Incidental isolation of Setaria equina microfilariae in preparations of equine peripheral blood mononuclear cells.

In the course of a vaccine experiment on horses, microfilariae were observed in cultures of peripheral blood mononuclear cells (PBMCs) isolated from eleven of fifteen study horses. The microfilariae were clearly viable as evidenced by their vigorous movements in the cultures, thus indicating that they had survived the Ficoll gradient purification and the cryopreservation method used for retaining the PBMCs. The microfilariae were identified as Setaria equina, which is a vector-borne filarial nematode that causes a relatively benign infection of equids in which the adult worms reside in the per

First report of the isolation and phylogenetic characterization of equine Setaria digitata from India based on mitochondrial COI, 12S rDNA, and nuclear ITS2 sequence data.

Equine ocular setariasis arising mainly from ectopic infestation of Setaria digitata is a common vision impairing ophthalmic disease in India, and the identification of this filarial nematode is based solely on morphology. However, morphological characters alone are inadequate to detect and differentiate S. digitata from its congeners. The present communication reports the first phylogenetic characterization of equine S. digitata from India based on sequences derived from the mitochondrial cytochrome c oxidase subunit 1 (COI), the mitochondrial small subunit ribosomal DNA (12S rDNA), and the n

Changes in the level of biogenic amines in experimentally infected and DEC treated proxy host mice.

The levels of biogenic amines, viz., epinephrine, histamine and dopamine were significantly altered in the blood and brain of the proxy host mice which were infected with the mf of Setaria digitata, a filarial parasite. When DEC, the drug of choice was administered to the infected mice, serotonine level was enhanced while the histamine level was decreased. The results were similar in normal mice which were given DEC.

Setaria labiatopapillosa (Filarioidea, Nematoda) in Moroccan cattle: atypical localization and morphological characterization of females and microfilariae by light and scanning electron microscopy.

Filarioid nematodes are parasites of the tissues and tissue spaces of all vertebrates except fish. Females produce microfilariae that enter the host's blood circulation or skin and may cause ocular and neurological pathology, leading to important implications in veterinary and public health. The present work is the first investigation on Setaria labiatopapillosa conducted in Morocco to characterize the morphological features of both adult and microfilaria forms. Two adult female nematodes were found free in the thoracic cavity of a slaughtered 3.5-year-old (6 teeth) Moroccan enhanced cross-bre

Biogenic amines, metabolites and monoamine oxidase in the filarial worm Setaria cervi.

Analysis of biogenic monoamines and their metabolites in Setaria cervi adults by reverse phase high performance liquid chromatography (HPLC) revealed dopamine as the major monoamine followed by norepinephrine and 5-hydroxytryptamine (5-HT). 5-Hydroxy indole acetic acid and tryptophan were also detected in significant amounts. A particulate-bound monoamine oxidase (MAO, EC 1.4.3.4.) catalysing the oxidative deamination of several amines was also demonstrated in both microfilariae and adults. The enzyme from the parasites exhibited unusually high Km values for various monoamines. Dopamine was ox

Periorchitis after tetramisole treatment in bulls implanted with Setaria labiatopapillos.

Thirteen mixed-breed beef bulls, 1 to 4 years old, were used to determine the effect of live and dead filarial nematodes, Setaria labiatopapillosa, placed in the vaginal cavity of the testes. When dead worms were used, granulomatous lesions developed on the tunica vaginalis parietalis in 7 of 8 testes. The lesions were similar to those seen in some clinical cases of periorchitis. Similar lesions developed in 5 of 6 testes after live worms were implanted in the vaginal cavity of the testes and tetramisole (8 mg/kg) was injected subcutaneously 6 days after implantation. When live worms were impl

Biochemical composition of microfiliarial and adult stages of Setaria cervi.

The microfilariae and adults of Setaria cervi, a filarial parasite of Indian water-buffalo (Bubalus bubalis Linn.) and of other ruminants were analysed for various biochemical constituents. Both stages of the parasite were rich in protein, carbohydrate, and lipids while nucleic acids were detectable in only small amounts. Microfilariae and adults contained 10 and 25% dry matter, 57.4 and 63.0% protein, 8.9 and 14.8% carbohydrate, and 12.8 and 9.2% lipid, respectively. Reducing sugars accounted for 52 and 67% respectively of the total carbohydrate in these two stages. Glycogen accounted for 95%

Molecular detection of Setaria tundra (Issaitshikoff & Rajewskaya, 1928) and Setaria cervi (Rudolphi, 1819) in red deer in Slovakia.

Filaroid nematodes Setaria tundra (Issaitshikoff & Rajewskaya, 1928) and Setaria cervi (Rudolphi, 1819) are internal parasites from family Onchocercidae with occurrence in the northern hemisphere. They have a considerably wide range of final host, including many species of family Cervidae. Intermediate hosts and vectors at the same time, are represented by the several mosquito species, mostly of genus Aedes. Infection of Setaria is relatively harmless and especially in wild cervids usually pass unnoticed. Although in some cases it can induce peritonitis which might be a life threatening condit

Outbreak of parasitic peritonitis in reindeer in Finland.

In 2003, there was an outbreak of peritonitis in reindeer in the southern and middle part of the Finnish reindeer herding area caused by the filarioid nematode Setaria species. In the province of Oulu, the proportion of reindeer viscera condemned owing to parasitic lesions increased from 4.9 per cent in 2001 to 40.1 per cent in 2003. In 2004, the focus of the outbreak moved approximately 100 km north. A total of 260 adult and pre-adult Setaria species nematodes were collected for morphological and molecular studies. The parasite was indistinguishable in terms of morphology and molecular biolog

Protein and antigenic composition of adult and microfilarial stages of Setaria cervi.

The protein and antigenic pattern of adult (female/male) and microfilarial stages of Setaria cervi, a bovine filarial parasite, was studied using certain immunochemical techniques. SDS-polyacrylamide gel electrophoretic analysis showed the presence of 35-40 protein bands in adults and 25-29 protein bands in microfilariae in molecular weight range of 10,000-200,000. Immunoelectrophoresis revealed the presence of 9-10 precipitin lines in adult and only 4 precipitin lines in microfilarial antigenic preparations. Crossed immunoelectrophoresis resolved these antigenic preparations further, and reve

Setaria cervi (Filarioidea, Onchocercidae) undressing in ungulates: altered morphology of developmental stages, their molecular detection and complete sequence cox1 gene.

This work introduces new morphological and molecular information on the filaroid nematode Setaria cervi (Rudolphi, 1819) obtained from 13 infected game ungulates out of 96 dissected. The hosts comprised the following: a single moose (Alces alces), ten red deer (Cervus elaphus) and two sika deer (Cervus nippon) originating from the western and northern regions of the Czech Republic. Based on the complete sequences of the gene encoding mitochondrial cytochrome c oxidase subunit 1 (cox1), all 20 females and four males belonged to the species S. cervi. We detected three developmental female stages

Characterization of the complete mitochondrial genome of Setaria digitata (Nematoda: Setariidae) from China.

Setaria digitata is a filarial parasite that causes fatal cerebrospinal nematodiasis in goats, horses and sheep, resulting in substantial economic losses to livestock farmers. In the present study, the complete mitochondrial (mt) genome of S. digitata from China was determined, characterized and compared with that of S. digitata from Sri Lanka. The identity of the mt genomes was 98.3% between S. digitata from China and Sri Lanka, and the complete mt genome sequence of S. digitata from China was slightly shorter (25 bp) than that from Sri Lanka. For the 12 protein genes, this comparison reveale
