Dietary sodium modulates nephropathy in Nedd4-2-deficient mice.

Salt homeostasis is maintained by tight control of Na+ filtration and reabsorption. In the distal part of the nephron the ubiquitin protein ligase Nedd4-2 regulates membrane abundance and thus activity of the epithelial Na+ channel (ENaC), which is rate-limiting for Na+ reabsorption. Nedd4-2 deficiency in mouse results in elevated ENaC and nephropathy, however the contribution of dietary salt to this has not been characterized. In this study we show that high dietary Na+ exacerbated kidney injury in Nedd4-2-deficient mice, significantly perturbing normal postnatal nephrogenesis and resulting i

Alterations in dietary sodium intake affect cardiovagal baroreflex sensitivity.

High dietary sodium intake has been linked to alterations in neurally mediated cardiovascular function, but the effects of high sodium on cardiovagal baroreflex sensitivity (cBRS) in healthy adults are unknown. The purpose of this study was to determine whether high dietary sodium alters cBRS and heart rate variability (HRV) and whether acute intravenous sodium loading similarly alters cBRS and HRV. High dietary sodium (300 mmol/day, 7 days) was compared with low dietary sodium (20 mmol/day, 7 days; randomized) in 14 participants (38 ± 4 yr old, 23 ± 1 kg/m2 body mass index, 7 women). Acute so

Hypercalciuria in parathyroid disorders: effect of dietary sodium control.

Moderate dietary Na restriction (80 mmol/d for 7 days) during constant Ca intake can reduce high urinary Ca excretion to normal levels in idiopathic hypercalciuria (IH). A similar protocol was used to test its effect in primary hyperparathyroidism (PHPT) and also in hypoparathyroid subjects (HOPT) during treatment with dihydrotachysterol (DHT). Nine subjects with PHPT, 10 with HOPT, and one with pseudo-HOPT were evaluated after Na-restricted (80 mmol/d) and Na-supplemented (200 mmol/d) diets for 7 days each with dietary Ca constant. Na restriction resulted in a decrease in mean urinary 24-hour

Dietary sodium induces a redistribution of the tubular metabolic workload.

KEY POINTS: Body Na+ content is tightly controlled by regulated urinary Na+ excretion. The intrarenal mechanisms mediating adaptation to variations in dietary Na+ intake are incompletely characterized. We confirmed and expanded observations in mice that variations in dietary Na+ intake do not alter the glomerular filtration rate but alter the total and cell-surface expression of major Na+ transporters all along the kidney tubule. Low dietary Na+ intake increased Na+ reabsorption in the proximal tubule and decreased it in more distal kidney tubule segments. High dietary Na+ intake decreased Na+

The health impacts of dietary sodium and a low-salt diet.

High salt intake is now endemic worldwide. It contributes to the generation and maintenance of high blood pressure, which is now the biggest risk factor for global disease. There is now compelling evidence to support salt reduction in hypertensives and a substantial body of evidence to support salt reduction in the general population to reduce risk of death from cardiovascular disease. In specific diseases such as heart failure and chronic kidney disease, guidelines support the World Health Organization target for reduced salt intake at 5 g daily. Achieving a diet that is lower in salt has cha

Dietary sodium alters aldosterone's effect on renal sodium transporter expression and distal convoluted tubule remodelling.

Aldosterone is responsible for maintaining volume and potassium homeostasis. Although high salt consumption should suppress aldosterone production, individuals with hyperaldosteronism lose this regulation, leading to a state of high aldosterone despite dietary sodium consumption. The present study examines the effects of elevated aldosterone, with or without high salt consumption, on the expression of key Na+ transporters and remodelling in the distal nephron. Epithelial sodium channel (ENaC) α-subunit expression was increased with aldosterone regardless of Na+ intake. However, ENaC β- and γ-s

Sodium space and intravascular volume: dietary sodium effects in cystic fibrosis and healthy adolescent subjects.

OBJECTIVE: To assess the physiologic response to salt depletion in subjects with cystic fibrosis (CF) and control male adolescents for sodium balance, sodium space, and stimulation of the renin-angiotensin-aldosterone axis. DESIGN: Seven subjects with CF and six controls received a salt-replete (150 or 290 mmol NaCl per day) diet and then a salt-deplete (10 mmol NaCl per day) diet while in a clinical research center. RESULTS: Space maintenance: CF subjects responded to salt depletion with a greater weight loss than did controls (1.9 vs 0.8 kg) and a decrease in 24Na+ space, whereas controls

Effect of Dietary Sodium on Blood Pressure: A Crossover Trial.

IMPORTANCE: Dietary sodium recommendations are debated partly due to variable blood pressure (BP) response to sodium intake. Furthermore, the BP effect of dietary sodium among individuals taking antihypertensive medications is understudied. OBJECTIVES: To examine the distribution of within-individual BP response to dietary sodium, the difference in BP between individuals allocated to consume a high- or low-sodium diet first, and whether these varied according to baseline BP and antihypertensive medication use. The role of dietary sodium in autoimmune diseases: The salty truth.

Autoimmune diseases are a group of heterogeneous condition that occur secondary to the intrinsic loss of tolerance to self- antigens. In genetically susceptible individuals, the complex interplay of environmental factors and epigenetic deregulations have been proposed to drive disease etiopathogenesis. Various environmental variables have been identified including viral infections, exposure to pollutants, stress and dietary factors. Sodium, a major constituent of salt is essential for mammalian physiology. However, high salt intake may play a role in the development of autoimmune diseases. Sev

Mineralocorticoid-stimulated renal acidification: the critical role of dietary sodium.

Recent in vitro studies of isolated distal nephron segments have demonstrated that mineralocorticoid hormone stimulates H+ secretion by both Na+-dependent and Na+-independent mechanisms, and the Na+-independent acidification mechanism has a greater capacity. These in vitro data suggest that mineralocorticoid administration in vivo might increase renal acid excretion when an augmentation in distal Na+ reabsorption is precluded by rigid restriction of dietary Na+; under these circumstances, virtually all Na+ delivered to the distal nephron is reabsorbed in the basal state. In the present studies

The Impact of Modern Industrialized Dietary Sodium Intake on the Plasma Proteome.

BACKGROUND: High dietary sodium intake is associated with cardiovascular disease. We investigated the influence of sodium intake on the plasma proteome. METHODS: Prospectively recruited normotensive participants underwent 2 controlled dietary sodium interventions to evaluate hormonal and proteomic (1,512 proteins) changes: sodium-restriction resembling ancestral hunter-gatherer intake (~10 mEq/day, ~230 mg/day) and sodium-loading resembling modern industrialized intake (~200 mEq/day, ~4,600 mg/day). Twenty-four hour urine collections were obtained after each diet. Plasma proteomic changes wer

Effects of dietary sodium and genetic background on angiotensinogen and Renin in mouse.

Elements of a renin-angiotensin system expressed along the entire nephron, including angiotensinogen secreted by proximal tubule and renin expressed in connecting tubule, may participate in the regulation of sodium reabsorption at multiple sites of the nephron. The response of this tubular renin-angiotensin system to stepwise changes in dietary sodium was investigated in 2 mouse strains, the sodium-sensitive inbred C57BL/6 and the sodium-resistant CD1 outbred. Plasma angiotensinogen was not affected by sodium regimen, whereas plasma renin increased 2-fold under low sodium. In both strains, the

Sodium reduction is the key ingredient in dietary treatment of hypertension - a randomized controlled trial on sodium, potassium and nitrate.

Dietary interventions are recommended as part of hypertension (HTN) treatment. Sodium reduction has proven effective, however supplements of potassium and/or nitrate are likely also of importance. We aimed to test how these three dietary components affect blood pressure (BP) individually and in combination in patients with HTN. A double-blind randomized controlled trial of 90 participants with HTN was conducted. Participants were randomized to eight groups receiving a hand-out diet and different combinations of supplementation of sodium chloride (130 mmol/day), potassium chloride (40 mmol/day)

Research Note: Evaluation of dietary administration of sodium chlorate and sodium nitrate for Histomonas meleagridis prophylaxis in turkeys.

Histomoniasis is currently a re-emerging disease of major significance for many commercial turkey and broiler breeder production companies because of the unavailability of drugs or vaccines. The protozoa Histomonas meleagridis (HM) requires the presence of enteric microflora to promote the disease. The objectives of this research note were to evaluate the effect of dietary administration of sodium chlorate (SC) and sodium nitrate (SN) in vitro and in vivo for HM prophylaxis in poults. A total of 128 day-of-hatch female poults obtained from a commercial hatchery were wing-tagged and randomly as

Reducing Dietary Sodium Exposure in Chronic Kidney Disease: Challenges and Opportunities.

Over 750 million individuals worldwide experience chronic kidney disease (CKD), which is a major contributor to cardiovascular disease and kidney failure as well as increased morbidity and mortality. Diet is a key lifestyle factor in CKD risk prevention and management. Overall public dietary quality and sodium intake have shown limited improvement despite decades of public health efforts. Excessive sodium consumption is a challenging dietary risk factor because exposure is largely passively embedded within the modern food environment. Furthermore, the consequences of overconsumption of dietary

Dietary Sodium Reduction Reveals Aldosterone Dysregulation in Patients With Essential Hypertension.

Dietary sodium reduction affects blood pressure (BP), the renin-angiotensin-aldosterone system (RAAS) and natriuretic peptides. Whether baseline levels of these hormones impact BP response to sodium reduction remains unknown. Aldosterone dysregulation relates to higher cardiovascular risk, but the relation to sodium reduction has not been uncovered. We conducted a randomized clinical trial of 72 patients with hypertension, who were randomised 2:1 to four weeks of dietary sodium reduction or a control group. 24-h BP and 24-h urine collection were performed and blood samples (renin, aldosterone,

Mineralocorticoid receptor modulation by dietary sodium influences NAFLD development in mice.

INTRODUCTION AND OBJECTIVES: Nonalcoholic-fatty-liver disease (NAFLD) is considered the hepatic manifestation of metabolic syndrome (MetS). Mineralocorticoid receptor (MR) activation is associated with increased risk of MetS but few studies have assessed the role of liver MR on NAFLD. We aimed to evaluate the effect of MR modulation by sodium intake in liver injury in experimental models of NAFLD. MATERIALS AND METHODS: C57BL/6J mice were fed either a high-fat-diet (HFD) or a choline/methionine deficient (MCD) diet with different sodium concentrations. Hepatic concentration of lipid species,
