Visual landmarks facilitate rodent spatial navigation in virtual reality environments.

Because many different sensory modalities contribute to spatial learning in rodents, it has been difficult to determine whether spatial navigation can be guided solely by visual cues. Rodents moving within physical environments with visual cues engage a variety of nonvisual sensory systems that cannot be easily inhibited without lesioning brain areas. Virtual reality offers a unique approach to ask whether visual landmark cues alone are sufficient to improve performance in a spatial task. We found that mice could learn to navigate between two water reward locations along a virtual bidirectiona

Distinct retinal pathways drive spatial orientation behaviors in zebrafish navigation.

Navigation requires animals to adjust ongoing movements in response to pertinent features of the environment and select between competing target cues. The neurobiological basis of navigational behavior in vertebrates is hard to analyze, partly because underlying neural circuits are experience dependent. Phototaxis in zebrafish is a hardwired navigational behavior, performed at a stage when larvae swim by using a small repertoire of stereotyped movements. We established conditions to elicit robust phototaxis behavior and found that zebrafish larvae deploy directional orienting maneuvers and reg

Spatial exploration and navigation in Down syndrome and Williams syndrome.

We know little about the ability to explore and navigate large-scale space for people with intellectual disability (ID). In this cross-syndrome study, individuals with Down syndrome (DS), individuals with Williams syndrome (WS) and typically developing children (TD; aged 5-11 years) explored virtual environments with the goal of learning where everything was within the environment (Experiment 1) or to find six stars (Experiment 2). There was little difference between the WS and DS groups when the goal was simply to learn about the environment with no specific destination to be reached (Experim

Anxiety and Spatial Navigation in Williams Syndrome and Down Syndrome.

Individuals with Down Syndrome (DS) and individuals with Williams syndrome (WS) present with poor navigation and elevated anxiety. The aim of this study was to determine the relationship between these two characteristics. Parent report questionnaires measured navigation abilities and anxiety in WS (N = 55) and DS (N = 42) as follows. Anxiety: Spence Children's Anxiety Scale and a novel measure of navigation anxiety. Navigation: Santa Barbara Sense of Direction Scale (SBSOD) and a novel measure of navigation competence. Most individuals were not permitted to travel independently. A relationship

Pathfinder: open source software for analyzing spatial navigation search strategies.

Spatial navigation is a universal behavior that varies depending on goals, experience and available sensory stimuli. Spatial navigational tasks are routinely used to study learning, memory and goal-directed behavior, in both animals and humans. One popular paradigm for testing spatial memory is the Morris water maze, where subjects learn the location of a hidden platform that offers escape from a pool of water. Researchers typically express learning as a function of the latency to escape, though this reveals little about the underlying navigational strategies. Recently, a number of studies hav

Spatial and temporal aspects of navigation in two neurological patients.

We present two cases (A.C. and W.J.) with navigation problems resulting from parieto-occipital right hemisphere damage. For both the cases, performance on the neuropsychological tests did not indicate specific impairments in spatial processing, despite severe subjective complaints of spatial disorientation. Various aspects of navigation were tested in a new virtual reality task, the Virtual Tübingen task. A double dissociation between spatial and temporal deficits was found; A.C. was impaired in route ordering, a temporal test, whereas W.J. was impaired in scene recognition and route continuat

Pharmacological stimulation of hippocampal GPR35 impairs spatial navigation, learning, and memory.

GPR35 is an orphan receptor expressed in astrocytes and neurons of the central nervous system (CNS), including the hippocampus; however, its role in cognitive functions is largely unknown. This study administered intra-hippocampal injections of GPR35 compound-10 (GPR35C10; a GPR35-selective agonist), ML194 (a GPR35-selective antagonist), and the combination of both to elucidate their pharmacological actions in the Barnes maze. Compared to the vehicle (10 % DMSO) and ML194 groups, animals treated with GPR35C10 showed a higher latency and longer distance to solve the maze (p < 0.05) (an effect p

Development and validation of the Spatial Navigation and Memory Questionnaire (SPAM).

INTRODUCTION: Spatial navigation deficits may provide a sensitive and specific marker of early Alzheimer's disease pathology. However, few spatial navigation assessment tools are available for clinical practice that are accessible, time efficient, and sensitive to cognitive aging. METHODS: Exploratory (n = 470) and confirmatory factor analyses (n = 470) were conducted to establish the factor structure of the Spatial Navigation and Memory Questionnaire (SPAM) in healthy older adults. A subset of participants (n = 445) completed the Virtual Supermarket Test (VST) to assess convergent validity.

The relationship between object-based spatial ability and virtual navigation performance.

Spatial navigation is a multi-faceted behaviour drawing on many different aspects of cognition. Visuospatial abilities, such as mental rotation and visuospatial working memory, in particular, may be key factors. A range of tests have been developed to assess visuospatial processing and memory, but how such tests relate to navigation ability remains unclear. This understanding is important to advance tests of navigation for disease monitoring in various disorders (e.g., Alzheimer's disease) where spatial impairment is an early symptom. Here, we report the use of an established mobile gaming app

Rats exposed prenatally to alcohol exhibit impairment in spatial navigation test.

Prenatal exposure to ethanol causes learning disabilities and low I.Q. scores. The objective of the present studies was to investigate whether exposure of rats to ethanol in utero, would induce a deficit in spatial memory in adult life. Pregnant rats were fed with an ethanol diet from day 1 of pregnancy till parturition. Control rats were either pair-fed with an isocaloric sucrose diet or were fed with lab-chow ad libitum. On the first day of birth, offspring exposed to ethanol in utero were placed with a control mother fed with lab-chow, while offspring of the lab-chow fed dams were placed wi

Spatial navigation after surgical resection of an acoustic neuroma: pilot study.

OBJECTIVE: To characterize navigation errors made by patients with the absence of vestibular function on one side owing to surgical resection of an acoustic neuroma. METHODS: Seventeen young (18-38 years) and 9 older healthy individuals (67-83 years), as well as 5 patients 2 to 20 months following surgery (37-61 years), were studied. They sidestepped laterally with eyes closed toward memorized targets located 1.25 m to their right or left. They stopped when they judged that they were in front of the target. The position of head and body markers was recorded in three dimensions with a six-came

Unraveling the Twist: Spatial Navigational Challenges in Cervical Dystonia.

BACKGROUND: Cervical dystonia (CD) is an intricate neurological condition with motor and nonmotor symptoms. These include disruptions in visual perception, self-orientation, visual working memory, and vestibular functions. However, the specific impact of CD on perceiving self-motion direction, especially with isolated visual or vestibular stimuli, remains largely unexplored. OBJECTIVE: This study aimed to examine the effects of CD on linear motion perception, hypothesizing impaired heading discrimination in both vestibular and visual tasks, and that such deficits correlate with the disease se

Human spatial navigation: Neural representations of spatial scales and reference frames obtained from an ALE meta-analysis.

Humans use different spatial reference frames (allocentric or egocentric) to navigate successfully toward their destination in different spatial scale spaces (environmental or vista). However, it remains unclear how the brain represents different spatial scales and different spatial reference frames. Thus, we conducted an activation likelihood estimation (ALE) meta-analysis of 47 fMRI articles involving human spatial navigation. We found that both the environmental and vista spaces activated the parahippocampal place area (PPA), retrosplenial complex (RSC), and occipital place area in the righ

Effects of hormonal contraceptives on spatial navigation in gonadally intact female rats.

Rats can either use place memory, response memory or rely on a single cue to navigate their environment. Which navigation strategy a female rat is biased towards is influenced by circulating levels of ovarian hormones. Hormonal contraceptive use is typically associated with changes in circulating levels of endogenous ovarian hormones. The aim of the current study was to examine the effects of synthetic hormones used in hormonal contraceptives on navigation strategies. A total of 61 gonadally intact female rats were tested in two Morris water maze tasks. Rats were administered ethinyl estradiol

Coordination of escape and spatial navigation circuits orchestrates versatile flight from threats.

Naturalistic escape requires versatile context-specific flight with rapid evaluation of local geometry to identify and use efficient escape routes. It is unknown how spatial navigation and escape circuits are recruited to produce context-specific flight. Using mice, we show that activity in cholecystokinin-expressing hypothalamic dorsal premammillary nucleus (PMd-cck) cells is sufficient and necessary for context-specific escape that adapts to each environment's layout. In contrast, numerous other nuclei implicated in flight only induced stereotyped panic-related escape. We reasoned the dorsal

Spatial Navigation in Children and Young Adults with Fetal Alcohol Spectrum Disorders.

BACKGROUND: Rodent studies have consistently shown that prenatal alcohol exposure (PAE) impairs performance on the Morris water maze (MWM), a test of spatial navigation. A previous study comparing boys with fetal alcohol syndrome (FAS) to controls found poorer performance on the virtual water maze (VWM), a human analogue of the MWM. We examined PAE effects on virtual navigation in both sexes using the VWM in a moderately exposed Detroit cohort (N = 104; mean = 19.4 year) and a heavily exposed Cape Town, South African cohort (N = 62; mean = 10.4 year). The effect of TOMM40 on spatial navigation in amnestic mild cognitive impairment.

The very long (VL) poly-T variant at rs10524523 ("523") of the TOMM40 gene may hasten the onset of late-onset Alzheimer's disease (LOAD) and induce more profound cognitive impairment compared with the short (S) poly-T variant. We examined the influence of TOMM40 "523" polymorphism on spatial navigation and its brain structural correlates. Participants were apolipoprotein E (APOE) ε3/ε3 homozygotes with amnestic mild cognitive impairment (aMCI). The homozygotes were chosen because APOE ε3/ε3 variant is considered "neutral" with respect to LOAD risk. The participants were stratified according to

Active and passive spatial learning in human navigation: acquisition of survey knowledge.

It seems intuitively obvious that active exploration of a new environment would lead to better spatial learning than would passive visual exposure. It is unclear, however, which components of active learning contribute to spatial knowledge, and previous literature is decidedly mixed. This experiment tests the contributions of 4 components to metric survey knowledge: visual, vestibular, and podokinetic information and cognitive decision making. In the learning phase, 6 groups of participants learned the locations of 8 objects in a virtual hedge maze by (a) walking, (b) being pushed in a wheelch

Are benzodiazepines really anxiolytic? Evidence from a 3D maze spatial navigation task.

The effects of diazepam and chlordiazepoxide were assessed in a 3D maze which is a modification of an 8-arm radial maze. Each arm of the maze is attached to a bridge radiating from a central platform. Animals exposed for the first time to the maze do not venture beyond the line that separate a bridge from an arm. The prime criteria set for an anxiolytic effect is whether mice would increase the frequency of entries onto arms and increase arm/bridge entries ratio. C57 mice readily cross the line on first exposure and make more than 8 arm visits onto arms on second exposure, while other strains

Spatial navigation as a digital marker for clinically differentiating cognitive impairment severity.

BACKGROUND: Spatial navigation impairments emerge early in Alzheimer's disease, but assessments targeting these deficits remain underutilised or impractical for cognitive screening. The Spatial Performance Assessment for Cognitive Evaluation (SPACE) is a newly developed digital tool that evaluates spatial navigation deficits associated with cognitive impairment. METHODS: We assessed spatial navigation ability using SPACE in 300 older adults recruited from memory clinics and the general community. Participants were classified across different levels of cognitive impairment using the Clinical D
