Allergenicity assessment of genetically-modified tobacco expressing salt tolerance cbl gene.

It is mandatory to assess the allergenic potential of genetically modified (GM) crops before their commercialization. Recently, a transgene [Calcineurin B-like (CBL) protein] has been introduced into tobacco plant to make the crop salt resistance. Therefore, it was felt necessary to assess the allergenic potential of the cbl gene product, which was introduced and expressed in Nicotiana tabacum (tobacco) plant and compared the allergenic effects with the wild-type (WT) counterpart. Bioinformatic analysis revealed that there was no significant sequence homology with known allergens. Also, no dif

Molecular mechanisms and regulatory networks of salt stress tolerance in plants.

Soil salinity is a major threat to crop productivity, sustainable agriculture, and global food security, with more than 833 million hectares of land affected worldwide. Salt stress restricts plant growth through osmotic stress, ion toxicity, oxidative damage, membrane disruption, reduced photosynthesis, and yield loss. Plants respond through coordinated regulatory networks that connect early stress perception with ion balance, osmotic adjustment, hormone signaling, transcript regulation, and protein modification. Recent advances have identified several sensory and signaling modules involved in

Populus euphratica PeIDD5 regulates PeNAC029 to suppress salt tolerance in poplars.

Populus euphratica downregulates the transcription factor PeNAC029 during prolonged salinity, thereby promoting long-term acclimation to salt stress. To elucidate this regulatory network, we identified PeIDD5, an INDETERMINATE DOMAIN (IDD) family member, as a direct transcriptional activator of PeNAC029. This study investigates the role of the PeIDD5-NAC029 transcriptional module in the salt tolerance of two contrasting poplars, P. euphratica (salt-tolerant) and P. × canescens (salt-sensitive). PeIDD5 directly binds the conserved cis-element TTTGTCC in the promoter regions of PeNAC029 and PcNA

Mechanism of enhanced salt tolerance in Saccharomyces cerevisiae by CRZ1 overexpression.

Achieving high-gravity fermentation in the industrial production of fuel ethanol, and enhancing the fermentation efficiency of high-salt raw materials, such as waste molasses, can significantly reduce wastewater output and process costs. Therefore, the development of hyperosmotic-tolerant industrial Saccharomyces cerevisiae strains, capable of resisting high-salt stress, offers both environmental and economic benefits. Our previous study highlighted the potential of CRZ1 overexpression as a strategy to improve the yeast strain's resistance to high-salt stress, however, the underlying molecular

RNAi-Mediated Suppression of OsBBTI5 Promotes Salt Stress Tolerance in Rice.

This study explores the impact of RNAi in terms of selectively inhibiting the expression of the OsBBTI5 gene, with the primary objective of uncovering its involvement in the molecular mechanisms associated with salt tolerance in rice. OsBBTI5, belonging to the Bowman-Birk inhibitor (BBI) family gene, is known for its involvement in plant stress responses. The gene was successfully cloned from rice, exhibiting transcriptional self-activation in yeast. A yeast two-hybrid assay confirmed its specific binding to OsAPX2 (an ascorbate peroxidase gene). Transgenic OsBBTI5-RNAi plants displayed insens

Improving salt tolerance of bean (Phaseolus vulgaris L.) with hydrogen sulfide.

The current study examined the H2S applications on growth, biochemical and physiological parameters of bean seedlings under saline conditions. The findings of the study indicated that salt stress decreased plant growth and development, photosynthetic activity, and mineral and hormone content [excluding abscisic acid (ABA)] in bean seedlings. Plant and root fresh mass and dry mass with H2S applications increased as compared to the control treatment at the same salinity level. Both salinity and H2S treatments significantly affected the net assimilation rate, stomatal conductance, transpiration r

Hydrogen sulfide improves salt tolerance through persulfidation of PMA1 in Arabidopsis.

A new interaction was found between PMA1 and GRF4. H2S promotes the interaction through persulfidated Cys446 of PMA1. H2S activates PMA1 to maintain K+/Na+ homeostasis through persulfidation under salt stress. Plasma membrane H+-ATPase (PMA) is a transmembrane transporter responsible for pumping protons, and its contribution to salt resistance is indispensable in plants. Hydrogen sulfide (H2S), a small signaling gas molecule, plays the important roles in facilitating adaptation of plants to salt stress. However, how H2S regulates PMA activity remains largely unclear. Here, we show a possible o

Effects, tolerance mechanisms and management of salt stress in grain legumes.

Salt stress is an ever-present threat to crop yields, especially in countries with irrigated agriculture. Efforts to improve salt tolerance in crop plants are vital for sustainable crop production on marginal lands to ensure future food supplies. Grain legumes are a fascinating group of plants due to their high grain protein contents and ability to fix biological nitrogen. However, the accumulation of excessive salts in soil and the use of saline groundwater are threatening legume production worldwide. Salt stress disturbs photosynthesis and hormonal regulation and causes nutritional imbalance

Overexpression of AmRosea1 Gene Confers Drought and Salt Tolerance in Rice.

Ectopic expression of the MYB transcription factor of AmROSEA1 from Antirrhinum majus has been reported to change anthocyanin and other metabolites in several species. In this study, we found that overexpression of AmRosea1 significantly improved the tolerance of transgenic rice to drought and salinity stresses. Transcriptome analysis revealed that a considerable number of stress-related genes were affected by exogenous AmRosea1 during both drought and salinity stress treatments. These affected genes are involved in stress signal transduction, the hormone signal pathway, ion homeostasis and th

Grafting Onto a Tolerant Rootstock Enhances Grapevine Tolerance to Combined Salt and Drought Stress.

Salt and drought (SD) stress often occur simultaneously, posing severe challenges to grapevine cultivation. Grafting resistant rootstocks is considered a simple and effective method for enhancing grapevine tolerance to environmental stress. This study assessed the performance of different resistant seedlings to SD stress, including the self-rooted seedlings of the SD-sensitive grapevine cultivar 'Shine Muscat' (SM), self-rooted seedlings of the highly SD-tolerant cultivar 'Thompson Seedless' (TS), and grafted seedlings SM/TS. We examined changes in growth, photosynthetic performance, osmotic r

Roles and mechanisms of halophilic/salt-tolerant microorganisms in mitigating plant salt stress and promoting growth.

Soil salinization is a major constraint on global agricultural productivity and food security, largely because excess salts disrupt osmotic balance, nutrient acquisition, ion homeostasis, and redox stability in plants. Halophilic and salt-tolerant microorganisms, including plant growth-promoting rhizobacteria, endophytes, and fungi, have emerged as promising biological tools for improving crop performance in saline environments. This review synthesizes current evidence on how these microorganisms mitigate salt stress and promote plant growth across diverse crop systems. The major mechanisms in

Overexpression of CdtCIPK21 from triploid bermudagrass reduces salt and drought tolerance but increases chilling tolerance in transgenic rice.

Calcineurin B-like-interacting protein kinase (CIPK) is a serine/threonine kinase, which transmits the Ca2+ signal sensed by CBL proteins. A CdtCIPK21 showing highly identical to OsCIPK21 in rice was isolated from triploid bermudagrass (Cynodon dactylon × Cynodon transvaalensis). CdtCIPK21 transcript could be detected in roots, rhizomes, stems, stolons, and leaves, with highest level in roots. It was induced by salinity, dehydration and chilling, but reduced by ABA treatment. Transgenic rice plants overexpressing CdtCIPK21 had decreased salt and drought tolerance as well as ABA sensitivity but

Foliar Spraying of NaHS Alleviates Cucumber Salt Stress by Maintaining N+/K+ Balance and Activating Salt Tolerance Signaling Pathways.

Hydrogen sulfide (H2S) is involved in the regulation of plant salt stress as a potential signaling molecule. This work investigated the effect of H2S on cucumber growth, photosynthesis, antioxidation, ion balance, and other salt tolerance pathways. The plant height, stem diameter, leaf area and photosynthesis of cucumber seedlings were significantly inhibited by 50 mmol·L-1 NaCl. Moreover, NaCl treatment induced superoxide anion (O2·-) and Na+ accumulation and affected the absorption of other mineral ions. On the contrary, exogenous spraying of 200 μmol·L-1 sodium hydrosulfide (NaHS) maintaine

TIME FOR COFFEE coordinates phase separation and chromatin regulation for salt tolerance.

The circadian clock enhances plant fitness by aligning internal timing with the external photoperiod, but how it coordinates spatiotemporal regulation with chromatin dynamics to improve salt tolerance is not well understood. Here, we show that the clock component TIME FOR COFFEE (TIC) forms biomolecular condensates in response to salt stress, but not to osmotic stress alone. Consistently, tic mutants were hypersensitive to salt stress yet displayed enhanced salt tolerance under 21-hour light-dark cycles matching their shortened circadian period, highlighting the importance of circadian clock i

Dual Repression by IPA1 Fine-Tunes OsbZIP79-Mediated Salt Tolerance in Rice.

Soil salinity severely impairs rice yield and quality, yet the molecular mechanisms of its salt tolerance remain unclear. In this study, we first discovered OsbZIP79, a novel basic leucine zipper (bZIP)-type transcription factor, as a positive regulator of salt tolerance in rice. We uncovered that OsbZIP79 positively regulates the expression of OsHAK26 (encoding a potassium transporter) and OsMPG1 (encoding a mannose-1-phosphate guanylyltransferase) to confer salt tolerance in rice. In vivo and in vitro evidence demonstrate that OsbZIP79 physically interacts with Ideal Plant Architecture1 (IPA

Exogenous spermidine (Spd) improves the tolerance to alkaline-salt stress in potato.

BACKGROUND: Potato (Solanum tuberosum L.) is the world's fourth most important staple crop, but its production is increasingly threatened by soil salinization, particularly alkaline-salt stress caused by excessive NaHCO3. Although exogenous spermidine (Spd) has been reported to alleviate abiotic stresses in various plants, its physiological and molecular mechanisms in conferring alkaline-salt tolerance in potato remain largely unknown. RESULTS: The results demonstrated that exogenous Spd enhances alkaline-salt tolerance in potato by increasing antioxidant enzyme activities and maintaining osm

ZmCASP1 is involved in Casparian strip formation and salt tolerance in maize.

Casparian strips (CSs) are lignin-based cell-wall modification located in the vascular plant roots. This structure acts as an impermeable barrier restricting the non-selective apoplastic transport of water and solutes. CS formation represents a highly ordered and tightly regulated biological process in plants. Nevertheless, the molecular mechanisms underlying this process in maize remain largely elusive. In this study, we performed a functional characterization of ZmCASP1 (CASPARIAN STRIP DOMAIN PROTEIN 1) in maize (Zea mays), which is ubiquitously expressed in all three root types. Sequence a
