Potential roles of the melatonin system in the promotion of ovarian estradiol secretion in the sea cucumber, Apostichopus japonicus.

Melatonin (MT), synthesized from tryptophan, modulates reproduction through its interaction with melatonin receptors (MTNRs). While its functions in vertebrates are well established, the MT system remains poorly understood in echinoderms. To elucidate MT synthesis regulation, we examined AjAsmt, a pivotal enzyme in MT biosynthesis, to determine its tissue-specific distribution in Apostichopus japonicus. AjAsmt was predominantly expressed in the nerve ring, polian vesicle, intestine, and muscle, with significantly reduced intestinal expression post-spawning, suggesting a role in ovarian develop

Calnexin interacts with B-cell receptor-associated protein 31 (Bap31) to mediate coelomocyte phagocytosis and Vibrio splendidus clearance in Apostichopus japonicus.

Calnexin serves as a lectin chaperone located on the endoplasmic reticulum membrane and functions in glycoprotein folding and synthesis quality control, as well as in Ca2+ storage. Calnexin is extensively documented to participate in host immunity in the endoplasmic reticulum. However, the functions and fundamental mechanisms of calnexin in the invertebrate innate defence remain largely unknown. In this research, the complete cDNA sequence for calnexin from Apostichopus japonicus (Ajcalnexin) was cloned, revealing a 1779 bp open reading frame that codes for 592 amino acids, 113 bp 5'-Untransla

Global N6-methyladenosine methylation analysis reveals the positive correlation between m6A modification and mRNA abundance during Apostichopus japonicus disease development.

N6-methyladenosine (m6A), the most abundant epitranscriptomic modification in eukaryotic messenger RNA (mRNA), plays important roles in regulation of gene expression for fundamental biological processes and diverse physiological functions, including combating with pathogen infection. Here, we were first profile transcriptome-wide m6A sequencing in four stages of skin ulceration syndrome-diseased Apostichopus japonicus following Vibrio splendidus infection, including Control (healthy), Early (small ulcer), Later (extensive ulcer), and Resistant (no ulcer) groups. Our results revealed that three

PPARα alleviates inflammation via inhibiting NF-κB/Rel pathway in Vibrio splendidus challenged Apostichopus japonicus.

Organisms trigger pro-inflammatory responses to resist the invasion of foreign pathogens in the early infection stage. However, excessive or chronic inflammation can also cause several diseases. We previously validated IL-17 from sea cucumbers mediated inflammatory response by the IL-17R-TRAF6 axis. But the anti-inflammatory effect was largely unknown in the species. In this study, the conserved PPARα gene was obtained from Apostichopus japonicus by RNA-seq and RACE approaches. The expression of AjPPARα was found to be significantly induced at the late stage of infection not only in Vibrio spl

Analysis of the gene transcription patterns and DNA methylation characteristics of triploid sea cucumbers (Apostichopus japonicus).

Breeding of polyploid aquatic animals is still an important approach and research hotspot for realizing the economic benefits afforded by the improvement of aquatic animal germplasm. To better understand the molecular mechanisms of the growth of triploid sea cucumbers, we performed gene expression and genome-wide comparisons of DNA methylation using the body wall tissue of triploid sea cucumbers using RNA-seq and MethylRAD-seq technologies. We clarified the expression pattern of triploid sea cucumbers and found no dosage effect. DEGs were significantly enriched in the pathways of nucleic acid

Transcriptomics reveals the temporal responses of sea cucumber (Apostichopus japonicus) to the challenge by bacterial peptidoglycans.

Sea cucumber (Apostichopus japonicus) is of considerable commercial and ecological value, yet disease outbreaks substantially constrain the sustainable development of its aquaculture industry. The molecular mechanisms underlying A. japonicus's response to pathogenic bacterial invasion remain incompletely elucidated to date, particularly with respect to its differential response to peptidoglycans derived from distinct Gram-specific bacteria. In this study, the transcriptomes of A. japonicus coelomocytes were analyzed in response to challenges involving two different peptidoglycans: one from Gra

Lvvibriocin-GK effectively reduced skin ulcer syndrome of Apostichopus japonicus by eliminating surface bacteria, modulating gut microbiota, and enhancing host immune responses.

Skin ulceration syndrome (SUS) is a major threat to the aquaculture of Apostichopus japonicus, particularly in southern China, where it has shown high mortality rates and infectious potential. Traditional antibiotic treatments often lead to challenges such as antibiotic resistance. Antimicrobial peptides (AMPs), which are vital elements of innate immunity, represent a promising alternative for treating SUS. In the study, a novel AMP named Lvvibriocin-GK identified in Litopenaeus vannamei was found to have a strong antibacterial activity against multiple Vibrio species that possibly cause SUS.

circRNA432 enhances the coelomocyte phagocytosis via regulating the miR-2008-ELMO1 axis in Vibrio splendidus-challenged Apostichopus japonicus.

Circular RNAs (circRNAs) are a kind of extensive and diverse covalently closed circular endogenous RNA, which exert crucial functions in immune regulation in mammals. However, the functions and mechanisms of circRNAs in invertebrates are largely unclarified. In our previous work, 261 differentially expressed circRNAs including circRNA432 (circ432) were identified from skin ulcer syndrome (SUS) diseased sea cucumber Apostichopus japonicus by RNA-seq. To better address the functional role of sea cucumber circRNAs, circ432 was first found to be significantly induced by Vibrio splendidus challenge

Identification of sex-specific splicing via comparative transcriptome analysis of gonads from sea cucumber Apostichopus japonicus.

Alternative splicing (AS) is an essential post-transcriptional regulation mechanism for sex differentiation and gonadal development, which has rarely been reported in marine invertebrates. Sea cucumber (Apostichopus japonicus) is an economically important marine benthic echinoderm with a potential XX/XY sex determination mechanism, whose molecular mechanism in the gonadal differentiation has not been clearly understood. In this study, we analyzed available RNA-seq datasets of male and female gonads to explore if AS mechanism exerts an essential function in sex differentiation and gonadal devel

Tissue inhibitor of metalloproteinases 1 is involved in ROS-mediated inflammation via regulating matrix metalloproteinase 1 expression in the sea cucumber Apostichopus japonicus.

Tissue inhibitors of metalloproteinases (TIMPs) serve as matrix metalloproteinase (MMP) inhibitors in the pathogenesis of inflammatory diseases in vertebrates. We cloned and characterised the TIMP1 gene from Apostichopus japonicus using RACE approaches (designated as AjTIMP1). For Vibrio splendidus-challenged sea cucumbers, the peak expression of AjTIMP1 mRNAs in coelomocytes was detected at 24 h (23.44-fold) and remained at high levels (4.01-fold) until 72 h. Similarly, AjTIMP1 expression was upregulated in primary coelomocytes exposed to 10 μg mL-1 LPS. AjTIMP1 was expressed in all tissues,

Integrated application of transcriptomics and metabolomics provides insights into sexual dimorphism in Apostichopus japonicus.

Sea cucumber (Apostichopus japonicus) is a commercially important aquaculture species in China. Research findings have indicated that adult male sea cucumbers with differentiated gonads have a greater growth advantage compared to females. Understanding the molecular basis of sexual dimorphism in A. japonicus is critical for developing sex control breeding strategies in aquaculture. In this study, we integrated transcriptomic and metabolomic analyses of testes and ovaries to elucidate differences in gene expression and metabolite profiles. Key sex-biased genes, including cyp2 subfamily members,

In Vivo Anticoagulant and Antithrombic Activity of Depolymerized Glycosaminoglycan from Apostichopus japonicus and Dynamic Effect-Exposure Relationship in Rat Plasma.

Glycosaminoglycan from Apostichopus japonicus (AHG) and its depolymerized fragments (DAHGs) are anticoagulant fucosylated chondroitin sulfate. The aim of this study was to further evaluate the anticoagulant and antithrombic activity of AHG and DAHGs, as well as reveal the dynamic relationship between exposure and effect in vivo. The results demonstrated that AHG100 (Mw~100 kDa), DAHG50 (Mw~50 kDa), and DAHG10 (Mw~10 kDa) exhibited potent anticoagulant activity by inhibiting intrinsic factor Xase complex (FXase) as well as antithrombin-dependent factor IIa (FIIa) and factor Xa (FXa). Identification of sex determination locus in sea cucumber Apostichopus japonicus using genome-wide association study.

BACKGROUND: Sex determination mechanisms are complicated and diverse across taxonomic categories. Sea cucumber Apostichopus japonicus is a benthic echinoderm, which is the closest group of invertebrates to chordate, and important economic and ecologically aquaculture species in China. A. japonicus is dioecious, and no phenotypic differences between males and females can be detected before sexual maturation. Identification of sex determination locus will broaden knowledge about sex-determination mechanism in echinoderms, which allows for the identification of sex-linked markers and increases th

Sex determination factor, a novel male-linked gene in the sea cucumber Apostichopus japonicus: Molecular characterization, expression patterns and effects of gene knockdown.

Apostichopus japonicus is a highly significant marine aquaculture species. Research findings have indicated that male sea cucumbers demonstrate a more rapid growth rate compared to females, underscoring the potential advantages of establishing an all-male population. In this study, we identified a specific protein-coding gene (ORFan) within a 4565 bp male fragment and named it sex determination factor (sdf). The sdf transcript exhibited ubiquitous expression in various adult male tissues, along with dynamic expression patterns in the testis across different developmental stages. Notably, knock

Development of Reliable Male-Specific Molecular Markers for Genetic Sex Identification in Sea Cucumber Apostichopus japonicus.

Apostichopus japonicus is an important marine aquaculture species in China, with high nutritional and economic value. In A. japonicus, there is no obvious sexual dimorphism in external appearance, and sex differentiation primarily relies on the observation of mature gonads after dissection, which leads to difficulties in sex identification. The confusion in sex identification greatly reduces breeding efficiency in the sea cucumber industry. Therefore, developing a reliable sex-specific marker is crucial. In this study, we identified 586 male-specific sequences through whole-genome assembly and

The influence of three antibiotics on the growth, intestinal enzyme activities, and immune response of the juvenile sea cucumber Apostichopus japonicus selenka.

The global abuse and misuse of antibiotics in the treatment and prevention of bacterial infections has resulted in the ubiquitous existence of these drugs in aquatic environments, which causes frequent antimicrobial resistance and pollution in ecosystems. However, the chronic effects of antimicrobial agents on aquatic animal growth and health have not been fully evaluated. In the present study, three typical antibiotics (tetracycline, erythromycin, and norfloxacin) were administered orally to juvenile sea cucumbers Apostichopus japonicus for 45 days, to mimic the long-term use of antibiotics.

A gut microbiota-stem cell axis mediates the anti-intestinal aging effect of fucoidan from Apostichopus japonicus.

BACKGROUND: Apostichopus japonicus is a traditional medicinal and culinary species, with existing anti-aging research primarily focusing on its bioactive peptides. In contrast, the anti-aging potential of its major polysaccharide, fucoidan (Aj-FUC), remains largely unexplored. Since the intestine is a central target in the aging process and the primary site for polysaccharide interaction, this study investigates the protective effects and underlying mechanisms of Aj-FUC against d-galactose (d-Gal)-induced intestinal senescence in mice. RESULTS: Aj-FUC significantly improved intestinal functio
