OBJECTIVE: Studies have found elevation in a key biomarker of aging (the senescence-associated secretory phenotype (SASP) index) in depression. We investigated whether sex moderates the association between the SASP and major depression in older adults.
METHODS: We included 423 older adults in a current major depressive episode and 140 adults with no history of depression. We measured the plasma levels of SASP biomarkers using multiplex immunoassays. The interaction effect between sex, depression diagnosis, and senescence markers were analyzed by general linear models, adjusted for confounding

Orthopedic degenerative diseases, including osteoarthritis (OA), intervertebral disc degeneration (IVDD), and osteoporosis (OP), are major causes of chronic pain and functional decline in aging populations worldwide. The senescence-associated secretory phenotype (SASP), a downstream but central effector of cellular senescence, has emerged as a key pathogenic mediator that links senescent cell accumulation to tissue degeneration in the musculoskeletal system. Comprising pro-inflammatory cytokines, chemokines, matrix-degrading enzymes, growth factors, and extracellular vesicle-associated signals

Bacterial genotoxins damage host cells by targeting their chromosomal DNA. In the present study, we demonstrate that a genotoxin of Salmonella Typhi, typhoid toxin, triggers the senescence-associated secretory phenotype (SASP) by damaging mitochondrial DNA. The actions of typhoid toxin disrupt mitochondrial DNA integrity, leading to mitochondrial dysfunction and disturbance of redox homeostasis. Consequently, it facilitates the release of damaged mitochondrial DNA into the cytosol, activating type I interferon via the cGAS-STING pathway. We also reveal that the GCN2-mediated integrated stress

BACKGROUND: Liver cancer is one of the most common malignancies in the world with a poor prognosis. Hepatocellular carcinoma (HCC) is the most prevalent primary liver cancer, accounting for 80-90% of cases. The initiation and progression of HCC are closely associated with chronic liver inflammation. In addition, HCC is often accompanied by cell senescence. Senescent hepatocytes can secrete various inflammatory factors, collectively called the senescence-associated secretory phenotype (SASP). The SASP has been confirmed to promote the occurrence of liver cancer by affecting the inflammatory mic

Cerebral cavernous malformation (CCM) is a cerebrovascular disease in which stacks of dilated haemorrhagic capillaries form focally in the brain. Whether and how defective mechanotransduction, cellular mosaicism and inflammation interplay to sustain the progression of CCM disease is unknown. Here, we reveal that CCM1- and CCM2-silenced endothelial cells expanded in vitro enter into senescence-associated secretory phenotype (SASP) that they use to invade the extracellular matrix and attract surrounding wild-type endothelial and immune cells. Further, we demonstrate that this SASP is driven by t

Epidemiological evidence links environmental pollution to chronic kidney disease (CKD). However, the molecular mechanisms by which diesel-derived pollutants trigger and sustain renal fibrosis are still unclear. In this study, we show that 1-aminopyrene (1-AP), a metabolite of diesel exhaust, leads to genomic instability and renal tubular epithelial cell (RTEC) senescence. Exposure of RTECs to sub-cytotoxic concentrations of 1-AP caused persistent DNA damage, as shown by comet assay and γ-H2AX foci. Sustained DNA damage activated the p21-mediated cellular senescence, followed by induction of SA

Thoracic aortic aneurysm (TAA) is an age-related and life-threatening vascular disease. Telomere shortening is a predictor of age-related diseases, and its progression is associated with premature vascular disease. The aim of the present work was to investigate the impacts of chronic hypoxia and telomeric DNA damage on cellular homeostasis and vascular degeneration of TAA. We analyzed healthy and aortic aneurysm specimens (215 samples) for telomere length (TL), chronic DNA damage, and resulting changes in cellular homeostasis, focusing on senescence and apoptosis. Compared with healthy thoraci

BACKGROUND: IGSF9, immunoglobulin superfamily member 9, has been reported to inhibit T cell proliferation and activation, thereby promoting tumor immune escape. Tumor-associated macrophages (TAMs), the most abundant tumor-infiltrating immune cells, play a crucial role in forming the immunosuppressive tumor microenvironment. We find that IGSF9 strongly binds to TAMs, however, how it affects TAMs function remains unreported.
METHODS: The spatial transcriptomics dataset (GSE189487) was analyzed to clarify the relationship among IGSF9, senescent TAMs, and T cells. RNA-seq revealed differentially

BACKGROUND: Pancreatic cancer (PC) exhibits an extremely poor prognosis due to its high heterogeneity. The senescence-associated secretory phenotype (SASP), a distinct secretory profile displayed by senescent cells, has been increasingly studied. However, the role of SASP in PC prognosis and treatment remains unclear.
METHODS: Transcriptomic sequencing data from PC patients were analyzed using consensus clustering based on SASP genes. A prognostic signature was subsequently constructed via Least Absolute Shrinkage and Selection Operator (LASSO) regression using survival-related SASP genes. Pa

Cellular senescence is a central driver of organismal ageing and related pathologies, primarily promoting chronic inflammation and tissue dysfunction through the senescence-associated secretory phenotype (SASP). Unlike senolytics, which aim to eliminate senescent cells, senomorphic agents offer a complementary therapeutic strategy by modulating the SASP without clearing the cells, thereby preserving their potential physiological functions. This review systematically elucidates the multi-target mechanisms of senomorphic agents, including the inhibition of key signaling pathways such as NF-κB, m

The male reproductive system experiences degradation with age, predominantly impacting the testes. Testicular aging can result in failure to produce physiological testosterone levels, normal sperm concentrations, or both. However, we cannot predict the onset of testicular aging in advance. Using single-cell RNA sequencing (scRNA-seq) from Gene Expression Omnibus (GEO) database, we conducted cell-cell communication network of human testis between older and young group, indicating Leydig cells' potential role in spermatogenesis microenvironment of aging testis. And we depicted the senescence-Ass

Cellular senescence is closely linked to endothelial dysfunction, a key factor in age-related vascular diseases. Senescent endothelial cells exhibit a proinflammatory phenotype known as SASP, leading to chronic inflammation (inflammaging) and vascular impairments. Albeit in a state of permanent growth arrest, senescent cells paradoxically display a high metabolic activity. The relationship between metabolism and inflammation is complex and varies across cell types and senescence inductions. While some cell types shift towards glycolysis during senescence, others favor oxidative phosphorylation

Cellular senescence has been considered a state of irreversible growth arrest upon exhaustion of proliferative capacity or exposure to various stresses. Recent studies have extended the role of cellular senescence to various physiological processes, including development, wound healing, immune surveillance, and age-related tissue dysfunction. Although cell cycle arrest is a critical hallmark of cellular senescence, an increased intracellular reactive oxygen species (ROS) production has also been demonstrated to play an important role in the induction of cellular senescence. In addition, recent

BACKGROUND: Prostate cancer develops through malignant transformation of the prostate epithelium in a stepwise, mutation-driven process. Although activator protein-1 transcription factors such as JUN have been implicated as potential oncogenic drivers, the molecular programs contributing to prostate cancer progression are not fully understood.
METHODS: We analyzed JUN expression in clinical prostate cancer samples across different stages and investigated its functional role in a Pten-deficient mouse model. We performed histopathological examinations, transcriptomic analyses and explored the s

Intratumoral heterogeneity in metastatic cancers complicates effective treatment, as distinct tumor subclones display varying drug sensitivities and metastatic capabilities, interacting through mechanisms that remain poorly understood. In this study, we utilized an isogenic ovarian cancer cell pair distinguished by differential β-catenin signaling: non-metastatic (NM) cells with low β-catenin and highly metastatic (HM) cells with elevated β-catenin signaling. Co-engrafting NM and HM cells synergistically enhances peritoneal metastasis compared to either subclone alone. Immunohistochemical anal

Uterine fibroids (or uterine leiomyoma, UFs) are one of the most prevalent benign uterine tumors with high proliferation and collagen synthesis capabilities. UFs are a significant worldwide health issue for women, affecting their physical and financial well-being. Risk factors for UFs include age, racial disparities, obesity, uterine infections, hormonal variation, and lifestyle (i.e., diet, exercise, stress, and smoking). Senescence and its associated secretory phenotypes (SASPs) are among the most salient changes accompanying the aging process. As a result, SASPs are suggested to be one of t

Intervertebral disc degeneration is an important contributor to chronic low back and neck pain. Although many environmental and genetic factors are known to contribute to disc degeneration, age is still the most significant risk factor. Recent studies have shown that senescence may play a role in age-related disc degeneration and matrix catabolism in humans and mouse models. Clearance of p16Ink4a-positive senescent cells reduces the degenerative phenotype in many age-associated diseases. Whether p16Ink4a plays a functional role in intervertebral disc degeneration and senescence is unknown. We

Periodontitis (PD) is a chronic inflammatory disorder marked by immune dysregulation and progressive tissue destruction. Cellular senescence and the senescence-associated secretory phenotype (SASP) have been increasingly recognized as pivotal drivers of chronic inflammation. However, their specific contributions to PD remain insufficiently clarified. In this study, integrative bioinformatic analyses were conducted across transcriptomic datasets, employing least absolute shrinkage and selection operator, support vector machine-recursive feature elimination, and eXtreme gradient boosting algorit

Chronic low-grade inflammation in gestational diabetes mellitus is one of the significant causes of placental dysfunction. However, the mechanisms by which abnormal glucose metabolism affects inflammation are not fully understood. p66Shc serves as a redox enzyme to regulate reactive oxygen species in mitochondria. Herein, we explore the interaction between p66Shc and OGT. High glucose induces O-GlcNAcylation of p66Shc, which promotes its protein stability and mitochondrial translocation, which induced elevated cytoplasmic mtDNA release and activation of the cGAS/STING to facilitate cellular se
