Superior antiglycation efficacy of silymarin from Silybum marianum in human αA-crystallin: Implications for diabetic cataract prevention.

Hyperglycemia activates the polyol pathway, producing fructose, which promotes glycation and denatures α-crystallin, ultimately leading to diabetic cataracts. This study compared the antiglycation effects of SMR and SBN, using a fructose-induced human αA-crystallin glycation model. Through fluorescence analysis, SDS-PAGE, and Western blotting methods, we found that glycation caused αA-crystallin to form fluorescent advanced glycation end products (AGEs), cross-linking AGEs, and Nε-carboxymethyllysine (CML). Results show that SMR (> 20 μg/mL) and SBN (> 100 μg/mL) effectively inhibited cross-li

Renoprotective effects of Silybum marianum (L.) Gaertn (Silymarin) on thioacetamide-induced renal injury: Biochemical and histopathological approach.

Thioacetamide (TAA), recognized as an experimental toxin, mainly causes acute liver damage through the production of free radicals. TAA as well induces renal dysfunction hence; renal failure is often related with the end-stage of the hepatic damage. The aim of the current study was to examine the protective effects of Silymarin (Sil) against TAA-induced kidney damage in this current study. The twenty eight rats were separated into four groups. Group 1 was performed as control (saline 0,5 mL intraperitonally i.p.). Group 2 was given to 50 mg/kg TAA (i.p.). Group 3 was administrated with TAA jus

[Identification of flavonoids 3-hydroxylase from [Silybum marianum (L.) Gaertn] and its application in enhanced production of taxifolin].

(2S)-taxifolin is an important flavonoid that has anti-inflammatory and anti-oxidation effects. It is widely used in pharmaceutical and nutraceutical industries. Flavone 3-hydroxylase (F3H) can catalyze the synthesis of (2S)-taxifolin and other 3-hydroxylated flavonoids from (2S)-eriodictyol. Due to the low catalytic efficiency of F3H, the titer of many 3-hydroxyflavones, such as taxifolin, synthesized by microbial method is relatively low. In this study, a SmF3H was identified from the transcriptome of Silybum marianum (L.) Gaertn. The results of fermentation showed that SmF3H can catalyze th…
