Role of maize kernels on the fitness of Spodoptera frugiperda (Lepidoptera: Noctuidae).

Recent outbreak of the fall armyworm, Spodoptera frugiperda (JE Smith), has caused considerable challenges to maize production, Zea mays L., in the world. Sustainable pest management through host plant resistance requires further confirmation of maize hybrids that reduce pest pressure during postflowering stage. In this study, the comparative resistance of kernels from 10 maize hybrids, including BC678, SC201, SC704, SC410, SC706, SC400, SC260, BK66, SC705, and BK65, was evaluated using the nutritional indices, demographic parameters, and enzymatic activities of Spodoptera frugiperda in relati

Spodoptera frugiperda FKBP-46 is a consensus p53 motif binding protein.

p53 protein, the central molecule of the apoptosis pathway, is mutated in 50% of the human cancers. Of late, p53 homologues have been identified from different invertebrates including Drosophila melanogaster, Caenorhabditis elegans, Squid, and Clams. We report the identification of a p53-like protein in Spodoptera frugiperda (Sf9) insect cells, which is activated during oxidative stress, caused by exposure to UV-B or H(2) O(2) , and binds to p53 consensus DNA binding motifs as well as other p53 cognate motifs. Sf9 p53 motif-binding protein is similar to murine and Drosophila p53 in terms of mo

Biogenic amines modulate cannibalistic behaviors in the fall armyworm, Spodoptera frugiperda.

BACKGROUND: Cannibalism, defined as the ingestion of an individual by another individual from the same species, is a phenomenon prevalent across various animal groups, particularly among the larvae of the fall armyworm, Spodoptera frugiperda. These larvae exhibit cannibalistic tendencies under specific environmental conditions. RESULTS: This study demonstrates that cannibalism is rarely observed during the first- and second-instar stages in S. frugiperda. The highest proportion of cannibalism was recorded among fifth- and sixth-instar larvae, with a clear increasing trend observed from the fo

Testing the joint effects hypothesis of elemental defense using Spodoptera exigua.

Metal hyperaccumulation may be an elemental defense, in which high concentrations of a metal in plant tissues decrease herbivore survival or growth rate. The Joint Effects Hypothesis suggests that a combination of metals, or a combination of a metal with an organic compound, may have an enhanced defensive effect. The enhancement may be additive or synergistic: in either case the concentration of a particular metal necessary to provide a defensive benefit for the plant is lowered. We tested the Joint Effects Hypothesis using Spodoptera exigua (beet armyworm) neonates fed artificial diets. Metal

A Glycoprotein Hormone Orchestrates Ovarian Development in the Fall Armyworm Spodoptera Frugiperda.

Glycoprotein hormones serve as critical regulators of diverse physiological processes in both vertebrates and invertebrates, yet their functional roles in insect reproduction remain poorly understood. Here, we identified two glycoprotein hormone subunits, SfLFT1 and SfLFT2, and their cognate receptor SfLFTR (a leucine-rich-repeat-containing GPCR) in Spodoptera frugiperda. Liposome-encapsulated siRNA-mediated knockdown of these genes severely disrupted ovarian development as evidenced by suppressed vitellogenin biosynthesis and reduced mature follicle formation. Coimmunoprecipitation and molecu

Functional analysis of Sirt5 in the effect of azadirachtin on the intestinal injury of Spodoptera litura.

Azadirachtin, a major botanical insecticide, is widely recognized as a sustainable alternative to synthetic pesticides, yet the molecular mechanisms underlying its effects on insect hosts remain poorly understood. Transcriptomic analysis, RT-PCR, and qPCR revealed significant upregulation of Sirt5 in the midgut following azadirachtin exposure. In the present study, we investigated the role of Sirt5, a key regulator of metabolic and stress responses, in mediating midgut damage in Spodoptera litura under azadirachtin stress. Functional assays showed that overexpression of Sirt5 in SpLi-221 cells

Functional characterization of carbon dioxide receptors in the fall armyworm, Spodoptera frugiperda.

BACKGROUND: Carbon dioxide (CO2) is an important chemosensory cue involved in host plant recognition and environmental navigation in moths. In insects, CO2 detection is generally mediated by specific gustatory receptors (GRs). This study aimed to identify and functionally characterize CO2-related GRs in the fall armyworm, Spodoptera frugiperda. RESULTS: Three candidate GR genes, designated SfruGR1, SfruGR2, and SfruGR3, were identified through genomic and transcriptomic analyses. Phylogenetic analysis showed that these receptors clustered with conserved insect CO2 GRs and formed three distinc

Effects of surfactin stress on gene expression and pathological changes in Spodoptera litura.

Spodoptera litura (S. litura) is a polyphagous pest of the family Lepidoptera, which causes damage and yields losses to many crops. The long-term use of chemical pesticides for control not only seriously threatens environmental health, but also causes S. litura to develop drug resistance. Therefore, there is an urgent need to develop environmentally safe and friendly biogenic pesticides. However, the mechanism of action of the secondary metabolite (surfactin) of Bacillus Vélezensis (B. vélezensis) on lepidopteran pests (S. litura) has not been reported yet. We found that several metabolites an

Effect of X-ray irradiation on development, flight, and reproduction of Spodoptera litura.

Spodoptera litura is an omnivorous pest that has spread globally. Because irradiation sterilization technology has a great potential for control of S. litura, the effect of 25-150 Gy doses of X-rays on pupal survival, flight and reproductive variables of adult moths were analyzed in this research. The X-ray irradiation with the dose of 25-150 Gy significantly affected the reproductive ability of females. Irradiating male pupae with 25-150 Gy doses of X-rays had no effect on mating, life span, or flight ability of adult moths, but significantly reduced survival and fecundity of their offspring,

Zea mays Volatiles that Influence Oviposition and Feeding Behaviors of Spodoptera frugiperda.

Fall armyworm (Spodoptera frugiperda) is a major global pest of many crops, including maize (Zea mays). This insect is known to use host plant-derived volatile organic compounds to locate suitable hosts during both its adult and larval stages, yet the function of individual compounds remains mostly enigmatic. In this study, we use a combination of volatile profiling, electrophysiological assays, pair-wise choice behavioral assays, and chemical supplementation treatments to identify and assess specific compounds from maize that influence S. frugiperda host location. Our findings reveal that met

Multi-Omic Analysis Identifies Key Genes Driving Testicular Fusion in Spodoptera litura.

The Spodoptera litura, a Lepidopteran pest known for its high fecundity, undergoes a complete metamorphosis, including a distinctive process during which the male testes fuse from two separate organs into a single entity, significantly enhancing its fertility. To elucidate the molecular mechanisms underlying this testicular fusion, this study employed an integrated multi-omics approach to investigate concurrent changes at the transcriptomic and proteomic levels. We identified a series of synchronized alterations on the peritestic larval membrane, including heme binding, peptidase activity, hyd

Potential insecticidal extracts from Artocarpus lacucha against Spodoptera litura (Lepidoptera: Noctuidae) larvae.

Artocarpus lacucha, a plant in the Moraceae family, has traditionally been used in Southeast Asian medicine to treat various ailments. This study investigated the insecticidal potential against Spodoptera litura of several compounds extracted from A. lacucha using a topical application method. A sequential extraction method was employed with A. lacucha stems to identify the most toxic crude extract by using hexane, dichloromethane, ethyl acetate, and methanol solvents. Subsequently, the most toxic crude extract was analyzed for chemical composition by HPLC, followed by the isolation process. A

Host detoxification and gut microbiota are associated with chlorantraniliprole resistance in Spodoptera frugiperda.

Spodoptera frugiperda is a major agricultural pest that causes severe damage in China. Chlorantraniliprole (CAP) is the primary insecticide used for control; however, it is yet unknown how much the intrinsic detoxification system and gut microbiota of the host contribute to resistance. In this study, we used a resistant strain (CR) with a 71.85-fold resistance ratio after 10 generations of CAP selection. The molecular and microbial changes associated with CAP resistance in S. frugiperda were systematically analyzed by integrating phenotypic and multi-omics data. Results demonstrated that detox

Construction of an infectious clone of Spodoptera frugiperda densovirus and its biological characteristics.

Densoviruses are highly pathogenic to their insect hosts and have great potential for biocontrol. Spodoptera frugiperda densovirus (SfDV) was isolated from diseased larvae of Spodoptera frugiperda, while its biological functions remain unclear. Herein, we successfully constructed an infectious clone of SfDV. The S. frugiperda larvae transfected with the infectious clone exhibited anorexia, stunted growth, and reduced activity. Histopathological analysis further showed that the epidermis, fat body and trachea were infected instead of muscle and midgut tissues. Transmission electron microscopy (

β2-tubulin regulates the development and migration of eupyrene sperm in Spodoptera frugiperda.

Spermatogenesis is the basis of sexual reproduction and is essential for the propagation of insect populations. Understanding the process of spermatogenesis and identifying key genes involved in sperm function can aid in developing pest genetic control methods. The testis-specific gene β2-tubulin (B2t) is crucial for spermatogenesis in insects possessing monomorphic spermatids. However, the role of B2t in lepidopteran dimorphic spermatogenesis remains unclear. In this study, we explored the effect of B2t in the development of eupyrene and apyrene sperm in the Spodoptera frugiperda, a major glo
