Structure identification and biological activity of a novel sulfated glycosaminoglycan-like polysaccharide from Sepia recurvirostra ink.

A novel sulfated glycosaminoglycan-like polysaccharide (SIP), with a molecular weight of 13.78 kDa, was isolated from Sepia recurvirostra ink. SIP displayed a sulfate content of 6.82 % and consisted of GlcNAc, GalNAc, GlcA and Fuc. The distinct structural sequence of SIP was identified by 1D/2D NMR assignments of the backbone released by deaminative treatment and the oligosaccharides generated by mild acid hydrolysis. The backbone was built up of disaccharide repeating blocks structured as [-D-GlcA-β1,4-L-Fuc-α1,3-]. Notably, the non-sulfated GalNAc was attached to C-3 of Fuc, while the 4, 6-O

Preparation of Phosphorylated Chitosan From the Cuttlebone of Sepia pharaonis and the Effect of Concentration on Oral Clinical Pathogens.

BACKGROUND: Chitosan, a biopolymer derived from chitin, has attracted scholarly interest because of its antibacterial, biocompatible, and biodegradable characteristics. We can phosphorylate the cuttlebone of Sepia pharaonis, a natural source of chitin, to enhance its antimicrobial properties. Phosphorylated chitosan is promising for treating oral infections, which are the causative agents of a variety of dental disorders. OBJECTIVES: The goal of this study is to look into how to make phosphorylated chitosan from cuttlebone and what effect different concentrations have on killing oral clinical

Reversion to developmental pathways underlies rapid arm regeneration in juvenile European cuttlefish, Sepia officinalis (Linnaeus 1758).

Coleoid cephalopods, including the European cuttlefish (Sepia officinalis), possess the remarkable ability to fully regenerate an amputated arm with no apparent fibrosis or loss of function. In model organisms, regeneration usually occurs as the induction of proliferation in differentiated cells. In rare circumstances, regeneration can be the product of naïve progenitor cells proliferating and differentiating de novo . In any instance, the immune system is an important factor in the induction of the regenerative response. Although the wound response is well-characterized, little is known about

Variability in the "stereotyped" prey capture sequence of male cuttlefish (Sepia officinalis) could relate to personality differences.

Studies of animal personality have shown consistent between-individual variation in behaviour in many social and non-social contexts, but hunting behaviour has been overlooked. Prey capture sequences, especially in invertebrates, are supposed to be quite invariant. In cuttlefish, the attack includes three components: attention, positioning, and seizure. The previous studies indicated some variability in these components and we quantified it under the hypothesis that it could relate to personality differences. We, therefore, analysed predation sequences of adult cuttlefish to test their associa

Phosphorylated chitosan from the internal bone of Sepia kobiensis (Hoyle, 1885) and their inhibition against oral pathogens.

Phosphorylated chitosan, derived from the internal bone of Sepia kobiensis (Hoyle, 1885), has been investigated for its antimicrobial properties against oral pathogens. Chitosan's solubility in water and its biological effectiveness were enhanced through the alteration of its composition with orthophosphoric acid and dimethylformamide (DMF). Structural analysis for both chitosan and phosphorylated chitosan was performed using FTIR spectroscopy, field emission scanning electron microscopy (FESEM), and X-ray diffraction (XRD). The antimicrobial efficacy of chitosan and phosphorylated chitosan wa

Cytomorphological aspects on the response of the branchial heart complex of Sepia officinalis L. (cephalopoda) to xenobiotics and bacterial infection.

The ovoid cells of the branchial heart complex of Sepia officinalis L. were investigated with respect to their role in detoxification processes. The electron microscopical localization of in situ injected ferritin in the endocytotic-lysosomal system, and the fluorescence microscopical localization of protein-bound Evans blue in the ovoid cells of in vivo incubated animals, indicate that foreign materials are eliminated from the hemolymph by the branchial heart tissue. In addition to the non-circulating ovoid cells of the branchial heart, hemocytes in the circulating blood and in the wall of th

Relations between total mercury, methylmercury and selenium in five tissues of Sepia officinalis captured in the south Portuguese coast.

Mercury, methylmercury and selenium were determined in digestive gland, branchial hearts, mantle, kidney and gills of Sepia officinalis from two areas of the south Portuguese coast. To the best of our knowledge these are the first data on Hg, MeHg and Se in branchial hearts, kidney and gills of cuttlefish. Digestive gland, branchial hearts and kidney presented higher levels of Hg and Se than mantle and gills. Methylmercury was significantly higher in digestive gland, branchial hearts and mantle. The enhanced levels of Hg in digestive gland and branchial heart reinforce the elevated storage cap

In vitro evidence and age-related changes for nicotinic but not muscarinic acetylcholine receptors in the central nervous system of Sepia officinalis.

Binding putative muscarinic ([3H]-NMS and [3H]-QNB) or nicotinic ([3H]-cytisine) acetylcholine receptors was quantitatively studied through the use of in vitro binding experiments on either membrane preparations or brain sections of juvenile (3 months), mature (15 months) or senescent (23 months) cuttlefish. No specific binding could be detected with muscarinic receptor ligands under any of the experimental conditions employed (ligand concentrations, buffers, ionic charges, types of tissue, i.e., brain sections or membrane preparations). On the other hand, [3H]-cytisine demonstrated a specific

Accumulation, transformation and tissue distribution of domoic acid, the amnesic shellfish poisoning toxin, in the common cuttlefish, Sepia officinalis.

Domoic acid (DA) is a phycotoxin produced by some diatoms, mainly from the Pseudo-nitzschia genus, and has been detected throughout the marine food web. Although DA has been frequently found in cephalopod prey such as crustaceans and fish, little is known about DA accumulation in these molluscs. This study presents the first data showing relevant concentrations of DA detected in the common cuttlefish, Sepia officinalis, which is one of the most studied cephalopod species in the world. Domoic acid was consistently found throughout 2003 and 2004 in the digestive gland of cuttlefish reaching conc

Size Matters: Observed and Modeled Camouflage Response of European Cuttlefish (Sepia officinalis) to Different Substrate Patch Sizes during Movement.

Camouflage is common throughout the phylogenetic tree and is largely used to minimize detection by predator or prey. Cephalopods, and in particular Sepia officinalis cuttlefish, are common models for camouflage studies. Predator avoidance behavior is particularly important in this group of soft-bodied animals that lack significant physical defenses. While previous studies have suggested that immobile cephalopods selectively camouflage to objects in their immediate surroundings, the camouflage characteristics of cuttlefish during movement are largely unknown. In a heterogenic environment, the v

First report of adults of Acanthocephaloides irregularis from cuttlefish Sepia officinalis (Cephalopoda) in Tunisia: SEM observations and molecular analysis.

During a parasitological survey of the common cuttlefish Sepia officinalis (Linnaeus) collected from the Bizerte Lagoon in northern Tunisia, adult acanthocephalans were found attached to the internal wall of cuttlefish stomach. The presence of adult acanthocephalans in cephalopods is unusual, given that definitive hosts for these obligate endoparasites are typically vertebrates. A detailed morphological examination of both sexes using light and scanning electron microscopy revealed high morphological similarities to Acanthocephaloides irregularis Amin, Oğuz, Heckmann, Tepe et Kvach, 2011. 1H NMR-based metabolomic analysis of cuttlefish, Sepia pharaonis (Ehrenberg, 1831) exposed to hypoxia stresses and post-anoxia recovery.

Oxygen deficiency (hypoxia and anoxia) is an emerging concern in estuarine and coastal ecosystems worldwide. Previous studies on Mollusca Cephalopoda have focused on the effects of hypoxia stress on physiological performance and survival, but there are few reports on the molecular mechanism, and the application of metabolomics in cephalopods remains unknown. In this study, a 1H nuclear magnetic resonance (NMR) based metabolomics approach was applied to investigate the metabolites profiles of Sepia pharaonis (Ehrenberg, 1831) during hypoxia and post-anoxia recovery. The results revealed that ob

Neuroprotective Effect of Taurine-Rich Cuttlefish (Sepia officinalis) Extract Against Hydrogen Peroxide-Induced Oxidative Stress in SH-SY5Y Cells.

Oxidative stress mediates the cell damage in several neurodegenerative diseases, some of which are Alzheimer's disease (AD), multiple sclerosis and Parkinson's disease (PD). In this study, we investigated whether the taurine-rich cuttlefish extract could exert a protective effect on damaged human neuroblastoma SH-SY5Y cells induced by hydrogen peroxide (H2O2). Our results revealed that pre-treatment with cuttlefish extract effectively increased the cell viability by protecting the cells from intracellular reactive oxygen species (ROS) induced by H2O2 exposure. Furthermore, apoptosis related pr

Extensive population subdivision of the cuttlefish Sepia officinalis (Mollusca: Cephalopoda) around the Iberian Peninsula indicated by microsatellite DNA variation.

The Atlantic Ocean-Mediterranean Sea junction has been proposed as an important phylogeographical area on the basis of concordance in genetic patterns observed at allozyme, mtDNA and microsatellite DNA markers in several marine species. This study presents microsatellite DNA data for a mobile invertebrate species in this area, the cuttlefish Sepia officinalis, allowing comparison of this relatively new class of DNA marker with previous allozyme results, and examination of the relative effects on gene flow of the Strait of Gibraltar and the Almería-Oran oceanographic front.
