In humans, like as in other mammals, the gonads, the internal genital ducts, and the external genital structures all develop from bipotential embryologic tissues. Male or female phenotype develops through a cascade of processes which initiate with sex determination and follow with sex differentiation. The karyotype (46, XY or 46, XX) of the embryo (genetic sex) determines whether primordial gonad differentiates into a testis or an ovary, respectively (gonadal differentiation). A Y-related gene, SRY, acts as a switch signal for testis differentiation. Testis development process involves several

Research on the biology and evolution of sex chromosomes has primarily focused on diploid XX/XY and ZW/ZZ systems. In contrast, the rise, evolution and demise of U/V systems has remained an enigma. Here we analyse genomes of nine brown algal species with different sexual systems to determine the history of their sex determination. U/V sex chromosomes emerged between 450 and 224 million years ago, when a region containing the pivotal male-determinant MIN ceased recombining. Seven ancestral genes within the sex-determining region show remarkable conservation over this vast evolutionary time, alt

AbstractSex chromosomes rapidly turn over in several taxonomic groups. Sex chromosome turnover is generally thought to start with the appearance of a new sex-determining gene on an autosome while an old sex-determining gene still exists, followed by the fixation of the new one. However, we do not know how prevalent the transient state is, where multiple sex-determining loci coexist within natural populations. Here, we removed a Y chromosome with a master male-determining gene DMY from medaka fish using high temperature-induced sex-reversed males. After four generations, the genomic characteris

Sex chromosome evolution remains an evolutionary puzzle despite its importance in understanding sexual development and genome evolution. The seemingly random distribution of sex-determining systems in reptiles offers a unique opportunity to study sex chromosome evolution not afforded by mammals or birds. These reptilian systems derive from multiple transitions in sex determination, some independent, some convergent, that lead to the birth and death of sex chromosomes in various lineages. Here we focus on turtles, an emerging model group with growing genomic resources. We review karyotypic chan

The ability to unequivocally identify the sex and reproductive status of individuals is crucial across many fields of study. Recent evidence indicates that avian sex determination is more flexible than once believed, with sex-reversed individuals identified in domestic bird populations-that is, individuals exhibiting gonadal and morphological traits of one sex while possessing the chromosomal make-up of the opposite sex. The presence of these individuals can challenge the reliability of traditional sexing methods that rely solely on external morphology, internal anatomy or genetic markers. The

Several New World atheriniforms have been recognized as temperature-dependent sex determined (TSD) and yet possess a genotypic sex determinant (amhy) which is primarily functional at mid-range temperatures. In contrast, little is known about the sex determination in Old World atheriniforms, even though such knowledge is crucial to understand the evolution of sex determination mechanisms in fishes and to model the effects of global warming and climate change on their populations. This study examined the effects of water temperature on sex determination of an Old World atheriniform, the cobaltca

The sex of an individual is determined at the time of fertilization. The mother passes on one sex chromosome, the X chromosome, and the father transmits the second sex chromosome, X or Y. Thus, an XX embryo becomes a female, whereas an XY individual becomes a male. A process known as "primary sex determination" allows the bipotential gonad to become a testis or an ovary in XY and XX embryos, respectively. In 1990, the Sry gene, located on the Y chromosome, was found to be necessary and sufficient to induce the male developmental program. At this time, the scientific community thought that othe

Next generation sequencing (NGS) method has been successfully used to identify sex-specific markers in many fish species. The Chinese dark sleeper (Odontobutis sinensis) is a commercially important aquaculture species in China. Here, by conducting whole genome sequencing of three females, three males and one female mixed pool, we initially identified 658 male-specific sequences and 242 female-specific sequences of Chinese dark sleeper. Based on polymerase chain reaction (PCR) confirmation, three markers showed 100 % male-specific amplification in 16 female and 16 male individuals from the Wuha

The Ussuri catfish Pseudobagrus ussuriensis belongs to the XY sex-determination system of male heterogamy, exhibiting typical male-biased dimorphism during growth and development. Therefore, all-male breeding possesses substantial economic significance. It is fundamental to all-male breeding to obtain XY pseudo-female and YY super-male fish. In this study, fry (6 days post-hatching, 2-3 mm total body length) were treated with 17-β estradiol, at graded doses of 50, 100, 150, and 200 µg/L for 40 days to induce sex reversal. After treatment, XY pseudo-females were screened using male-specific mol

Mechanisms for sex determination are highly diverse among animals and can evolve rapidly across taxonomic groups. This fundamental process dictates an animal's sexual fate and ultimately its development. Recent research has suggested that cephalopods follow a ZZ/Z0 sex determination (where males are homozygous and females are hemizygous) that originated at least 480 million years ago, making it one of the oldest conserved sex determination systems known for animals.1 By combining phenotypic sex data with three genomic datasets from the highly divergent cephalopod clade Nautiloidea (including t

Sex determination in eukaryotes defines whether an organism develops as male or female and is shaped by genetic and environmental factors. In the malaria parasite Plasmodium, recent data suggest that sex determination is decided at two critical checkpoints. First, chromatin remodeling activates the master regulator AP2-G, triggering a transcriptional cascade that commits the parasites to sexual development and drives the expression of genes crucial for male and female gametocytes. Second, post-transcriptional processes - including the stabilization and suppression of transcript via RNA-binding

The ribosome consists of protein and RNA components. Deletion of genes encoding specific ribosomal proteins has revealed that heterogeneity in the ribosome must exist in vertebrates; however, this has not been tested for ribosomal RNA (rRNA). In zebrafish (Danio rerio), the '45S-M' ribosomal RNA-encoding locus undergoes massive extrachromosomal amplification during oocyte growth and ovary differentiation and is distinct from the regular ribosomal DNA (rDNA) locus encoding somatic rRNA (45S-S). Although the 45S-M rDNA locus falls within the only described sex-linked region in multiple wild zebr

Therian mammals have among the oldest and most conserved sex-determining systems known to date. Any deviation from the standard XX/XY mammalian sex chromosome constitution usually leads to sterility or poor fertility, due to the high differentiation and specialization of the X and Y chromosomes. Nevertheless, a handful of rodents harbor so-called unusual sex-determining systems. While in some species, fertile XY females are found, some others have completely lost their Y chromosome. These atypical species have fascinated researchers for over 60 years, and constitute unique natural models for t

The water skinks Eulamprus tympanum and Eulamprus heatwolei show thermally induced sex determination where elevated temperatures give rise to male offspring. Paradoxically, Eulamprus species reproduce in temperatures of 12-15 °C making them outliers when compared with reptiles that use temperature as a cue for sex determination. Moreover, these two species are among the very few viviparous reptiles reported to have thermally induced sex determination. Thus, we tested whether these skinks possess undetected sex chromosomes with thermal override. We produced transcriptome and genome data for E.

Vertebrate sex differentiation follows a conserved suite of developmental events: the bipotential gonads differentiate and shortly thereafter sex specific traits become dimorphic. However, this may not apply to squamates, a diverse vertebrate lineage comprising of many species with thermosensitive sexual development. Of the three species with data on the relative timing of gonad differentiation and genital dimorphism, the females of two (Niveoscincus ocellatus and Barisia imbricata) exhibit a phase of temporary pseudohermaphroditism or TPH (gonads have differentiated well before genital dimorp

The sex ratio of Siberian crane chicks (Grus leucogeranus Pallas) from the captive population ofthe Oka Crane Breeding Center was analyzed with the use of molecular sex marker EE0.6 in 2009-2014. We determined the sex of 84 birds bred from 12 dams by means of artificial insemination and natural breeding. The total primary sex ratio was 40:44, and the secondary sex ratio was 36:39 with a minor predominance of females. The mortality rate of embryos was the same for both sexes. The primary and secondary sex ratio among the first eggs in clutches showed the same trends as those observed in general

Apostichopus japonicus is an important marine aquaculture species in China, with high nutritional and economic value. In A. japonicus, there is no obvious sexual dimorphism in external appearance, and sex differentiation primarily relies on the observation of mature gonads after dissection, which leads to difficulties in sex identification. The confusion in sex identification greatly reduces breeding efficiency in the sea cucumber industry. Therefore, developing a reliable sex-specific marker is crucial. In this study, we identified 586 male-specific sequences through whole-genome assembly and
