Development of a Green, Quick, and Efficient Method Based on Ultrasound-Assisted Extraction Followed by HPLC-DAD for the Analysis of Bioactive Glycoalkaloids in Potato Peel Waste.

α-Solanine and α-chaconine are the two most predominant glycoalkaloids (GAs) present in potato. Potato peel contains a high concentration of GAs, which are especially interesting for application in the pharmaceutical industry due to their different beneficial properties (such as anticarcinogenic, anti-inflammatory, antiallergic, antipyretic, antiviral, fungicide, and antibiotic activities, among others); so, potato peel waste can be valorized by extracting these biologically active compounds. For this, a green, quick, and efficient miniaturized analytical approach based on ultrasound-assisted

Developmental toxicology of potato alkaloids in the frog embryo teratogenesis assay--Xenopus (FETAX).

Potatoes frequently contain growth inhibitors and toxic compounds including digestive enzyme inhibitors, lectins and glycoalkaloids. The literature suggests that Solanum alkaloids have the ability to induce neurological damage such as spina bifida and other malformations. As part of a programme of improvement in the safety of potatoes using molecular plant genetics and parallel food safety evaluation, we evaluated the effect of several potato glycoalkaloids and aglycones in the frog embryo teratogenesis assay--Xenopus (FETAX) with and without metabolic activation by Aroclor 1254-induced rat li

Toxic Risks of Nightshade Species: A comprehensive review of the documented toxicity of Atropa belladonna, Solanum dulcamara, and Solanum nigrum.

The Solanaceae family comprises numerous species of economic and pharmacological importance, including the toxic deadly nightshade (Atropa belladonna), bittersweet nightshade (Solanum dulcamara), and black nightshade (Solanum nigrum). These plants are native to Europe, North Africa, and Western Asia, and have been used since ancient times in both ritual practices and medicine. A. belladonna contains potent tropane alkaloids (atropine, hyoscyamine, and scopolamine) and is widely utilized in modern pharmacology, although accidental poisonings still occur, often due to confusion with edible plant

Normal and atypical butyrylcholinesterases in placental development, function, and malfunction.

1. In utero exposure to poisons and drugs (e.g., anticholinesterases, cocaine) is frequently associated with spontaneous absorption and placental malfunction. The major protein interacting with these compounds is butyrylcholinesterase (BuChE), which attenuates the effects of such xenobiotics by their hydrolysis or sequestration. Therefore, we studied BuChE expression during placental development. 2. RT-PCR revealed both BuChEmRNA and acetylcholinesterase (AChE) mRNA throughout gestation. However, cytochemical staining detected primarily BuChE activity in first-trimester placenta but AChE activ

Synthesis and insecticidal activities of novel solanidine derivatives.

BACKGROUND: Potato (Solanum tuberosum) is the fourth culture in the world and is widely used in the agri-food industries. They generate by-products in which α-chaconine and α-solanine, the two major solanidine-based glycoalkaloids of potato, are present. As secondary metabolites, they play an important role in the protection system of potato and are involved in plant protection against insects. To add value to these by-products, we described here new glycoalkaloids that could have phytosanitary properties. RESULTS: Solanidine, as a renewable source, was modified with an azido linker and coupl

Tissue distribution of human acetylcholinesterase and butyrylcholinesterase messenger RNA.

Cholinesterase inhibitors occur naturally in the calabar bean (eserine), green potatoes (solanine), insect-resistant crab apples, the coca plant (cocaine) and snake venom (fasciculin). There are also synthetic cholinesterase inhibitors, for example man-made insecticides. These inhibitors inactivate acetylcholinesterase and butyrylcholinesterase as well as other targets. From a study of the tissue distribution of acetylcholinesterase and butyrylcholinesterase mRNA by Northern blot analysis, we have found the highest levels of butyrylcholinesterase mRNA in the liver and lungs, tissues known as t

Central diabetes insipidus following digestion Solanum indicum L. concentrated solution.

In Taiwan, Solanum indicum L. has been used in folk medicine for the treatment of inflammation, toothache, ascites, edema, and wound infection. The plant is rich in solanine, an alkaloidal glycoside. We report a 43-year-old man who developed polyuria and polydipsia after taking seven doses of concentrated solution of Solanum indicum L. over two weeks. A water deprivation test and a low serum antidiuretic hormone level helped to confirm a diagnosis of central diabetes insipidus. We suggest that excessive doses of Solanum indicum L. may cause central diabetes insipidus.

Risk assessment of glycoalkaloids in feed and food, in particular in potatoes and potato-derived products.

The European Commission asked EFSA for a scientific opinion on the risks for animal and human health related to the presence of glycoalkaloids (GAs) in feed and food. This risk assessment covers edible parts of potato plants and other food plants containing GAs, in particular, tomato and aubergine. In humans, acute toxic effects of potato GAs (α-solanine and α-chaconine) include gastrointestinal symptoms such as nausea, vomiting and diarrhoea. For these effects, the CONTAM Panel identified a lowest-observed-adverse-effect level of 1 mg total potato GAs/kg body weight (bw) per day as a referenc

Ma-khwaen essential oil vapour suppresses potato sprouting under tropical ambient storage via GA3-ABA modulation, oxidative stress and carbohydrate metabolism.

BACKGROUND: Sprouting is a major postharvest challenge in potato storage, accelerating physiological ageing, starch mobilisation and α-solanine accumulation, thereby reducing processing quality and food safety. With the phase-out of hazardous synthetic sprout inhibitors, safe and residue-free alternatives are needed. This study evaluated the vapour-phase efficacy of three essential oils (EOs; lime peel, holy basil and ma-khwaen) applied at a nominal loading of approximately 70 μL L-1 (v/v) as overnight fumigation to 'Spunta' potato tubers stored under ambient conditions (28 ± 4 °C, 65-83% rela

Effect of carbohydrates on the production of thaxtomin A by Streptomyces acidiscabies.

Several Streptomyces species cause plant diseases, including S. scabies, S. acidiscabies and S. turgidiscabies, which produce common scab of potato and similar diseases of root crops. These species produce thaxtomins, dipeptide phytotoxins that are responsible for disease symptoms. Thaxtomins are produced in vivo on diseased potato tissue and in vitro in oat-based culture media, but the regulation of thaxtomin biosynthesis is not understood. S. acidiscabies was grown in a variety of media to assess the impact of medium components on thaxtomin A (ThxA) production. ThxA biosynthesis was not corr

Preparation of methyl salicylate nanoemulsion and its application in sprouting inhibition on potato tubers.

Sprouting of potato tubers is accompanied by the accumulation of toxic solanine. In this study, a methyl salicylate (MeSA) nanoemulsion was prepared using ultrasonic emulsification, and its effect on potato tuber sprouting was investigated. Results showed that the optimum preparation conditions of MeSA nanoemulsion were as follows: surfactant concentration, 5.02 %; MeSA concentration, 9.79 %; ultrasonic power, 428.03 W; and ultrasonic time, 3.79 min. The results of Fourier transform infrared spectroscopy and rheological properties of the emulsion showed that MeSA was successfully embedded in t

Comparative metabolite profiling of Solanum tuberosum against six wild Solanum species with Colorado potato beetle resistance.

The Colorado potato beetle Leptinotarsa decemlineata (Say) (CPB) is a coleopteran herbivore that feeds on the foliage on Solanum species, in particular, potato. Six resistant wild Solanum species were identified, and two of these species had low levels of glycoalkaloids. Comparative analysis of the untargeted metabolite profiles of the foliage using UPLC-qTOF-MS was done to find metabolites shared between the wild species but not with Solanum tuberosum (L.) to identify resistance-related metabolites. It was found that only S. tuberosum produced the triose glycoalkaloids solanine and chaconine.

Glycoalkaloid and calystegine levels in table potato cultivars subjected to wounding, light, and heat treatments.

Potato tubers naturally contain a number of defense substances, some of which are of major concern for food safety. Among these substances are the glycoalkaloids and calystegines. We have here analyzed levels of glycoalkaloids (α-chaconine and α-solanine) and calystegines (A₃, B₂, and B₄) in potato tubers subjected to mechanical wounding, light exposure, or elevated temperature: stress treatments that are known or anticipated to induce glycoalkaloid levels. Basal glycoalkaloid levels in tubers varied between potato cultivars. Wounding and light exposure, but not heat, increased tuber glycoalka

Antifungal activity of secondary plant metabolites from potatoes (Solanum tuberosum L.): Glycoalkaloids and phenolic acids show synergistic effects.

AIMS: To study the antifungal effects of the potato secondary metabolites α-solanine, α-chaconine, solanidine and caffeic acid, alone or combined. METHODS AND RESULTS: Resistance to glycoalkaloids varied among the fungal species tested, as derived from minimum inhibitory concentrations assays. Synergistic antifungal activity between glycoalkaloids and phenolic compounds was found. Changes in the fluidity of fungal membranes caused by potato secondary plant metabolites were determined by calculation of the generalized polarization values. The results partially explained the synergistic effect

Review of the inhibition of biological activities of food-related selected toxins by natural compounds.

There is a need to develop food-compatible conditions to alter the structures of fungal, bacterial, and plant toxins, thus transforming toxins to nontoxic molecules. The term 'chemical genetics' has been used to describe this approach. This overview attempts to survey and consolidate the widely scattered literature on the inhibition by natural compounds and plant extracts of the biological (toxicological) activity of the following food-related toxins: aflatoxin B1, fumonisins, and ochratoxin A produced by fungi; cholera toxin produced by Vibrio cholerae bacteria; Shiga toxins produced by E. co

Comparative transcriptome and metabolome profiling uncovers tissue-specific mechanisms in potato flesh and skin.

Potato (Solanum tuberosum L.), a globally cultivated tuber crop, is crucial for food security. The metabolic specialization between tuber skin and flesh directly influences tuber stress resistance and quality. Transcriptome-metabolome analysis of two cultivars (IAW30 and IAY31) revealed tissue-specific metabolic profiles and molecular mechanisms. Flesh accumulates primary metabolites such as saccharides, amino acids and derivatives, and vitamins, whereas skin is enriched in secondary metabolites including glycoalkaloids and flavonoids. Marker metabolite analysis identified four flesh-specific
