Dimethylsilanediol from silicone elastomers: Analysis, release from biopharmaceutical process equipment, and clearance studies.

Polysiloxanes are considered one of the most important commercial families of synthetic elastomers. They are frequently employed in biopharmaceutical manufacturing equipment as flexible single-use solutions due to superior material properties and compatibility with diverse sterilization methods. Extractables and leachables (E&L) testing is essential in qualifying such equipment, involving extraction studies to assess the potential release of compounds from plastic components for risk assessment. Silicone releases oligomeric siloxanes and small hydrolysis products, with dimethylsilanediol (DMSD

On the mechanism of marine fouling-prevention performance of oil-containing silicone elastomers.

For many decades, silicone elastomers with oil incorporated have served as fouling-release coating for marine applications. In a comprehensive study involving a series of laboratory-based marine fouling assays and extensive global field studies of up to 2-year duration, we compare polydimethylsiloxane (PDMS) coatings of the same composition loaded with oil via two different methods. One method used a traditional, one-pot pre-cure oil addition approach (o-PDMS) and another method used a newer post-cure infusion approach (i-PDMS). The latter displays a substantial improvement in biofouling preve

Tough Body Temperature-Triggered Shape Memory Silicone-Based Elastomers via Segment Sequence Regulation Strategy.

Developing high-strength, tough, shape-memory, and self-healing PDMS elastomers is crucial for flexible electronics and biosensing. Nevertheless, synergistically improving all these properties is challenging due to their intrinsically low glass transition temperature, amorphous nature, and weak intermolecular interactions. Herein, a molecular strategy by regulating the segment sequences is proposed to create multifunctional PDMS elastomers with tunable mechanical properties. Specifically, well-designed chain segment sequences can optimize the microphase separation structure. This strategy endo

Use of hydrophilic pharmaceutical excipients to modulate release of metal ions from silicone elastomers.

Silicone elastomers based on polydimethylsiloxane are biocompatible and non-biodegradable thermosetting polymers used in various drug delivery applications, including subdermal implants, vaginal rings, and intrauterine devices. Without exception, all marketed silicone elastomer drug delivery products provide sustained or controlled release of highly hydrophobic small drug molecules, since drug solubility in the silicone matrix is a prerequisite for molecular diffusion and release. We are interested in developing multipurpose silicone elastomer vaginal rings for local administration of metal io

Adhesion of maxillofacial silicone elastomer to a fiber-reinforced composite resin framework.

PURPOSE: Recently, fiber-reinforced composite resin (FRC) has been introduced as a framework material for maxillofacial silicone prostheses. The purpose of this research was to study the tensile bond strength between a room temperature-polymerized maxillofacial silicone elastomer and a unidirectional FRC. MATERIALS AND METHODS: Three different bonding agents were compared. Specimens were loaded in tension mode according to ISO 22401 in a universal testing device with a crosshead speed of 10 mm/min until bonding failure occurred. The influence of the surface characteristics (ground vs intact)

Comparative studies on the adhesion of a silicone elastomer to a chromium-cobalt dental alloy.

Rocatec, Kevloc and Siloc bonding systems were used to bond Molloplast-B silicone elastomer to a metal alloy denture base material. The mechanical strength of the bonds was examined using tensile tests. Two groups of tests were performed, 30 samples after one day storage in water and 30 samples after 6 months in water at 37 degrees C. At both testing times, the Rocatec system exhibited high reproducibility and the highest mean results. For the Kevloc system the values were lower while the Siloc system yielded the lowest values. The strength of samples, bonded with the Rocatec system, was impro

Safety and intracardiac function of a silicone-polyurethane elastomer designed for vascular use.

No ideal prosthetic heart valve exists. While polyurethane copolymers possess excellent physical properties, thrombosis and embolism remain a problem and compounds designed to be less thrombogenic have been prone to biodegradation and failure ('cracking'). We tested a new material which has an elastomeric silicone applied to the polymer surface. A hydrophilic film of protamine and gelatin is covalently bonded to the silicone--this obviates the need for preclotting and should permit endothelial growth. The material was tested by implantation during cardiopulmonary bypass as patches in the mitra

Endoscopic rehabilitation of vocal cord paralysis with a silicone elastomer suspension implant.

OBJECTIVES: Because of the side effects of Teflon, the risk of infection from the use of collagen, autologous fat resorption, and the lack of alternative substances, injection laryngoplasty tends to be replaced by laryngeal framework surgery as the method of choice for the treatment of unilateral vocal cord recurrent paralysis (LP). The aim of this study was to evaluate the results, for morbidity and voice quality, of treating this paralysis by injection of a silicone suspension elastomer implant (SSEI). STUDY DESIGN: The study was retrospective, and 19 patients were included. Average follow-

Replacement of the first metatarsophalangeal joint with a silicone elastomer ball-shaped spacer.

We report the use of a silicone elastomer ball-shaped spacer for the treatment of hallux valgus and hallux rigidus. One hundred and eleven operations were performed in 77 patients over a 5-year period. The best results were obtained in patients with hallux rigidus. Good results were obtained in hallux valgus, but it was found that in patients with severe preoperative deformity the correction obtained was inadequate, and an additional procedure would be required for full correction. There were no cases of silicone synovitis.

Silicone elastomer films loaded with 5-fluorouracil for time-dependent drug release.

Silicone elastomer composite films loaded with the antitumor drug 5-fluorouracil (5-FU) were developed for controlled release applications. Polydimethylsiloxane (PDMSOH) films were crosslinked using tetraethoxysilane and room-temperature vulcanization catalysts, with glycerol incorporated as a hydrophilic modifier to regulate permeability of the hydrophobic silicone matrix. Dimethyl sulfoxide was used to solubilize 5-FU during film preparation. Film microstructure and drug distribution were analyzed by scanning electron microscopy and Raman spectroscopic imaging. Among the tested formulations,

Development of Silicone Elastomer-Based Composite Films Containing Ibuprofen and Functional Additives.

Silicone elastomers are attractive matrices for transdermal drug delivery systems, but the controlled release of poorly water-soluble drugs from hydrophobic silicone networks remains challenging. Medical-grade silicone elastomers are generally regarded as chemically stable, biologically inert, and highly biocompatible polymer matrices, which supports their use in biomedical and pharmaceutical materials. Here, ibuprofen-loaded silicone/polyol composite films were prepared from hydroxyl-terminated polydimethylsiloxane (PDMS-OH) using glycerol- and 1,2-propylene glycol-derived alkoxysilane cross-

Central venous silicone elastomer catheter placement by basilic vein cutdown in neonates.

There is a need for central venous access in small premature infants and other neonates when enteral feeding is not tolerated or is contraindicated. We placed 83 small (0.635-mm od) silicone elastomer catheters by basilic vein cutdown through a subcutaneous tunnel in 79 patients during a 12-month period. Thirty-five patients (44%) weighed less than 1,000 g. Each patient on whom the procedure was attempted had successful placement of a catheter, and they remained in place a mean of 20 days (range three to 82). Patients had a mean weight gain of 15 g per day of catheter use. Sixty-two catheters

Hydrophilic modification of silicone elastomer films: thermal, mechanical and theophylline permeability properties.

The improvement of the hydrophilicity of silicone rubber (SR) is sought in many biomedical applications. In the present work, we have prepared neat films coming from condensation-type SR, modified either through blending or end-linking reaction with low molecular weight poly(ethylene glycol) (PEG). The films were studied with respect to their water uptake capacity, stability of embedment of ethylenoxy moieties, and mechanical and thermal properties. Subsequently, we prepared drug-loaded films with theophylline, a relatively hydrophilic model drug, and studied the release performance of these s

A dynamic mechanical method for determining the silicone elastomer solubility of drugs and pharmaceutical excipients in silicone intravaginal drug delivery rings.

The silicone elastomer solubilities of a range of drugs and pharmaceutical excipients employed in the development of silicone intravaginal drug delivery rings (polyethylene glycols, norethisterone acetate, estradiol, triclosan, oleyl alcohol, oxybutynin) have been determined using dynamic mechanical analysis. The method involves measuring the concentration-dependent decrease in the storage modulus associated with the melting of the incorporated drug/excipient, and extrapolation to zero change in storage modulus. The study also demonstrates the effect of drug/excipient concentrations on the mec

A prodrug strategy for sustained release of lactic acid from silicone elastomer vaginal rings.

Lactic acid is the most abundant organic weak acid in the healthy human vagina and plays a pivotal role in maintaining an acidic vaginal environment protective against exogenous bacteria and viruses. However, in dysbiotic or non-optimal vaginal environments, significantly decreased concentrations of lactobacilli result in reduced lactic acid production, increased vaginal pH, and enhanced risk of sexually transmitted infections (including human immunodeficiency virus), and bacterial vaginosis. Various gel-based products are marketed to administer lactic acid vaginally for the treatment of bacte

Evaluation of Antifungal Activity of MaxilloFacial Silicone Elastomer Reinforced with Chitosan Nanoparticles-An In vitro Study.

BACKGROUND: Microbial and fungal growth, particularly Candida albicans, is a major issue with the usage of maxillofacial silicone material, which can lead to chronic infection, inflammation, and silicone material degradation, for which the development of an antimicrobial silicone elastomer has become crucial. METHODOLOGY: This study was designed to determine the efficacy of adding chitosan nanoparticles (1.5%, 3%, and 5% by weight) to silicone matrix on antifungal activity. Eighty specimens were fabricated with varying concentrations of chitosan and separated into four groups [C1 (control), C

Orbital inflammation as a late complication of frontalis silicone elastomer sling operation for congenital ptosis.

Frontalis suspension with the use of a silicone elastomer is one of the preferred procedures for correcting moderate-to-severe ptosis with poor levator function. This procedure involves the attachment of the upper eyelid to the frontalis muscle by a silicone sling. Although this surgery has been performed for many years, information regarding long-term outcomes and complications is relatively limited. We report a case of orbital inflammation 15 years after a frontalis silicone sling suspension operation for congenital ptosis.
