Induction of larval settlement in the serpulid polychaete Hydroides elegans (Haswell): role of bacterial extracellular polymers.

Larval settlement in the marine polychaete Hydroides elegans is effectively mediated upon contact with the surface of marine bacterial films. Using the bacterium Roseobacter litoralis as a model strain, the effect of bacterial extracellular polymers (exopolymers) on larval settlement of H. elegans was investigated. Bioassays with exopolymer fractions dissociated from bacterial films evoked the initial stages of the larval settlement process, i.e. larvae slowed down, secreted a mucous thread and crawled over the surface. This response is typical of larvae that encounter an attractive bacterial

A denitrifying community associated with a major, marine nitrogen fixer.

The diazotrophic cyanobacterium, Trichodesmium, is an integral component of the marine nitrogen cycle and contributes significant amounts of new nitrogen to oligotrophic, tropical/subtropical ocean surface waters. Trichodesmium forms macroscopic, fusiform (tufts), spherical (puffs) and raft-like colonies that provide a pseudobenthic habitat for a host of other organisms including marine invertebrates, microeukaryotes and numerous other microbes. The diversity and activity of denitrifying bacteria found in association with the colonies was interrogated using a series of molecular-based methodol

Brucellosis outbreak in a Swedish kennel in 2013: determination of genetic markers for source tracing.

Brucellosis is a highly infectious zoonotic disease but rare in Sweden. Nonetheless, an outbreak of canine brucellosis caused by an infected dog imported to Sweden was verified in 2013. In total 25 dogs were tested at least duplicated by the following approaches: real-time PCR for the detection of Brucella canis, a Brucella genus-specific real-time PCR, selective cultivation, and microscopic examination. The whole genome of B. canis strain SVA13 was analysed regarding genetic markers for epidemiological examination. The genome of an intact prophage of Roseobacter was detected in B. canis strai

Evolutionary Conservation of the Copper-Dependent Thiol Oxidase Activity of Selenium-Binding Proteins.

Human selenium-binding protein 1 (SELENBP1) is a methanethiol oxidase (MTO), converting methanethiol (MT) to hydrogen sulfide (H2S), hydrogen peroxide (H2O2) and formaldehyde (HCHO). SELENBP1 has orthologs in all domains of life. RdMTO, an orthologous enzyme recently identified in the marine bacterium Roseobacter denitrificans, was postulated to require the cysteine residue closest to its C-terminus, Cys448, for MT binding and to oxidize MT to sulfane sulfur (S0) rather than to H2S. SELENBP1 and RdMTO exhibit ~53% sequence identity, with Cys448 (numbered Cys466 in SELENBP1) and amino acids req

Re-examining the iron paradox: the influence of growth medium on regulation and functionality of the tropodithietic acid (TDA) biosynthetic pathway in Phaeobacter piscinae.

Tropodithietic acid (TDA) is a secondary metabolite with antimicrobial and iron-binding properties, produced by several marine Roseobacter group bacteria. Although classified as a siderophore, TDA is also produced under iron-replete conditions. To address this paradox, we fused the tdaCDE promoter region of Phaeobacter piscinae S26 with a promoter-less gfp reporter gene to monitor the transcription of TDA biosynthetic genes under different conditions and validated its accuracy using RT-qPCR. In both complex ($\frac{1}{2}$YTIO) and low-complexity (IOCGH) marine media, iron supplementation repre

Identification of a functional DddX-like enzyme that catalyzes dimethylsulfoniopropionate cleavage in marine bacteria.

Dimethylsulfoniopropionate (DMSP), a ubiquitous organosulfur compound in marine environments, plays an important role in global sulfur cycling and climate regulation. Many marine microorganisms catabolize DMSP through DMSP lytic enzymes, producing the infochemical and potential climate-cooling gas dimethyl sulfide (DMS). The marine Roseobacter group (MRG) is one of the most abundant heterotrophic bacteria groups in marine ecosystems and an active participant in DMSP cycling. To expand our knowledge of the diversity of DMSP lytic enzymes, we identified and characterized a functional DddX-like e

Possible links between white plague-like disease, scleractinian corals, and a cryptochirid gall crab.

White plague (WP) is a highly destructive coral disease that rapidly kills susceptible coral species by mass tissue lysis. The pathogen and underlying causes of this disease are not known. In this laboratory-based study, we examined a small coral-associated gall crab from the family Cryptochiridae in terms of a possible association with WP-like lesions. A series of experiments was conducted after observations that 2 scleractinian coral species, Diploria labyrinthiformis and Pseudodiploria strigosa, developed signs of WP-like disease within a laboratory holding aquarium and that small gall crab

Friends and foes: symbiotic and algicidal bacterial influence on Karenia brevis blooms.

Harmful Algal Blooms (HABs) of the toxigenic dinoflagellate Karenia brevis (KB) are pivotal in structuring the ecosystem of the Gulf of Mexico (GoM), decimating coastal ecology, local economies, and human health. Bacterial communities associated with toxigenic phytoplankton species play an important role in influencing toxin production in the laboratory, supplying essential factors to phytoplankton and even killing blooming species. However, our knowledge of the prevalence of these mechanisms during HAB events is limited, especially for KB blooms. Here, we introduced native microbial communiti

Changes in the Urinary Microbiome After Transurethral Resection of Non-muscle-Invasive Bladder Cancer: Insights from a Prospective Observational Study.

BACKGROUND: This study aimed to characterize the urinary and tumor microbiomes in patients with non-muscle-invasive bladder cancer (NMIBC) before and after transurethral resection of the bladder tumor (TURBT). METHODS: This single-center prospective study included 26 samples from 11 patients with low-grade Ta papillary NMIBC. Urine samples were collected at the index TURBT and at a 1-year follow-up cystoscopy. The metagenomic analysis of bacterial and archaeal populations was performed based on the highly variable V3-V4 region of the 16S rRNA gene. RESULTS: Phylogenetic alpha diversity of th

Natural Benzo/Acetophenones as Leads for New Synthetic Acetophenone Hybrids Containing a 1,2,3-Triazole Ring as Potential Antifouling Agents.

Marine biofouling is a natural process that represents major economic, environmental, and health concerns. Some booster biocides have been used in biofouling control, however, they were found to accumulate in environmental compartments, showing negative effects on marine organisms. Therefore, it is urgent to develop new eco-friendly alternatives. Phenyl ketones, such as benzophenones and acetophenones, have been described as modulators of several biological activities, including antifouling activity (AF). In this work, acetophenones were combined with other chemical substrates through a 1,2,3-

16S rDNA sequencing combined with metabolomic probes to investigate the effects of Salmonella Pullorum on gut microbes and metabolites in broilers.

Pullorum disease (PD) caused by Salmonella Pullorum (SP) results in high mortality in chicks and potential carriers in adult chickens, negatively affecting growth and egg production. This study identified SP infection in 100-day-old White Plymouth Rock hens by serum plate agglutination and fecal and anal swab polymerase chain reaction. SP-infected broilers were classified into positive (P) and negative (N) groups using hematoxylin-and-eosin staining, metabolome sequencing, and 16S rDNA to investigate the effects of SP infection on the metabolites and microorganisms in the cecum of broilers. Multi-omics uncovers nutrient stress-driven interactions in a synthetic Prymnesium parvum holobiont, with vitamin B12-limitation revealing mutualism.

Microalgal-bacterial interactions are central to nutrient cycling and ecosystem functioning in marine habitats, yet the mechanisms structuring these associations under defined nutrient constraints remain poorly resolved. Using a synthetic 15-member bacterial community (SynCom) and controlled nitrogen (N), phosphorus (P), and vitamin B12-limitation, we investigated how nutrient scarcity shapes the physiology, metabolism, and transcriptional activity of the harmful alga Prymnesium parvum and its associated microbiota. Under N- and P-limitation, the SynCom had minimal impact on algal growth despi

Coordinated regulation of trimethylamine catabolism in abundant marine bacteria.

Methylated amines (MAs) are ubiquitous in the marine environment, constituting an important part of the marine dissolved organic nitrogen pool and contributing to the formation of cloud condensation nuclei in the marine atmosphere. The aerobic trimethylamine (TMA) oxidation pathway represents the primary route for TMA utilization in marine environments and has been characterized in the model marine Roseobacter group bacterium Ruegeria pomeroyi DSS-3. However, the regulatory mechanism underlying this pathway has remained unclear. Transcriptome analysis revealed coordinated induction of the enti
