Nitrate reduction associated with respiration in Sinorhizobium meliloti 2011 is performed by a membrane-bound molybdoenzyme.

The purification and biochemical characterization of the respiratory membrane-bound nitrate reductase from Sinorhizobium meliloti 2011 (Sm NR) is reported together with the optimal conditions for cell growth and enzyme production. The best biomass yield was obtained under aerobic conditions in a fed-batch system using Luria-Bertani medium with glucose as carbon source. The highest level of Sm NR production was achieved using microaerobic conditions with the medium supplemented with both nitrate and nitrite. Sm NR is a mononuclear Mo-protein belonging to the DMSO reductase family isolated as a

Role of extracellular compounds in Cd-sequestration relative to Cd uptake by bacterium Sinorhizobium meliloti.

The role of bacterially derived compounds in Cd(II) complexation and uptake by bacterium Sinorhizobium meliloti wild type (WT) and genetically modified ExoY-mutant, deficient in exopolysaccharide production, was explored combining chemical speciation measurements and assays with living bacteria. Obtained results demonstrated that WT- and ExoY-strains excreted siderophores in comparable amounts, while WT-strain produced much higher amount of exopolysaccharides and less exoproteins. An evaluation of Cd(II) distribution in bacterial suspensions under short term exposure conditions, showed that mo

Deficiency of a Sinorhizobium meliloti BacA mutant in alfalfa symbiosis correlates with alteration of the cell envelope.

The BacA protein is essential for the long-term survival of Sinorhizobium meliloti and Brucella abortus within acidic compartments in plant and animal cells, respectively. Since both the S. meliloti and B. abortus bacA mutants have an increased resistance to bleomycin, it was hypothesized that BacA was a transporter of bleomycin and bleomycin-like compounds into the bacterial cell. However, our finding that the S. meliloti bacA mutant also has an increased sensitivity to detergents, a hydrophobic dye, ethanol, and acid pH supported a model in which BacA function affects the bacterial cell enve

VisN and VisR are global regulators of chemotaxis, flagellar, and motility genes in Sinorhizobium (Rhizobium) meliloti.

The known 41 flagellar, chemotaxis, and motility genes of Sinorhizobium (Rhizobium) meliloti contained in the "flagellar regulon" are organized as seven operons and six transcription units that map to a contiguous 45-kb chromosomal region. By probing gene expression on Western blots and with lacZ fusions, we have identified two master regulatory genes, visN and visR, contained in one operon. The gene products probably form a heterodimer, VisNR, acting as a global transcription activator of other flagellar genes. The related 27-kDa VisN and VisR proteins are LuxR-type proteins with typical liga

Characterization of Brucella abortus sigma factor sigma54 (rpoN): genetic complementation of Sinorhizobium meliloti ntrA mutant.

The intracellular pathogen Brucella abortus has an alternative sigma factor sigma54 (RpoN) highly similar to Sinorhizobium meliloti NtrA. RpoN was described to be required for the transcription of a wide range of genes involved in diverse physiological functions including the regulation of virulence-related factors in both plants and animal pathogens. B. abortus rpoN gene restored the normal growth of an S. meliloti ntrA mutant in minimal media with succinic acid as a sole carbon source as well as the formation of functional nodules in alfalfa, thus revealing that the gene is functional. Identification of a Sinorhizobium meliloti YbgC-like thioesterase that contributes to the production of the infochemical 2-tridecanone.

Sinorhizobium meliloti is a soil bacterium that can establish beneficial symbiosis with legume plants. The fadD gene encodes a long-chain fatty acyl-coenzyme A (CoA) synthetase. Inactivation of FadD in S. meliloti leads to a pleiotropic phenotype, including the overproduction of several volatile methylketones (MKs). One of them, 2-tridecanone (2-TDC), was found to act as an infochemical that affects important bacterial traits and hampers plant-bacteria interactions. Knowledge about bacterial genes involved in MK production is limited. In wild tomato species, MK synthesis requires intermediates

XoxF and the Calvin-Benson cycle mediate lanthanide-dependent growth on methanol in Bradyrhizobium and Sinorhizobium.

Nodule-forming bacteria play crucial roles in plant health and nutrition by providing fixed nitrogen to leguminous plants. Despite the importance of this relationship, how nodule-forming bacteria are affected by plant exudates and soil minerals is not fully characterized. Here, the effects of plant-derived methanol and lanthanide metals on the growth of nitrogen-fixing Rhizobiales are examined. Prior work has demonstrated that select Bradyrhizobium strains can assimilate methanol only in the presence of lanthanide metals; however, the pathway enabling assimilation remains unknown. In this stud

Internalization of a thiazole-modified peptide in Sinorhizobium meliloti occurs by BacA-dependent and -independent mechanisms.

BacA proteins play key roles in the chronic intracellular infections of Sinorhizobium meliloti, Brucella abortus and Mycobacterium tuberculosis within their respective hosts. S. meliloti, B. abortus and M. tuberculosis BacA-deficient mutants have increased resistance to the thiazole-modified peptide bleomycin. BacA has been previously hypothesized, but not experimentally verified, to be involved in bleomycin uptake. In this paper, we show that a BacA-dependent mechanism is the major route of bleomycin internalization in S. meliloti. We also determined that the B. abortus and S. meliloti BacA p

The effect of Sinorhizobium meliloti volatilomes and synthetic long-chain methylketones on soil and Medicago truncatula microbiomes.

Bacterial volatile compounds play important roles in intra- and interkingdom interactions but little is known about their effects on soil and plant microbiomes. The legume symbiont Sinorhizobium meliloti (Sm) releases volatile methylketones (MKs), one of which acts as an infochemical among bacteria and hampers plant-bacteria interactions. Inactivation of the fatty acyl-CoA ligase FadD in Sm moderately enhances MK production. To further explore the ecological role of MKs on soil and plant bacterial communities, we aimed at obtaining an MK-overproducing Sm strain by deleting the 3-oxoacyl-CoA th

Identification of Sinorhizobium (Ensifer) medicae based on a specific genomic sequence unveiled by M13-PCR fingerprinting.

A collection of nodule isolates from Medicago polymorpha obtained from southern and central Portugal was evaluated by M13-PCR fingerprinting and hierarchical cluster analysis. Several genomic clusters were obtained which, by 16S rRNA gene sequencing of selected representatives, were shown to be associated with particular taxonomic groups of rhizobia and other soil bacteria. The method provided a clear separation between rhizobia and co-isolated non-symbiotic soil contaminants. Ten M13-PCR groups were assigned to Sinorhizobium (Ensifer) medicae and included all isolates responsible for the form

Structural characterization of the lipid A component of Sinorhizobium sp. NGR234 rough and smooth form lipopolysaccharide. Demonstration that the distal amide-linked acyloxyacyl residue containing the long chain fatty acid is conserved in rhizobium and Sinorhizobium sp.

A broad-host-range endosymbiont, Sinorhizobium sp. NGR234 is a component of several legume-symbiont model systems; however, there is little structural information on the cell surface glycoconjugates. NGR234 cells in free-living culture produce a major rough lipopolysaccharide (LPS, lacking O-chain) and a minor smooth LPS (containing O-chain), and the structure of the lipid A components was investigated by chemical analyses, mass spectrometry, and NMR spectroscopy of the underivatized lipids A. The lipid A from rough LPS is heterogeneous and consists of six major bisphosphorylated species that

Glycerol-Based Cryopreservation of Sinorhizobium/Ensifer Strains from Sweet Acacia (Vachellia farnesiana [L.] Wight & Arn.).

The genus Sinorhizobium (Ensifer) exhibits remarkable metabolic versatility and adaptive capacity, establishing effective nitrogen-fixing symbioses with diverse legume crops, including the pioneer shrub Vachellia farnesiana. This association is crucial for ecological restoration in arid and nutrient-deficient Mexican dryland ecosystems and for the remediation of contaminated sites. However, the prolonged preservation of specialized native strains through continuous serial subculturing risks genomic instability and the loss of extrachromosomal replicons harboring essential nod, nif, and heavy m

Lanthanide-Dependent Methanol Dehydrogenase XoxF Confers a Competitive Advantage to Sinorhizobium meliloti During Symbiosis with Medicago sativa.

The recent discovery of the lanthanide (Ln)-dependent methanol dehydrogenase (Ln-MDH) XoxF has expanded the spectrum of bacteria recognized for methylotrophic metabolism. Many bacteria, including rhizobia, have historically escaped being categorized as methylotrophs because they exclusively produce XoxF-type Ln-MDHs and entirely lack the long-studied calcium-dependent MDH MxaFI. We report that the XoxF-type Ln-MDH encoded by the smb20173 gene is the sole MDH that supports methylotrophic growth of Sinorhizobium meliloti. The lanthanides that consistently supported growth of S. meliloti in minim

pH dependence of cyanide and imidazole binding to the heme domains of Sinorhizobium meliloti and Bradyrhizobium japonicum FixL.

Equilibrium and kinetic properties of cyanide and imidazole binding to the heme domains of Sinorhizobium meliloti and Bradyrhizobium japonicum FixL (SmFixLH and BjFixLH) have been investigated between pH5 and 11. KD determinations were made at integral pH values, with the strongest binding at pH9 for both ligands. KD for the cyanide complexes of BjFixLH and SmFixLH is 0.15±0.09 and 0.50±0.20μM, respectively, and 0.70±0.01mM for imido-BjFixLH. The association rate constants are pH dependent with maximum values of 443±8 and 252±61M(-1)s(-1) for cyano complexes of BjFixLH and SmFixLH and (5.0±0.3

Independent activity of the homologous small regulatory RNAs AbcR1 and AbcR2 in the legume symbiont Sinorhizobium meliloti.

The legume symbiont Sinorhizobium meliloti expresses a plethora of small noncoding RNAs (sRNAs) whose function is mostly unknown. Here, we have functionally characterized two tandemly encoded S. meliloti Rm1021 sRNAs that are similar in sequence and structure. Homologous sRNAs (designated AbcR1 and AbcR2) have been shown to regulate several ABC transporters in the related α-proteobacteria Agrobacterium tumefaciens and Brucella abortus. In Rm1021, AbcR1 and AbcR2 exhibit divergent unlinked regulation and are stabilized by the RNA chaperone Hfq. AbcR1 is transcribed in actively dividing bacteria

An ABC-type cobalt transport system is essential for growth of Sinorhizobium meliloti at trace metal concentrations.

We report expression and mutant phenotypes for a gene cluster in Sinorhizobium meliloti, designated cbtJKL, that has been shown to encode an ABC-type cobalt transport system. Transcription of cbtJKL initiated 384 nucleotides upstream from the cbtJ translation start codon, and the resulting 5' region contained a putative B(12)riboswitch. Expression of the cbtJKL genes appeared to be controlled by (cobalt-loaded) cobalamin interacting at the B(12)riboswitch, since (i) a putative B(12)riboswitch was located within this large upstream region, (ii) cbtJ transcription was repressed upon addition of

Distinct and indispensable roles of cell viability and extracellular polymeric substances in Pb mineralization by Sinorhizobium sp. C10.

Microbially induced phosphate mineralization (MIPP) is a promising strategy for immobilizing lead (Pb) in contaminated environments. This study aimed to determine whether cell viability and EPS are indispensable for Pb mineralization by Sinorhizobium sp. C10, a strain previously shown to form lead phosphate minerals. We compared live versus heat-inactivated cells, employed a mild sodium citrate method to strip EPS while preserving cell viability, and determined Pb removal efficiency, alkaline phosphatase (ALP) activity, inorganic phosphate (PO43-) release, and mineral phase composition. Toxicity and osmoprotective activities of analogues of glycine betaine obtained by solid phase organic synthesis towards Sinorhizobium meliloti.

Seven analogues of the bacterial osmoprotectant glycine betaine (GB, trimethylammonioacetate), in which the methyl groups of the Me3N+ moiety are replaced by various substituents, were obtained by SPOS using Wang resin. Their biological activities (osmoprotection vs toxicity), appeared closely related to their uptake efficiency and their catabolism in the betaine-demethylating model bacterium Sinorhizobium meliloti.

Similarity to peroxisomal-membrane protein family reveals that Sinorhizobium and Brucella BacA affect lipid-A fatty acids.

Sinorhizobium meliloti, a legume symbiont, and Brucella abortus, a phylogenetically related mammalian pathogen, both require the bacterial-encoded BacA protein to establish chronic intracellular infections in their respective hosts. We found that the bacterial BacA proteins share sequence similarity with a family of eukaryotic peroxisomal-membrane proteins, including the human adrenoleukodystrophy protein, required for the efficient transport of very-long-chain fatty acids out of the cytoplasm. This insight, along with the increased sensitivity of BacA-deficient mutants to detergents and cell
