Opsins are the light receptors in retinal rod and cone photoreceptor cells that initiate vision in response to a light stimulus. Rhodopsin is the opsin in rods, and the influence of mutations that disrupt its structure and function has been characterized in detail. Less is known about cone opsins, the opsins in cones, and their gene arrays. Cones are responsible for color vision and high visual acuity and operate under most lighting conditions. There are up to three types of cone opsins (L-, M-, and S-opsin) in most vertebrates, each defined by their distinct spectral sensitivities. Disruption

Photoreceptor cell differentiation in the rat retina was studied in vivo and in vitro, using an immunohistochemical method to demonstrate opsin-like immunoreactivity. Cells in a dissociated monolayer culture expressed some properties characteristic of rat rod cells developing in vivo, including a ciliary structure and opsin-like immunoreactivity. Immunoblot analysis revealed that cultured retinal cells synthesize a polypeptide with the same molecular weight as that synthesized by the intact retina. Although the outer segment (OS) was not present in the culture, immunoreactive cells possessed a

A quantitative analysis of photoreceptor properties was performed in the retina of the nocturnal deer mouse, Peromyscus maniculatus, using pigmented (wildtype) and albino animals. The aim was to establish whether the deer mouse is a more suitable model species than the house mouse for photoreceptor studies, and whether oculocutaneous albinism affects its photoreceptor properties. In retinal flatmounts, cone photoreceptors were identified by opsin immunostaining, and their numbers, spectral types, and distributions across the retina were determined. Rod photoreceptors were counted using differe

We report the isolation and characterisation of a novel opsin cDNA from the retina and pineal of the common carp (Cyprinus carpio L.). When a comparison of the amino acid sequences of salmon vertebrate ancient opsin (sVA) and the novel carp opsin are made, and the carboxyl terminus is omitted, the level of identity between these two opsins is 81% and represents the second example of the VA opsin family. We have therefore termed this C. carpio opsin as carp VA opsin (cVA opsin). We show that members of the VA opsin family may exist in two variants or isoforms based upon the length of the carbox

A rod-specific antiserum was used to immunolabel elements within the retina and pineal of the adult Djungarian hamster and Welsh Mountain sheep. In the retina immunostaining was localized to the outer segments and perikarya of photoreceptor cells, while in the pineal limited numbers of labelled pinealocytes were scattered throughout the gland. An enzyme-linked immunosorbent assay (ELISA) was then used to obtain a quantitative measure of rod opsin in total eye and pineal extracts from the Djungarian hamster. Total rod opsin (+/- SEM) in the eye was measured by absorbance spectroscopy (1.88 +/-

The pineal organ of the frog, Rana esculenta and R. temporaria, was studied by opsin immunocytochemistry using two polyclonal antibovine rhodopsin and the monoclonal antichicken opsin antibodies OS-2 (detecting blue and green pigments) and COS-1 (detecting green and red pigments). Four types of photoreceptor cells were distinguished. The large outer segments of the numerous electron-dense photoreceptor cells ("large pineal rods") were immunoreactive with the rhodopsin and OS-2 antibodies, but reacted weakly with antibody COS-1. Some electron-dense photoreceptors with smaller outer segments ("s

Several retinal photoreceptor proteins involved in phototransduction have also been found in the mammalian pineal. This study demonstrates that rat and human pineals express protein kinases that are identical to the corresponding rod photoreceptor rhodopsin kinases. The deduced amino acid sequence of rat and human rhodopsin kinases have 84% sequence similarity to the earlier reported sequence of the bovine retinal enzyme, with complete conservation of the topological regions containing the position of the catalytic domain and sites of posttranslational modifications. Rat pineal also expresses

We previously characterized five visual opsin genes of American chameleon (Anolis carolinensis). Here we report its nonvisual opsin gene orthologous to the chicken pineal gland-specific opsin (p-opsin) gene. In the pure-cone American chameleon retina, all visual opsins including rod opsin are expressed. In both pineal and parietal eye, three visual opsins as well as P-opsin are expressed. Although opsins are detected in the pineal glands of a wide variety of vertebrates, Southern analysis suggests that the P-opsin gene is used mainly by birds and reptiles.

The retinal pigment epithelium (RPE)-dependent visual cycle provides 11-cis-retinal to opsins in the photoreceptor outer segments to generate functional visual pigments that initiate phototransduction in response to light stimuli. Both RPE65 isomerase of the visual cycle and the rhodopsin visual pigment have recently been identified as critical players in mediating light-induced retinal degeneration. These findings suggest that the expression and function of RPE65 and rhodopsin need to be coordinately controlled to sustain normal vision and to protect the retina from photodamage. However, the

The mechanisms that specify photoreceptor cell-fate determination, especially as regards to short-wave-sensitive (S) versus medium-wave-sensitive (M) cone identity, and maintain their nature and function, are not fully understood. Here we report the importance of general transcription factor II-I repeat domain-containing protein 1 (GTF2IRD1) in maintaining M cone cell identity and function as well as rod function. In the mouse, GTF2IRD1 is expressed in cell-fate determined photoreceptors at postnatal day 10. GTF2IRD1 binds to enhancer and promoter regions in the mouse rhodopsin, M- and S-opsin

The fovea is a region of the human retina specialized for high-acuity vision, with a high density of cones and no rod photoreceptors. Recent studies using retinal organoids indicate that retinoic acid (RA) may play a key role in foveal development: low RA levels in the fovea, maintained by CYP26A1, lead to early cell cycle exit for progenitors and a low rod-to-cone ratio. To test this model in fetal human development, we studied human fetal retina in 3D cultures in the presence of RA and RA inhibitors. We find that inhibition of RA reduces retinal progenitor proliferation and rod development,

The vertebrate visual system contains a sophisticated physiological assembly of highly-specialized proteins that mediate light stimuli to be processed by the brain. Two types of cells, the rod and cone cells, are involved in detecting dim and bright light, respectively. The rod cell contains rhodopsin whose structure and function has been extensively studied and the molecular interactions involving rhodopsin and the other proteins of the phototransduction cascade have been dissected in great detail, although several mechanistic features remain to be determined. The cone phototransduction proce

It is unclear whether normal human skin tissue or abnormal scarring are photoreceptive. Therefore, this study investigated photosensitivity in normal skin tissue and hypertrophic scars. The expression of opsins, which are photoreceptor proteins, in normal dermal fibroblasts (NDFs) and hypertrophic scar fibroblasts (HSFs) was examined. After exposure to blue light (BL), changes in the expression levels of αSMA and clock-related genes, specifically PER2 and BMAL1, were examined in both fibroblast types. Opsins were expressed in both fibroblast types, with OPN3 exhibiting the highest expression l

PURPOSE: Accumulation of free opsin by mutations in rhodopsin or insufficiencies in the visual cycle can lead to retinal degeneration. Free opsin activates phototransduction; however, the link between constitutive activation and retinal degeneration is unclear. In this study, the photoresponses of Xenopus rods rendered constitutively active by vitamin A deprivation were examined. Unlike their mammalian counterparts, Xenopus rods do not degenerate. Contrasting phototransduction in vitamin A-deprived Xenopus rods with phototransduction in constitutively active mammalian rods may provide new unde

The non-visual photoreceptor opsin 3 (OPN3) performs various functions by directly accepting light and activating G protein-coupled receptor signalling in non-visual tissues. OPN3, which is expressed in brown adipocytes, induces the expression of uncoupling protein 1 to generate heat upon exposure to blue light. There have been few reports on OPN3 expression in ruminants, and the mRNA sequence registered in a database as bovine OPN3 is the only predicted sequence. Therefore, we examined OPN3 expression in cattle. In the bovine retina, amplified fragments of the visual opsins, rhodopsin and ops

Snakes are a valuable yet understudied taxon for investigating evolutionary adaptations in the vertebrate retina. They possess up to three visual pigments: a short-wavelength-sensitive opsin (SWS1), a medium/long-wavelength-sensitive opsin (LWS), and rhodopsin (RH1). Nocturnal snakes have duplex retinas containing both rod and cone photoreceptors, whereas diurnal caenophidian ("advanced") snakes exhibit simplex "all-cone" retinas, lacking morphologically typical rods. In this study, we analyzed photoreceptor morphology in the retinas of caenophidian snakes using high-resolution scanning electr

RHO (Rod opsin) encodes a G-protein coupled receptor that is expressed exclusively by rod photoreceptors of the retina and forms the essential photopigment, rhodopsin, when coupled with 11-cis-retinal. Many rod opsin disease -mutations cause rod opsin protein misfolding and trigger endoplasmic reticulum (ER) stress, leading to activation of the Unfolded Protein Response (UPR) signal transduction network. Chop is a transcriptional activator that is induced by ER stress and promotes cell death in response to chronic ER stress. Here, we examined the role of Chop in transgenic mice expressing huma

In normal retina the developing photoreceptor cells first show presence of opsin over the distal ends of the ciliary protrusions. In a fully differentiated cell intense activity is seen over the rod outer-segment discs; some activity is also seen over the Golgi zone and near the distal ends of the inner segments but the other parts of the receptor cell appear negative. In the pigment epithelium opsin is seen only over phagosomes containing rod outer segment debris. In the homozygous rds mutant retina, developing receptor cells show opsin activity over the ciliary protrusions as in the normal.

A dominant form of human congenital nightblindness is caused by a gly90-->asp (G90D) mutation in rhodopsin. G90D has been shown to activate the phototransduction cascade in the absence of light in vitro. Such constitutive activity of G90D rhodopsin in vivo would desensitize rod photoreceptors and lead to nightblindness. In contrast, other rhodopsin mutations typically give rise to nightblindness by causing rod cell death. Thus, the proposed desensitization without rod degeneration would be a novel mechanism for this disorder. To explore this possibility, we induced mice to express G90D opsin i

The G90D rhodopsin mutation is known to produce congenital night blindness in humans. This mutation produces a similar condition in mice, because rods of animals heterozygous (D+) or homozygous (D+/+) for this mutation have decreased dark current and sensitivity, reduced Ca(2+), and accelerated values of tau(REC) and tau(D), similar to light-adapted wild-type (WT) rods. Our experiments indicate that G90D pigment activates the cascade, producing an equivalent background light of approximately 130 Rh* rod(-1) for D+ and 890 Rh* rod(-1) for D+/+. The active species of the G90D pigment could be un
