Fire as a regeneration filter in the Arctic: effects of heat and smoke on seed germination in 25 common plant species.

BACKGROUND AND AIMS: The rapid warming of the Arctic is increasing the frequency, intensity, and spatial extent of fires. Because fire has historically been rare in this region, most Arctic plant species are unlikely to have evolved traits that confer tolerance to fire, and the consequences for early life-history stages such as seed germination remain largely unknown. Here, we experimentally tested the effects of two key fire-related cues, heat shock and smoke exposure, on the germination traits of 25 widespread Arctic plant species. METHODS: Seeds collected across the Arctic were subjected t

Lipidomics of orchid seeds: from development to storage.

Orchid seeds primarily contain lipid reserves, yet details of their deposition, in quantity and quality, remain unknown. Deposition begins as carbohydrates, which are eventually converted into fatty acids during seed development. This research aimed to dissect the pattern of lipid deposition in seeds of Dactylorhiza fuchsii and Disa uniflora, from pollination to seed storage, including at five stages of seed maturation, germination capacity and relationships among fatty acids (FAs). The capsules were harvested, cleaned and measured. Extracted seeds were analysed using differential scanning cal

Getting by With a Little Help From Fungal Friends: Seed-Associated Endophytes Contribute to Seed Defense Against Fungal Pathogens in Keystone Tropical Trees.

Soil-borne fungal pathogens are known to be major causes of seed mortality in tropical forests; however, not all fungal infections of seeds are necessarily lethal. It has been hypothesized that seed-associated fungi may contribute to seed defense and persistence in soil seed banks and are especially important for non-dormant seeds with limited physical and chemical defenses. This study examined microbially mediated seed defense in two fig (Ficus) species from a tropical forest in the Western Ghats, India. One species, Ficus callosa has a free-standing habit (seeds germinate in the soil), and t

Advances Towards Ex Situ Conservation of Critically Endangered Rhodomyrtus psidioides (Myrtaceae).

Rhodomyrtus psidioides (G.Don) Benth. (Myrtaceae) is a critically endangered rainforest species from the east coast of Australia, where populations have severely and rapidly declined due to the effects of repeated myrtle rust infection. With very limited material available in the wild and freezing-sensitive seeds that have prevented storage in a seed bank, ex situ conservation of this exceptional species has proven difficult. Material from a seed orchard grown at the Australian Botanic Garden Mount Annan was successfully used to initiate three new accessions into tissue culture from cuttings,

Humidity and temperature interact in breaking physical dormancy in Fabaceae seeds from the Brazilian semiarid region.

PREMISE: In semiarid ecosystems such as the Brazilian Caatinga, physical seed dormancy (PY) is critical for plant persistence when water is unpredictable, but how soil moisture and temperature regulate PY release and seed longevity in species of native Fabaceae that have different growth forms is unknown. METHODS: Seeds of the herbaceous species Centrosema pascuorum and Crotalaria retusa and tree species Mimosa ophthalmocentra, M. tenuiflora, and Piptadenia retusa were collected in the Caatinga and tested in the field and laboratory to assess dormancy, viability, and germination. Seeds were e

FinTech and financial instability. Is this time different?

We study the development of FinTech, defined as a set of innovations and an economic sector that apply recently developed digital technologies to financial services, with particular focus on payment and lending platforms, and digital asset management and online trading apps. We use mixed methods, including a theoretical exercise on the main balance sheet interactions involved in FinTech banking, and empirical insights from fieldwork in Latin America and the United States. Our analysis corroborates previous literature identifying several systemic risks in FinTech payment and lending platforms.

Sensitive protein misfolding cyclic amplification of sporadic Creutzfeldt-Jakob disease prions is strongly seed and substrate dependent.

Unlike variant Creutzfeldt-Jakob disease prions, sporadic Creutzfeldt-Jakob disease prions have been shown to be difficult to amplify in vitro by protein misfolding cyclic amplification (PMCA). We assessed PMCA of pathological prion protein (PrPTSE) from 14 human sCJD brain samples in 3 substrates: 2 from transgenic mice expressing human prion protein (PrP) with either methionine (M) or valine (V) at position 129, and 1 from bank voles. Brain extracts representing the 5 major clinicopathological sCJD subtypes (MM1/MV1, MM2, MV2, VV1, and VV2) all triggered seeded PrPTSE amplification during se

Genetic and neuroattenuation phenotypic characteristics and their stabilities of SA14-14-2 vaccine seed virus.

Japanese encephalitis (JE) live attenuated vaccine SA14-14-2 is the most widely used JE vaccine in the world. Large-scale clinical trials have demonstrated satisfactory safety and efficacy profiles. The establishment of genetic and attenuated neurovirulence characteristics and their stabilities of SA14-14-2 virus are important in relation to vaccine safety in humans. Therefore, several researchers have studied and analyzed the full-length gene sequences of the SA14-14-2 virus strain. However, sequencing results have shown a significant difference. Here, we further studied the full-length seque

Intrinsic functional clustering of ventral premotor F5 in the macaque brain.

Neurophysiological and anatomical data suggest the existence of several functionally distinct regions in the lower arcuate sulcus and adjacent postarcuate convexity of the macaque monkey. Ventral premotor F5c lies on the postarcuate convexity and consists of a dorsal hand-related and ventral mouth-related field. The posterior bank of the lower arcuate contains two additional premotor F5 subfields at different anterior-posterior levels, F5a and F5p. Anterior to F5a, area 44 has been described as a dysgranular zone occupying the deepest part of the fundus of the inferior arcuate. Finally, area G

In situ seed storage affects germination responses to fire-related cues in Cerrado species.

BACKGROUND AND AIMS: Fire-related cues can influence germination and seedling establishment in fire-prone ecosystems by breaking physical dormancy (PY) or stimulating germination in species with water-permeable seeds. However, most studies rely on fresh seeds and overlook how soil seed bank storage may change germination responses to fire cues. We investigated whether in situ seed storage affects germination responses to heat shock and smoke in Cerrado species. METHODS: Seeds of 13 Cerrado species, dispersed at the beginning of either the dry or rainy season, were collected and exposed to hea

Preparation and immune efficacy evaluation of inactivated lumpy skin disease virus vaccine.

Lumpy Skin Disease (LSD), historically restricted to Africa, has in recent years exhibited a marked expansion into several Asian and European countries, resulting in substantial economic impacts on the global cattle industry. In August 2019, the first confirmed outbreak of LSD was documented in the Xinjiang region of China. In response to this emerging threat, our study established primary, basic, and working seed banks utilizing the LSDV/MZGD/2020 strain, which was isolated from Meizhou, Guangdong. These seed banks underwent rigorous validation via sterility assays and evaluation of prolifera

Characterization of tau prion seeding activity and strains from formaldehyde-fixed tissue.

Tauopathies such as Alzheimer's disease (AD) feature progressive intraneuronal deposition of aggregated tau protein. The cause is unknown, but in experimental systems trans-cellular propagation of tau pathology resembles prion pathogenesis. Tau aggregate inoculation into mice produces transmissible pathology, and tau forms distinct strains, i.e. conformers that faithfully replicate and create predictable patterns of pathology in vivo. The prion model predicts that tau seed formation will anticipate neurofibrillary tau pathology. To test this idea requires simultaneous assessment of seed titer

Molecular aspects of the antagonistic interaction of smoke-derived butenolides on the germination process of Grand Rapids lettuce (Lactuca sativa) achenes.

Smoke-derived compounds provide a strong chemical signal to seeds in the soil seed bank, allowing them to take advantage of the germination niche created by the occurrence of fire. The germination stimulatory activity of smoke can largely be attributed to karrikinolide (KAR(1) ), while a related compound, trimethylbutenolide (TMB), has been shown to have an inhibitory effect on germination. The aim of this study was to characterize the interaction of these potent fire-generated compounds. Dose-response analysis, leaching tests and a detailed transcriptome study were performed using highly KAR(

Strigolactones: how far is their commercial use for agricultural purposes?

Strigolactones are a class of natural and synthetic compounds that in the past decade have been exciting the scientific community not only for their intriguing biological properties but also for their potential applications in agriculture. These applications range from their use as hormones to modify and/or manage plant architecture, to their use as stimulants to induce seed germination of parasitic weeds and thus control their infestation by a reduced seed bank, to their use as 'biostimulants' of plant root colonisation by arbuscular mycorrhizal fungi, improving plant nutritional capabilities

Development of germplasm repositories to assist conservation of endangered fishes: Examples from small-bodied livebearing fishes.

Germplasm repositories are a necessary tool for comprehensive conservation programs to fully preserve valuable genetic resources of imperiled animals. Cryopreserved germplasm can be used in the future to produce live young for integration into other conservation projects, such as habitat restoration, captive breeding, and translocations; thus compensating for genetic losses or negative changes that would otherwise be permanent. Although hundreds of cryopreservation protocols for various aquatic species have been published, there are great difficulties in moving such research forward into appli

Prion-like transmission and propagation of human β-amyloid to the bank vole rodent model.

Over the past decades, growing experimental and observational evidence has suggested that Aβ and pTau, the hallmarks of Alzheimer's disease (AD), may spread through the nervous system via a prion-like mechanism. Here, we investigated the transmissibility of Aβ and pTau by inoculating bank voles, a wild-type rodent highly susceptible to prion diseases, with brain homogenates from four sporadic and five familial AD-affected patients. We observed that (i) neo-formed Aβ deposits and pTau inclusions were induced in recipient vole brains; (ii) Aβ pathology appeared to follow a specific neurotropic d

Varietal diversity and conservation status of banana, taro, pumpkin, and mustard green in mountainous areas of Northwest Vietnam.

This study assesses the agrobiodiversity richness, traditional knowledge, and conservation status associated with four key crops-banana, taro, pumpkin, and mustard green-in Northwest Vietnam. Using the 5-cell method, 133 farmer varieties were identified in Mai Son and Sa Pa districts. Mai Son showed the highest level of varietal diversity in banana and pumpkin, while Sa Pa had the higher richness in mustard green and taro. Ethnic groups, particularly Thai and Dao households, played a key role in maintaining this diversity. The findings reveal significant vulnerability to varietal loss, particu
