Response of Hemolytic and Photosynthetic Activity of Chattonella marina Complex Under Variable N:P Stoichiometry.

Chattonella marina is an ichthyotoxic, bloom-forming raphidophyte known for its hemolytic activity. However, the mechanisms by which nitrogen (N) and phosphorus (P) limitation influence this hemolytic toxicity remain poorly understood. In this study, both N and P limitation reduced growth, photosynthetic efficiency (Fv/Fm, YII, rETRmax), and the expression of nutrient-uptake, tetrapyrrole/chlorophyll biosynthesis genes. Nevertheless, the two nutrients produced opposite effects on toxicity: N limitation lowered hemolytic activity and ROS levels to near zero, whereas P limitation kept both relat

Effect of Heterologous Expression of Key Enzymes Involved in Astaxanthin and Lipid Synthesis on Lipid and Carotenoid Production in Aurantiochytrium sp.

Aurantiochytrium sp., a heterotrophic microorganism, has received increasing attention for its high production of polyunsaturated fatty acids and has been widely applied in various industries. This study intended to optimize the carotenoid synthesis pathway in Aurantiochytrium sp. by metabolic engineering to increase the carotenoid content. Multi-sourced key enzyme genes involved in lipid synthesis (LPAAT and DGAT) and astaxanthin synthesis (crtZ and crtW) were selected to construct single-gene expression vectors and transformed into Aurantiochytrium sp. The results showed that the overexpress

Combining high pressure and electric fields towards Nannochloropsis oculata eicosapentaenoic acid-rich extracts.

Nannochloropsis oculata is naturally rich in eicosapentaenoic acid (EPA). To turn this microalga into an economically viable source for commercial applications, extraction efficiency must be achieved. Pursuing this goal, emerging technologies such as high hydrostatic pressure (HHP) and moderate electric fields (MEF) were tested, aiming to increase EPA accessibility and subsequent extraction yields. The innovative approach used in this study combined these technologies and associated tailored, less hazardous different solvent mixtures (SM) with distinct polarity indexes. Although the classical

Efficient solvothermal wet in situ transesterification of Nannochloropsis gaditana for biodiesel production.

In situ transesterification of wet microalgae is a promising, simplified alternative biodiesel production process that replaces multiple operations of cell drying, extraction, and transesterification reaction. This study addresses enhanced biodiesel production from Nannochloropsis gaditana at elevated temperatures. Compared with the previously reported in situ transesterification process of conducting the reaction at a temperature ranging from 95 to 125 °C, the present work employs higher temperatures of at least 150 °C. This relatively harsh condition allows much less acid catalyst with or wi

Physiological and transcriptomic responses of Heterosigma akashiwo to nitrogen deficiency and urea utilization.

Nitrogen (N) is an essential nutrient for phytoplankton growth. The availability of ambient N and its chemical forms are critically important for the formation of harmful algal blooms (HABs). Although previous studies have explored the transcriptomic responses of Heterosigma akashiwo to N conditions, these analyses were largely descriptive and lacked in-depth characterization of the molecular mechanisms underlying urea utilization and N deficiency responses. To address this gap, we conducted a comprehensive physiological and transcriptomic investigation of this globally distributed HAB species

Kinetic and thermodynamic evaluation of lipid and pigment extraction from Nannochloropsis oculata by ultrasound-assisted and conventional methods.

Conventional extraction of lipids and pigments involves the use of toxic solvents and long extraction times potentially leading to degradation of the target compounds. Ultrasound-assisted extraction (UAE) has emerged as a promising alternative, capable of enabling high yields under milder conditions and shorter extraction times. This study investigated the extraction kinetics of lipids, chlorophylls, and carotenoids from Nannochloropsis oculata using 75% (v/v) ethanol at 20–60 °C, with and without ultrasound, and compared the results with conventional extraction. Optimal extraction conditions

Biology and systematics of heterokont and haptophyte algae.

In this paper, I review what is currently known of phylogenetic relationships of heterokont and haptophyte algae. Heterokont algae are a monophyletic group that is classified into 17 classes and represents a diverse group of marine, freshwater, and terrestrial algae. Classes are distinguished by morphology, chloroplast pigments, ultrastructural features, and gene sequence data. Electron microscopy and molecular biology have contributed significantly to our understanding of their evolutionary relationships, but even today class relationships are poorly understood. Haptophyte algae are a second

The regulatory function of Blastocystis spp. on the immune inflammatory response in the gut microbiome.

Blastocystis spp. is a unicellular organism that resides in digestive tract of various vertebrates, with a worldwide distribution and a variable prevalence. For many years, Blastocystis spp. was considered a cyst of a flagellate, a fungus, or a saprophyte yeast of the digestive tract; in 1996, it is placed in the group of stramenopiles (heterokonts). Since its new classification, many questions have arisen around this protist about its role as a pathogen or non-pathogen organism. Recent evidence indicates that Blastocystis spp. participates in the immune inflammatory response in the intestinal

Tryptophanol enhances nitrogen assimilation in marine diatoms.

Diatoms exhibit high competitive capacity in nitrogen assimilation, but the underlying mechanisms remain unclear. Here, we identify a non-ribosomal peptide synthase-like gene (PtNRPS1) with an atypical domain structure (A-T-R1-R2) in the marine diatom Phaeodactylum tricornutum, crucial for short-term nitrogen assimilation. In vitro enzyme assays show PtNRPS1 catalyzes conversion of L-tryptophan to tryptophanol, a tryptophan-derived indole compound that promotes diatom growth at concentrations far lower than indole-3-acetic acid. Transcriptomic, metabolomic analyses, and stable-isotope analyses

Integrated analysis of protein sequence and structure redefines viral diversity and the taxonomy of the Flaviviridae.

The Flaviviridae are a family of non-segmented positive-sense enveloped RNA viruses containing significant pathogens including hepatitis C virus and yellow fever virus. Recent large-scale metagenomic surveys have identified many diverse RNA viruses related to classical orthoflaviviruses and pestiviruses but quite different genome lengths and configurations, and with a hugely expanded host range that spans multiple animal phyla, including molluscs, cnidarians and stramenopiles,, and plants. Grouping of RNA-directed RNA polymerase (RdRP) hallmark gene sequences of flavivirus and 'flavi-like' vir

PlasmoFP: leveraging deep learning to predict protein function of uncharacterized proteins across the malaria parasite genus.

The first malaria parasite Plasmodium falciparum genome published in 2002 jump-started functional studies, but a large fraction of all predicted proteins remains partially annotated and of 'unknown function'. Here, we introduce Plasmodium Function Predictor (PlasmoFP), deep learning models designed specifically for species of genus Plasmodium. Innovatively, PlasmoFP models are trained on structure-function relationships of the phylogenetically relevant SAR (Stramenopiles, Alveolate, and Rhizarians) supergroup proteins, addressing challenges to annotating Plasmodium proteins due to their low se

B-vitamin supplementation unlocks DHA production of Aurantiochytrium limacinum on molasses-based fermentation residue.

Various marine protists inhabiting high-salinity environments can convert organic nutrients into high-value biomolecules or biomass, making them suitable agents for valorizing saline organic residue from fermentation waste streams, such as condensed molasses fermentation solubles (CMS). This study established a screening method to identify mixotrophic or heterotrophic marine protists suitable for CMS valorization and to assess their bioconversion potential. Growth-performance and nutrient-replacement assays were evaluated based on relative colony coverage on agar plates and final cell density

tArp Is an Actin-Related Protein Required for Ciliary Function That Is Conserved in Ciliates and Related Eukaryotic Groups.

Cells exhibit a wide range of behaviors due to their diverse molecular systems, which have been established through the molecular evolution of individual species. Protein families generated through gene duplications may be involved in this process. The actin cytoskeleton is no exception. The majority of eukaryotes express not only actin, but also several types of actin-related proteins (Arps). The ciliate Tetrahymena thermophila possesses a species-specific Arp, named tArp, which localizes to cilia. However, the evolutionary origin of the gene and its detailed cellular functions remain unclear

Ultrasound-assisted pretreatment using ternary deep eutectic solvent and lipid extraction from Nannochloropsis oceanica.

Nannochloropsis sp. contains high levels of polyunsaturated fatty acids, particularly eicosapentaenoic acid (EPA). There is a need to establish highly efficient extraction processes to replace traditional organic solvents. First, the optimal DES system Choline chloride:1,4-butanediol:lactic acid = 1:2:1 was selected for Nannochloropsis oceanica. Response surface methodology optimized pretreatment conditions to 73% ultrasonic power (876 W), 17 min duration, and a 1:22 g/g solid-to-liquid ratio. Subsequently, the optimal methanol/hexane ratio was 1:3 v/v and the optimal extraction time was 15 mi

Growth medium sterilization using decomposition of peracetic acid for more cost-efficient production of omega-3 fatty acids by Aurantiochytrium.

Aurantiochytrium can produce significant amounts of omega-3 fatty acids, specifically docosahexaenoic acid and docosapentaenoic acid. Use of a glucose-based medium for heterotrophic growth is needed to achieve a high growth rate and production of abundant lipids. However, heat sterilization for reliable cultivation is not appropriate to heat-sensitive materials and causes a conversion of glucose via browning (Maillard) reactions. Thus, the present study investigated the use of a direct degradation of Peracetic acid (PAA) for omega-3 production by Aurantiochytrium. Polymer-based bioreactor and

Biomass Growth and Fatty Acid Production by the Marine Thraustochytrium sp. RT2316-16 in Chemically Defined Media.

The biomass and lipid production responses of the psychrophilic marine thraustochytrid Thraustochytrium sp. RT2316-16 were assessed in chemically defined media comprising glucose, up to 17 amino acids and up to 9 B-vitamins and mineral salts. Compared to the control medium with all amino acids and B-vitamins (biomass concentration: 7.1 ± 0.1 g L-1; total lipid content: 30.4 ± 0.5% of the DW), the growth of RT2316-16 was reduced by more than 50% in the medium that lacked cyanocobalamin or pyridoxamine. The total lipid content of the biomass grown in the absence of vitamins was 63% lower than in
