This was a cohort study of in vitro fertilization (IVF) subjects at the University of Utah, Salt Lake City (UT, USA) utilizing partner sperm. Cycles where both the hamster egg penetration test (HEPT) and semen analysis were performed within 2 years prior to IVF cycles were stratified into four groups based on a normal or an abnormal HEPT and morphology. The mean conventional and intracytoplasmic sperm injection (ICSI) fertilization rates were calculated in each group. We performed a univariate analysis on the primary outcome comparing clinically interesting subjects. We performed a cost-effect

Out of the publications of the last three years concerning the influence of the epididymis on the male fertility those with practical relevance were reviewed. Several qualities of the spermatozoa were influenced by the epididymis: the metabolism, the functions of cell membrane, the fertilizing capacity, the ability to bind to zonae pellucidae of the oocytes and the motility. The motility depends on the electrolytes, androgenes, cAMP-system and the forward motility protein in the epididymis. Furthermore investigations exist dealing with the inhibition of the maturation of spermatozoa during the

After natural or artificial insemination, the spermatozoon starts a journey from the site of deposition to the place of fertilization. However, only a small subset of the spermatozoa deposited achieves their goal: to reach and fertilize the egg. Factors involved in controlling sperm transport and fertilization include the female reproductive tract environment, cell-cell interactions, gene expression, and phenotypic sperm traits. Some of the significant determinants of fertilization are known (i.e., motility or DNA status), but many sperm traits are still indecipherable. One example is the infl

BACKGROUND: Fertilization ensures the transmission of genetic material across generations through a series of precisely coordinated physiological and molecular events. To fertilize an oocyte, a spermatozoon must pass through the cumulus cell layer, penetrate the zona pellucida (ZP), and ultimately adhere to and fuse with the oolemma (the oocyte plasma membrane). Upon fusion, the oocyte initiates mechanisms to block additional sperm entry at both the ZP and oolemma. These processes are highly dynamic in space and time, posing substantial technical barriers to their mechanistic dissection. Nonet

Fertilization involves dynamic sperm-egg interactions, yet has been primarily studied in static samples. Here, we use high-resolution live imaging to capture fertilization from the moment of sperm binding in zona-intact mouse oocytes. We identify two phases of sperm remodeling: a static phase, during which sperm remain beneath the oocyte cortex as DNA decondensation and histone loading occur, and a mobile phase characterized by stereotyped sperm movement. Initial displacement away from the spindle is driven by cytoplasmic streaming, with manipulations in mouse indicating that sperm movement re

This Opinion piece is offered as a cursory overview of sperm development, function, and transport through the eyes of an equine veterinarian. My professional background is predominantly clinical in nature, but my fascination with sperm function and preservation has led to a fairly sizeable review of the scientific literature over the years in hopes of extracting laboratory findings that have application to my daily activities in the clinical arena. Spermatozoa are quite unique among cellular types with regard to both form and function, and represent the only endogenously derived cell type that

Mammalian fertilization is a complex process that involves different steps of interaction between the male and female gametes. In spite of its relevance, the molecular mechanisms underlying this process still remain to be elucidated. The present review describes the contribution of our laboratory to the understanding of mammalian fertilization using Cysteine-RIch Secretory Proteins (CRISP) as model molecules. Substantial evidence obtained from in vitro assays and knockout models shows that epididymal CRISP1 associates with the sperm surface with two different affinities during maturation, and

PURPOSE: Platelet-activating factor (PAF) is a potent signaling phospholipid that is found in mammalian sperm and has a positive correlation with fertility. Whereas PAF is present in human sperm, there are no relational reports on its content and the cells fertilization potential. Therefore, the study objective was to determine if PAF content in capacitated-induced sperm is related to fertilization potential as determined by the sperm penetration assay (SPA).
METHODS: Endogenous sperm lipids were measured for PAF content by a specific radioimmunoassay following insemination of zona pellucida-

A model system of hamster oocyte and human sperm interaction was used to assess sperm entry into the perivitelline space after partial zona dissection. The procedure of zona dissection was standardized by creating slits which included one fourth, one eighth, one eighth X 2, or one sixteenth of the zonal circumference. Manipulated eggs were allowed to interact with 1 X 10(6) sperm/mL for 3 hours. A single large or medium slit was equally effective in permitting sperm entry into 46% and 47% of the manipulated eggs, respectively. However, the longer slit doubled the average number of sperm detect

In the poultry industry, every chick or poult produced has high economic value; therefore, selection of male breeders capable of transmitting desirable traits to their offspring has an important role in fertility and hatching success. Following either natural mating or artificial insemination, sperm must go through a sequence of steps to fertilize the egg: 1. Go from the site of deposition (vagina) to the Sperm Storage Tubules (SST); 2. Enter and stay inside the SST; 3. Reacquire motility; and 4. Bind to and penetrate the egg. To perform these tasks successfully, sperm must be alive, motile, a

The objective was to examine, in vitro, the motility, acrosomal proteolytic activity (APA), and penetrating ability of fowl sperm recovered from the testis and epididymis, as well as the proximal, middle, and distal vas deferens, to assess the potential fertilizing ability of sperm as a function of maturation. A motile sperm separation technique was used to estimate sperm motility with Accudenz, a gelatin slide technique was used to measure the diameter of the halo around the acrosome of individual sperm as an indication of APA, and a sperm-inner perivitelline layer (IPL) interaction assay was

Human fertilization begins when a capacitated spermatozoon binds to the zona pellucida (ZP) surrounding a mature oocyte. Defective spermatozoa-ZP interaction contributes to male infertility and is a leading cause of reduced fertilization rates in assisted reproduction treatments (ARTs). Human ejaculate contains millions of spermatozoa with varying degrees of fertilization potential and genetic quality, of which only thousands of motile spermatozoa can bind to the ZP at the fertilization site. This observation suggests that human ZP selectively interacts with competitively superior spermatozoa

The objective of this study was to determine the age effect of a broiler breeder flock on duration of fertility and number of spermatozoa penetrating the perivitelline layer overlying the germinal disc (SP/mm(2) GDIPVL). Moreover, in the second half of the flock's reproductive life, the effect of using ejaculates of young roosters (CA2) in artificial insemination (AI) on the above parameters of fertility was estimated. The commercial flock of broiler breeder hens (n = 100) was inseminated six times from 31 to 62 weeks of age. Additional inseminations, with ejaculates of roosters aged 31 and 36

In the ctenophore Beroe ovata, action potentials and sperm-induced egg electrical responses were recorded, in unfertilized and in vitro inseminated eggs, respectively. The unfertilized egg had a resting membrane potential of -65.6 +/- 1.3 mV and a membrane resistance of 1.45 +/- 0.09 M omega. When depolarized beyond a threshold of about -20 mV, the membrane displayed an action potential with a fast phase and then a peak reaching 38 +/- 0.6 mV, followed by a positive declining plateau which lasted about 20 sec and ended by a biphasic repolarization of the membrane. The action potential was main

Spermatozoa were recovered from the uterine horns or vagina of mated rabbit does 12-20 hr p.c. and their fertilizing capacity tested in an in-vitro system. Uterine spermatozoa gave consistently high fertilization (93-100%) throughout. Vaginal spermatozoa gave good results (81-85%) 12-14 hr p.c. but fertilization then declined with time. Vaginal spermatozoa recovered from mated females with or without ligation of the uterine horns were equally able to fertilize eggs. Fragmentation of uncleaved eggs was noted in experiments with vaginal spermatozoa, but never with uterine spermatozoa. The former

In avian species, male fertility significantly impacts reproductive success. This study investigates the relationship between proctodeal gland size in male Japanese quails and sperm function, as well as female fertility duration. Six hundred adult Japanese quails were selected and housed in individual cages. Males (n = 300) were divided into 6 groups (50/group) based on the size of their proctodeal glands. Females (n = 300) were randomly assigned to corresponding groups. After acclimatization, sperm kinematics and the frequency and weight of foam discharge were evaluated. Males were paired wit

The efficiency of cervical mucus in filtering out single, multiple and associated abnormalities of human spermatozoa was determined. Twenty semen samples which gave a normal in vitro cervical mucus penetration test (CMPT) were analysed before and after migration using a detailed classification system (13 categories). The % of normal forms was significantly increased in cervical mucus (59.5 vs 33.2%), whereas the % of sperm with single, multiple or associated abnormalities of the midpiece or of the flagellum were found to decrease significantly in cervical mucus. Sperm with single or multiple a

The value of various sperm parameters and the zona-free hamster egg sperm penetration assay (SPA) in predicting human in vitro fertilization (IVF) failure and subsequent successful fertilization with zona drilling was assessed. In 19 couples, throughout 31 IVF cycles, a total of 153 oocytes failed to be fertilized. In subsequent 12 cycles with zona drilling, 33 of 131 (25%) were fertilized. The incidence of teratospermia and asthenospermia was significantly higher in the study group than in the control, 74% versus 32% and 42% versus 5%, respectively. Although the mean values for the performanc

Male infertility is a widely debated issue that affects males globally. There are several mechanisms involved. Oxidative stress is accepted to be the main contributing factor, with sperm quality and quantity affected by the overproduction of free radicals. Excess reactive oxygen species (ROS) cannot be controlled by the antioxidant system and, thus, potentially impact male fertility and hamper sperm quality parameters. Mitochondria are the driving force of sperm motility; irregularities in their function may lead to apoptosis, alterations to signaling pathway function, and, ultimately, comprom

A series of studies was conducted to examine the role of copulatory dysfunction, spermatotoxicity, and/or impaired fertilization in the reduced rates of fertility and implantation observed in females mated to acrylamide-treated male rats. In initial experiments, males were gavaged with 0, 5, 15, 30, 45, or 60 mg/kg acrylamide (ACR) for 5 days and then mated serially to naive females. ACR treatment reduced fertility and increased pre- and post-implantation loss, primarily over the first 3 weeks post-treatment. The effects at Week 1 appeared to result from an interference in sperm transport as d
