Combined increases in atmospheric CO2 and warming temperatures impact photosynthesis in complex yet mechanistically predictable ways. Seminal work in this area showed that temperatures above a thermal optimum reduce photosynthesis primarily by increasing photorespiration, but that elevated CO2 and higher temperature can act synergistically to raise the thermal optimum of photosynthesis and increase absolute photosynthetic rates. The modeling work that led to this prediction outlined both the synergistic effects of CO2 and temperature and hypothesized scenarios that could deviate from theory. H

Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco), the key CO2-fixing enzyme in photosynthesis, is notorious for its low carboxylation activity. However, the difficulty in rationally engineering a fast Rubisco over the past decades brings a question whether a constraint exists in Rubisco's catalytic mechanism. In this study, we show that altering a single amino acid at position 398 in Form II Rubisco doubles its catalytic efficiency. The T398S and T398A mutations of the Form II Rubisco from the symbiont of Riftia pachyptila increases activity by 61 % and 74 %, respectively. The T398A m

Despite the declarations and collective measures taken to eradicate hunger at World Food Summits, food security remains one of the biggest issues that we are faced with. The current scenario could worsen due to the alarming increase in world population, further compounded by adverse climatic conditions, such as increase in atmospheric temperature, unforeseen droughts and decreasing soil moisture, which will decrease crop yield even further. Furthermore, the projected increase in yields of C3 crops as a result of increasing atmospheric CO2 concentrations is much less than anticipated. Thus, the

RIBULOSE-1,5-BISPHOSPHATE CARBOXYLASE/OXYGENASE (RUBISCO) and the Calvin-Benson-Bassham (CBB) cycle assimilate most of the inorganic carbon on earth. Conserved CP12 proteins control diurnal C-assimilation of the CBB cycle. Their function originated in cyanobacteria that deactivate GLYCERALDEHYDE 3-PHOSPHATE DEHYDROGENASE (GAPDH) and PHOSPHORIBULOKINASE (PRK) during darkness by redox-regulated complex-formation with CP12. This mechanism indicates metabolic regulation of C-assimilation. We measured C-assimilation in vivo at the day/night transition by dynamic 13CO2 labeling and 1-C-positional mo

Poplar are important forestry species in China, but the Botryosphaeria dothidea pathogen causes serious economic losses worldwide. To identify candidate B. dothidea resistance proteins and explore the molecular mechanisms involved in poplar-pathogen interactions, proteomic responses of stem samples from resistant and susceptible poplar ecotypes to B. dothidea were investigated using nanoflow liquid chromatography-tandem mass spectrometry with label-free quantitative analysis. We identified 588 proteins, divided into 21 biological process categories including 48 oxidoreductases, 72 hydrolytic e

RuBisCO (Ribulose-1,5-bisphosphate carboxylase/oxygenase) is the primary enzyme responsible for carbon fixation in the Calvin-Benson-Bassham cycle, yet it remains one of the most inefficient enzymes in nature. Its slow catalytic rate, low specificity for CO₂ over O₂, and susceptibility to inhibition and heat stress limit photosynthetic efficiency and crop productivity, particularly under fluctuating environmental conditions. This review highlights current strategies to enhance RuBisCO performance. These include direct mutagenesis, ancestral protein reconstruction, and the incorporation of chim

Rubisco fixes CO2 through the carboxylation of ribulose 1,5-bisphosphate (RuBP) during photosynthesis, enabling the synthesis of organic compounds. The natural diversity of Rubisco properties represents an opportunity to improve its performance and there is considerable research effort focusing on better understanding the properties and regulation of the enzyme. This chapter describes a method for large-scale purification of Rubisco from leaves. After the extraction of Rubisco from plant leaves, the enzyme is separated from other proteins by fractional precipitation with polyethylene glycol fo

Future climate scenarios suggest that crop plants will experience environmental changes capable of affecting their productivity. Among the most harmful environmental stresses is drought, defined as a total or partial lack of water availability. It is essential to study and understand both the damage caused by drought on crop plants and the mechanisms implemented to tolerate the stress. In this study, we focused on four cultivars of tomato, an economically important crop in the Mediterranean basin. We investigated the biochemical mechanisms of plant defense against drought by focusing on protei

Rubiscolin-6 (Tyr-Pro-Leu-Asp-Leu-Phe) is produced by a pepsin digest of spinach d-ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO) and known to act as an agonist on δ-opioid receptor. Here, we showed that administration of rubiscolin-6 reduced immobility time in the tail suspension test in restraint-stressed mice without effect on locomotor activity. The antidepressant-like effect of rubiscolin-6 was blocked by a δ-opioid receptor antagonist, naltrindole. These results indicate that rubiscolin-6 exerts antidepressant-like effect through activation of δ-opioid receptor.

Plants adapted to cool habitats have been suggested to possess Rubisco with a high carboxylation capacity and low CO2 affinity, thereby facilitating efficient photosynthesis at low temperatures. Rubisco with a low CO2 affinity is believed to be less abundant owing to its concomitant high carboxylation capacity. This study aimed to provide information on these subjects by examining plants with early-spring persistent leaves and alpine plants. In these plants, the turnover rates of Rubisco carboxylation under CO2-saturated conditions (kcatc) were low to moderate, similar to those of the control

Underwater CO2 concentration fluctuates extremely in natural water bodies. Under low CO2, the unique CO2 concentrating mechanism in aquatic plants, bicarbonate use, can suppress photorespiration. However, it remains unknown (1) to what extent bicarbonate use reduces photorespiration, (2) how exactly photorespiration varies between bicarbonate-users and CO2-obligate users under CO2-fluctuated environments, and (3) what are differences in Rubisco characteristics between these two types of aquatic plants. In the present study, the bicarbonate user Ottelia alismoides and its phylogenetically close

Information on the kinetic properties of Rubisco, a key enzyme for photosynthesis, is scarce in land plants that emerged early during the evolutionary process. This study examined the carboxylase activity and abundance of Rubisco in five conifers, two lycopods, and three control C3 crops. The turnover rates of Rubisco carboxylation (kcatc) under saturated-CO2 conditions in conifers and lycopods were comparable to those in the control C3 crops. Rubisco carboxylase activity under CO2-unsaturated conditions (vcu) was also measured using reaction mixtures saturated with a N2 gas containing CO2 and

Photorespiration is an essential metabolic process in C3 plants, yet it imposes significant carbon losses of up to 30% or more. Synthetic biology has recently enabled the engineering of diverse photorespiratory bypasses to overcome this limitation. In this review, we categorize these bypasses into three major types based on glycolate carbon retention and CO2 release. The first are chloroplast-localized carbon-releasing bypasses, which shift CO2 release from mitochondria to chloroplasts, thereby establishing a localized CO2-concentrating mechanism around Rubisco. The second are carbon-neutral b

An unbiased, quantitative view of biomolecules in a living cell is a prerequisite for accurate modeling approaches and informs our understanding of cellular metabolism at scale. In this work, we used the total protein approach (TPA), in which the total protein mass of a given proteomics sample is used as a calibrator for absolute protein quantification, to determine protein abundances during the Chlamydomonas reinhardtii diurnal cycle. We use external, independently measured quantitative markers (metals, pigments) to assess the absolute protein abundances in unlabeled whole cell extracts. We c

Carbon dioxide (CO2) assimilation by the enzyme Ribulose-1,5-bisphosphate Carboxylase/Oxygenase (Rubisco) underpins biomass accumulation in photosynthetic bacteria and eukaryotes. Despite its pivotal role, Rubisco has a slow carboxylation rate ([Formula: see text]) and is competitively inhibited by oxygen (O2). These traits impose limitations on photosynthetic efficiency, making Rubisco a compelling target for improvement. Interest in Form II Rubisco from Gallionellaceae bacteria, which comprise a dimer or hexamer of large subunits, arises from their nearly fivefold higher [Formula: see text]

Carboxysomes are anabolic bacterial microcompartments that play an essential role in CO2 fixation in cyanobacteria. This self-assembling proteinaceous organelle uses a polyhedral shell constructed by hundreds of shell protein paralogs to encapsulate the key CO2-fixing enzymes Rubisco and carbonic anhydrase. Deciphering the precise arrangement and structural organization of Rubisco enzymes within carboxysomes is crucial for understanding carboxysome formation and overall functionality. Here, we employed cryoelectron tomography and subtomogram averaging to delineate the 3D packaging of Rubiscos

Destruction of guard cell nuclei in epidermis isolated from leaves of pea, maize, sunflower, and haricot bean, as well as destruction of cell nuclei in leaves of the aquatic plants waterweed and eelgrass were induced by cyanide. Destruction of nuclei was strengthened by illumination, prevented by the antioxidant alpha-tocopherol and an electron acceptor N,N,N ,N -tetramethyl-p-phenylenediamine, and removed by quinacrine. Diatoms use multiple mechanisms of dissolved inorganic carbon (DIC) uptake to maintain CO2 supply to RuBisCO (ribulose-1,5-bisphosphate carboxylase/oxygenase), but how these different pathways are coordinated in response to rapid changes in their environment remains unclear. By measuring the dynamic changes in phycosphere carbonate chemistry around single diatom cells, we show that DIC uptake is rapidly modulated. We find that external carbonic anhydrase (eCA) plays a major role in DIC uptake in large centric diatoms at ambient seawater CO2, but there is a strong switch to direct HCO3- uptake

Bacterial microcompartments (BMCs) are pseudo-organelles that sequester metabolic enzymes, intermediates, and/or gases within the bacterial cytosol. One model BMC is the carboxysome (CB). CBs facilitate rubisco-driven fixation of CO2, increasing efficiency and maximizing the phosphoglycerate output in CB-containing bacteria. The α-CBs are of particular interest due to their small size and relative simplicity, making them ideal targets for bioengineering applications. These CBs were the first BMC observed and have been a long-studied model; however, they are challenging to study in native syste

The small chloroplastic protein CP12 has multiple functions, including the regulation of enzymes in the Calvin-Benson-Bassham cycle. Here, we investigated its role in the acclimation of Chlamydomonas reinhardtii to varying CO2 availability. We show that phosphoribulokinase can interact with CP12 in conditions where the Calvin-Benson-Bassham cycle is active. Compared to the wild type, at high CO2 C. reinhardtii ΔCP12 mutants have less phosphoribulokinase and ribulose-1,5-bisphosphate (RuBP), indicating that the regeneration of RuBP is regulated, in part, by CP12. Chlamydomonas reinhardtii has
