Sonication of removed implants improves microbiological diagnosis of postoperative spinal infections.

PURPOSE: In total joint replacement, culturing of the sonication fluid of removed implants has proven to be more sensitive than conventional periprosthetic tissue culture for the microbiological diagnosis of prosthetic joint infection. However, its role in postoperative spinal implant infection (PSII) is not well investigated. Therefore, the aim of this study was to determine the validity of sonication in detecting infection following instrumented spine surgery. METHODS: In this prospective controlled consecutive cohort study, patients undergoing spinal revision between September 2016 and Mar

Sonication of Arthroplasty Implants Improves Accuracy of Periprosthetic Joint Infection Cultures.

BACKGROUND: There is evidence that sonication of explanted prosthetic hip and knee arthroplasty components with culture of the sonication fluid may enhance diagnostic sensitivity. Previous studies on the use of implant sonicate cultures have evaluated diagnostic thresholds but did not elaborate on the clinical importance of positive implant sonicate cultures in the setting of presumed aseptic revisions and did not utilize consensus statements on periprosthetic joint infection (PJI) diagnosis when defining their gold standard for infection. QUESTIONS/PURPOSES: (1) How do implant sonicate cultu

Novel quantitative autophagy analysis by organelle flow cytometry after cell sonication.

Autophagy is a dynamic process of bulk degradation of cellular proteins and organelles in lysosomes. Current methods of autophagy measurement include microscopy-based counting of autophagic vacuoles (AVs) in cells. We have developed a novel method to quantitatively analyze individual AVs using flow cytometry. This method, OFACS (organelle flow after cell sonication), takes advantage of efficient cell disruption with a brief sonication, generating cell homogenates with fluorescently labeled AVs that retain their integrity as confirmed with light and electron microscopy analysis. These AVs could

The enhancement of hydration of fluidized bed combustion ash by sonication.

The use of limestone to control SO2 emissions in fluidized bed combustors (FBC) results in high CaO content in the ashes. This presents challenges for their disposal including significant exothermic behavior and uncontrolled expansion in the landfill. Hydration of the ashes is required to convert the CaO, but the current two-step hydration process is not very effective. In the present work a new technique using ultrasound to promote the hydration was examined. Initial work was done using an ultrasonic bath and subsequently an ultrasonic probe. Hydration levels greater than 80% in 20-40 min wit

The Significance of Sonication in the Detection of Peri-Implant Infections.

Objective: The current gold standard for detection of implant-related infections is the intraoperative collection of tissue samples. However, false-negative results frequently occur, particularly in infections caused by biofilm-forming bacteria. As a complementary method, sonication has therefore been established for detecting implant-associated infections, especially in periprosthetic joint infections. In trauma surgery, this technique is still rarely used. The aim of this study is to evaluate the diagnostic significance of sonication after osteosynthesis. Methods: A retrospective single-cent

Differentiating intracellular from extracellular alkaline phosphatase activity in soil by sonication.

Differentiating intracellular from extracellular enzyme activity is important in soil enzymology, but not easy. Here, we report on an adjusted sonication method for the separation of intracellular from extracellular phosphatase activity in soil. Under optimal sonication conditions [soil:water ratio  =  1/8 (w/v) and power density  =  15 watt ml(-1)], the activity of alkaline phosphomonoesterase (phosphatase) in a Haplic Cambisol soil increased with sonication time in two distinct steps. A first plateau of enzyme activity was reached between 60 and 100 s, and a second higher plateau after 300 s

Improved colony forming unit recovery from aggregating lactobacilli via mild probe sonication.

Colony forming unit (CFU)-based enumeration of culturable bacterial populations is essential for microbiological research, probiotic quality control, and regulatory compliance. However, this is compromised in highly aggregative lactobacilli strains because individual viable cells within an aggregate often coalesce to produce a single colony. This study evaluated physical and chemical strategies to enhance CFU recovery by dispersing bacterial aggregates while preserving bacterial membrane integrity. Probe sonication, bath ultrasonication, and treatments with sodium chloride (NaCl), lithium chlo

Debunking top-down borophene: a multimodal reassessment of boron sonication and milling.

The rapid growth of reports claiming the top-down synthesis of free-standing borophene from boron has generated substantial ambiguity regarding the true nature of the resulting materials. Here, representative sonication- and milling-based processing routes were investigated through multimodal structural, chemical, and optical characterization. Boron powders were treated by bath sonication in water, hydrogen peroxide, dimethylformamide, and isopropanol, as well as by ball milling under air and argon atmospheres. In all cases, no evidence of known borophene polymorphs was observed. Instead, the

ASAP─Automated Sonication-Free Acid-Assisted Proteomes─from Cells and FFPE Tissues.

Formalin-fixed, paraffin-embedded (FFPE) tissues are an invaluable resource for retrospective studies, but protein extraction and subsequent sample processing steps have been shown to be challenging for mass spectrometry (MS) analysis. Streamlined high-throughput sample preparation workflows are essential for efficient peptide extraction from complex clinical specimens such as fresh frozen tissues or FFPE. Overall, proteome analysis has gained significant improvements in the instrumentation, acquisition methods, sample preparation workflows, and analysis pipelines, yet even the most recent FFP

Development of a Low-Frequency Piezoelectric Ultrasonic Transducer for Biological Tissue Sonication.

The safety of ultrasound exposure is very important for a patient's well-being. High-frequency (1-10 MHz) ultrasound waves are highly absorbed by biological tissue and have limited therapeutic effects on internal organs. This article presents the results of the development and application of a low-frequency (20-100 kHz) ultrasonic transducer for sonication of biological tissues. Using the methodology of digital twins, consisting of virtual and physical twins, an ultrasonic transducer has been developed that emits a focused ultrasound signal that penetrates into deeper biological tissues. For t

Usefulness of sonication procedure in mesh infection diagnosis associated with hernia repair.

BACKGROUND: The use of prosthetic meshes is a common practice in hernia repair surgery. However, infection can appear as an important complication where antibiotic selection must be directed by the etiology of the infection. In recent years, sonication has appeared as an important tool for the diagnosis of many biomaterial-associated infections. Here, we evaluated our experience with this methodology for the diagnosis of mesh infection. METHODS: We retrospectively reviewed the microbiological records between 2015 and 2019 looking for sonicated meshes in the microbiology laboratory. All sample

Tip-Sonication-Induced Dilute Alloying of Metal Pairs with Limited Bulk-Immiscibility.

Alloying metals with unfavorable bulk mixing thermodynamics remains challenging because conventional synthesis often favors elemental segregation rather than compositionally mixed nanoparticles. This study aimed to determine whether direct tip sonication can promote compositional intermixing in metal pairs with limited bulk miscibility and to distinguish sonication-specific effects from conventional thermal and chemical effects. Using Au-Mo as a model system, tip sonication of gold acetate and molybdenum hexacarbonyl in oleic acid successfully produced Au-Mo intermixed nanoparticles with an av

Novel method for reduction of virus load in blood plasma by sonication.

BACKGROUND: Aim of the present study is the evaluation of ultrasound as a physical method for virus inactivation in human plasma products prior to transfusion. Our study is focused on achieving a high level of virus inactivation simultaneously leaving blood products unaltered, measured by the level of degradation of coagulation factors, especially in third world countries where virus contamination of blood products poses a major problem. Virus inactivation plays an important role, especially in the light of newly discovered or unknown viruses, which cannot be safely excluded via prior testing.

Meta-analysis of sonication prosthetic fluid PCR for diagnosing periprosthetic joint infection.

Periprosthetic joint infection (PJI) is a catastrophic complication following total joint arthroplasty. Until now, the diagnosis of PJI is still confronted with difficulties, which is characterized by technical limitations. The question of whether sonication fluid PCR can provide high value in the diagnosis of PJI remains unanswered. This meta-analysis included 9 studies that evaluated PCR assays of sonication fluid for the diagnosis of PJI. The pooled sensitivity, specificity, Positive likelihood ratio (PLR), Negative likelihood ratio (NLR) and Diagnostic odds ratio (DOR) were 0.75 (95% confi
