Secondary metabolites of Silene claviformis Litv. and the anti-inflammatory activity of its butanol extract.

In this study, we present the result of chemical investigation of aerial parts of Silene claviformis Litv., where we isolated four known natural secondary metabolites: two triterpenoids cycloastragenol (1), astrasieversianin X(2), as well as two steroid glycosides lineolon (3), 3-O-acetate sibirgenin (4), for the first time for this plant. The compound 4 was isolated from this plant for the first time as a natural substance. The chemical structures of these compounds were determined through comprehensive spectroscopic analyses, including 1D and 2D NMR spectroscopy, LC-MS, and comparison with p

The Sister Chromatid Division of the Heteromorphic Sex Chromosomes in Silene Species and Their Transmissibility towards the Mitosis.

Young sex chromosomes possess unique and ongoing dynamics that allow us to understand processes that have an impact on their evolution and divergence. The genus Silene includes species with evolutionarily young sex chromosomes, and two species of section Melandrium, namely Silene latifolia (24, XY) and Silene dioica (24, XY), are well-established models of sex chromosome evolution, Y chromosome degeneration, and sex determination. In both species, the X and Y chromosomes are strongly heteromorphic and differ in the genomic composition compared to the autosomes. It is generally accepted that fo

Antimalarial Effect of the Root of Silene macrosolen A. Rich (Caryophyllaceae) on Plasmodium-berghei-Infected Mice.

BACKGROUND: Malaria remains a major public health problem globally. Poor access to antimalarial drugs compounded with rapidly evolving drug resistance encourages researchers to continuously look for new drugs. Of importance, traditionally used medicines of plant origin are the highest priority as the ethnobotanical claim can be used as an important clue for its safety and efficacy profiles. Silene macrosolen A. Rich (Caryophyllaceae) has been traditionally used for malaria treatment in Ethiopia. Therefore, this study was aimed to evaluate the in vivo antimalarial activity of the plant against

High-quality genome assembly and linkage map for a rapidly evolving plant species: Silene uniflora.

The genus Silene is an important model system for fields as diverse as sex chromosome evolution, speciation, and disease ecology. However, genomic resources remain scarce in the genus. Here, we present a near chromosome-scale genome assembly and high-density linkage map for S. uniflora, a hermaphroditic/gynodioecious species which is an important model for rapid adaptation to anthropogenic disturbance and the role of phenotypic plasticity in adaptive evolution. Using a combination of long-read and Hi-C sequencing technologies, we generated a 1,268 Mb genome assembly with a scaffold N50 of 40.7

Chemical and Biological Characterization of Metabolites from Silene viridiflora Using Mass Spectrometric and Cell-Based Assays.

A comprehensive metabolite profiling of the medicinal plant Silene viridiflora using an UHPLC-ESI-MS/MS method is described for the first time. A total of 71 compounds were identified and annotated, the most common of which were flavonoids, triterpene glycosides, and ecdysteroids. The three major compounds schaftoside, 26-hydroxyecdysone, and silviridoside can be chosen as the markers for the assessment of the quality of S. viridiflora preparations. The methanol extract and a variety of metabolites identified in S. viridiflora were screened for their cytotoxic and Wnt pathway-inhibiting activi

Experimental manipulation of floral display size does not affect the distribution of paternal diversity in Silene dioica.

PREMISE: Various factors, including pollinator visitation patterns and pollen carryover, govern paternal diversity and its distribution within maternal plants. Floral display size may have opposite effects on fruit- and plant-level paternal diversity, because plants with large displays may receive more pollinator visits but also more sequential probes per visit. METHODS: To investigate the effect of floral display size on the distribution of paternal diversity in Silene dioica, we genotyped and assigned paternity for 1320 seeds sampled in several fruits per plant in control and manipulated fe

Methylcytosine oxidation derivatives are preferentially associated with repetitive chromatin and mark active rDNA in Silene latifolia.

INTRODUCTION: DNA methylation and its oxidative derivatives are key components of epigenetic regulation, yet their roles in plant repetitive DNA remain poorly understood. Here, we investigate the genomic distribution of oxidized cytosine modifications: 5-hydroxymethylcytosine (5hmC), 5-formylcytosine (5fC) and 5-carboxycytosine (5caC) within the repeat-rich genome of Silene latifolia, a dioecious plant with heteromorphic sex chromosomes. MATERIALS AND METHODS: Using low-coverage oxi-mC sequencing combined with RepeatExplorer2 clustering, ChIP-seq Mapper analysis, genome mapping, and immunoFIS

Heterosis in crosses between remnant populations of the rare prairie forb Silene regia: Implications for restoration genetics.

PREMISE: Calls to adopt seed-sourcing strategies in biological restoration that preserve local adaptation while maximizing genetic variation (e.g., regional admixture provenancing) are increasingly common. Heterosis, the increased fitness of progeny from crosses between populations relative to progeny from crosses within populations, could provide an added benefit under such strategies. METHODS: We quantified heterosis in crosses between three small remnant populations of the rare prairie forb Silene regia. We measured early fitness components (seed number per fruit, germination, and juvenile

Identification of Acidic Triterpenoid Saponins from Silene vulgaris Using a Methylation-Based Isolation and LC-MS Analysis Strategy.

The Silene genus plants in the Caryophyllaceae family are a large genus with over 850 species. It has been known that Silene genus plants contain triterpenoid saponins. These saponins have glucuronic acid in the sugar chain and are difficult to separate in chromatography. In this study, a strategy was developed to clarify the distribution of triterpenoid saponins in whole plants of Silene vulgaris by isolation of neutralized saponins using methylation reactions, which were used as standard substances for LC-MS analysis, and elucidating their characteristic MS and MS/MS fragment patterns. Histone post-translational modifications in Silene latifolia X and Y chromosomes suggest a mammal-like dosage compensation system.

Silene latifolia is a model organism to study evolutionary young heteromorphic sex chromosome evolution in plants. Previous research indicates a Y-allele gene degeneration and a dosage compensation system already operating. Here, we propose an epigenetic approach based on analysis of several histone post-translational modifications (PTMs) to find the first epigenetic hints of the X:Y sex chromosome system regulation in S. latifolia. Through chromatin immunoprecipitation we interrogated six genes from X and Y alleles. Several histone PTMS linked to DNA methylation and transcriptional repression

Synthesis and Photochemistry of Tris(trimethoxysilyl)acyl-silanes and 1,4-Tetrakis(silyl)-1,4-bisacylsilanes.

In this contribution, we present the synthesis of two groups of novel acylsilanes 1-6. Compounds 1 and 2 represent tris(trimethoxysilyl)acylsilanes, and compounds 3-6 are 1,4-tetrakis(silyl)-1,4-bisacylsilanes. All isolated compounds were characterized by infrared (IR) and nuclear magnetic resonance (NMR) spectroscopy and X-ray crystallography. Additionally, these compounds were further analyzed by ultraviolet/visible (UV/vis) spectroscopy and their longest wavelength absorption bands were assigned by density functional theory (DFT) calculations. Evolution of a plant sex chromosome driven by expanding pericentromeric recombination suppression.

Recombination suppression around sex-determining gene(s) is a key step in evolution of sex chromosomes, but it is not well understood how it evolves. Recently evolved sex-linked regions offer an opportunity to understand the mechanisms of recombination cessation. This paper analyses such a region on Silene latifolia (Caryophyllaceae) sex chromosomes, where recombination was suppressed in the last 120 thousand years ("stratum 3"). Locating the boundaries of the stratum 3 in S. latifolia genome sequence revealed that this region is far larger than assumed previously-it is about 14 Mb long and in

A CLAVATA3-like Gene Acts as a Gynoecium Suppression Function in White Campion.

How do separate sexes originate and evolve? Plants provide many opportunities to address this question as they have diverse mating systems and separate sexes (dioecy) that evolved many times independently. The classic "two-factor" model for evolution of separate sexes proposes that males and females can evolve from hermaphrodites via the spread of male and female sterility mutations that turn hermaphrodites into females and males, respectively. This widely accepted model was inspired by early genetic work in dioecious white campion (Silene latifolia) that revealed the presence of two sex-deter

Heavier Carbonyl Olefination: The Sila-Wittig Reaction.

The Wittig reaction is one of the most versatile tools in the repertoire of organic chemists. Thus, a broad variety of carbonyl compounds can be converted to tailor-made alkenes with phosphorus ylides under mild conditions. However, no comparable reaction has been reported for silanones, the silicon congeners of ketones. Here, we demonstrate for the first time the successful application of the Wittig olefination to iminosilylsilanone 1. The selective formation of a series of silenes (R2Si═CR2) via the sila-Wittig reaction revealed an unprecedented approach to otherwise elusive compounds. In ad

Evolution of sex-determination in dioecious plants: From active Y to X/A balance?

Sex chromosomes in plants have been known for a century, but only recently have we begun to understand the mechanisms behind sex determination in dioecious plants. Here, we discuss evolution of sex determination, focusing on Silene latifolia, where evolution of separate sexes is consistent with the classic "two mutations" model-a loss of function male sterility mutation and a gain of function gynoecium suppression mutation, which turned an ancestral hermaphroditic population into separate males and females. Interestingly, the gynoecium suppression function in S. latifolia evolved via loss of f

International intercomparison for criticality dosimetry: the case of biological dosimetry.

The Institute of Radiation Protection and Nuclear Safety (IRSN) organized a biological dosimetry international intercomparison with the purpose of comparing (i) dicentrics yield produced in human lymphocytes; (ii) the gamma and neutron dose estimate according to the corresponding laboratory calibration curve. The experimental reactor SILENE was used with different configurations: bare source 4 Gy, lead shield 1 and 2 Gy and a 60Co source 2 Gy. An increasing variation of dicentric yield per cell was observed between participants when there were more damages in the samples. Doses were derived fr

A Striking Mode of Activation of Carbon Disulfide with a Cooperative Bis(silylene).

The reactivity of the 1,4-substituted bis(silylenyl)terphenylene 1, 1,4-[ortho-(LSi)C6 H4 ]2 C6 H4 , (L=RC(NtBu)2 , R=Ph, Mes) towards CS2 is reported. It results in a dearomatization of the phenylene ring, affording the 1,3-substituted cyclohexadiene derivative 2. According to DFT calculations, a transient silene containing a Si=C bond capable of π(C=C) addition at the aromatic phenylene ring is a key intermediate. In contrast, addition of CS2 to the biphenyl-substituted mono-silylene ortho-(LSi)C6 H4 -C6 H5 3 leaves the aromatic π-system intact and forms, in a [1+2] cycloaddition reaction, t
