TBBPA downregulates thyroid receptor and estrogen receptor mRNA levels in goldfish gonadal tissue.

Tetrabromobisphenol A (TBBPA) is a brominated flame retardant that has been shown to be a potential thyroid disrupting chemical. Currently, TBBPA is not included in the UN's list of endocrine disruptors and adverse effects of TBBPA in mammals has not been fully investigated. However, there is clear evidence that TBBPA exerts adverse health effects on reproduction of aquatic species. Therefore, it is important to provide more information on potential endocrine disruptive effects of TBBPA in vertebrate species. In this study we investigated the effect of TBBPA on transcript levels of estrogen re

Effect of Tetrabromobisphenol A on expression of biomarkers for inflammation and neurodevelopment by the placenta.

OBJECTIVE: Polybrominated diphenyl ethers (PBDEs) enhance basal and bacteria-stimulated production of proinflammatory cytokines by the placenta and may also increase the risk of preterm birth and neurodevelopmental disorders. Whether or not other brominated flame retardants such as Tetrabromobisphenol A (TBBPA) have similar properties is unclear. Therefore, we evaluated the effects of TBBPA on the production of steroids, as well as biomarkers for inflammation, oxidative stress, and neurodevelopment by the placenta. METHODS: Placental explant cultures were established from women undergoing ele

Reprotoxic Effect of Tris(2,3-Dibromopropyl) Isocyanurate (TBC) on Spermatogenic Cells In Vitro.

Tris(2,3-dibromopropyl) isocyanurate (TBC) belongs to the class of novel brominated flame retardants (NFBRs) that are widely used in industry. It has commonly been found in the environment, and its presence has been discovered in living organisms as well. TBC is also described as an endocrine disruptor that is able to affect male reproductive processes through the estrogen receptors (ERs) engaged in the male reproductive processes. With the worsening problem of male infertility in humans, a mechanism is being sought to explain such reproductive difficulties. However, so far, little is known ab

Competing pathway involved in allylic acetoxylation of androst-5-en-17-one, and oxidation of allylic alcohols with chromium oxides.

Allylic acetoxylation of androst-5-en-17-one (1) with bromine and silver acetate gave 6 alpha- and 6 beta-acetoxyandrost-4-en-17-ones [4 (3%) and 5 (12%)] and 5 alpha-bromo-6 beta-acetoxy steroid 8 (4%) along with the expected product 4 beta-acetoxy derivative 2 (45%). Treatment of 5 alpha,6 beta-bromide 12, an intermediate of the acetoxylation reaction, with silver acetate also produced the acetates 2, 4, 5, and 8 in relative yields similar to those above. These results indicate that the 6-acetates 4 and 5 are produced through a competing pathway involving formation of a bridged carbonium ion

Exposure of Female Rats to an Environmentally Relevant Mixture of Brominated Flame Retardants Targets the Ovary, Affecting Folliculogenesis and Steroidogenesis.

Brominated flame retardants (BFRs) are incorporated into various consumer products to prevent flame propagation. These compounds leach into the domestic environment, resulting in chronic exposure and contamination. Pregnancy failure is associated with high levels of BFRs in human follicular fluid, raising serious questions regarding their impact on female reproductive health. The goal of this study is to elucidate the effects of an environmentally relevant BFR mixture on female rat ovarian functions (i.e., folliculogenesis and steroidogenesis). A BFR dietary mixture formulated to mimic the rel

A multipronged approach to inform ecological risks of emerging organic contaminants in surface water and sediment in the estuarine-receiving environment of Tauranga Harbour, Bay of Plenty, New Zealand.

Increasing coastal urbanisation is a continuous stressor on coastal ecosystems. Emerging organic contaminants (EOCs) comprise thousands of chemicals whose environmental fate and effects are not all well understood. A study of 96 EOCs was undertaken in Tauranga Harbour, New Zealand, with surficial sediments collected and passive sampling devices (PSDs) deployed at 12 sites. Per- and polyfluorinated alkyl substances (PFAS), glyphosate/aminomethylphosphonic acid (AMPA), and chloroxylenol were detected in PSDs only, while nonylphenol, bisphenol A, ketoprofen, salicylic acid, meclofenamic acid, UV

Industrial phenolic compounds targeting 11β-hydroxysteroid dehydrogenase 2: A comparative in silico and structure-activity investigation in humans and rats.

Industrial phenolic compounds (IPCs) are widespread environmental contaminants with known endocrine-disrupting potential. This study aimed to investigate the inhibition of 13 IPCs on human and rat 11β-hydroxysteroid dehydrogenase 2 (11β-HSD2), a crucial enzyme responsible for cortisol inactivation. The inhibitory potential of IPCs was evaluated using enzymatic screening. Direct enzyme binding was assessed via surface plasmon resonance (SPR). Intracellular cortisol-to-cortisone conversion was tested in human placental BeWo cells. Additionally, structure-activity relationships, 3D-QSAR analyses,

Significant enhancement of inhibitory strength of bisphenol A analogs against human and rat 5α-reductase 1 upon halogenation of the benzene ring: Potentially disrupting neurosteroid biosynthesis.

Bisphenol A (BPA) halogenation derivatives, formed via radical or electrophilic substitution, constitute a class of emerging contaminants, with brominated variants dominating the flame-retardant market. Their effect on steroid 5α-reductase 1 (SRD5A1) activity in neural and testicular cells remains unclear. This study examined inhibitory effects of seven BPA analogs on SRD5A1, focusing on dihydrotestosterone synthesis. Using SRD5A1 in human brain SF126 cell and rat testicular microsomes, we found an inhibitory potency gradient. For human SRD5A1: tetrabromobisphenol A (12.71 μM) > tetrachlorobis

The Role of Plant-Derived Natural Products as a Regulator of the Tyrosine Kinase Pathway in the Management of Lung Cancer.

One of the most common malignant tumors worldwide is lung cancer, and it is associated with the highest death rate among all cancers. Traditional treatment options for lung cancer include radiation, chemotherapy, targeted therapy, and surgical resection. However, the survival rate is low, and the outlook is still dreadfully dire. The pursuit of a paradigm change in treatment approaches is, therefore, imperative. Tyrosine kinases (TKs), a subclass of protein kinases, regulate vital cellular function by phosphorylating tyrosine residues in proteins. Mutations, overexpression, and autocrine parac

How histone modifications influence cellular radiosensitivity: Pharmaceutically targeting epigenetic regulators as a promising avenue to overcome radioresistance.

Cancer remains the leading cause of mortality worldwide. Radiotherapy (RT), a cornerstone of oncological treatment for over a century, has achieved great success in various cancers. However, radioresistance remains the primary factor leading to the failure of radiotherapy. Histone modifications in cancer cells are known to play a pivotal role in regulating radiosensitivity by modulating chromatin structure, either by loosening or tightening it. Here, we provide a comprehensive summary of the link between aberrant histone modifications and radiation resistance across various cancer and normal t
