Sodium benzoate, a common food preservative-induced Insulin resistance and atrophy in C2C12 myotubes.

Sodium benzoate (SB), a widely used food preservative, has been reported to possess several therapeutic benefits including treating neurodegenerative disorders as well as negative effects such as altering glucose homeostasis. However, limited attention has been given to its role in inducing insulin resistance (IR). Therefore, current study was designed to investigate the relationship between SB and insulin resistance using C2C12 myotubes and to explore the possible underlying mechanisms. Myotubes were exposed to SB (20 mM) for 24 h and glucose consumption and uptake assays along with confocal

Batrachotoxinin-A 20-alpha-benzoate: a new radioactive ligand for voltage sensitive sodium channels.

Batrachotoxinin-A 20-alpha-benzoate (BTX-B), an analog of the potent depolarizing agent batrachotoxin (BTX), was prepared by selective esterification of naturally occurring batrachotoxin-A with benzoic acid. BTX-B depolarized rat phrenic nerve-diaphragm preparations with a time course and concentration dependence virtually indistinguishable from that of BTX. A specific, saturable component of equilibrium binding of [3H]BTX-B to mouse cerebral cortex homogenates was measured, described by an equilibrium dissociation constant of 0.7 microM and a maximum number of binding sites of 90 pmol per gra

Stability Study of a 100 mg/mL Sodium Benzoate Solution in Water for Parenteral Use.

CONTEXT: Sodium benzoate is mainly used as a preservative in food and pharmaceuticals and as a treatment for hyperammonemia in urea-cycle disorders. It is typically diluted in 5 or 10% dextrose solutions for intravenous administration to patients. While the stability of oral and injectable sodium benzoate formulations has been studied, there is limited research on its stability as an injectable solution under International Council for Harmonisation (ICH) conditions. AIM: This study evaluates the stability of a sodium benzoate injectable formulation in sterile water (100 mg/mL). METHODS: A 10

Effects of sodium benzoate on cognitive function in neuropsychiatric disorders: a systematic review and meta-analysis.

UNLABELLED: We performed a systematic review and meta-analysis on sodium benzoate's effects on cognitive function and other psychiatric symptoms in individuals with neuropsychiatric disorders. We searched PubMed, Embase, Cochrane Library, and PsychInfo databases until September 2023. A random-effects meta-analysis was performed within a frequentist framework. To investigate the potential sources of heterogeneity, we performed subgroup analyses based on sex, dose, diagnosis, and risk of bias of the included studies. Trial sequential analyses were performed to investigate the statistical power o

Sodium benzoate for the treatment of hepatic encephalopathy in humans and animals: a systematic review and meta-analysis.

BACKGROUND AND AIM: Hepatic encephalopathy (HE) is a life-threatening condition where brain function is impaired mainly due to high systemic ammonia levels. HE is associated with a high 1-year mortality. No universally accepted guidelines for the treatment of HE exist. Nitrogen scavengers, such as sodium benzoate (SB), have been proven very effective to treat hyperammonemia in patients with urea cycle defects, in acute and chronic settings. We hypothesized that SB can also be an effective treatment of HE caused by end-stage liver disease or portosystemic shunting, as long as liver function is

Effects of Sodium Benzoate Treatment in Combination with An Extinction Training on the Maintenance of Cocaine-Supported Memory.

Activation of N-methyl-D-aspartate (NMDA) receptor can facilitate the extinction of various maladaptive memories. Sodium benzoate (NaB) has been known to enhance a naturally occurring full agonist on the glycine binding site of the NMDA receptor. This study aimed to test whether systemic NaB treatment can affect the extinction of a cocaine-supported memory, the cocaine-induced conditioned place preference (CPP). Following the establishment of the cocaine (10 mg/kg/conditioning × 3)-induced CPP, an extinction protocol, consisting of two consecutive extinction training bouts at an 8-h interval,

Efficacy and safety of i.v. sodium benzoate in urea cycle disorders: a multicentre retrospective study.

BACKGROUND: The efficacy and safety of intra-venous (i.v.) sodium benzoate for treating acute episodes of hyperammonemia in urea cycle enzyme disorders (UCD) is well known. However, published data do not provide a clear picture of the benefits and risks of this drug. We report a retrospective multicentre study on the use of i.v. sodium benzoate in patients treated for UCD between 2000 and 2010 in the 6 French reference centres for metabolic diseases. RESULTS: Sixty-one patients with UCDs - 22 ornithine transcarbamylase (20 confirmed, 2 suspected), 18 arginino-succinate synthetase, 15 carbamoy

Protective effect of resveratrol on oxidative stress in rats exposed to elevated doses of sodium benzoate.

BACKGROUND: Sodium benzoate is a widely used food preservative permitted in Ukraine with an acceptable daily intake of 5 mg/kg body weight. However, exposure to elevated doses may induce oxidative imbalance and metabolic disturbances. Therefore, the search for effective protective agents against sodium benzoate-induced oxidative stress remains relevant. Resveratrol is a natural polyphenolic compound known for its antioxidant, anti-inflammatory, and cytoprotective properties. OBJECTIVE: To evaluate the effect of resveratrol on free radical processes and antioxidant defense parameters in rats e

Free and esterified coenzyme A in the liver and muscles of chronically hyperammonemic mice treated with sodium benzoate.

Ammonia toxicity and relative sodium benzoate toxicity alters the energy metabolism, leading to a decrease of adenosine triphosphate and free coenzyme A levels. The object of the present study was to analyze the hepatic and muscular acyl-coenzyme A profiles in chronically hyperammonemic mice treated with varying doses of the sodium benzoate. An enzymatic method was used for the measurement of free coenzyme A, acetyl-coenzyme A, and medium and long chain acyl-coenzyme A. Untreated chronic hyperammonemia resulted in a decrease in acetyl-coenzyme A and an increase in the long chain acyl-coenzyme

Effect of sodium benzoate on cerebral and hepatic energy metabolites in spf mice with congenital hyperammonemia.

The sparse-fur (spf) mutant mouse has an X-linked deficiency of ornithine transcarbamylase and develops congenital hyperammonemia similar to that seen in human patients. We studied the effect of sodium benzoate (2.5, 5 and 10 mmol/kg body wt) on ammonia, glutamine and glutamate, as well as various intermediates of energy metabolism in brain and liver of normal CD-1/Y and hyperammonemic spf/Y mice. The ammonia concentration of brain was decreased with 2.5 mmol sodium benzoate in spf/Y mice, whereas higher doses resulted in a significant increase in both liver and brain. Cerebral glutamine conte

The mode of action of ethanol on batrachotoxinin-A benzoate binding to sodium channels in mouse brain cortex.

Since ethanol has local anesthetic activity its effect was examined in vitro on the scorpion toxin-enhanced specific binding of [3H]batrachotoxinin A 20-alpha-benzoate ([3H]BTX-B) to the gating complex in sodium channel preparations from mouse brain cortex by equilibrium and kinetic experiments. Ethanol inhibited the specific binding of [3H]BTX-B in a vesicular preparation with an IC50 value of 310 mM and a Hill number of 1.0. Ethanol increased the equilibrium dissociation constant of batrachotoxin in a concentration dependent manner without changing the maximal binding capacity and decreased

Inhibition of binding of [3H]batrachotoxinin A 20-alpha-benzoate to sodium channels by local anesthetics.

The effects of several local anesthetics on the binding of ligands to receptors associated with voltage-sensitive sodium channels in rat brain synaptosomes have been examined. In the presence of 0.3 microM scorpion toxin, the 13 local anesthetics tested inhibited the specific binding of [3H]batrachotoxinin A 20 alpha-benzoate [( 3H]BTX-B), a ligand which binds to a receptor site responsible for the activation of sodium channel ion flux, in a dose-dependent fashion, with KD values ranging from 1.2 microM for tetracaine to 1.58 mM for benzocaine. A plot of log KD from these binding experiments a

Cerebrospinal fluid glycine in nonketotic hyperglycinemic: effect of treatment with sodium benzoate and a ventricular shunt.

In three infants with nonketotic hyperglycinemia, glycine was increased three-to fourfold in plasma, 13- to 28-fold in lumbar spinal fluid, and was higher yet in ventricular fluid. Oral sodium benzoate lowered cerebrospinal fluid (CSF) glycine by greater than 40%, but did not change the abnormal plasma: CSF ratio. An adult control, made hyperglycinemic with oral glycine, had a normal plasma: CSF ratio. Treatment of one patient with sodium benzoate from birth did not prevent mental retardation; the degree of brain stem depression was a function of CSF glycine in another patient. The persistance

Poor outcome for neonatal-type nonketotic hyperglycinemia treated with high-dose sodium benzoate and dextromethorphan.

Neonatal-type nonketotic hyperglycinemia treatment remains unsatisfactory, even if started early. A review of six patients who underwent treatment for neonatal-type nonketotic hyperglycinemia in our hospital is presented. All patients were treated with a standardized protocol. Medical histories were retrieved from case notes. All six patients had elevated cerebrospinal fluid plasma glycine levels initially. All but one had received sodium benzoate and dextromethorphan from 1 month of age. All suffered from intractable seizures and severe mental retardation, and only two patients remain alive.
