Thiamin dynamics during the adult life cycle of Atlantic salmon (Salmo salar).

Thiamin is an essential water-soluble B vitamin known for its wide range of metabolic functions and antioxidant properties. Over the past decades, reproductive failures induced by thiamin deficiency have been observed in several salmonid species worldwide, but it is unclear why this micronutrient deficiency arises. Few studies have compared thiamin concentrations in systems of salmonid populations with or without documented thiamin deficiency. Moreover, it is not well known whether and how thiamin concentration changes during the marine feeding phase and the spawning migration. Therefore, samp

Experimentally induced marine flexibacteriosis in Atlantic salmon smolts Salmo salar. II. Pathology.

The fish disease marine flexibacteriosis is characterised by necrotic lesions on the body, head, fins, and occasionally gills, with erosive lesions on the external surface as the prominent clinical sign. In Australia, the main species affected are Atlantic salmon Salmo salar and rainbow trout Oncorhynchus mykiss in sea-cage culture in Tasmania. Using a dose-dependent trial to determine pathology, 2 forms of the disease were noted in Atlantic salmon. The acute form occurs within 2 to 3 d after inoculation at high doses (1 x 10(8) cells ml(-1)) and is characterised by the disintegration of the e

Retinoic acid cross-talk with calcitriol activity in Atlantic salmon (Salmo salar).

Vitamins A (VA) and D (VD) are metabolised by vertebrates to bioactive retinoic acid (RA) and calcitriol (CTR). RA and CTR involvement in bone metabolism requires fine-tuned regulation of their synthesis and breakdown. In mammals antagonism of VA and VD is observed, but the mechanism of interaction is unknown. We investigated VA-VD interactions in Atlantic salmon (Salmo salar L.) following i.p. injection of RA and/or CTR. VA metabolites, CTR, calcium (Ca), magnesium (Mg) and phosphorus (P) were determined in plasma. Expression of bone matrix Gla protein (mgp), collagen 1 alpha2 chain (col1a2)

Purification and characterization of pancreatic elastase from North Atlantic salmon (Salmo salar).

An elastase I-like enzyme was purified to homogeneity from the pyloric caeca of North Atlantic salmon (Salmo salar) and compared with porcine elastase I. The molecular weight and isoelectric point were estimated to be 27 kDa and over 9.3, respectively. The pH optimum was between 8.0 and 9.5, and the enzyme was unstable at pH values below 4. Kinetic properties examined using Suc-(Ala)3-p-nitroanilide showed that the catalytic efficiency of salmon elastase was about 2.5 times higher than that of porcine elastase. Furthermore, the salmon enzyme was less stable at lower pH values and temperatures

Vibrio frigidus sp. nov., Isolated from the Gills of Atlantic Salmon (Salmo salar).

A novel strain, designated F74T, was isolated from the gills of a healthy Atlantic salmon, Salmo salar L., at a land-based aquaculture farm located on the Reykjanes peninsula in Iceland. The bacteria were facultative anaerobic, Gram-stain-negative curved rods that were motile and required NaCl for growth. They were sensitive to the vibriostatic agent O/129 and showed growth at 0 °C. The predominant fatty acids were C16:1 ω7c and/or C15 iso 2-OH (summed feature 3) (53.5%) and 16:0 (16.7%). The digital genomic DNA G + C content was 41.6%. Comparative analysis of the 16S rRNA gene revealed that t

Behavioural thermoregulation modulates circadian clock gene expression in juvenile Atlantic salmon (Salmo salar).

Environmental temperature is a key factor regulating physiological processes and circadian organization in ectothermic vertebrates. In fish, behavioural thermoregulation optimize physiological performance and may contribute to the synchronization of biological clocks, however, its effects on central and peripheral circadian oscillators remain poorly understood. In the present study, we investigated the daily rhythms of thermal preference and their influence on circadian organization in juvenile Atlantic salmon (Salmo salar). Fish were maintained either at a constant temperature or in a multi-c

Immunomodulatory and cellular effects of Aeromonas salmonicida melanin in Atlantic salmon (Salmo salar).

Aeromonas salmonicida subspecies salmonicida is a Gram-negative fish pathogen that synthesizes and secretes melanin, a dark pigment with a complex molecular structure. Melanin is formed by polymerized phenolic and/or indole monomers, which possess antioxidant, radical scavenging, photo-protective, and even semiconducting properties. The role of melanin in A. salmonicida pathogenesis is unknown. In this study, we evaluated the effect of A. salmonicida melanin on Atlantic salmon (Salmo salar) head kidney and spleen. Atlantic salmon were intraperitoneally injected with A. salmonicida melanin (100

New insight into the biological activity of Salmo salar NK-lysin antimicrobial peptides.

NK-lysin is a potent antimicrobial peptide (AMP) with antimicrobial activity against bacteria, fungi, viruses, and parasites. NK-lysin is a type of granulysin, a member of the saposin-like proteins family first isolated from a pig's small intestine. In previous work, for the first time, we identified four variants of nk-lysin from Atlantic salmon (Salmo salar) using EST sequences. In the present study, we reported and characterized two additional transcripts of NK-lysin from S. salar. Besides, we evaluated the tissue distribution of three NK-lysins from S. salar and assessed the antimicrobial,

Vertebrate ancient (VA) opsin and extraretinal photoreception in the Atlantic salmon (Salmo salar).

A member of a new photopigment family first isolated from teleost fish, vertebrate ancient (VA) opsin, has recently been shown to form a functional photopigment and to be expressed within a subset of horizontal and amacrine cells of the inner retina. These sites of expression (and structural features) of VA opsin suggest that this photopigment might mediate non-image-forming light-detection tasks. We attempted to gain support for this hypothesis by examining the expression of VA opsin within the central nervous system (CNS) (pineal and deep brain) of the Atlantic salmon Salmo salar. In additio

Ocular Manifestations of Infectious Salmon Anaemia Virus Infection in Atlantic Salmon (Salmo salar).

Eyes are essential sensory organs needed by teleost Atlantic salmon for high visual acuity and survival in both the wild and in aquaculture settings. In this work, we assessed the ocular manifestations of Infectious Salmon Anaemia Virus (ISAV) infection in Atlantic salmon by a cohabitation-mediated infection assay and histological and immunohistochemical approaches. The findings reveal that Atlantic salmon with a systemic ISAV infection displayed ISAV antigen accumulation in specific ocular tissues and significant ocular morphological changes that could compromise visual acuity. Comparative multi-transcriptomic analysis of Atlantic salmon (Salmo salar) under distinct pathogen infections.

Atlantic salmon is among the most widely farmed species globally. The large-scale cultivation of salmon inevitably results in the emergence of diverse diseases, highlighting the critical need to investigate host-pathogen interaction. While numerous transcriptomic analyses have been reported for infected Atlantic salmon, there is a notable lack of comprehensive studies directly comparing transcriptomic responses across different pathogens. Furthermore, differences in bioinformatic methodologies among individual studies present challenges for integrating immunological characteristics in response

Comparative ploidy response to experimental hydrogen peroxide exposure in Atlantic salmon (Salmo salar).

While research into the growth, survival, nutrition and, more recently, disease susceptibility of triploid Atlantic salmon has expanded, there remains an overall lack of studies assessing the response of triploids to chemical treatments. It is essential that the response of triploids to disease treatments be characterised to validate their suitability for commercial production. This study aimed to investigate and compare the stress and immune responses of triploid and diploid Atlantic salmon following an experimental treatment with hydrogen peroxide (H2O2). A dose response test was first under

Multigenic Delineation of Lower Jaw Deformity in Triploid Atlantic Salmon (Salmo salar L.).

Lower jaw deformity (LJD) is a skeletal anomaly affecting farmed triploid Atlantic salmon (Salmo salar L.) which leads to considerable economic losses for industry and has animal welfare implications. The present study employed transcriptome analysis in parallel with real-time qPCR techniques to characterise for the first time the LJD condition in triploid Atlantic salmon juveniles using two independent sample sets: experimentally-sourced salmon (60 g) and commercially produced salmon (100 g). A total of eleven genes, some detected/identified through the transcriptome analysis (fbn2, gal and g

Increased reactivity and monoamine dysregulation following stress in triploid Atlantic salmon (Salmo salar).

Artificial triploid salmonids are sterile and therefore commercially bred to prevent genetic interactions between wild and domestic fish strains. The full biological effects of having an extra chromosome set are largely unknown, but triploids are considered to be more sensitive to sub-optimal environmental conditions and to be stressed by the presence of diploid conspecifics. Brain serotonergic and dopaminergic activity are known to regulate the stress response in vertebrates, but monoamine systems in diploid and triploid fish have yet to be compared. Here we study monoamine neurochemistry in

Water contaminated with Didymosphenia geminata generates changes in Salmo salar spermatozoa activation times.

Didimosphenia geminata ("didymo"), has become a powerful and devastating river plague in Chile. A system was developed in D. geminata channels with the purpose evaluating the effects of water polluted with didymo on the activation of Atlantic salmon (Salmo salar) spermatozoa. Results indicate that semen, when activated with uncontaminated river water had an average time of 60±21s. When using Powermilt, (a commercial activator), times of 240±21s are achieved, while rivers contaminated with D. geminata achieve a motility time of 30±12s. Interestingly enough, the kinetic parameters of VSL, VCL an

Modelling salmon lice, Lepeophtheirus salmonis, reproduction on farmed Atlantic salmon, Salmo salar L.

The aim of this study was to model sea lice levels and the effect on reproduction by a stochastic simulation model and to evaluate the uncertainty of lice estimates based upon counts. Two empirical data sets were examined to parameterize the models. An overall fit of the data to the Poisson distribution was found and thus was used as the base of the stochastic models. In the model, salmon lice reproduction is not linear with the number of adult females and at low lice loads a smaller proportion of the adult female lice will reproduce. Depending on the variance structure, it was estimated that

Transcriptional effects of PFOS in isolated hepatocytes from Atlantic salmon Salmo salar L.

The main aim of the current in vitro experiment was to search for makers for PFOS exposure in isolated hepatocytes from Atlantic salmon Salmo salar, based on genes responding to PFOS exposure in other animals. Primary cell cultures of hepatocytes were exposed to four concentrations of PFOS (2.1-6.2-15.1-25.0 mg/L) for 24 and 48 h and the transcriptional levels of 12 genes encoding proteins known to respond to PFOS were quantified with real-time RT-PCR. The 12 examined genes were caspase 3B (apoptosis), GSH-Px and HSP70 (cellular stress), CYP1A, CYP3A, GST and UGT (P450 and phase II enzymes), a

Paralytic shellfish poisoning toxins induce xenobiotic metabolising enzymes in Atlantic salmon (Salmo salar).

Paralytic shellfish poisoning (PSP) toxins have been implicated as the causative agent of a number of fish kills. Exposure experiments indicate that fish are susceptible to PSPs by intraperitoneal (i.p.) and oral administration, while sampling of fish affected by toxic blooms reveals that these toxins can be accumulated. In spite of the potential impact to marine fisheries, little research has been conducted on the potential metabolism and detoxification of PSPs in marine fishes. Previous work by this group has shown that the xenobiotic metabolising enzyme (XME) cytochrome P-450 (CYP1A) is ind

Effects of repeated anaesthesia on gill and general health of Atlantic salmon, Salmo salar.

Fish are the second most widely utilized vertebrate group used for scientific procedures in the United Kingdom, but the development and application of 3Rs (the principles of replacement, reduction, and refinement) in aquaculture disease research lags behind methodologies in place for mammalian studies. With a need for individual monitoring and non-lethal sampling, the effect of repeat anaesthesia on experimental fish needs to be better understood. This study analyses the effect of repeat anaesthesia with MS-222, metomidate and AQUI-S upon the gill and general health of post-smolt Atlantic salm
