Adolescent social defeat decreases spatial working memory performance in adulthood.

BACKGROUND: Adolescent social stress is associated with increased incidence of mental illnesses in adulthood that are characterized by deficits in cognitive focus and flexibility. Such enhanced vulnerability may be due to psychosocial stress-induced disruption of the developing mesocortical dopamine system, which plays a fundamental role in facilitating complex cognitive processes such as spatial working memory. Adolescent rats exposed to repeated social defeat as a model of social stress develop dopaminergic hypofunction in the medial prefrontal cortex as adults. To evaluate a direct link bet

Reduced nucleus accumbens enkephalins underlie vulnerability to social defeat stress.

Enkephalins, endogenous ligands for delta opioid receptors (DORs), are highly enriched in the nucleus accumbens (NAc). They are implicated in depression but their role in the NAc, a critical brain region for motivated behavior, is not fully investigated. To provide insight into enkephalin function we used a chronic social defeat stress paradigm, where animals are either categorized as susceptible or resilient to stress based on their performance in a social interaction test. Compared to controls, susceptible animals showed reduced enkephalin levels in the NAc. Such decrease in enkephalin level

Effects of chronic social defeat stress on MAP kinase cascade.

Chronic psychological and social stress can cause psychiatric disorders in humans. In this study, we analyzed the mitogen-activated protein kinase (MAPK) cascade in the hippocampus of chronically socially defeated rats. The rats that were subjected to social defeat every day for 5 weeks showed physiological and behavioral changes, including a reduced rate of weight gain, enlarged adrenal glands, and increased immobility in the forced swim test without concomitant changes in locomotor activity in the open field test. Altered body weight and enlarged adrenal glands are typical symptoms of human

[An emotional stress model using witnessing social defeat scenes in mice].

Chronic exposure to stress can lead to a variety of mental disorders such as depression. There are many treatment-resistant patients who do not respond adequately to the standard pharmacological treatments. Therefore, the development of novel therapeutic agents is highly expected. In rodents, socially defeated animals that were exposed to repeated physical aggression from other individuals are widely used in this field of research. The social defeat is considered as a stress that mimics human social stress. On the other hand, emotional stress, but not physical stress, is likely to contribute t

Submitting to defeat: social anxiety, dominance threat, and decrements in testosterone.

Although theory suggests a link between social anxiety and social dominance, direct empirical evidence for this link is limited. The present experiment tested the hypothesis that socially anxious individuals, particularly men, would respond to a social-dominance threat by exhibiting decrements in their testosterone levels, an endocrinological change that typically reflects pronounced social submission in humans and other animals. Participants were randomly assigned to either win or lose a rigged face-to-face competition with a confederate. Although no zero-order relationship between social anx

Dose-dependent modulation of social and motivational behavior by lurasidone in a rat model of social defeat.

Chronic psychosocial stress is a key risk factor for psychiatric disorders, often causing motivational and social impairments poorly responsive to conventional pharmacotherapies. The present study investigated the effects of chronic lurasidone treatment on behavioral and molecular alterations induced by prolonged social defeat (SD) stress in adult male rats. Rats underwent resident-intruder SD paradigm for approximately seven weeks and treated orally with lurasidone (1 or 3 mg/kg/day) during the final three weeks. Motivational drive was assessed using sucrose self-administration under fixed an

Social defeat interacts with Disc1 mutations in the mouse to affect behavior.

DISC1 (Disrupted-in-schizophrenia 1) is a strong candidate susceptibility gene for psychiatric disease that was originally discovered in a family with a chromosomal translocation severing this gene. Although the family members with the translocation had an identical genetic mutation, their clinical diagnosis and presentation varied significantly. Gene-environment interactions have been proposed as a mechanism underlying the complex heritability and variable phenotype of psychiatric disorders such as major depressive disorder and schizophrenia. We hypothesized that gene-environment interactions

Christensenella minuta mitigates behavioral and cardiometabolic hallmarks of social defeat stress.

Psychological stress during early development and adolescence may increase the risk of psychiatric and cardiometabolic comorbidities in adulthood. The gut microbiota has been associated with mental health problems such as depression and anxiety and with cardiometabolic disease, but the potential role of the gut microbiota in their comorbidity is not well understood. We investigated the effects and mode of action of the intestinal bacterium Christensenella minuta DSM 32891 on stress-induced mental health and cardiometabolic disturbances in a mouse model of social defeat stress. We demonstrate t

Splenic denervation attenuates repeated social defeat stress-induced T-lymphocyte inflammation.

BACKGROUND: Post-traumatic stress disorder (PTSD) is a devastating psychological disorder. Patients with PTSD canonically demonstrate an increased risk for inflammatory diseases, as well as increased sympathetic tone and norepinephrine (NE) outflow. Yet, the exact etiology and causal nature of these physiologic changes remain unclear. Previously, we demonstrated that exogenous NE alters mitochondrial superoxide in T-lymphocytes to produce a pro-inflammatory T-helper 17 (TH17) phenotype, and observed similar TH17 polarization in a preclinical model of PTSD. Therefore, we hypothesized sympatheti

Molecular Adaptations to Social Defeat Stress and Induced Depression in Mice.

Chronic stress is a risk factor for major depression. Social defeat stress is a well-validated murine model of depression. However, little is known about the gene activity dynamics during the development of a depression-like state. We analyzed the effects of social defeat stress of varying duration (10 and 30 days) on the behavioral patterns and prefrontal-cortex transcriptome of C57BL/6 mice. The 10-day exposure to social defeat stress resulted in a high level of social avoidance with no signs of depression-associated behavior. Most animals exposed to 30 days of social defeat stress demonstra

Vicarious Social Defeat Stress Induces Depression-Related Outcomes in Female Mice.

BACKGROUND: Stress is a prevailing risk factor for mood-related illnesses, wherein women represent the majority of those affected by major depression. Despite the growing literature suggesting that affective disorders can arise after a traumatic event is vicariously experienced, this relationship remains understudied in female subjects at the preclinical level. Thus, the objective of the current investigation was to examine whether exposure to emotional and/or psychological stress (ES) mediates depression-related outcomes in female mice. METHODS: Female C57BL/6 mice (8 weeks old, null parity)

Social defeat stress switches the neural system mediating benzodiazepine conditioned motivation.

Benzodiazepines have been demonstrated to have a high abuse liability in persons suffering from anxiety but have demonstrated mixed abuse liability findings in preclinical models. We hypothesized that by modeling anxiety in a male C57BL/6 mouse model it would be possible to reveal a preference for benzodiazepines within this subpopulation through negative reinforcement. Using the Tube Test of Social Dominance and the Resident/Intruder Paradigm we investigated whether animals identified as dominant or submissive/defeated would differentially display a preference for midazolam (a short acting be

Effects of Qingyangshen glycosides on neuroplasticity in a mouse model of social defeat.

BACKGROUND: Qingyangshen (Cynanchum otophyllum C.K. Schneid) is a folk drug for treating depression and other mental disorders induced by social defeat stress. Neuroplasticity in the hippocampus is essential for the modulation of cognition and emotion, and its impairment may contribute to the development and progression of depression. Our previous studies have found that Qingyangshen glycosides (QYS) can improve depression-like behavior in social failure mouse models, mainly through PGC-1α/FNDC5/BDNF signaling pathways activation, but its effects and mechanisms on hippocampal neuroplasticity r

Social Buffering is Dependent on Mutual Experience in Adolescent Male Mice Exposed to Social Defeat Stress.

BACKGROUND: Individuals who experience emotional, physical, or sexual abuse as children suffer from higher rates of major depressive disorder, drug abuse, and suicide. Early life interventions such as peer support groups can be beneficial to adolescents who experience trauma, suggesting that social support is important in facilitating rehabilitation and promoting resiliency to stress. Although there are some animal paradigms that can model how peer-peer interactions influence stress-reactivity, less is known about how individual stress experiences influence the effectiveness of social bufferin

Social Housing Conditions Modulate the Long-Lasting Increase in Cocaine Reward Induced by Intermittent Social Defeat.

Social defeat is considered the most representative animal model for studying the consequences of social stress. Intermittent social defeat (ISD) has proved to enhance the response to cocaine hedonic properties. In the present research, we evaluated if different social housing conditions, as housing with a familiar conspecific or with a female, exert a protective effect modulating the negative consequences of ISD as the increased sensitivity to cocaine and the induction of anxiety-like behavior. To achieve this objective, non-stressed or ISD OF1 male mice were divided into five different exper

Cardiometabolic and anxiogenic consequences of chronic social defeat stress in male mice.

Social adversity, such as that which occurs during subjugation to a lower social status, has profound psychological and cardiometabolic consequences that are conserved across species. Clinically, such adversity often arises from being of a lower socioeconomic status and may contribute to health disparities in cardiometabolic and affective disorders. To develop a better understanding of the cardiometabolic consequences of social adversity, we employ chronic social defeat stress (CSDS) in adult male mice. CSDS results in increases in body mass, that are accompanied by elevated lean and fluid mas

Chronic social defeat stress induces meningeal neutrophilia via type I interferon signaling.

Animal models of stress and stress-related disorders are also associated with blood neutrophilia. The mechanistic relevance of this to symptoms or behavior is unclear. We used cytometry, immunohistochemistry, whole tissue clearing, and single-cell sequencing to characterize the meningeal immune response to chronic social defeat (CSD) stress in mice. We find that chronic, but not acute, stress causes meningeal neutrophil accumulation, and CSD increases neutrophil trafficking in vascular channels emanating from skull bone marrow (BM). Transcriptional analysis suggested CSD increases type I inter

Oxytocin prevents the increase of cocaine-related responses produced by social defeat.

The neuropeptide oxytocin (OXT) plays a critical role in the regulation of social and emotional behaviors. OXT plays a role in stress response and in drug reward, but to date no studies have evaluated its implication in the long-lasting increase of the motivational effects of cocaine induced by repeated social defeat (RSD). During the social defeat procedure, 1 mg/kg of OXT was administered 30 min before each episode of RSD. Three weeks after the last defeat, the effects of cocaine on the conditioned place preference (CPP), locomotor sensitization and the self-administration (SA) paradigms wer

Sympathetic Release of Splenic Monocytes Promotes Recurring Anxiety Following Repeated Social Defeat.

BACKGROUND: Neuroinflammatory signaling may contribute to the pathophysiology of chronic anxiety disorders. Previous work showed that repeated social defeat (RSD) in mice promoted stress-sensitization that was characterized by the recurrence of anxiety following subthreshold stress 24 days after RSD. Furthermore, splenectomy following RSD prevented the recurrence of anxiety in stress-sensitized mice. We hypothesize that the spleen of RSD-exposed mice became a reservoir of primed monocytes that were released following neuroendocrine activation by subthreshold stress. METHODS: Mice were subject

Effects of chronic social defeat stress on social behavior and cognitive flexibility for early and late adolescent.

This study investigated the risk to social behavior and cognitive flexibility induced by chronic social defeat stress (CSDS) during early and late adolescence (EA and LA). Utilizing the "resident-intruder" stress paradigm, adolescent male Sprague-Dawley rats were exposed to CSDS during either EA (postnatal days 29-38) or LA (postnatal days 39-48) to explore how social defeat at different stages of adolescence affects behavioral and cognitive symptoms commonly associated with psychiatric disorders. After stress exposure, the rats were assessed for anxiety-like behavior in the elevated plus maze
