Lotus japonicus THIC is expressed in all organs, and the encoded protein catalyzes thiamine biosynthesis. Loss of function produces chlorosis, a typical thiamine-deficiency phenotype, and mortality. To investigate thiamine's role in symbiosis, we focused on THI1, a thiamine-biosynthesis gene expressed in roots, nodules, and seeds. The thi1 mutant had green leaves, but formed small nodules and immature seeds. These phenotypes were rescued by THI1 complementation and by exogenous thiamine. Thus, THI1 is required for nodule enlargement and seed maturation. On the other hand, colonization by arbus

Legume plants commonly associate with both arbuscular mycorrhizal (AM) fungi and rhizobia and thus enhance the acquisition of phosphorus (P) and nitrogen (N) nutrition. Inoculation with AM fungi can promote nodulation and N2 fixation of legume plants; however, the underlying mechanisms remain poorly understood. Here, root exudates collected from AM-colonised soybean plants showed greater accumulation of the specific flavonoids (daidzein and genistein) and phenolic acids (benzoic acid and p-Hydroxybenzoic acid), and significantly promoted nodulation. Furthermore, the exudates from AM-colonised

Legume plants can acquire mineral nitrogen (N) either through their roots or via a symbiotic interaction with N-fixing rhizobia bacteria housed in root nodules. To identify shoot-to-root systemic signals acting in Medicago truncatula plants at N deficit or N satiety, plants were grown in a split-root experimental design in which either high or low N was provided to half of the root system, allowing the analysis of systemic pathways independently of any local N response. Among the plant hormone families analyzed, the cytokinin trans-zeatin accumulated in plants at N satiety. Cytokinin applicati

Symbiosis Receptor-like Kinase (SYMRK) is indispensable for the development of phosphate-acquiring arbuscular mycorrhiza (AM) as well as nitrogen-fixing root nodule symbiosis, but the mechanisms that discriminate between the two distinct symbiotic developmental fates have been enigmatic. In this study, we show that upon ectopic expression, the receptor-like kinase genes Nod Factor Receptor 1 (NFR1), NFR5, and SYMRK initiate spontaneous nodule organogenesis and nodulation-related gene expression in the absence of rhizobia. Furthermore, overexpressed NFR1 or NFR5 associated with endogenous SYMRK

PREMISE: Prairies are among the most threatened biomes due to changing patterns of climate and land use, yet information on genetic variation in key species that would inform conservation is often limited. We assessed evidence for the geographic scale of population-level variation in growth of two species of prairie clover and of their symbiotic associations with nitrogen-fixing bacteria.
METHODS: Seed representing two species, Dalea candida and D. purpurea, from the same five source populations were planted into an experimental site in Minnesota. We assessed variation within and among source

BACKGROUND: In the current agricultural model, the massive use of chemical fertilizer causes environmental and economic losses. Inoculation of plant-growth-promoting (PGP) nitrogen-fixing bacteria is an alternative to the use of nitrogen, phosphorus and potassium fertilizers. In this study, rhizobia strains isolated from common bean (Phaseolus vulgaris) root nodules were evaluated in an effort to identify an efficient nitrogen-fixing rhizobia strain able to improve bean germination and growth.
RESULTS: Common bean plants were collected from seven sites in southern Brazil, and 210 native rhizo

Although it is well established that nitrate exerts an inhibitory effect on nodulation and biological nitrogen fixation in soybean, the underlying mechanism remains unclear. In soybean plants, nitrate is assimilated into asparagine (Asn) and glutamine (Gln); their systemic circulation within the plant may contribute to the reduced N-fixation capacity of nodules. To investigate the effects of nitrate, Asn, and Gln on soybean nodulation and biological N fixation, a unilateral nodulated double-root soybean system was used. The non-nodulated side roots were supplied with nitrate (14 mM), Asn (20 m

Reactive oxygen species play a crucial role in various stages of the legume-rhizobia symbiosis, from initial nodulation signaling to nodule senescence. However, how rhizobial redox-related proteins regulate symbiotic nodulation in legumes remains largely unknown. By combining transcriptomics, proteomics, and biochemical and molecular genetics, we investigated the role of the Sinorhizobium fredii Q8 enzyme 3-mercaptopyruvate sulfurtransferase (3MST). Although 3MST was not the primary enzyme responsible for hydrogen sulfide (H2S) production under our conditions, its absence significantly impaire

The symbiotic relationship between the legume Medicago sativa and the soil bacteria Sinorhizobium meliloti results in the formation of nitrogen-fixing root nodules. Traditional destructive methods, including paraffin sectioning, vibratome sectioning, and cryosectioning, have been applied to visualize how bacteria occupy the nodule, making it extremely difficult to obtain reliable three-dimensional information. These approaches are often combined with fluorescent labeling or staining, which can introduce additional stress affecting plant growth and nodule formation. MicroCT has emerged as a rel

Reactive sulfur species (RSS) function as strong antioxidants and are involved in various biological responses in animals and bacteria. Few studies; however, have examined RSS in plants. In the present study, we clarified that RSS are involved in root nodule symbiosis in the model legume Lotus japonicus. Polysulfides, a type of RSS, were detected in the roots by using a sulfane sulfur-specific fluorescent probe, SSP4. Supplying the sulfane sulfur donor Na2S3 to the roots increased the amounts of both polysulfides and hydrogen sulfide (H2S) in the roots and simultaneously decreased the amounts

The root system of barley plants is composed of embryogenic, seminal roots as well as lateral and nodal roots that are formed postembryonically from seminal roots and from the basal part of shoots, respectively. Due to their distinct developmental origin, seminal and nodal roots may differ in function during plant development; however, a clear comparison between these two root types has not yet been undertaken. In this study, anatomical, proteomic and physiological traits were compared between seminal and nodal roots of similar developmental stages. Nodal roots have larger diameter, larger met

White lupin (Lupinus albus) is a legume species that develops two remarkable root adaptations to nutrient limitation: nitrogen-fixing nodules and dense cluster roots specialized for phosphate acquisition. These traits, together with the species' agronomic relevance and strong developmental plasticity, make white lupin a powerful model for studying local and systemic regulation of root development under nutrient stress. This chapter provides a comprehensive collection of experimental procedures designed to investigate cluster root formation, nodulation, and associated physiological responses at

Nitrate reductase (NR) is a key enzyme in nitrate assimilation, yet its function within nodules remains poorly understood. In Medicago truncatula, 3 NR genes, MtNR1, MtNR2, and MtNR3, exhibit distinct evolutionary origins and regulatory features. Phylogenetic analyses indicate that NR3-type genes, originated from a duplication of NR1 within the inverted repeat-lacking clade (IRLC) legumes, have lost the conserved phosphorylation sites critical for post-translational regulation. To assess the functional significance of these isoforms, we characterized single and double nr mutants obtained throu

Microorganisms belonging to root and soil provide a wide range of services and benefits to the plant by promoting plant growth and controlling phytopathogens. This study aimed to isolate endophytic bacteria from the root nodules of chickpea (Cicer arietinum L.) and determine their potential in improving plant growth. A total of nineteen different bacterial morphotypes were isolated from root nodules of chickpea and characterized in vitro for plant growth promotion abilities. All bacterial isolates were able to produce indole acetic acid at varying levels, out of which MCA19 was screened as the

Legume plants have the capacity to incorporate atmospheric nitrogen by establishing an endosymbiotic interaction with soil bacteria resulting in the formation of nitrogen-fixing nodules. Bacteria are internalized through a tightly regulated process that requires membrane remodelling and vesicle trafficking, which are controlled by small GTPases. Members of the ARF family of GTPases mediate vesicle budding in a wide range of biological processes; however, the modulation of ARF members, their subcellular localization and the formation of complexes with other proteins during the root nodule symbi

This section details the optimized methods for single-cell RNA sequencing (scRNA-seq) in Medicago truncatula roots and nodules. We describe a detailed experimental workflow, comprising plant growth and bacteria application, protoplast isolation, quality control, and library processing according to 10× Genomics®, suitable for both long- and short-read sequencing. This protocol can serve as a starting point for establishing robust, high-quality scRNA-seq workflows to study nodulation and root development in legumes.

The symbiotic interaction between soybean (Glycine max) and Bradyrhizobium japonicum results in the formation of root nodules, specialized organs that house nitrogen-fixing bacteria converting atmospheric N2 into plant-accessible ammonia (NH3). Accurate quantification of nodule traits is essential for understanding host-microbe interactions and genetic determinants of nodulation. However, traditional manual or semi-quantitative approaches are labor-intensive, subjective, and unsuitable for large-scale studies. Here, we present a high-throughput phenotyping pipeline based on the YOLO deep learn

Legume root nodules harbor rhizobia and other non-nodulating endophytes known as nodule-associated bacteria (NAB) whose role in the legume symbiosis is still unknown. We analysed the genetic diversity of 34 NAB isolates obtained from the root nodules of faba bean grown under various soil conditions in Egypt using 16S rRNA and concatenated sequences of three housekeeping genes. All isolates were identified as members of the family Enterobacteriaceae belonging to the genera Klebsiella, Enterobacter and Raoultella. We identified nine enterobacterial genospecies, most of which have not been previo
