Solubilization of aster-forming proteins from yeast: possible constituents of spindle pole body and reconstitution of asters in vitro.

Spindle pole bodies (SPBs) attached to nuclei were isolated from Saccharomyces cerevisiae; they nucleated microtubules to form asters in vitro. This aster-forming activity was stable in 30% dimethylsulfoxide or 10% 2-propanol and could be solubilized with KCl. The KCl extract contained many protein components, which aggregated upon dialysis against a low concentration salt solution. When incubated with tubulin, the aggregates formed asters. Measurements of the elongation rates of the astral microtubules indicated that the microtubules were nucleated from the SPBs or from the aggregates reconst

Meiotic progression in multinuclear mouse spermatocytes without the spindle pole clustering motor protein KIFC1 or cytokinesis forms single-cell late-stage spermatids.

Mammalian spermatogenesis is a highly dynamic process involving male germ cell self-renewal and terminal differentiation into progenitor cells that produce testicular sperm in coordination with testicular somatic support cells. Multinuclear giant cells identified in mutant, toxin-exposed or genetically altered mice post-meiosis, reflect abnormal spermiogenesis. Here, analysis of fixed isolated testicular cells immunolabeled for microtubules, molecular motors, spindle binding proteins, and DNA from non-transgenic and GFP-CETN2-expressing CB6F1 or C57BL mice identify polynuclear primary spermato

Evaluating serum extra spindle pole bodies-like 1 protein vs p53 antibody for hepatitis B virus-related hepatocellular carcinoma diagnosis.

BACKGROUND: Hepatitis B virus (HBV) infection is a leading cause of global hepatocellular carcinoma (HCC). Conventional biomarkers such as alpha-fetoprotein (AFP) demonstrate suboptimal sensitivity and specificity. Emerging evidence suggests that serum extra spindle pole bodies like 1 (ESPL1) protein and p53 antibody may improve diagnostic accuracy. AIM: To assess and compare the diagnostic performance of serum ESPL1 protein and p53 antibody in HBV-related HCC (HBV-HCC). METHODS: This case-control study from the First Affiliated Hospital of Guangxi Medical University enrolled 30 patients wit

Development and Validation of an Extra Spindle Pole Bodies-like 1-Based Diagnostic and Prognostic Model for Hepatitis B Virus-Related Hepatocellular Carcinoma: Retrospective Cohort Study.

BACKGROUND: Early diagnosis of hepatocellular carcinoma (HCC) in patients with chronic hepatitis B virus (HBV) is challenging. Models that combine novel biomarkers with clinical features may improve both early diagnosis and risk stratification, but few have been systematically validated. OBJECTIVE: This study aimed to develop and validate an extra spindle pole bodies-like 1 (ESPL1)-based model for diagnostic discrimination of HBV-related HCC and longitudinal risk stratification in patients with chronic HBV infection. METHODS: Patients with chronic HBV were consecutively recruited from the Fi

An Asp-CaM complex is required for centrosome-pole cohesion and centrosome inheritance in neural stem cells.

The interaction between centrosomes and mitotic spindle poles is important for efficient spindle formation, orientation, and cell polarity. However, our understanding of the dynamics of this relationship and implications for tissue homeostasis remains poorly understood. Here we report that Drosophila melanogaster calmodulin (CaM) regulates the ability of the microcephaly-associated protein, abnormal spindle (Asp), to cross-link spindle microtubules. Both proteins colocalize on spindles and move toward spindle poles, suggesting that they form a complex. Our binding and structure-function analys

Expansion microscopy reveals characteristic ultrastructural features of pathogenic budding yeast species.

Candida albicans is the most prevalent fungal pathogen associated with candidemia. Similar to other fungi, the complex life cycle of C. albicans has been challenging to study with high-resolution microscopy due to its small size. Here, we employed ultrastructure expansion microscopy (U-ExM) to directly visualise subcellular structures at high resolution in the yeast and during its transition to hyphal growth. N-hydroxysuccinimide (NHS)-ester pan-labelling in combination with immunofluorescence via snapshots of various mitotic stages provided a comprehensive map of nucleolar and mitochondrial s

CPAP insufficiency leads to incomplete centrioles that duplicate but fragment.

Centrioles are structures that assemble centrosomes. CPAP is critical for centrosome assembly, and its mutations are found in patients with diseases such as primary microcephaly. CPAP's centrosomal localization, its dynamics, and the consequences of its insufficiency in human cells are poorly understood. Here we use human cells genetically engineered for fast degradation of CPAP, in combination with superresolution microscopy, to address these uncertainties. We show that three independent centrosomal CPAP populations are dynamically regulated during the cell cycle. We confirm that CPAP is crit

Flagellar duplication and migration during the Trichomonas vaginalis cell cycle.

Trichomonas vaginalis is a flagellated protozoan, a representative of 1 of the earliest known eukaryotic lineages. Trichomonas vaginalis lacks centrioles but possesses basal bodies. We report here the cell cycle-dependent flagellar dynamics of T. vaginalis. By immunofluorescence, we found that T. vaginalis flagella duplicated during S-phase, segregated toward the nuclear poles, and then emanated from the spindle poles at mitosis. This behavior strongly parallels that of centrioles and other spindle pole-associated structures variously termed centrosomes, spindle pole bodies, or microtubule org

A bifunctional ARHGEF17-AS1 locus encodes MSEP, a protein that supports mitotic spindle organization.

Bifunctional genomic loci can produce both coding and noncoding outputs, yet the proteins encoded by many such loci remain unexplored. Here, we identify MSEP, a previously uncharacterized ∼54-kDa protein translated from an open reading frame embedded within the antisense RNA locus ARHGEF17-AS1. MSEP localizes to the mitotic spindle and spindle poles, and disruption of its start codon increases spindle abnormalities. MSEP interacts with the spindle-assembly factor TPX2, and its depletion is accompanied by reduced TPX2 abundance. MSEP undergoes cell-cycle-dependent phosphorylation that is regula

Sequential changes in calcium transients during M phase regulate cardiomyocyte proliferation.

Heart muscle growth and regeneration require the proliferation of cardiomyocytes. Rapid pulsatile increases in cytosolic Ca2+ concentration, called calcium transients (CaTs), trigger cardiomyocyte contractions, but how cardiomyocytes adapt Ca2+ signaling during proliferation is largely unknown. Here, we show that cardiomyocyte proliferation requires changes in Ca2+ signaling. Cardiomyocytes undergo a sequence of CaT changes during M phase: CaT amplitudes begin to decline in prometaphase, reach a minimum in metaphase, rise during anaphase, and return to the original state in daughter cardiomyoc

NuMA-related LIN-5, ASPM-1, calmodulin and dynein promote meiotic spindle rotation independently of cortical LIN-5/GPR/Galpha.

The spindle apparatus dictates the plane of cell cleavage, which is critical in the choice between symmetric or asymmetric division. Spindle positioning is controlled by an evolutionarily conserved pathway, which involves LIN-5/GPR-1/2/Galpha in Caenorhabditis elegans, Mud/Pins/Galpha in Drosophila and NuMA/LGN/Galpha in humans. GPR-1/2 and Galpha localize LIN-5 to the cell cortex, which engages dynein and controls the cleavage plane during early mitotic divisions in C. elegans. Here we identify ASPM-1 (abnormal spindle-like, microcephaly-associated) as a novel LIN-5 binding partner. ASPM-1, t

Aurora A phosphorylation of WD40-repeat protein 62 in mitotic spindle regulation.

Mitotic spindle organization is regulated by centrosomal kinases that potentiate recruitment of spindle-associated proteins required for normal mitotic progress including the microcephaly protein WD40-repeat protein 62 (WDR62). WDR62 functions underlie normal brain development as autosomal recessive mutations and wdr62 loss cause microcephaly. Here we investigate the signaling interactions between WDR62 and the mitotic kinase Aurora A (AURKA) that has been recently shown to cooperate to control brain size in mice. The spindle recruitment of WDR62 is closely correlated with increased levels of

Pericentrin: critical for spindle orientation.

Mutations in the pericentrin (PCNT) gene cause Majewski osteodysplastic primordial dwarfism type II (MOPDII). Recent work reveals that a discrete set of centrosome proteins require PCNT for their robust localization to mitotic spindle poles. Critically, this complex is crucial for mitotic spindle orientation and involved in the pathogenesis of MOPDII.

Human ASPM participates in spindle organisation, spindle orientation and cytokinesis.

BACKGROUND: Mutations in the Abnormal Spindle Microcephaly related gene (ASPM) are the commonest cause of autosomal recessive primary microcephaly (MCPH) a disorder characterised by a small brain and associated mental retardation. ASPM encodes a mitotic spindle pole associated protein. It is suggested that the MCPH phenotype arises from proliferation defects in neural progenitor cells (NPC). RESULTS: We show that ASPM is a microtubule minus end-associated protein that is recruited in a microtubule-dependent manner to the pericentriolar matrix (PCM) at the spindle poles during mitosis. The effects of diazepam on mitosis and the microtubule cytoskeleton. I. Observations on the diatoms Hantzschia amphioxys and Surirella robusta.

The effects of diazepam (DZP) on mitosis and the microtubule (MT) cytoskeleton in the live diatoms Hantzschia amphioxys and Surirella robusta were followed using time-lapse video microscopy. Similarly treated cells were fixed and later examined for immunoflouresence staining of MTs or for transmission electron microscopy. DZP treatment (250 microM) had no effect on interphase cells but affected mitosis, resulting in the majority of prometaphase and metaphase chromosomes releasing from one or both spindle poles and collecting irregularly along the central spindle. Chromosomes remaining attached

Disjunction of conjoined twins: Cdk1, Cdh1 and separation of centrosomes.

Accurate transmission of chromosomes from parent to progeny cell requires assembly of a bipolar spindle. Centrosomes (spindle pole body in yeast) are critical for the biogenesis of this complex mitotic apparatus since they confer bipolarity on the spindle and serve as the site of microtubule polymerization. In each division cycle, the centrosome is duplicated and the sister-centrosomes move away from each other, forming the two poles of the spindle. While the structure and the duplication of centrosomes have been investigated extensively, the understanding of the control of their segregation r

The spindle-associated antisense RNA ARHGEF17-AS1 functionally contributes to mitotic spindle integrity.

Long non-coding RNAs (lncRNAs) are increasingly recognized as regulators of mitotic progression, yet few have been shown to act directly at the mitotic spindle. Here we identify ARHGEF17-AS1 as a metaphase-enriched antisense RNA that localizes to the mitotic spindle and is associated with spindle pole integrity. ARHGEF17-AS1 interacts with RPS3 and the dynein motor complex and is associated with RPS3 recruitment to the mitotic spindle and with the RPS3-dynein interaction. Depletion of ARHGEF17-AS1 reduces this interaction and decreases RPS3 accumulation at spindle poles. ARHGEF17-AS1 depletion

Segregation of holocentric chromosomes at meiosis in the nematode, Caenorhabditis elegans.

The meiotic segregation of the holocentric chromosomes of Caenorhabditis elegans in both spermatogenesis and oogenesis is described. The extended kinetochore typical of the mitotic chromosome could not be differentiated on meiotic bivalents; instead microtubules appeared to project into the chromatin. The meiotic spindles formed during spermatogenesis contain centrioles and asters, while in oogenesis the spindles are acentriolar and barrel shaped. The formation of the acentriolar spindle was studied in fixed specimens by anti-tubulin immunofluorescence. Microtubule arrays were seen first to ac
