Myopathy and encephalopathy in chick embryos experimentally infected with Akabane virus.

Chick embryos infected with Akabane virus by the yolk sac route at 6 days of incubation developed polymyositis and encephalitis. At 3 to 7 days after inoculation, skeletal muscles had myotubule degeneration, clumping of muscle cell nuclei, and infiltration of heterophils; dysplasia and aplasia were evident at 9 to 15 days after inoculation. Changes in the cerebral neostriatum and optic lobes at 2 to 11 days after inoculation included necrosis of primordial nervous tissue, hemorrhages, and hyperplasia of the vascular endothelial cells. Cavities were in nervous tissue subsequent to encephalitis.

Teratogenicity of Australian Simbu serogroup and some other Bunyaviridae viruses: the embryonated chicken egg as a model.

The use of embryonated chicken eggs as a model for assessing the teratogenic potential of animal viruses was investigated with 12 members of the Bunyaviridae family. Infection of 4-day-old embryonated chicken eggs via the yolk sac with 10 of the viruses resulted in deaths or congenital deformities that were similar to those observed in Akabane virus infections of fetal ruminants and included arthrogryposis, scoliosis, mandible defects, and retarded development. Statistical analysis showed that the viruses fell into three main groupings, namely, those that caused both death and deformities (Aka

Genomic Insights into Neglected Orthobunyaviruses: Molecular Characterization and Phylogenetic Analysis.

Orthobunyaviruses are a diverse group of segmented RNA viruses with significant but underexplored public and veterinary health implications. This study provides a genomic, phylogenetic, and ecological analysis of neglected Orthobunyaviruses using next-generation sequencing and computational predictions. We identified unique phylogenetic relationships, with Tanga virus forming a distinct lineage linked to zoonotic, human-associated, or non-vertebrate viruses across segments. GC content analysis revealed segment-specific patterns: higher GC content in the S segment suggests genomic stability and

Experimental infection of suckling mice by subcutaneous inoculation with Oropouche virus.

Oropouche virus, of the family Bunyaviridae, genus Orthobunyavirus, serogroup Simbu, is an important causative agent of arboviral febrile illness in Brazil. An estimated 500,000 cases of Oropouche fever have occurred in Brazil in the last 30 years, with recorded cases also in Panama, Peru, Suriname and Trinidad. We have developed an experimental model of Oropouche virus infection in neonatal BALB/c mouse by subcutaneous inoculation. The vast majority of infected animals developed disease on the 5th day post infection, characterized mainly by lethargy and paralysis, progressing to death within

Re-emerging Oropouche virus infections: insights into maternal-fetal crosstalk, coinfections, and expanding public health threats.

Oropouche virus (OROV), a re-emerging orthobunyavirus of the Simbu serogroup, is increasingly recognized as a significant arboviral threat across the Americas. Although historically underrecognized, OROV infection raises urgent concerns due to reported co-infections with other arboviruses and possible novel nonvector transmission, including vertical and sexual routes. Recent reports link OROV infection during pregnancy with fetal demise, miscarriage, and congenital anomalies in newborns. Drawing on emerging epidemiological, clinical, and virological studies, we review recent findings on matern

Salient lesions in domestic ruminants infected with the emerging so-called Schmallenberg virus in Germany.

The so-called Schmallenberg virus (SBV), first detected in a German town of the same name in October 2011, is a novel emerging orthobunyavirus in Europe causing malformations and severe economic loss in ruminants. This report describes lesions in 40 sheep, 2 goats, and 16 cattle naturally infected with SBV as determined by real-time quantitative reverse transcription polymerase chain reaction. The most common macroscopic changes were arthrogryposis, vertebral malformations, brachygnathia inferior, and malformations of the central nervous system, including hydranencephaly, porencephaly, hydroce

Mutation of a conserved lysine in the RdRp fingers domain broadly attenuates orthobunyaviruses.

Orthobunyaviruses, comprising more than 130 species, constitute a significant threat to human and animal health, yet no effective broad-spectrum treatments or vaccines are currently available. Here, we initially identified an evolutionarily constrained lysine residue, highly conserved across all 22 Orthobunyavirus serogroups, within the α30 helix of the RNA-dependent RNA polymerase (RdRp). Functional interrogation of this site by charge-reversal substitution (K-to-E) substantially impaired polymerase activity and led to marked attenuation of Ebinur Lake virus (EBIV), as evidenced by impaired r

Schmallenberg virus, a novel orthobunyavirus infection in ruminants in Europe: potential global impact and preventive measures.

In autumn 2011, Schmallenberg virus was the first orthobunyavirus detected in Europe. The virus belongs to the Simbu serogroup. Like other orthobunyaviruses, it is apparently transmitted by arthropod vectors, primarily by biting midges (Culicoides spp.). Ruminants and new-world camelids (alpacas) are susceptible to infection. Adult animals may develop mild disease, if any. However, transplacental infection can lead to severe congenital malformations such asarthrogryposis, malformation of the vertebral column (kyphosis, lordosis, scoliosis, torticollis) and of the skull (macrocephaly, brachygna

Radioimmune assays and molecular studies that place Anopheles B and Turlock serogroup viruses in the Bunyavirus genus (Bunyaviridae).

Molecular analyses indicate that Turlock virus (TUR, Turlock serogroup) and Boraceia virus (BOR, Anopheles B serogroup) have virion RNA species and polypeptides comparable in size to those of members of the Bunyavirus genus and unlike those of members of the newly defined Phlebovirus, Nairovirus, or Uukuvirus genera (Bunyaviridae). The 11 terminal 3' end nucleotides of the three virion RNA species of both BOR and TUR viruses (HOUCAUCACAUG...) are identical in sequence to the 3' end sequences of the viral RNA species of snowshoe hare (SSH) and La Crosse bunyaviruses (LAC, California serogroup,

The N-terminal domain of Schmallenberg virus envelope protein Gc is highly immunogenic and can provide protection from infection.

Schmallenberg virus (SBV) is transmitted by insect vectors, and therefore vaccination is one of the most important tools of disease control. In our study, novel subunit vaccines on the basis of an amino-terminal domain of SBV Gc of 234 amino acids ("Gc Amino") first were tested and selected using a lethal small animal challenge model and then the best performing formulations also were tested in cattle. We could show that neither E. coli expressed nor the reduced form of "Gc Amino" protected from SBV infection. In contrast, both, immunization with "Gc Amino"-encoding DNA plasmids and "Gc-amino"

Akabane virus.

Akabane virus, an arthropod-borne Bunyavirus, is the major cause of epizootics of congenital malformations in ruminants in Australia, Japan, Korea, and Israel, and is suspected to be a cause of sporadic outbreaks elsewhere. Blood-sucking insects, such as biting midges, transmit the virus horizontally to vertebrates. Climatic factors influence the seasonal activity and geographic range of the vector population and, therefore, occurrence of related disease. Inoculated ruminants seroconvert rapidly after a short subclinical viremia. Infection is of consequence only if ruminants are pregnant and n

A New Threat on the Rise: Oropouche Viral Infection.

The Oropouche virus (OROV), a member of the Simbu group's Orthobunyaviruses, is a vector-borne virus with three single-stranded negative-sense RNA segments and a spherical lipid sheath. It is mainly spread by Culicoides paraensis midges and, to a lesser extent, Culex quinquefasciatus mosquitoes. OROV, first reported in Trinidad and Tobago in 1955, has since spread to Brazil, Colombia, and Peru, causing serological epidemics and recently invading nonendemic regions. The severity of the disease can vary from cold symptoms to severe illness, such as meningitis. Recent outbreaks in Acre and Brazil

Insect-specific viruses and arboviruses in adult male culicids from Midwestern Brazil.

Viruses were identified from male anthropophilic mosquitoes from Mato Grosso (MT) State, Midwest Brazil from February 2017 to January 2018. Mosquitoes tested included Aedes (Stegomyia) aegypti (1139 males; 84 pools), Culex quinquefasciatus (9426 males; 179 pools), Culex sp. (3 males; 3 pools) and Psorophora albigenu (1 male; 1 pool) collected from four cities of MT. Pools were subjected to viral RNA extraction followed by RT-PCRs specific for ten flaviviruses, five alphaviruses and Simbu serogroup of orthobunyaviruses. Positive pools were passaged three times in VERO cells (alphavirus and orth

Encephalomyelitis in mice experimentally infected with Akabane virus.

Lesions in the central nervous system of mice, induced by intracerebral injection of Akabane virus, were observed by the fluorescent antibody technique and histological method. Fluorescent antigens were recognized in the cytoplasm of nerve cells, but were not detected exactly in any other part. Fluoresced nerve cells were distributed almost all over the central nervous system, especially in medulla oblongata and spinal cord. The appearance of fluorescent antigens was followed by histological changes. So-called Nissl's acute severe degeneration was observed in nerve cells in the area where the

A novel approach using IFNAR1 KO mice for assessing Akabane virus pathogenicity and vaccine efficacy.

Akabane virus (AKAV) is a Simbu serogroup virus that can cause congenital abnormalities in ruminants. In 2010, an AKAV-7 strain exhibiting different characteristics and belonging to a distinctive genogroup compared to previous AKAVs was isolated in South Korea. Although this novel pathogenic AKAV-7 has been discovered, in vivo studies on AKAV-7 are currently insufficient due to limitations of using large animals and suckling mice. Therefore, the development of a novel small animal model for AKAV studies is necessary. Type I interferon receptor knock out (IFNAR1 KO) mice are widely employed as

Experimental infection of pregnant goats with Akabane virus.

Ten pregnant goats were inoculated intravenously with a newly isolated strain of Akabane virus. As a result, vertical infection and experimental reproduction of congenital morphological abnormality were accomplished. Fetuses were removed from two of them 10 days after inoculation and used for recovery of the virus. Clinically, the goats were free from any abnormal sign. Mild viremia was demonstrated in one of them inoculated with the virus at 30 days of pregnancy, but the fetus removed from this goat was negative for virus recovery. No viremia was demonstrated in the other one inoculated with

Deformities of chick embryos in experimental Akabane virus infection.

A material containing 10(3.0) approximately 10(5.0) TCID50 of Akabane virus was inoculated into 6-day-old chick embryos by the yolk sac route. Death of embryos did not increase in the course of embryonic development till 18 days of age, as compared with control groups. Later than 18 days of age, however, the numbers of dead and peeping but unhatched embryos increased, making the hatching rate significantly low. Deformities, such as arthrogryposis and hydranencephaly, appeared in almost all the dead and unhatched embryos. They were severe in dead embryos, considerably severe in peeping unhatche
