Sexually dimorphic effects of lipopolysaccharide on spatial memory.

Deficits in hippocampal-dependent memory tasks following lipopolysaccharide (LPS) administration are frequently reported. However, prior work has predominantly been conducted with males. Given recent reports showing sex-related differences in the cognitive effects of an acute LPS challenge, the present study evaluated whether sex differences in spatial learning and memory exist. Adult female and male C57BL/6 J mice were evaluated for spatial learning and memory in a hidden platform version of the water maze following administration of LPS (0.25 mg/kg) prior to the first day of training. Navigation strategies predict spatial memory and hippocampal recruitment.

Navigation in the real world may rely on a multitude of strategies with distinct advantages and disadvantages; however, the characterization of spontaneous strategy use is scarce. We explore data across three groups engaged in virtual navigation: patients with epilepsy undergoing invasive electrophysiological recordings, two groups undergoing fMRI scanning: young adults at genetic risk for Alzheimer's disease (APOE-ε4 carriers), and healthy controls. We establish metrics to quantitatively characterize participants' trajectories, reflecting putative strategies: straightness, deviation to bounda

Cholinergic modulation of spatial learning, memory and navigation.

Spatial learning, including encoding and retrieval of spatial memories as well as holding spatial information in working memory generally serving navigation under a broad range of circumstances, relies on a network of structures. While central to this network are medial temporal lobe structures with a widely appreciated crucial function of the hippocampus, neocortical areas such as the posterior parietal cortex and the retrosplenial cortex also play essential roles. Since the hippocampus receives its main subcortical input from the medial septum of the basal forebrain (BF) cholinergic system,

Human hippocampus and viewpoint dependence in spatial memory.

Virtual reality was used to sequentially present objects within a town square and to test recognition of object locations from the same viewpoint as presentation, or from a shifted viewpoint. A developmental amnesic case with focal bilateral hippocampal pathology showed a massive additional impairment when tested from the shifted viewpoint compared with a mild, list length-dependent, impairment when tested from the same viewpoint. While the same-view condition could be solved by visual pattern matching, the shifted-view condition requires a viewpoint independent representation or an equivalent

Spatial organization of tissue-resident memory T cells.

Tissue-resident memory T (TRM) cells durably reside in peripheral tissues, poised to deliver rapid local immune protection. TRM cells are found in most organs, and they are a core component of immunological memory, complementing circulating memory T cell populations. By definition, TRM cells can persist in tissues for months to years without antigen stimulation. However, demonstrating both residency and antigen absence in reality is challenging. Here, we discuss the strengths and challenges to our canonical definitions of TRM cells. We examine how early priming and subsequent tissue imprinting

Improved spatial memory for physical versus virtual navigation.

Objective. Virtual reality (VR) has become a key tool for researching spatial memory. Virtual environments offer many advantages for research in terms of logistics, neuroimaging compatibility etc. However, it is well established in animal models that the lack of physical movement in VR impairs some neural representations of space, and this is considered likely to be true in humans as well. Furthermore, it is unclear how big the disruptive effect stationary navigation is-how much does physical movement during encoding and recall affect human spatial memory and representations of space? What eff

Locomotion, active choice, and spatial memory in children.

In two studies, children between 4 and 6 years old were tested on a radial search task requiring nonredundant sampling of eight identically labeled positions in a room. In the training phase (5 days), children made free choices by walking between positions, were passively transported in a pushchair, actively directed their own route from a pushchair, or were led on foot to positions selected by the experimenter. When tested (whether walking or directing while seated in a pushchair), children who had either walked independently or directed the experimenter while being pushed performed competent

Increased PKA signaling disrupts recognition memory and spatial memory: role in Huntington's disease.

Huntington's disease (HD) patients and mouse models show learning and memory impairment even before the onset of motor symptoms. However, the molecular events involved in this cognitive decline are still poorly understood. Here, using three different paradigms, the novel object recognition test, the T-maze spontaneous alternation task and the Morris water maze, we detected severe cognitive deficits in the R6/1 mouse model of HD before the onset of motor symptoms. When we examined the putative molecular pathways involved in these alterations, we observed hippocampal cAMP-dependent protein kinas

Spatial memory following prenatal alcohol exposure: more than a material specific memory deficit.

Spatial memory deficits have been reported following prenatal alcohol exposure and animal studies have demonstrated hippocampal vulnerability to alcohol. This study examined spatial memory in children diagnosed with fetal alcohol syndrome (FAS) or fetal alcohol effects (FAE) and matched controls. Spatial memory was examined with location recall measures. Since visual perceptual skills and nonspecific memory impairment could impact spatial memory, tasks assessing perception and verbal memory were administered. Analyses revealed group differences on perceptual and verbal and spatial memory tasks

The effect of spatial boundaries on memory in a virtual environment.

Crossing a spatial boundary, such as a doorway, often signals the ending of one episode and the beginning of another, segmenting ongoing experience into events. When conducted in real environments, this 'doorway effect' differentially affects memory for objects encountered within and between events. The evidence for this occurring in immersive virtual reality (VR) environments is mixed. The present study investigated the 'doorway effect' in a VR environment and also examined how event memory is affected when segmentation processes are disrupted by interference tasks. Ninety participants explor

Distributed encoding and retrieval of spatial memory in the hippocampus.

To determine whether memory is processed in a localized or distributed manner by the hippocampus, we inactivated small regions of the structure in pretrained rats before a retention test. Ibotenic acid-induced lesions removing 40% of the hippocampal tissue disrupted retrieval of spatial memory in a water maze but failed to affect new learning or retrieval of a task that was acquired postoperatively. Partial inactivation of the hippocampus by local intrahippocampal 5-aminomethyl-3-hydroxyisoxazole muscimol infusion also impaired retrieval but not new learning. This impairment was temporary; inf

Spatial learning and memory in the tortoise (Geochelone carbonaria).

A single tortoise (Geochelone carbonaria) was trained in an eight-arm radial maze, with the apparatus and general procedures modeled on those used to demonstrate spatial learning in rats. The tortoise learned to perform reliably above chance, preferentially choosing baited arms, rather than returning to arms previously visited on a trial. Test sessions that examined control by olfactory cues revealed that they did not affect performance. No systematic, stereotyped response patterns were evident. In spite of differences in brain structure, the tortoise showed spatial learning abilities comparab

The chemotherapeutic agent paclitaxel selectively impairs learning while sparing source memory and spatial memory.

Chemotherapeutic agents are widely used to treat patients with systemic cancer. The efficacy of these therapies is undermined by their adverse side-effect profiles such as cognitive deficits that have a negative impact on the quality of life of cancer survivors. Cognitive side effects occur across a variety of domains, including memory, executive function, and processing speed. Such impairments are exacerbated under cognitive challenges and a subgroup of patients experience long-term impairments. Episodic memory in rats can be examined using a source memory task. In the current study, rats rec

Active navigation enhances children's spatial but not episodic memory.

This paper uses a Yoked experimental design to investigate the selective advantage of active spatial navigation in 4- to 10-year-old children. Participants were presented with a map game in which they were asked to find the shortest way for a monster to get across town while collecting its five monster friends on the way. Active control was associated with better recall for path length and the area between paths, whereas no differences were observed for the number of corners. Further, active navigation did not enhance episodic memory. Our results indicate that the benefits of active control du

Spatial working memory across psychiatric disorders in young adults.

OBJECTIVE: Impairments in spatial working memory (SWM) have been observed in several disparate psychiatric conditions. The present study investigated how SWM varied across a range of psychiatric disorders in a large sample of young adults using the SWM test from the Cambridge Neuropsychological Test Automated Battery. METHODS: A sample of 575 participants aged 18-29 years was enrolled from the general community and evaluated for psychiatric disorders using several screening evaluations. Performance on the SWM test was assessed, with outcome measures of strategy and total errors. Using Z-score

Impaired allocentric spatial memory in patients with affective disorders.

BACKGROUND: Memory disturbances are frequent in unipolar depression (UD) and bipolar disorder (BD) and may comprise important predisposing and maintaining factors. Previous studies have demonstrated hippocampal abnormalities in UD and BD but there is a lack of studies specifically assessing hippocampus-dependent memory. METHODS: We used a virtual task to assess hippocampus-dependent (allocentric) vs non-hipppocampal (egocentric) spatial memory in remitted and partially remitted patients with UD or BD (N = 22) and a healthy control group (N = 32). Participants also completed a range of standar

Medial prefrontal theta phase coupling during spatial memory retrieval.

Memory retrieval is believed to involve a disparate network of areas, including medial prefrontal and medial temporal cortices, but the mechanisms underlying their coordination remain elusive. One suggestion is that oscillatory coherence mediates inter-regional communication, implicating theta phase and theta-gamma phase-amplitude coupling in mnemonic function across species. To examine this hypothesis, we used non-invasive whole-head magnetoencephalography (MEG) as participants retrieved the location of objects encountered within a virtual environment. We demonstrate that, when participants a

Orientation dependence of spatial memory acquired from auditory experience.

The present study investigated whether memory for a room-sized spatial layout learned through auditory localization of sounds exhibits orientation dependence similar to that observed for spatial memory acquired from stationary viewing of the environment. Participants learned spatial layouts by viewing objects or localizing sounds and then performed judgments of relative direction among remembered locations. The results showed that direction judgments following auditory learning were performed most accurately at a particular orientation in the same way as were those following visual learning, i

Effects of complete monocular deprivation in visuo-spatial memory.

Monocular deprivation has been associated with both specific deficits and enhancements in visual perception and processing. In this study, performance on a visuo-spatial memory task was compared in congenitally monocular individuals and sighted control individuals viewing monocularly (i.e., patched) and binocularly. The task required the individuals to view and memorize a series of target locations on two-dimensional matrices. Overall, congenitally monocular individuals performed worse than sighted individuals (with a specific deficit in simultaneously maintaining distinct spatial representati

Egocentric and allocentric spatial memory in idiopathic cervical dystonia.

OBJECTIVE: To investigate whether impaired spatial memory contributes to cognitive deficits in idiopathic cervical dystonia (ICD). METHODS: The authors tested 16 untreated patients with ICD and 16 healthy controls with four variants of a classic spatial memory task, in which subjects were requested to recall the locations of 16 toy objects either immediately after their presentation or after an unfilled delay of 10 minutes. By varying the spatial relationship between subject and toy object array, we tested encoding and memory of spatial relationships between subject and toy objects (egocentri
