The second messenger system(s) mediating the secretion of atrial natriuretic peptide (ANP) from the isolated rat heart during rapid cardiac pacing.

This study demonstrates that rapid cardiac pacing elevates Atrial Natriuretic Peptide (ANP) levels, independently from atrial stretch. The second messenger system mediating this response was examined. The phosphoinositide system, generally regarded to be important in mediating ANP release, was shown to play only a modulating role during rapid cardiac pacing. The main mediator would appear to be calcium, and a non-calmodulin dependent, calcium mediated system controlling ANP release during rapid cardiac pacing is suggested.

A review of the possible relevance of inositol and the phosphatidylinositol second messenger system (PI-cycle) to psychiatric disorders--focus on magnetic resonance spectroscopy (MRS) studies.

Myo-inositol is an important part of the phosphatidylinositol second messenger system (PI-cycle). Abnormalities in nerve cell myo-inositol levels and/or PI-cycle regulation has been suggested as being involved in the pathophysiology and/or treatment of many psychiatric disorders including bipolar disorder, major depressive disorder, panic disorder, obsessive-compulsive disorder, eating disorders and schizophrenia. This review examines the metabolism and biochemical importance of myo-inositol and the PI-cycle. It relates this to the current in vivo evidence for myo-inositol and PI-cycle involve

Phosphoinositide second messenger system is enriched in striosomes: immunohistochemical demonstration of inositol 1,4,5-trisphosphate receptors and phospholipase C beta and gamma in primate basal ganglia.

The neurochemical organization of the basal ganglia has been studied extensively with respect to neurotransmitters, neuropeptides, and their receptors. The chemoarchitecture of the striatum has been found particularly striking, because it distinguishes many substances by their relative distributions within the striosome and matrix compartments of the striatum. Very little is yet known about the differential distribution of second messenger systems in the basal ganglia, however, and no information is available about whether the distribution of second messenger systems is related to the prominen

The Panoptes system uses decoy cyclic nucleotides to defend against phage.

Bacteria combat phage infection using antiphage systems and many systems generate nucleotide-derived second messengers upon infection that activate effector proteins to mediate immunity1. Phages respond with counter-defences that deplete these second messengers, leading to an escalating arms race with the host. Here we outline an antiphage system we call Panoptes that indirectly detects phage infection when phage proteins antagonize the nucleotide-derived second-messenger pool. Panoptes is a two-gene operon, optSE, wherein OptS is predicted to synthesize a nucleotide-derived second messenger a

Noradrenaline induces the polyploidization of smooth muscle cells: the synergism of second messengers.

The effect of noradrenaline (NA) on DNA replication of cultured smooth muscle cells (SMC) isolated from rat aorta was examined. It was found that 10 microM NA significantly increased (approximately by twofold) the frequency of tetraploid cells. Cultivation of 4C cells isolated by flow cytometric cell sorting revealed that they were true polyploid cells. This receptor-mediated effect of NA was blocked only by simultaneous action of alpha- and beta-adrenoreceptor antagonists. SMC polyploidization was also stimulated by simultaneous application of direct activators of "second messenger" systems f

Structural mechanism of SAM-AMP and SAM-AMP2 synthesis by the type III-D2 CRISPR effector complex.

The type III-D2 CRISPR-Cas system comprises multiple Cas subunits and a CRISPR RNA, and is likely an evolutionary intermediate between the well-studied type III-A and III-E systems. Here we show that the type III-D2 complex synthesizes two distinct second messengers, SAM-AMP and SAM-AMP2, from S-adenosylmethionine (SAM) and ATP in response to target RNA recognition. We determined cryo-electron microscopy structures of the type III-D2 effector complex in different functional states, providing mechanistic insights into target RNA cleavage and second messenger synthesis. The structures reveal how

Role of messenger system in orthodontic tooth movement.

Various in vivo and in vitro studies have established that arachidonic acid metabolites i.e., prostaglandins and leukotriens play a key role in the conversion of orthodontic pressure stimuli into a cell mediated response. However, considering the different response of osteoblasts and osteoclasts to the same chemical stimuli that leads to the deposition of bone on tension side and resorption on pressure side it has been suggested that it is the messenger system that modulates the behaviour of cells. The messenger system translates a wide array of external stimuli (first messenger) in to a narro

Some Metabolites Act as Second Messengers in Yeast Chronological Aging.

The concentrations of some key metabolic intermediates play essential roles in regulating the longevity of the chronologically aging yeast Saccharomyces cerevisiae. These key metabolites are detected by certain ligand-specific protein sensors that respond to concentration changes of the key metabolites by altering the efficiencies of longevity-defining cellular processes. The concentrations of the key metabolites that affect yeast chronological aging are controlled spatially and temporally. Here, we analyze mechanisms through which the spatiotemporal dynamics of changes in the concentrations o

Injectable hybrid hydrogels enhance macrophage communication via second messenger amplification.

Macrophages play a pivotal role in tissue regeneration and immune regulation through efferocytosis and other mechanisms, with their intracellular communication largely dependent on second messengers (e.g., cyclic adenosine monophosphate, cAMP). However, the broad-spectrum nature of these second messengers renders related interventions susceptible to uncontrollable systemic and off-target effects. Here, we developed an injectable apoptosis-mimicking hybrid hydrogel (PAS@Gel) that enables specific and coordinated macrophage intervention of both first and second messengers involved in efferocytos

Inhibitors of second messenger pathways and Ca(2+)-induced exposure of phosphatidylserine in red blood cells of patients with sickle cell disease.

The present work investigates the contribution of various second messenger systems to Ca(2+)-induced phosphatidylserine (PS) exposure in red blood cells (RBCs) from sickle cell disease (SCD) patients. The Ca(2+) dependence of PS exposure was confirmed using the Ca(2+) ionophore bromo-A23187 to clamp intracellular Ca(2+) over 4 orders of magnitude in high or low potassium-containing (HK or LK) saline. The percentage of RBCs showing PS exposure was significantly increased in LK over HK saline. This effect was reduced by the Gardos channel inhibitors, clotrimazole and charybdotoxin. Nevertheless,

Free radicals in inflammation: second messengers and mediators of tissue destruction.

In recent years it has become increasingly apparent that, in man, free radicals play a role in a variety of normal regulatory systems, the deregulation of which may play an important role in inflammation. As examples, we discuss the second messenger roles of: NO in the regulation of vascular tone, O2.- in fibroblast proliferation and H2O2 in the activation of transcription factors such as NF kappa B. Other control mechanisms, the physiological function of which may be perturbed in inflammation, include: the oxidative modification of low density lipoprotein, the oxidative inactivation of alpha-

Hepatocanalicular organic-anion transport is regulated by protein kinase C.

In order to investigate the regulation of canalicular organic-anion transport, we used a hepatocyte transport assay in which canalicular secretion of a model organic anion, dinitrophenyl-glutathione (GS-DNP), was measured in the presence of stimulators and inhibitors of the Ca2+/protein kinase C (PKC) second-messenger system and of the cyclic AMP (cAMP) second-messenger system. Vasopressin (24 nM) and the phorbol ester phorbol 12-myristate 13-acetate (1 microgram/ml), both stimulators of PKC, stimulated GS-DNP efflux by 65 +/- 36% and 55 +/- 28% respectively, whereas staurosporine (10 microM),

Proliferative response of smooth muscle cells in hypertension.

A significant increase (up to 20% from about 10% in normals) in the number of smooth muscle cells (SMC) with tetraploid DNA content was found in the media and intima of human hypertensive aorta. A similar process was detected during normal human vessel aging. It was found that SMC from normal human aorta and normotensive rat aorta, which were able to incorporate 3H-thymidine, had diminished proliferative potency and a tendency to polyploidization in primary culture. We failed to detect a similar phenomenon in SMC obtained from aorta from spontaneously hypertensive rats. It was found that 10 mu

Host-derived stimuli alter intracellular calcium levels in Pseudomonas aeruginosa, revealing its potential role as a second messenger.

Calcium signaling is a fundamental cellular communication mechanism, well established in eukaryotes, that relies on calcium ions (Ca2+) acting both as first and second messengers to regulate diverse life-sustaining processes. In bacteria, although the significance of Ca2+ regulation is recognized, it is commonly attributed to its role as a first messenger, while the mechanistic basis for its capacity as a second messenger remains elusive. Previously, we have shown that Pseudomonas aeruginosa, an opportunistic pathogen causing life-threatening infections in humans, possesses the key prerequisit

[Proliferation of smooth muscle cells in hypertension].

The aortic intima and media isolated from hypertensives showed a significantly larger number (up to 20%) of smooth muscle cells (SMC) with tetraploid DNA content. The similar process was shown to be also a part of normal human vessel maturation. Normotensive human and rat aortic SMCs were found to accumulate 3H-thymidine, have a lower proliferative ability and they were apt to polyploidize in the primary culture. Such a population could not be detected in the aorta of spontaneously hypertensive rats. It was ascertained that 10 microM of noradrenaline significantly increased (approximately by 2

Chemical control of growth and morphological characteristics of anaplastic glioma cells.

Nerve growth factor (NGF) and glia maturation factor (GMF) reverse some of the transformed characteristics of T9 glioma cells (Marushige et al., Cancer Res 47: 4109-4115, 1987). As an attempt to define the mechanisms of such actions, various chemical agents which modulate second messenger systems were examined for their effects on growth and morphological characteristics of these cells. Administration of bromo-cAMP, forskolin and methylisobutylxanthine retarded cell growth and induced formation of long, branching processes which were similar to those induced by GMF. Perturbation of Ca2(+)-medi

NAADP as a second messenger: neither CD38 nor base-exchange reaction are necessary for in vivo generation of NAADP in myometrial cells.

Nicotinic acid adenine dinucleotide phosphate (NAADP) has recently been shown to act as a second messenger controlling intracellular Ca(2+) responses in mammalian cells. Many questions remain regarding this signaling pathway, including the role of the ryanodine receptor (RyR) in NAADP-induced Ca(2+) transients. Furthermore, the exact metabolic pathway responsible for the synthesis of NAADP in vivo has not been determined. Here, we demonstrate that the NAADP mediated Ca(2+) release system is present in human myometrial cells. We also demonstrate that human myometrial cells use the NAADP second

Clinically accessible cell signaling: second messengers in sepsis and trauma.

Inflammatory mediators of trauma and sepsis transduce cellular events through cell surface receptors initiating intricate membrane and cytosolic reaction cascades that funnel through surprisingly few checkpoints in order to provoke a cellular response. As critical care surgeons, we can explore these cell signalling systems. The purpose of this article is to delineate the six known second messenger pathways relevant to surgical sepsis and trauma. Our comprehension of these signaling systems may offer us an opportunity to blunt post-traumatic cellular injury and promote a constructive response.
