showed that the two peptides were nonc

Regulatory role of external calcium on Pythium porphyrae (Oomycota) zoospore release, development and infection in causing red rot disease of Porphyra yezoensis (Rhodophyta).

Formation of zoosporangia and cleavage of zoosporangial cytoplasm to zoospores in Pythium porphyrae is absolutely dependent on extracellular calcium. Calcium ion could be substituted neither by monovalent nor divalent cations tested. Increased concentrations of extracellular calcium did not affect the release of zoospores from zoosporangia but inhibited the zoospore motility. Chelating calcium ion by EGTA has decreased the ability of encysted zoospores to germinate and form appressoria. The increased external calcium-ion concentration has decreased the infectivity of Porphyra yezoensis thalli

Evaluation of food grade solvents for lipid extraction and impact of storage temperature on fatty acid composition of edible seaweeds Laminaria digitata (Phaeophyceae) and Palmaria palmata (Rhodophyta).

This study evaluated the impact of different food- and non-food grade extraction solvents on yield and fatty acid composition of the lipid extracts of two seaweed species (Palmaria palmata and Laminaria digitata). The application of chloroform/methanol and three different food grade solvents (ethanol, hexane, ethanol/hexane) revealed significant differences in both, extraction yield and fatty acid composition. The extraction efficiency, in terms of yields of total fatty acids (TFA), was in the order: chloroform/methanol>ethanol>hexane>ethanol/hexane for both species. Highest levels of polyunsa

Immunochemistry of biliproteins.

Biliproteins were extracted from representatives of the Cyanophyta, Rhodophyta, and Cryptophyta and purified. Both purified and crude biliproteins were used to stimulate rabbit antibody directed specifically against the biliproteins. The antigenic and immunogenic inter-relationships of these proteins were investigated by the Ouchterlony double diffusion technique. C-phycocyanins from all sources were found to be antigenically and immunogenically related and apparently also related to allophycocyanin but not to any of the phycoerythrins. Larger antigenic differences among phycoerythrins from di

Enhancing the dispersibility of Gelidium amansii-derived microfibrillated cellulose through centrifugal fractionation.

Hydrothermal pretreatment is useful for microfibrillated cellulose (MFC) preparation due to its safety, but the remaining hemicellulose might affect MFC properties. This study aimed to investigate the effect of centrifugation time on hemicellulose removal and the physicochemical properties of MFC obtained after hydrothermal pretreatment and micro-fibrillation. In this study, centrifugation was applied to the MFC suspension at varying duration times. Composition analysis and Fourier transform infrared spectra indicated that fractionated MFC has no hemicellulose content after 10, 20, and 30 min

Unique alcohol dehydrogenases involved in algal sugar utilization by marine bacteria.

Marine algae produce complex polysaccharides, which can be degraded by marine heterotrophic bacteria utilizing carbohydrate-active enzymes. The red algal polysaccharide porphyran contains the methoxy sugar 6-O-methyl-D-galactose (G6Me). In the degradation of porphyran, oxidative demethylation of this monosaccharide towards D-galactose and formaldehyde occurs, which is catalyzed by a cytochrome P450 monooxygenase and its redox partners. In direct proximity to the genes encoding for the key enzymes of this oxidative demethylation, genes encoding for zinc-dependent alcohol dehydrogenases (ADHs) w

Elevated carbon dioxide stimulates efficient organic carbon consumption for the unicellular alga Galdieria.

Unicellular algae are appealing for nutritional and biotechnological utility, but show wide variation across strains and can be challenging to produce. Species in the thermo-acidophilic genus Galdieria use diverse organic carbon sources for fermentative growth, which can include waste-stream feedstocks, while having complete amino acid compositions for human nutrition. Here, we investigated the metabolic dynamics of Galdieria to catalog organic carbon conversion to biomass. Tested strains had enhanced growth upon 3% CO2 supplementation, triggering efficient glucose uptake to reach ~5 ± 0.3 g d

The red alga Tsunamia transpacifica (Stylonematophyceae) from plastic drift shows adaptation to its uncommon habitat in ultrastructure and soluble low molecular weight carbohydrate composition.

The recently described red alga Tsunamia transpacifica (Stylonematophyceae) was previously isolated from plastic drift found at the pacific coast, but the natural habitat remains unknown. Here, we investigate ultrastructural details and the low molecular weight soluble carbohydrate composition to get further insight into the adaptation to this uncommon habitat. By means of high pressure freeze fixation, followed by freeze substitution, we could detect an up to 2-µm-thick cell wall surrounded by a distinct layer of extracellular polymeric substances (EPS), likely responsible for the adhering ca

Acaricidal and repellent activity of terpenoids from seaweeds Collected in Pernambuco, Brazil.

n-Hexane and dichloromethane extracts obtained from Laurencia dentroidea (Rhodophyta) and Canistrocarpus cervicornis (Phaeophyta) were investigated for their acaricidal and repellent properties against Tetranychus urticae under laboratory conditions. The two extracts displayed moderate toxicity and good repellent proprieties, and were significantly more toxic (36-fold) than the positive control (eugenol), whereas eugenol was tenfold more repellent than either seaweed extract. The sesquiterpenoid elatol (1) was isolated from L. dentroidea and the diterpenoid seco-dolastane (4R,9S,14S)-4alpha-ac

Impacts of light limitation on corals and crustose coralline algae.

Turbidity associated with elevated suspended sediment concentrations can significantly reduce underwater light availability. Understanding the consequences for sensitive organisms such as corals and crustose coralline algae (CCA), requires an understanding of tolerance levels and the time course of effects. Adult colonies of Acropora millepora and Pocillopora acuta, juvenile P. acuta, and the CCA Porolithon onkodes were exposed to six light treatments of ~0, 0.02, 0.1, 0.4, 1.1 and 4.3 mol photons m-2 d-1, and their physiological responses were monitored over 30 d. Not All Children Are the Same: Differences in the Microbiome Assembly During Early Development of Seaweeds.

Microbial symbionts play key roles in macroalgal development, yet the processes structuring early-life microbiomes remain poorly understood. Using laboratory outgrowth experiments and 16S rRNA gene amplicon sequencing we compared microbiome acquisition and assembly during the early development of three distinct macroalgae: Ulva australis (Chlorophyta), Hormosira banksii (Phaeophyceae) and Delisea pulchra (Rhodophyta). All species established distinct bacterial communities within the first week of outgrowth, with significant shifts in community composition and structure associated with major de

Region-specific carbon metabolism underlies floridean starch accumulation in rhizoid-like filament cells of Pyropia yezoensis.

BACKGROUND: Pyropia yezoensis thalli exhibit pronounced cellular differentiation throughout their growth and development. In particular, basal rhizoid-like filament cells (R-region cells) differ significantly from other thallus cells in cell morphology, photosynthetic physiology and carbon metabolism. Previous studies have shown that abundant floridean starch granules accumulate in R-region cells, whereas only few floridean starch granules are present in neighbouring vegetative cells (V-region cells). However, the specific mechanism underlying floridean starch accumulation in R-region cells re

The effects of temperature and CO2 enrichment on the red seaweed Asparagopsis taxiformis from Southern California with implications for aquaculture.

The red alga Asparagopsis taxiformis has recently been recognized for its unique ability to significantly reduce methane emissions from ruminant animals when fed in small quantities. The main obstacle in using this seaweed as a methane-mitigating feed supplement is the lack of commercially available biomass. Little is known about how best to grow this red alga on a commercial scale, as there are few published studies that have investigated the factors that influence growth, physiology, and overall performance. This study examined the effects of temperature and CO2 enrichment on the growth, pho
