In vitro studies indicate a quinone is involved in bacterial Mn(II) oxidation.

Manganese(II)-oxidizing bacteria play an integral role in the cycling of Mn as well as other metals and organics. Prior work with Mn(II)-oxidizing bacteria suggested that Mn(II) oxidation involves a multicopper oxidase, but whether this enzyme directly catalyzes Mn(II) oxidation is unknown. For a clearer understanding of Mn(II) oxidation, we have undertaken biochemical studies in the model marine alpha-proteobacterium, Erythrobacter sp. strain SD21. The optimum pH for Mn(II)-oxidizing activity was 8.0 with a specific activity of 2.5 nmol x min(-1) x mg(-1) and a K (m) = 204 microM. The activit

Biodegradation and toxicity attenuation of bisphenol A by Sphingopyxis granuli XYQ201: mechanism elucidation and wastewater application.

Bisphenol A (BPA) is a widespread endocrine-disrupting contaminant in wastewater, and microbial biodegradation is a promising approach for its removal. However, BPA-degrading bacteria with clarified degradation products, reduced estrogenic activity after degradation, and demonstrated performance in real wastewater remain limited. In this study, a BPA-degrading bacterium, Sphingopyxis granuli XYQ201, was isolated from municipal wastewater and shown to utilize BPA as the sole carbon source. Strain XYQ201 completely removed 50 mg/L BPA within 38 h under laboratory conditions. Four major degradati

Biocontrol potential of endophytic Sphingobium xenophagum J-1 against Fusarium oxysporum causing root rot in Paris polyphylla var. yunnanensis.

Paris polyphylla var. yunnanensis (PPY) is a valuable medicinal plant frequently threatened by root rot caused by pathogenic fungi such as Fusarium oxysporum. Traditional chemical control methods pose environmental and resistance risks, highlighting the need for sustainable alternatives. This study investigated the antifungal potential of the endophytic bacterium Sphingobium xenophagum J-1, isolated from healthy PPY, against F. oxysporum in vitro. The ethyl acetate crude extract of J-1 fermentation broth exhibited significant concentration-dependent inhibition of spore germination, with a 94.6

Effluent decontamination by the ibuprofen-mineralizing strain, Sphingopyxis granuli RW412: Metabolic processes.

The high global consumption of ibuprofen and its limited elimination by wastewater treatment plants (WWTPs), has led to the contamination of aquatic systems by this common analgesic and its metabolites. The potentially negative environmental and public health effects of this emerging contaminant have raised concerns, driving the demand for treatment technologies. The implementation of bacteria which mineralize organic contaminants in biopurification systems used to decontaminate water or directly in processes in WWTPs, is a cheap and sustainable means for complete elimination before release in

Novosphingobium aromaticivorans: a potential initiator of primary biliary cirrhosis.

Primary biliary cirrhosis (PBC) is characterized by a T-cell-mediated destruction of bile duct epithelial cells that line the small intrahepatic bile ducts. The targets of activated T-lymphocytes are the dihydrolipoamide acetyltransferase components of the 2 oxo acid dehydrogenases, enzyme complexes that are important in oxidative energy metabolism. Pyruvate dehydrogenase is the best known of these. Its dihydrolipoamide acetyltransferase component is referred to as PDC-E2. A major question in understanding the pathogenesis of PBC is why PBC patients lose their tolerance to antigens that are fo

A novel metronidazole-degrading bacterium Sphingopyxis sp. K5: isolation, characterization, and application potential in soil and wastewater remediation.

Metronidazole is a widely used nitroimidazole antibiotic in clinical practice. Following its widespread use, it enters the environment via medical wastewater, posing a potential risk of toxicity to ecosystems and living organisms. Microbial degradation is an efficient method of accurately eliminating target pollutants from the environment. Here, a metronidazole degrader, Sphingopyxis sp. K5, was isolated from medical wastewater sludge. Strain K5 could degrade 58.7 μ M metronidazole after 4 days of incubation with an initial OD600 of 1.5. The optimal degradation conditions were 35°C and pH 8.0,

Biosynthesis of poly-beta-hydroxyalkanoate by Brevundimonas vesicularis LMG P-23615 and Sphingopyxis macrogoltabida LMG 17324 using acid-hydrolyzed sawdust as carbon source.

Poly-beta-hydroxyalkanoate (PHA) is a biodegradable polymer accumulated in intracellular granules by different bacterial species. Its physical and chemical properties are similar to those of petroleum-derived plastics. Material generated by the acid hydrolysis of wood was evaluated for use in the bacterial synthesis of PHA. Acid-hydrolyzed sawdust was prepared and adjusted to pH 7. Mineral salts with carbon:nitrogen (C:N) proportions of 100:1, 100:3.5, 100:10, 100:30, or 100:50 and trace elements were added and these solutions were inoculated with a bacterial strain Brevundimonas vesicularis L

Stringent response-mediated ferroptosis-like death resistance underlies Novosphingobium persistence during ciprofloxacin stress.

Antibiotics, as emerging hazardous materials in the environment, pose significant risks to ecosystems and contribute to the spread of antibiotic-resistant bacteria. Although extensive knowledge has been accumulated on antibiotic-resistance mechanisms in individual bacteria, less is understood about how the bacterial communities respond to antibiotic exposure under natural environmental conditions, where nutrient supplies are often limited and fluctuating. Here, we report that Novosphingobium dominated in a wetland bacterial community under 1 µg/mL ciprofloxacin (CIP) exposure and persisted dur

Unusual Relationship between Iron Deprivation and Organophosphate Hydrolase Expression.

Organophosphate hydrolases (OPH), hitherto known to hydrolyze the third ester bond of organophosphate (OP) insecticides and nerve agents, have recently been shown to interact with outer membrane transport components, namely, TonB and ExbB/ExbD. In an OPH negative background, Sphingopyxis wildii cells failed to transport ferric enterobactin and showed retarded growth under iron-limiting conditions. We now show the OPH-encoding organophosphate degradation (opd) gene from Sphingobium fuliginis ATCC 27551 to be part of the iron regulon. A fur-box motif found to be overlapping with the transcriptio

Microbial degradation of nondesorbable organic compounds on biochars by extracellular reactive oxygen species.

Knowledge of microbial degradation of biochar-adsorbed organic pollutants is essential for recovering adsorption performance of biochars and reducing secondary pollution in soil remediation. In previous study, desorption of organic compounds from biochars was perceived as a prerequisite for the microbial degradation. However, microbial degradation of the nondesorbable organic compounds on biochars has not been studied. Therefore, degradation of nondesorbable naphthalene (NAPH), phenanthrene (PHEN) and pyrene (PYR) on a wood chip-derived biochar (WBC700) by Sphingobium yanoikuyae B1 was investi

Changes in the gut microbiota of pseudoxanthoma elasticum patients.

OBJECTIVE: Pseudoxanthoma elasticum (PXE) is a rare autosomal disorder with a variable phenotype that may be modulated by environmental factors. Plasma vitamin K (VK) levels may be involved in the ectopic calcification process observed in PXE. Since VK2 is predominantly produced by the gut microbiota, we hypothesized that changes in the gut microbiota of PXE patients might exacerbate the calcification process and disease symptoms. METHODS: Twenty PXE patients were included in the study and 60 gut microbiota profiles from the Biofortis laboratory database were used as controls. RESULTS: The R

Isolation and Characterization of Sphingopyxis sp. Q841: A New Zearalenone- Degrading Strain.

Zearalenone (ZEN) is a mycotoxin exhibiting estrogenic activity that poses a considerable risk to human health. In this study, a ZEN-degrading bacterium isolated from soil was identified as Sphingopyxis sp. and designated strain Q841. Q841 completely degraded 5 mg/L of ZEN within 4 h in mineral medium. It showed the highest activity within the tested ranges (25-37°C, pH 4.0-9.0), with optimal activity at pH 9.0 and 37°C. Assays of degradation efficiency in different fractions indicated that the ZEN-degrading activity was mainly associated with intracellular components rather than with the extr

Precise Transcription Control Drives Optimized Catabolism of β-5-Type Lignin-Derived Dimers in Sphingobium lignivorans SYK-6.

Sphingobium lignivorans SYK-6, a promising platform for lignin valorization, can degrade dehydrodiconiferyl alcohol (DCA), which is a β-5-type dimer. We found that phcR (a gene encoding a MarR-type transcriptional regulator) negatively regulates the transcription of DCA catabolism genes. Furthermore, the previously identified DesR was observed to be involved in the regulation of a specific DCA catabolism gene. The effector molecule of PhcR was identified as a DCA metabolite: 5-(2-carboxyvinyl)-2-(4-hydroxy-3-methoxyphenyl)-7-methoxy-2,3-dihydrobenzofuran-3-carboxylic acid. The disruption of ph

Kinetic and Structural Characterization of the First B3 Metallo-β-Lactamase with an Active-Site Glutamic Acid.

The structural diversity in metallo-β-lactamases (MBLs), especially in the vicinity of the active site, has been a major hurdle in the development of clinically effective inhibitors. Representatives from three variants of the B3 MBL subclass, containing either the canonical HHH/DHH active-site motif (present in the majority of MBLs in this subclass) or the QHH/DHH (B3-Q) or HRH/DQK (B3-RQK) variations, were reported previously. Here, we describe the structure and kinetic properties of the first example (SIE-1) of a fourth variant containing the EHH/DHH active-site motif (B3-E). SIE-1 was ident

Discovery of novel enzyme genes involved in the conversion of an arylglycerol-β-aryl ether metabolite and their use in generating a metabolic pathway for lignin valorization.

Microbial conversions known as "biological funneling" have attracted attention for their ability to upgrade heterogeneous mixtures of low-molecular-weight aromatic compounds obtained by chemical lignin depolymerization. β-hydroxypropiovanillone (HPV) and its analogs can be obtained by chemoselective catalytic oxidation of lignin using 2,3-dichloro-5,6-dicyano-1,4-benzoquinone/tert-butyl nitrite/O2, followed by cleavage of arylglycerol-β-aryl ether with zinc. Sphingobium sp. strain SYK-6 can degrade HPV generated by the catabolism of arylglycerol-β-aryl ether through 2-pyrone-4,6-dicarboxylate
