Methods in molecular biology (Clifton, N.J.)2024eng
Mammalian meiosis is a highly specialized cell division process, resulting in the production of genetically unique haploid cells. However, the molecular mechanisms governing meiosis remain largely unknown, primarily due to the difficulty in isolating pure sub-populations of spermatocytes. Definitive molecular, biochemical, and functional investigations of the meiosis process require the isolation of these individual homogeneous sub-populations of spermatocytes. Here, we present an approach that enables the purification of homogeneous spermatocytes from mouse testis at desired sub-stages. This

An MRL/MpJ strain of mice, including Ipr mutants, reveals the complex pathological manifestations of collagen disease, such as systemic vasculitis, glomerulonephritis, arthritis and sialoadenitis, in association with several autoimmune factors. Studies involving this mouse strain have shown that it exhibits a much-enhanced healing response compared with other mouse strains, together with reduced scarring in the periphery. Recently, unique characteristics were found in the testis of the MRL/MpJ mouse: metaphase-specific apoptosis (MSA) of meiotic spermatocytes, heat stress resistance in spermat

During meiotic prophase I, mammalian spermatocytes must synthesize large amounts of recombination and synapsis proteins despite global transcriptional suppression at the leptotene/zygotene (L/Z) stages. Here, we identify eukaryotic translation elongation factor 1 gamma (eEF1G), highly expressed in spermatogenic cells, as a factor essential for sustaining translation during this transcriptionally quiescent period. Germ cell-specific Eef1g knockout causes complete male infertility due to zygotene arrest, characterized by defects in recombination intermediate stabilization and synapsis. Mechanist

Bisphenol A (BPA) is widely exposed in populations worldwide and has negative effects on spermatogenesis both in animals and humans. The homeostasis of the actin cytoskeleton in the spermatogenic epithelium is crucial for spermatogenesis. Actin cytoskeleton destruction in the seminiferous epithelium is one of the important reasons for BPA-induced spermatogenesis disorder. However, the underlying molecular mechanisms remain largely unexplored. Herein, we explored the role and mechanism of Rsad2, an interferon-stimulated gene in BPA-induced actin cytoskeleton disorder in mouse GC-2 spermatocyte

BACKGROUND: Spermatogenesis is a temperature-sensitive process, and elevation in temperature hampers this process quickly and significantly. We studied the molecular effects of testicular heating on piRNAs and gene expression in rat testicular germ cells.
METHODS: We generated a cryptorchid rat model by displacing the testis from the scrotal sac (34 °C) to the abdominal area (37 °C) and sacrificed animals after 1 day, 3 days, and 5 days. Pachytene spermatocytes and round spermatids were purified using elutriation centrifugation and percoll gradient methods. We performed transcriptome sequenci

In this study, we examined the mechanisms of cadmium exposure-induced endoplasmic reticulum (ER) stress response and apoptosis in spermatocytes. Responses to cadmium toxicity were investigated using spermatocytes overexpressing p50ATF6, ATF4, and spliced XBP1s, belonging to the 3 unfolded protein response pathways. The ER stress and apoptosis response to cadmium were most strongly stimulated through the activating transcription factor 6 (ATF6) pathway; in contrast, siRNA-induced inhibition of protein expression could reduce apoptosis under stressful conditions. The male infertility of cattleyak resulted from spermatogenic arrest has greatly restricted the effective utilization of the heterosis from crossbreeding of cattle and yak. Based on our previous studies, the significant divergences of the transcriptomic and proteomic sequencing between yak and cattleyak prompt us to investigate the critical roles of microRNAs in post-transcriptional regulation of gene expression during spermatogenesis. TUNEL-POD analysis presented sharply decreased spermatogenic cell types and the increased apoptotic spermatogonia in cattleyak. The STA-PUT velocity sedimentati

Sexually reproducing organisms use meiosis to generate haploid gametes and faithfully transmit their genome to the next generation. In comparison to oogenesis in many organisms, spermatogenesis is particularly sensitive to small temperature fluctuations, and spermatocytes must develop within a very narrow isotherm [1-4]. Although failure to thermoregulate spermatogenetic tissue and prolonged exposure to elevated temperatures are linked to male infertility in several organisms, the mechanisms of temperature-induced male infertility have not been fully elucidated [5]. Here, we show that upon exp

Phospholipid hydroperoxide glutathione peroxidase (GPx4) is an intracellular antioxidant enzyme that directly reduces peroxidized phospholipids. GPx4 is strongly expressed in the mitochondria of testis and spermatozoa. We previously found a significant decrease in the expression of GPx4 in spermatozoa from 30% of infertile human males diagnosed with oligoasthenozoospermia (Imai, H., Suzuki, K., Ishizaka, K., Ichinose, S., Oshima, H., Okayasu, I., Emoto, K., Umeda, M., and Nakagawa, Y. (2001) Biol. Reprod. 64, 674-683). To clarify whether defective GPx4 in spermatocytes causes male infertility,

Spermatocytic tumors are a rare type of testicular cancer, comprising <1% of all testicular malignancies. This type of cancer typically affects males in their 60s and 70s and rarely metastasizes; however, it poses a threat to the health of affected individuals if left untreated. The present study describes the case of a 68-year-old male patient with this type of tumor, including a presentation of his initial symptoms, treatment and subsequent monitoring. A male patient, aged 68 years, visited the authors' clinic with an asymptomatic mass in the right testicle. The mass had been progressively i

The proliferative activity and other characteristics of germ cells in the vitamin A-deficient (VAD) rat testis were investigated. In the VAD testis, A spermatogonia and preleptotene spermatocytes were found. The A spermatogonia in the VAD testis showed a bromodeoxyuridine (BrdU) labeling index of 6.6 +/- 1.1% and a mitotic index of 2.8 +/- 0.5%. After continuous labeling with BrdU for up to four days, the ultimate labeling index of A spermatogonia was 11.6 +/- 2.5%, which is less than expected. It is concluded that in the VAD rat testis, many of the proliferating A spermatogonia degenerate. Du

Cadmium (Cd) can cause testis toxicity, and we have demonstrated Cd induced ferroptosis in testis. However, the underlying toxic mechanism has not yet been fully elucidated. Here, we performed in vitro experiments on a mouse spermatocyte cell line. GC–2spd cells were divided into control, Cd, and Cd+ferroptosis inhibitor groups and cultured in high-glucose DMEM for 36 h. We conducted metabolome analysis, RNA sequencing, western blot, and immunofluorescence on GC-2spd cells to determine whether Cd exposure induced ferroptosis in spermatocyte and explore the potential mechanism. The results show

BACKGROUND: Nanoplastics (NPs), owing to their small size and ability to cross the blood-testis barrier, pose a potential risk to the male reproductive system. However, the molecular mechanisms underlying NP-induced reproductive toxicity, particularly cell senescence, remain poorly understood.
OBJECTIVES: This study aims to investigate the mechanisms by which polystyrene nanoplastics (PS-NPs) induce spermatocyte senescence, with a particular focus on the role of YTHDF3-mediated m6A RNA methylation in regulating the cGAS-NF-κB signaling pathway.

BACKGROUND: Caseinolytic peptidase P (ClpP) plays a key role in maintaining cellular homeostasis for mitochondrial quality control. However, the specific function of ClpP during meiosis and its subcellular localization in spermatocytes remain poorly understood.
OBJECTIVE: To investigate the function of ClpP in spermatocyte meiosis.
MATERIALS AND METHODS: ClpP expression was examined in mouse spermatocytes, and tamoxifen was utilized to achieve spatiotemporal-specific deletion of Clpp mediated by Ddx4-CreERT2 in spermatocytes. We analyzed the meiotic progression of Clpp conditional KO (ClppcK

Meiosis, a special cell division, occurs in germ cells to produce gametes. It is vital for reproduction and fostering genetic diversity in offspring. Understanding and characterizing meiotic events are essential for elucidating genetic disorders. Here, we demonstrate a classical technique employing mouse spermatocyte chromosome spreads to study mammalian meiosis. Spermatocytes are isolated from seminiferous tubules of the testis and gently released on a depression slide. These cells are then immersed in a hypotonic solution to introduce swelling. Subsequent incubation in a sucrose solution fac

Spermatocytic tumour (ST) accounts for 1% of testicular germ cell tumours. It is an indolent neoplasm with good prognosis. In approximately 6% of STs, sarcomatous dedifferentiation may occur, portending an aggressive behaviour and representing a significant diagnostic challenge that can lead to its misdiagnosis. Herein, we report the clinicopathological features of a patient with a sarcomatoid spermatocytic tumor, initially diagnosed as mixed germ cell tumour, who was referred to our institution with lung metastases mainly composed of rhabdomyosarcomatous elements. This case report illustrates

The transition from spermatogonia to spermatocytes and the initiation of meiosis are key steps in spermatogenesis and are precisely regulated by a plethora of proteins. However, the underlying molecular mechanism remains largely unknown. Here, we report that Src homology domain tyrosine phosphatase 2 (Shp2; encoded by the protein tyrosine phosphatase, nonreceptor type 11 [Ptpn11] gene) is abundant in spermatogonia but markedly decreases in meiotic spermatocytes. Conditional knockout of Shp2 in spermatogonia in mice using stimulated by retinoic acid gene 8 (Stra8)-cre enhanced spermatogonial di

Varicocele is one of the most important causes of male infertility, as this condition leads to a decline in sperm quality. It is generally believed that the presence of varicocele induces an increase in reactive oxygen species levels, leading to oxidative stress and sperm apoptosis; however, the specific pathogenic mechanisms affecting spermatogenesis remain elusive. Prokineticin 2 (PK2), a secretory protein, is associated with multiple biological processes, including cell migration, proliferation, and apoptosis. In the testis, PK2 is expressed in spermatocytes under normal physiological condi

The purification of individual male germ cell populations is integral for the molecular and biochemical characterisation of specific spermatogenic phases. Although a number of more contemporary techniques have been developed, velocity sedimentation using the STAPUT method remains as a gold standard for this purpose. The gentle nature of the technique, wherein germ cell subpopulations are separated by sedimentation at unit gravity, results in the isolation of viable and high-purity cells. We provide an updated and simplified step-by-step version of the STAPUT protocol for the purification of mo
