Supramolecular solvent-based ultrasound-assisted extraction of fangchinoline and tetrandrine from the roots of Stephaniae tetrandrae.

A supramolecular solvent (SUPRAS) based ultrasound-assisted extraction method was developed for the extraction of fangchinoline and tetrandrine from Radix Stephaniae tetrandrae. SUPRASs were prepared in aqueous solutions containing hexafluorobutanol (HFB) and alkanols for the first time. HFB was used as a coacervation agent and density-regulating agent for the SUPRAS preparation. The biosurfactants, namely hexanol, octanol, and decanol were selected as the amphiphiles. The possible mechanism of the new SUPRAS formation was studied. The hydrogen-bond and hydrophobic interactions between HFB and

Isoquinoline alkaloids from Stephania japonica var. discolor and their neuroprotective effects against OGD/R-induced oxidative injury.

Stephania japonica var. discolor is a traditional medicinal plant reported to contain alkaloids. In this study, eight isoquinoline alkaloids were isolated from the whole plant, including four previously undescribed compounds (1-3, 6). These compounds were evaluated for neuroprotective effects using an oxygen-glucose deprivation/reperfusion (OGD/R) model in PC-12 cells. Several of the isolated alkaloids significantly improved cell viability and reduced intracellular reactive oxygen species (ROS) levels under OGD/R conditions. These results suggest that the neuroprotective activity of these comp

Cepharanthin, an alkaloid from Stephania cepharantha, inhibits increased pulmonary vascular permeability in an ovine model of sepsis.

Sepsis is a life-threatening event when it occurs in patients suffering from smoke inhalation injury. Pneumonia is one of the most frequent sources of infection in sepsis. Activated leukocytes likely play a role in the pathogenesis of sepsis. Cepharanthin is a biscoclaurine alkaloid that reportedly inhibits the activation of neutrophils. In this study, we investigated the effects of cephranthin on a post-smoke inhalation model of sepsis in sheep. Female sheep (n = 15) were surgically prepared for the study. After 5 days recovery from the operative procedures, tracheostomy was performed in all

Tetrandrine and fangchinoline, bisbenzylisoquinoline alkaloids from Stephania tetrandra can reverse multidrug resistance by inhibiting P-glycoprotein activity in multidrug resistant human cancer cells.

The overexpression of ABC transporters is a common reason for multidrug resistance (MDR) in cancer cells. In this study, we found that the isoquinoline alkaloids tetrandrine and fangchinoline from Stephania tetrandra showed a significant synergistic cytotoxic effect in MDR Caco-2 and CEM/ADR5000 cancer cells in combination with doxorubicin, a common cancer chemotherapeutic agent. Furthermore, tetrandrine and fangchinoline increased the intracellular accumulation of the fluorescent P-glycoprotein (P-gp) substrate rhodamine 123 (Rho123) and inhibited its efflux in Caco-2 and CEM/ADR5000 cells.
