Evidence for the use of a high-resolution magnetic map by a short-distance migrant, the Alpine newt (Ichthyosaura alpestris).

Newts can use spatial variation in the magnetic field (MF) to derive geographic position, but it is unclear how they detect the 'spatial signal', which, over the distances that newts move in a day, is an order of magnitude lower than temporal variation in the MF. Previous work has shown that newts take map readings using their light-dependent magnetic compass to align a magnetite-based 'map detector' relative to the MF. In this study, time of day, location and light exposure (required by the magnetic compass) were varied to determine when newts obtain map information. Newts were displaced from

The road not taken: Evolution of tetrodotoxin resistance in the Sierra garter snake (Thamnophis couchii) by a path less travelled.

The repeated evolution of tetrodotoxin (TTX) resistance provides a model for testing hypotheses about the mechanisms of convergent evolution. This poison is broadly employed as a potent antipredator defence, blocking voltage-gated sodium channels (Nav ) in muscles and nerves, paralysing and sometimes killing predators. Resistance in taxa bearing this neurotoxin and a few predators appears to come from convergent replacements in specific Nav residues that interact with TTX. This stereotyped genetic response suggests molecular and phenotypic evolution may be constrained and predictable. Here, we

ITRAQ-based quantitative proteomic analysis of Cynops orientalis limb regeneration.

BACKGROUND: Salamanders regenerate their limbs after amputation. However, the molecular mechanism of this unique regeneration remains unclear. In this study, isobaric tags for relative and absolute quantification (iTRAQ) coupled with liquid chromatography tandem mass spectrometry (LC-MS/MS) was employed to quantitatively identify differentially expressed proteins in regenerating limbs 3, 7, 14, 30 and 42 days post amputation (dpa). RESULTS: Of 2636 proteins detected in total, 253 proteins were differentially expressed during different regeneration stages. Among these proteins, Asporin, Cadher

Noxious newts and their natural enemies: Experimental effects of tetrodotoxin exposure on trematode parasites and aquatic macroinvertebrates.

The dermal glands of many amphibian species secrete toxins or other noxious substances as a defense strategy against natural enemies. Newts in particular possess the potent neurotoxin tetrodotoxin (TTX), for which the highest concentrations are found in species within the genus Taricha. Adult Taricha are hypothesized to use TTX as a chemical defense against vertebrate predators such as garter snakes (Thamnophis spp.). However, less is known about how TTX functions to defend aquatic-developing newt larvae against natural enemies, including trematode parasites and aquatic macroinvertebrates. Her

The effects of thalidomide and two analogues on the regenerating forelimb of the newt.

Oral administration (3 mg/day) of thalidomide during the dedifferentiation and early limb-bud stages of newt forelimb regeneration produced a variety of specific limb deformities. Proximal and preaxial skeletal elements were the most severely malformed, e.g. preaxial hemimelia, severe proximal deformities, and preaxial polydactyly. Likewise, oral, daily doses (3 mg) of the teratogenic analogue, EM12, on days 7 and 8 following bilateral amputation caused the same incidence and type of forelimb abnormalities as did thalidomide. Conversely, the non-teratogenic analogue, EM87, when orally administ

Retinoic Acid-Induced Limb Duplications.

Retinoic acid (RA) and the family of molecules based on vitamin A known as retinoids have remarkable effects on limb regeneration in salamanders and newts and cause whole limb duplications in a concentration-dependent manner. They respecify all three axes of the limb-the proximodistal, the anteroposterior, and the dorsoventral axis. As a result, complete limbs can be induced to regenerate from distal amputation planes producing two limbs in tandem. Here, we describe the basic methods for undertaking these experiments as well as the use of new synthetic retinoids which have retinoic acid recept

Variation in vertebrae shape across small-bodied newts reveals functional and developmental constraints acting upon the trunk region.

The salamander vertebral column is largely undifferentiated with a series of more or less uniform rib-bearing presacral vertebrae traditionally designated as the trunk region. We explored regionalization of the salamander trunk in seven species and two subspecies of the salamander genus Lissotriton by the combination of microcomputed tomography scanning and geometric morphometrics. The detailed information on trunk vertebral shape was subjected to a multidimensional cluster analysis and a phenotypic trajectory analysis. With these complementary approaches, we observed a clear morphological reg

Daily melatonin injections entrain the circadian change of synaptic ribbon number in the pineal organ of the Japanese newt.

The effect of melatonin on the number of synaptic ribbons (SR) in the pineal photoreceptor cells was examined in the Japanese newt using electron microscopy. Under a light-dark cycle of LD 12:12 (lights on at 07:00), a subcutaneous injection of 10 microg of melatonin at two times during the day (at 11:00 and 11:30) did not increase the number of SR. However, daily injections of 10 microg of melatonin in constant darkness at 11:00 for 7 days did reset the phase of the circadian rhythm in SR number. The time of melatonin injection, corresponding to the onset of subjective night, preceded the dai

The occurrence of melanosomes in the newt glomus cell.

Combined light and electron microscopic observations of the carotid labyrinth of the newt, Cynops pyrrhogaster, revealed the occurrence of glomus cells with melanosomes (MG cells), a finding so far unreported in the amphibian carotid labyrinth. These melanosomes, which derived from melanophores, were electron-dense and usually oval (250-700 nm in diameter). They occurred in groups enclosed by a limiting membrane, and the diameter of clusters was approximately 1.5 microns. These clusters were surrounded by a large number of dense-cored vesicles (60-100 nm in diameter), which were predominantly

Ranavirus-associated mass mortality in wild amphibians, the Netherlands, 2010: a first report.

In 2010, a mass die-off of over 1000 wild water frogs (Pelophylax spp.) and at least 10 common newts (Lissotriton vulgaris) occurred in a pond in The Netherlands. Haemorrhagic disease with hepatomegaly and splenomegaly was evident. Microscopically, multiple organs presented cells with multifocal intracytoplasmic inclusion bodies, in which ranavirus-like particles were demonstrated ultrastructurally. All specimens examined tested positive for ranavirus by PCR. The sequence obtained showed a 100% identity with the one deposited for common midwife toad virus (CMTV). This is the first report of ra

Glycoconjugate expression in the olfactory bulb of the premetamorphic larva of the Japanese sword-tailed newt (Cynops ensicauda).

We examined the organization of the olfactory organ and assessed the lectin histochemistry to investigate the glycoconjugate distribution of the olfactory bulb in premetamorphic larvae of Cynops ensicauda. The nasal cavity was an oval chamber that contained olfactory epithelium and a primitive vomeronasal organ. Secretory products were found in the supporting cells of the two sensory epithelia and in the respiratory cells. Ten lectins bound to the olfactory and vomeronasal nerve fibers as well as to the glomeruli in the olfactory bulb. The binding intensity in larvae was weaker than that repor

The regeneration of newt limbs deformed in nature.

The regeneration of abnormal limbs from a wild newt population was examined. The abnormalities were: mirror symmetrical reduplications of digit parts, deformed wrists, or limbs with supernumerary digits. Normal regeneration resulted after amputation of the abnormal parts, regardless of the original deformity. The results suggest that the abnormalities are probably the result of local trauma to the limbs.

Patterns of regional variation in the vertebral column of terrestrial salamanders.

Regional variation in the vertebral column of several species of salamanders (families Ambystomatidae, Salamandridae and Plethodontidae) is analyzed. Measurements of three dimensions, centrum length, prezygapophyseal width, and transverse process length, provide the data. Ontogenetic, interspecific, intergeneric and interfamilial patterns of positional variation are diagrammed and discussed. Distinctive patterns of variation characterize the families, genera, and to a lesser extent, the species. The patterns of ambystomatid salamanders are the most generalized, and probably reflect derivation

Differential Immune Responses Correlate With Chytridiomycosis Severity in Italian Crested Newts.

In the midst of the current biodiversity crisis, amphibians are severely threatened by emerging diseases such as chytridiomycosis. Characterizing the mechanisms that underlie susceptibility to this disease is fundamental to improve amphibian conservation. Using a comprehensive multi-omics approach, this study investigates the impact of an exposure to the chytrid fungus Batrachochytrium salamandrivorans (Bsal) on the gene expression of Italian crested newts (Triturus carnifex) and on the bacterial symbionts that constitute their skin microbiota. Exposure to Bsal affected multiple components of

Distribution of accessory and hypoglossal nerves in the hindbrain and spinal cord of lungless salamanders, family Plethodontidae.

Study of the innervation of the musculature related to feeding behavior in plethodontid salamanders by means of the horseradish peroxidase (HRP) technique has demonstrated the existence of a true spinal accessory nerve which innervates neck musculature, enters the brain via the ganglion of IX/X cranial nerves and has its motor neurons within the nucleus of the second spinal nerve. Further, it has been shown that the first spinal nerve, being strictly motor, alone constitutes the ramus hypoglossus and is, therefore, homologous to the hypoglossus of amniotes.

miR-20487-5p/SERCA1/MAPK/ERK Pathway Regulates Newt Limb Regeneration.

Cynops orientalis, a member of the Salamandridae family, can regenerate severely injured limbs throughout its lifespan. The molecular mechanisms regulating limb regeneration in C. orientalis remain largely unknown. SERCA1 (sarcoplasmic/endoplasmic reticulum calcium ATPase 1) is a major sarco/endoplasmic reticulum calcium pump in skeletal muscle, and our previous transcriptomic sequencing results showed increased SERCA1 mRNA expression during the early stages of newt limb regeneration. In the present study, we investigated the role and regulatory mechanism of SERCA1 during newt limb regeneratio

Daily and circadian variations of the pineal and ocular melatonin contents and their contributions to the circulating melatonin in the Japanese newt, Cynops pyrrhogaster.

Daily and circadian variations of melatonin contents in the diencephalic region containing the pineal organ, the lateral eyes, and plasma were studied in a urodele amphibian, the Japanese newt (Cynops pyrrhogaster), to investigate the possible roles of melatonin in the circadian system. Melatonin levels in the pineal region and the lateral eyes exhibited daily variations with higher levels during the dark phase than during the light phase under a light-dark cycle of 12 h light and 12 h darkness (LD12:12). These rhythms persisted even under constant darkness but the phase of the rhythm was diff

J10 lactate-induced polycythemia in newts.

Specimens of Triturus cristatus carnifex (Laurenti) raised for about 3 weeks in a 3 gm/liter solution of sodium lactate present a slight alkalosis and a mean increase of about 35% in the red blood cell count in comparison to control animals raised in identically oxygenated water; analogous treatment with an equal molar concentration of sodium pyruvate produces only alkalosis. Since lactate alone determines an increase in the number of erythrocytes as does hypoxia, and hypoxia always induces an increase in plasma lactate levels, it can be deduced that erythropoietin (ESF) production is stimulat
