Notch signalling is traditionally mediated through direct interaction between the Notch receptor and its transmembrane ligands, presented on the surface of adjacent cells. While both canonical and non-canonical pathways activate Notch signalling in signal-receiving cells, it remains unclear whether Notch itself can function as a ligand to activate receptors in neighboring cells. Here, we report that in the Drosophila testis, Notch expressed in spermatogonial progenitors, functions as a ligand for the phagocytic receptor Draper on neighboring somatic cyst cells. This Reverse-Notch signalling tr

Mouse spermatogenesis entails the maintenance and self-renewal of spermatogonial stem cells (SSCs), which require a complex web-like signaling network transduced by various cytokines. Although brain-derived neurotrophic factor (BDNF) is expressed in Sertoli cells in the testis, and its receptor tropomyosin receptor kinase B (TrkB) is expressed in the spermatogonial population containing SSCs, potential functions of BDNF for spermatogenesis have not been uncovered. Here, we generate BDNF conditional knockout mice and find that BDNF is dispensable for in vivo spermatogenesis and fertility. Howev

Mammalian spermatogenesis produces numerous sperm for a long period based on a highly potent stem cell system, which relies on a special microenvironment, or niche, that has not yet been identified. In this study, using time-lapse imaging of green fluorescent protein-labeled undifferentiated spermatogonia (A(undiff)) and three-dimensional reconstitution, we revealed a biased localization of A(undiff) to the vascular network and accompanying Leydig and other interstitial cells, in intact testes. Differentiating spermatogonia left these niche regions and dispersed throughout the basal compartmen

Cryptorchid testes from adult mice were incubated in calf serum-supplemented medium. There was an effective differentiation of adult type A spermatogonia up to the pachytene stage of meiotid division whcih resembled the process of spermatogenesis in vivo. In the absence of calf serum, type A spermatogonia did not differentiate at all. They differentiated when the serum was present for the first day, but was absent for the rest of cultivation. These results indicate that serum is necessary for only the early process of spermatogenesis from type A spermatogonia in vitro and not for the further p

Spermatogenesis is a continual process that relies on the activities of undifferentiated spermatogonia, which contain spermatogonial stem cells (SSCs) that serve as the basis of spermatogenesis. The gene expression pattern and molecular control of fate decisions of undifferentiated spermatogonia are not well understood. Rho guanine nucleotide exchange factor 15 (ARHGEF15, also known as EPHEXIN5) is a guanine nucleotide-exchange factor (GEF) that activates the Rho protein. Here, we reported that ARHGEF15 was expressed in undifferentiated spermatogonia and spermatocytes in mouse testes; however,

Maintenance and self-renewal of the spermatogonial stem cell (SSC) population in the testis are dictated by the expression of a unique suite of genes. In manipulating gene expression through loss-of-function approaches, we can identify important regulatory mechanisms that dictate spermatogonial fate decisions. One such approach is RNA interference (RNAi), which uses natural cellular responses to small interfering RNAs to decrease levels of a targeted transcript. RNAi is performed in primary cultures of undifferentiated spermatogonia, and can be paired with techniques such as spermatogonial tra

The male infertility of cattleyak resulted from spermatogenic arrest has greatly restricted the effective utilization of the heterosis from crossbreeding of cattle and yak. Based on our previous studies, the significant divergences of the transcriptomic and proteomic sequencing between yak and cattleyak prompt us to investigate the critical roles of microRNAs in post-transcriptional regulation of gene expression during spermatogenesis. TUNEL-POD analysis presented sharply decreased spermatogenic cell types and the increased apoptotic spermatogonia in cattleyak. The STA-PUT velocity sedimentati

Male fertility depends on the regulatory balance between germ cell self-renewal and differentiation, and the spatial and temporal patterns of this balance must be maintained throughout the life cycle. Retinoic acid and its receptors are important factors in spermatogenesis. Spermatogonia cells can self-proliferate and differentiate and have unique meiotic capabilities; they halve their genetic material and produce monomorphic sperm to pass genetic material to the next generation. A number of studies have found that the spermatogenesis process is halted in animals with vitamin A deficiency and

Cryopreservation of spermatogonia (SGs) is crucial for aquaculture, broodstock innovation, and the long-term conservation of aquatic genetic resources. This study developed an efficient cryopreservation protocol for spotted sea bass SGs and evaluated their post-thaw viability and transplantation potential. Initially, we characterized testicular tissue from 6-month-old male spotted sea bass, identifying various germ cell stages and strong Lmvasa expression in SGs. An optimized method for isolating and enriching SGs using Percoll density gradient centrifugation yielded a highly viable (99.6%) SG

Undifferentiated spermatogonia, which form shortly after birth, consist of spermatogonial stem cells and progenitor spermatogonia that maintain homeostasis. As the origin of spermatogenesis, undifferentiated spermatogonia must preserve genome integrity. Paradoxically, we demonstrate that massive spontaneous DNA damage, potentially generated by formaldehyde, arises during the formation of undifferentiated spermatogonia, posing a significant threat to genome integrity. We further reveal that BRCA1 is essential for the timely repair of this spontaneous DNA damage. BRCA1 loss leads to a dramatic r

In the mammalian testis, the mitotic complements of spermatogenic cells are spermatogonia, including spermatogonial stem cells (SSCs) which form the basis of life-long spermatogenesis and male fertility. Thus, investigating spermatogonia and subdivisions thereof is essential to increase our understanding of male germline development and infertility. This protocol describes the isolation of spermatogonia from both adult and developing [postnatal day 6 (P6)] mouse testes. Cell suspensions of the adult mouse testis from the Id4-Egfp transgenic mouse line are obtained through a two-step enzymatic

Sertoli cell nodule consists of clusters of seminiferous tubules composed of immature Sertoli cells and, in some cases, spermatogonia. We report a case of macroscopic Sertoli cell nodule that occurred in the descended left testis of a 26-year-old man. The nodule was centrally located and measured 1.7cm in greatest dimension, the largest such lesion reported to date. Spermatogonia were prominent, a feature that is particularly common in the larger nodules. Macroscopic Sertoli cell nodules (greater than 0.5cm) typically occur in a descended testis. The age range of patients in cases reported sin

AIM: To investigate whether estrogen stimulates the proliferation of spermatogonia or induces spermatogenesis in cryptorchid mice.
METHODS: Mice were surgically rendered cryptorchid, then treated with different doses of 17beta-estradiol (E2) s.c. once a day. Mice were killed at sexual maturity (45 days of age), and histological analysis and immunofluorescence were performed. Serum follicle stimulating hormone (FSH), estradiol, testosterone and luteinizing hormone (LH) were measured.
RESULTS: Low doses of E2 had no notable effect on spermatogonia, but at higher doses, E2 stimulated the prolif

The following conclusions which show the importance of time of medical treatment resulted from the examination of 288 testicular biopsies (including 46 controls) by statistical analysis in correlation to age. 1. There is no difference in the number of spermatogonia per tubule cross section of descended and undescended testes in children from birth to 2 years of age. 2. The number of spermtogonia in descended testes is not different in children from birth to 2 years in comparison to children aged 2-6 years. 3. A statistically significant decrease of spermatogonia takes place in the undescended

Spermatogonial stem cells (SSCs) hold great promise for treating male infertility, but their clinical translation is impeded by the lack of optimal conditions to maintain their undifferentiated state in vitro. In this study, we focused on epigenetic regulators upregulated during differentiation as potential targets. Through a small-molecule screen targeting such conserved regulators, we found that PRMT5 inhibition suppressed mouse SSC differentiation and enhanced their proliferation in a GDNF-deficient, differentiation-prone microenvironment in vitro. Using SSC transplantation assays, we confi

The proliferation of spermatogonia directly affects spermatogenesis and male fertility, but its underlying molecular mechanisms are poorly understood. In this study, Smoothened (Smo), the central transducer of Hedgehog signaling pathway, was characterized in medaka (Oryzias latipes), and its role and underlying mechanisms in the proliferation of spermatogonia were investigated. Smo was highly expressed in spermatogonia. In ex vivo testicular organ culture and a spermatogonial cell line (SG3) derived from medaka mature testis, Smo activation promoted spermatogonia proliferation, while its inhib

Robust methods have been developed that leverage next-generation sequencing (NGS) to measure abundance of all mRNAs (RNA-seq) in samples as small as individual cells in order to study the testicular transcriptome in mammals. In this chapter, we present robust options for implementing bioinformatics workflows for the analysis of bulk RNA-seq from aggregate samples of hundreds to millions of cells and single-cell RNA-seq from individual cells. We also provide detailed protocols for using the R packages DESeq2 and Seurat, important parameters for successful implementation, and considerations for

Changes in zebrafish testicular gene expression induced by follicle-stimulating hormone (Fsh) or anti-Mullerian hormone (Amh) suggested that Amh inhibition and Fsh stimulation of spermatogenesis involved up and downregulation, respectively, of prostaglandin (PG) signaling. We found that Sertoli cells contacting type A undifferentiated (Aund) and differentiating (Adiff) spermatogonia expressed a key enzyme of PG production (Ptgs2); previous work showed that Sertoli cells contacting Adiff and B spermatogonia and spermatocytes showed ptges3b expression, an enzyme catalyzing PGE2 production. A bulk of evidence suggests that d-aspartate (d-Asp) regulates steroidogenesis and spermatogenesis in vertebrate testes. This review article focuses on intracellular signaling mechanisms elicited by d-Asp possibly via binding to the N-methyl-d-aspartate receptor (NMDAR) in both Leydig cells, and spermatogonia. In Leydig cells, the amino acid upregulates androgen production by eliciting the adenylate cyclase-cAMP and/or mitogen-activated protein kinase (MAPK) pathways. d-Asp treatment enhances gene and protein expression of enzymes involved in the steroidogenic cascade. d-Asp also directly affe

STUDY QUESTION: Does a combined approach allow for the unequivocal detection of human germ cells and particularly of spermatogonia in vitro?
SUMMARY ANSWER: Based on our findings, we conclude that an approach comprising: (i) the detailed characterization of patients and tissue samples prior to the selection of biopsies, (ii) the use of unambiguous markers for the characterization of cultures and (iii) the use of biopsies lacking the germ cell population as a negative control is the prerequisite for the establishment of human germ cell cultures.
WHAT IS KNOWN ALREADY: The use of non-specific
