The genotoxicity of particulate and soluble chromate in sperm whale (physeter macrocephalus) skin fibroblasts.

Hexavalent chromium is a marine pollutant of concern, both for the health of ocean ecosystems and for public health. Hexavalent chromium is known to induce genotoxicity in human and other terrestrial mammals. It is also known to be present in both water and air in the marine environment. However, currently there are limited data concerning both chromium levels and its toxicological effects in marine mammals. This study investigated the cytotoxic and genotoxic effects of soluble and particulate hexavalent chromium in sperm whale skin fibroblasts. Both forms of hexavalent chromium induced concen

Morphological and proteomic characterization of sperm in a beluga whale (Delphinapterus leucas):a case report.

The study of cetacean reproductive physiology is important for their conservation; however, limited information is available on the reproductive characteristics of these species. The present study aims to characterize beluga whale (Delphinapterus leucas) sperm by analyzing their morphology, dimensions and shape, and proteome. Epididymal fluid samples (from each epididymis) were collected from a beluga whale housed in an aquarium that died (~ 60 years old). The results indicated that, in most cases, the sperm head has an ovoid shape with a slightly tapered anterior end. However, significant div

Fatal ingestion of floating net debris by two sperm whales (Physeter macrocephalus).

In 2008 two male sperm whales (Physeter macrocephalus) stranded along the northern California coast with large amounts of fishing net scraps, rope, and other plastic debris in their stomachs. One animal had a ruptured stomach, the other was emaciated, and gastric impaction was suspected as the cause of both deaths. There were 134 different types of nets in these two animals, all made of floating material, varying in size from 10 cm(2) to about 16 m(2). The variability in size and age of the pieces suggests the material was ingested from the surface as debris rather than bitten off from active

Postmortem investigations on winter stranded sperm whales from the coasts of Belgium and The Netherlands.

During winter 1994-95, four and three sperm whales (Physeter macrocephalus) were stranded along the Belgian and the Dutch coasts, respectively. Necropsies and tissue samplings were collected 24 hrs post mortem. Lesions on several whales included round and linear skin scars, ventral skin abrasions, acute skin ulcers, acute ulcerative stomatitides, acute to chronic external otitides, and passive visceral congestion. In addition, these sperm whales appeared to be debilitated with severe weight deficit, had blubber thickness reduction, the absence of abdominal fat, and the intestinal tracts were a

Titration behavior of individual tyrosine residues of myoglobins from sperm whale, horse, and red kangaroo.

The titration behavior of individual tyrosine residues of myoglobins has been studied by observing the pH dependence of the chemical shifts of Czeta and Cgamma of these residues in natural abundance of 13C Fourier transform NMR spectra (at 15.18 MHz, in 20-mm sample tubes, at 37 degrees) of cyanoferrimyoglobins from sperm whale, horse, and red kangaroo. A comparison of the pH dependence of the spectra of the three proteins yielded specific assignments for the resonance of Tyr-151 (sperm whale) and Tyr-103 (sperm whale and horse). Selective proton decoupling yielded specific assignments for Cze

From the jaws of the "Leviathan": A sperm whale tooth from the Valencina Copper Age Megasite.

During the excavations undertaken in 2018 at the Nueva Biblioteca sector of the Valencina Copper Age mega-site, in south-west Spain, an exceptional sperm-whale tooth was found inside a non-burial pit. This remarkable object is the first of its kind ever found for Late Prehistoric Iberia. Due to its rarity and importance, a multidisciplinary study was carried out, including photogrammetric 3D modelling, as well as taphonomic, paleontological, technological and contextual analysis. This led to a full characterisation of the artefact through the analysis of its bioerosion traces, anthropogenic ma

Can genetic differences explain vocal dialect variation in sperm whales, Physeter macrocephalus?

Sperm whale social groups can be assigned to vocal clans based on their production of codas, short stereotyped patterns of clicks. It is currently unclear whether genetic variation could account for these behavioural differences. We studied mitochondrial DNA (mtDNA) variation among sympatric vocal clans in the Pacific Ocean, using sequences extracted from sloughed skin samples. We sampled 194 individuals from 30 social groups belonging to one of three vocal clans. As in previous studies of sperm whales, mtDNA control region diversity was low (π = 0.003), with just 14 haplotypes present in our

Influence of mercury and selenium chemistries on the progression of cardiomyopathy in pygmy sperm whales, Kogia breviceps.

More than half of pygmy sperm whales (Kogia breviceps) that strand exhibit signs of cardiomyopathy (CMP). Many factors may contribute to the development of idiopathic CMP in K. breviceps, including genetics, infectious agents, contaminants, biotoxins, and dietary intake (e.g. selenium, mercury, and pro-oxidants). This study assessed trace elements in K. breviceps at various stages of CMP progression using fresh frozen liver and heart samples collected from individuals that stranded along US Atlantic and Gulf coasts between 1993 and 2007. Acoustic property reconstruction of a pygmy sperm whale (Kogia breviceps) forehead based on computed tomography imaging.

Computed tomography (CT) imaging and sound experimental measurements were used to reconstruct the acoustic properties (density, velocity, and impedance) of the forehead tissues of a deceased pygmy sperm whale (Kogia breviceps). The forehead was segmented along the body axis and sectioned into cross section slices, which were further cut into sample pieces for measurements. Hounsfield units (HUs) of the corresponding measured pieces were obtained from CT scans, and regression analyses were conducted to investigate the linear relationships between the tissues' HUs and velocity, and HUs and densi

Structural and functional characterization of sperm whale myoglobin mutants: role of arginine (E10) in ligand stabilization.

1H NMR and ligand-binding data were used to assess the role of residue Arg(E10) in ligand stabilization of several site-directed mutants, all carrying the His(E7) to Val substitution, obtained using a synthetic sperm whale myoglobin gene. Arg(E10) was previously found to form a hydrogen bond with the ligand in fluoro-, azido- and cyanomet derivatives of Aplysia limacina myoglobin, which lacks the distal His(E7) [Qin, J., La Mar, G. Kinetic and equilibrium studies of the ligand binding reactions of eight electrophoretic components of sperm whale ferrimyoglobin.

The reactions of eight electrophoretic components of sperm whale ferrimyoglobin with fluoride, azide, and cyanide have been studied. There do not appear to be significant differences in rate constants or equilibrium constants among the various components. We conclude that at pH 7.0 in 0.05 M potassium phosphate these ligand binding kinetics and equilibria are insensitive to the net charge on the protein. The variation of the azide equilibrium constant with ionic strength from mu 0.01 M to 0.11 M is not in accord with the predictions of the Debye-Hückel theory. On the other hand, azide associat

Migration to breeding areas by male sperm whales Physeter macrocephalus from the Northeast Atlantic Arctic.

Mature male sperm whales (Physeter macrocephalus) primarily inhabit high latitude regions, travelling to tropical/temperate waters for breeding, where females and juveniles reside in cohesive social groups. Though mating is known to occur at low latitudes, the timing, duration, and routes of adult male migrations between feeding and breeding areas are poorly known. To study movement patterns of adult male sperm whales, 29 individuals were equipped with satellite transmitters in the Northeast Atlantic Arctic (69-79°N). Twelve of these animals undertook southward migrations. Departures from nort

Coinfection and vertical transmission of Brucella and Morbillivirus in a neonatal sperm whale (Physeter macrocephalus) in Hawaii, USA.

The viral genus Morbillivirus and the bacterial genus Brucella have emerged as important groups of pathogens that are known to affect cetacean health on a global scale, but neither pathogen has previously been reported from endangered sperm whales (Physeter macrocephalus). A female neonate sperm whale stranded alive and died near Laie on the island of Oahu, Hawaii, US, in May of 2011. Congestion of the cerebrum and enlarged lymph nodes were noted on the gross necropsy. Microscopic findings included lymphoid depletion, chronic meningitis, and pneumonia, suggesting an in utero infection. Cerebru

Identification and characterization of broad-spectrum cathelicidin family antimicrobial peptides from the pygmy sperm whale Kogia breviceps.

Cathelicidins are crucial antimicrobial peptides (AMPs) that play a key role in the innate immunity of marine vertebrates. The pygmy sperm whale (Kogia breviceps), which resides in environments populated by diverse microbial communities, faces complex pathogenic threats; however, its cathelicidin family AMPs remain uncharacterized. In this study, we identified three novel cathelicidin peptides-Kb4-1, Kb4-2, and Kb5 from the whale (Kogia breviceps). All three peptides exhibited broad-spectrum and potent antibacterial activity, with Kb5, an amphipathic α-helical peptide, showing the strongest an

Bony outgrowths on the jaws of an extinct sperm whale support macroraptorial feeding in several stem physeteroids.

Several extinct sperm whales (stem Physeteroidea) were recently proposed to differ markedly in their feeding ecology from the suction-feeding modern sperm whales Kogia and Physeter. Based on cranial, mandibular, and dental morphology, these Miocene forms were tentatively identified as macroraptorial feeders, able to consume proportionally large prey using their massive teeth and robust jaws. However, until now, no corroborating evidence for the use of teeth during predation was available. We report on a new specimen of the stem physeteroid Acrophyseter, from the late middle to early late Mioce

Cyanide binding to Lucina pectinata hemoglobin I and to sperm whale myoglobin: an x-ray crystallographic study.

The x-ray crystal structures of the cyanide derivative of Lucina pectinata monomeric hemoglobin I (L. pectinata HbI) and sperm whale (Physeter catodon) myoglobin (Mb), generally taken as reference models for monomeric hemoproteins carrying hydrogen sulfide and oxygen, respectively, have been determined at 1.9 A (R-factor = 0. 184), and 1.8 A (R-factor = 0.181) resolution, respectively, at room temperature (lambda = 1.542 A). Moreover, the x-ray crystal structure of the L. pectinata HbI:cyanide derivative has been studied at 1.4-A resolution (R-factor = 0.118) and 100 K (on a synchrotron source

Buoyancy regulation through modulation of onboard gas volume by resting sperm whales (Physeter macrocephalus).

Sperm whales rest while drifting under the surface in a unique stereotypical vertical position. This position is thought to enable them to sleep effectively by buffering them from surface wave action, while avoiding the costs of transiting to deeper depths. While resting, sperm whales release bubbles, hypothesised to help them regulate their buoyancy. As sperm whales are breath-hold divers, if they slowly drift up while resting, the gas in their lung will slowly expand following Boyle-Mariotte's gas law. Hence, releasing bubbles would enable them to counteract the expanding diving gas and achi

Stable isotopes provide insight into population structure and segregation in eastern North Atlantic sperm whales.

In pelagic species inhabiting large oceans, genetic differentiation tends to be mild and populations devoid of structure. However, large cetaceans have provided many examples of structuring. Here we investigate whether the sperm whale, a pelagic species with large population sizes and reputedly highly mobile, shows indication of structuring in the eastern North Atlantic, an ocean basin in which a single population is believed to occur. To do so, we examined stable isotope values in sequential growth layer groups of teeth from individuals sampled in Denmark and NW Spain. In each layer we measur
